Preparation and Property of Antibacterial Nucleus with the Combination of Nano-Silver and Antibiotics

To improve the yield and quality of pearls in freshwater pearl culture and the survival rates after nucleus implanting surgery, pearl farmers used artificial pearl nuclear transplantation techniques to raise pearls. To address the common problem of Staphylococcus aureus and Escherichia coli infection in oyster farming, a new prophylactic method by using compound antibiotics to prepare the medicine coated pearl nucleus was put forward based on existing research results of the nanosilver antibacterial nucleus. Single-factor experiment, multi-factor experiment, orthogonal experiment, SPSS analysis of variance was used to optimize the antibacterial formulation on the assumption that the contaminated probability of these two pathogenic bacteria was the same. The result showed that the optimal ratio of compound antibiotics was 0.0075g/ml of the flavomycin solution and 0.01g/mL of the terramycin solution; the inhibition zones diameter of both pathogenic bacteria was more than 2.6cm in vitro, which was higher than the nanosilver antibacterial nucleus of 0.9cm in vitro. Indicating that the addition of this compound antibiotic formula for the nanosilver antibacterial nucleus could reduce the usage of antibiotics under the premise of maintaining antibacterial effectiveness, and could preferably inhibit the pathogenic microorganisms in the postoperative infection period. This also indicates that compound antibiotics coated antibacterial nanosilver nucleus would be applied more widely.

The present study was included the assessment of the antimicrobial activity of AgNPs synthesized by Punica granatum peel extract against pathogenic bacteria by testing warm aqueous P. granatum peel extract and silver nanoparticles. Punica granatum indicated potency for AgNP extracellular nanobiosynthesis after addition of silver nitrate (AgNO3) 4mM to the extract supernatant, in both concentrations (100mg and 50mg). The biogenic AgNPs showed potency to inhibit both gram-negative and gram-positive bacterial growth. Zons of inhibition in (mm) was lesser in gram-positive than gram-negative bacteria. The resulted phytogenic AgNPs gave higher biological activity than warm aqueous Punica granatum peel extract. The inhibition zone of the phytogenic AgNPs on E. coli reached 17.53, 22.35, and 26.06 mm at (0.1, 0.5, and 1) mg/ml respectively. While inhibition zones of Punica warm aqueous extract reached 5.33, 10.63, and 16.08 mm at the same concentrations. phytogenic AgNPs gave smaller inhibition zones in gram-positive than gram- negative. Cytotoxic activity of the phytogenic AgNPs was assayed in vitro agaist human blood erythrocytes (RBCs), spectroscopic results showed absorbance at 540 nm hemolysis was observed. In general, AgNPs showed least RBCs hemolysis percentage, at 1 mg/ml concentration, hemolysis percentage was (4.50%). This study, concluded that the Punica granatum peel extract has the power of synthses of AgNPs characterized by broad spectrum antimicrobial activity with cyto-toxicity proportional to AgNPs concentration.

The trial was conducted at the Centro de Producción Agrícola of ICfA in Cuyuta, Escuintla-Guatemala in order to generate information on the yield and quality of edible mulberry (Morus sp.) roughage treatments consisted of three harvest frequencies (6; 9 and 12 weeks and three fertilization levels of nitrogen (0.40 and 80 kg/ha). A complete randomized block experimental design, with a factorial arrangement (3x3) was used. The cutting height was 0.3 m above the ground level with two sequencies: from August 2nd to September 13 th and from September 13th to December 6th, 1990. In both cases, the 12 week frequency cutting and 80 kg of N were superior to the others (P<=0.01), yielding 6.87 and 6.15 t/ha of dry matter respectively. The yields at 9 weeks were statistically higher than those at 6 weeks. The highest protein percentage of the whole plant, leaves and stalks were produced at 6 weeks. The dry matter digestibility values in vitro showed little variability among treatments, with averages for whole plants, leaves and stalks of 65, 91 and 41 %, respectively. The preceeding data suggests that the mulberry has an excellent potential as a balanced supplement (protein and energy) for dual purpose cattle on the Southern coast of Guatemala.

Probiotics are live microorganisms which when consumed in large number together with a food promote the health of the consumer. The aim of this study was to evaluate in vitro probiotic properties of lactic acid bacteria (LAB) isolated from traditional Ethiopian fermented Teff injera dough, Ergo, and Kocho products. A total of 90 LAB were isolated, of which 4 (4.44%) isolates showed 45.35–97.11% and 38.40–90.49% survival rates at pH values (2, 2.5, and 3) for 3 and 6 h, in that order. The four acid-tolerant isolates were found tolerant to 0.3% bile salt for 24 h with 91.37 to 97.22% rate of survival. The acid-and-bile salt-tolerant LAB isolates were found inhibiting some food-borne test pathogenic bacteria to varying degrees. All acid-and-bile-tolerant isolates displayed varying sensitivity to different antibiotics. The in vitro adherence to stainless steel plates of the 4 screened probiotic LAB isolates were ranged from 32.75 to 36.30% adhesion rate. The four efficient probiotic LAB isolates that belonged to Lactobacillus species were identified to the strain level using 16S rDNA gene sequence comparisons and, namely, were Lactobacillus plantarum strain CIP 103151, Lactobacillus paracasei subsp. tolerans strain NBRC 15906, Lactobacillus paracasei strain NBRC 15889, and Lactobacillus plantarum strain JCM 1149. The four Lactobacillus strains were found to be potentially useful to produce probiotic products.

Midas marrowstem kale (Brassica oleracea L.) was grown in different row width associations with United 106 corn (Zea maize L.) in two studies and ensiled in different moisture blends with corn stover in another. Highest dry matter yields were obtained where a single row of kale was grown at 30 cm to the side of a corn row. This combination also provided the lowest moisture content feed and the highest in vitro digestibility and crude protein content. Changing the corn row width had no significant effect upon yield, plant height, in vitro digestibility, kale leaf or corn ear content. Blending kale with corn stover to provide a silage of about 70% moisture increased the digestibility and protein content of the feed and provided a silage that kept well in storage.

Background: In the past few decades focus of research has been toward herbal medicines because of growing bacterial resistance and side effects of antimicrobial agents. The extract derived from the plants may increase the efficacy of antibiotics when used in combination against pathogenic bacteria. In the current study, the synergistic antibacterial efficacy of plant extracts in combination with antibiotics has been assessed on selected periodontal pathogens. Methods: Ethanolic extracts were prepared from Salvadora persica (Miswak) and Cinnamomum zeylanicum (Ceylon cinnamon), by the soxhalate method. Plaque samples were collected from clinical periodontitis patients to isolate and grow the periodontal pathobionts under favorable conditions. Susceptibility of bacteria to the extracts was assessed by gauging the diameter of the inhibition zones. Minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of plant extracts were determined against each bacterium. Synergistic activity of plants extract in combination with antibiotics against the bacteria was also assessed by measuring the diameter of the inhibition zones. Results: Ethanolic extract of both the plants showed an inhibitory effect on the proliferation and growth of all four strains of periodontal pathobionts. Maximum antibacterial activity was exhibited by C. zeylanicum against Tannerella forsythia (MIC = 1.56 ± 0.24 mg/mL, MBC = 6.25 ± 0.68 mg/mL), whereas among all the studied groups the minimum activity was reported by C. zeylanicum against Aggregatibacter actinomycetemcomitans the (MIC = 12.5 ± 3.25 mg/mL, MBC = 75 ± 8.23 mg/mL). Combination of herbal extracts with different antibiotics revealed a synergistic antibacterial effect. The best synergism was exhibited by S. persica with metronidazole against A. actinomycetemcomitans (27 ± 1.78). Conclusions: Current in vitro study showed variable antibacterial activity by experimented herbal extracts against periodontal pathobionts. The synergistic test showed significant antibacterial activity when plant extracts were combined with antibiotics.

Although many studies have been conducted on somatic cell nuclear transfer, there are only a few reports on cryopreservation of reconstructed embryos after nuclear transplantation. The objective of this study was to examine in vitro or in vivo development of vitrified blastocysts obtained by nuclear transfer. Nuclear transfer was carried out according to the procedure of Goto et al. (1999 Anim. Sci. J. 70, 243–245), and conducted using abattoir-derived oocytes and cumulus cells derived by ovum pickup from Holstein and Japanese Black cows. Embryos were vitrified as described by Saito et al. (1998 Cryobiol. Cryotech. 43, 34–39). The vitrification solution (GESX solution) was based on Dulbecco's PBS containing 20% glycerol (GL), 20% ethylene glycol (EG), 0.3 M sucrose (Suc), 0.3 M xylose (Xyl), and 3% polyethylene glycol (PEG). The blastocysts were equilibrated in three steps, with 10% GL, 0.1 M Suc, 0.1 M Xyl, and 1% PEG for 5 min (1); with 10% GL, 10% EG, 0.2 M Suc, 0.2 M Xyl, and 2% PEG for 5 min (2) and GESX solution (3). After transfer to GESX, equilibrated embryos were loaded to 0.25-mL straws and plunged into liquid nitrogen for 1 min. The vitrified blastocysts were warmed in water (20°C) and diluted in 0.5 M and 0.25 M sucrose for 5 min each. Equilibration and dilution procedures were conducted at room temperature (25–26°C). After dilution, the vitrified blastocysts were cultured in TCM-199 supplemented with 20% fetal calf serum and 0.1 mM β-mercaptoethanol at 38.5°C under gas phase of 5% CO2 in air. In Experiment 1, survival rates after vitrification were compared between the nuclear transfer and the IVF blastocysts. Survival rates of vitrified nuclear transfer blastocysts (n = 60, Day 8) at 24 and 48 h were 70.0% and 56.7%, respectively, and those of vitrified IVF blastocysts (n = 41) were 82.9% and 82.9%, respectively. There were no significant differences in survival rates at 24 and 48 h between the two groups. In Experiment 2, one (VIT-single) or two (VIT-double) vitrified and one (nonVIT-single) or two (nonVIT-double) nonvitrified reconstructed blastocysts per animal were transferred into Holstein dry cows. The result of Experiment 2 is shown in Table 1. This experiment demonstrated that the vitrification method in this study can be used for cloned embryo cryopreservation but the production rate should be improved. Table 1. Three spring barley (Hordeum vulgare L.) and four annual ryegrass (Lolium multiflorum Lam.) types were intercropped to evaluate the potential yield and quality of forage which can be produced under irrigation in southern interior B.C. All treatments were intercrops; when barley cultivar effects are described they are averaged over the ryegrasses and vice versa. Barley cultivars differed in grain maturity. Ryegrass cultivars included diploid and tertraploid Italian and Westerwolds types. Intercrops including late grain maturing barley cultivars (Samson and Virden) increased the yield of the first silage cut (both by 25% over 2 yr) compared to Diamond, a medium-maturing cultivar adapted to the area. Intercrops containing the semi-dwarf barley, Samson, produced more digestible forage including higher in vitro digestible dry matter (IVDDM) and lower ADF and lignin. Annual ryegrass yield in the first cut intercropped with Samson was almost twice that with either Diamond or Virden, showing that Samson barley is less competitive. Second-cut yield (ryegrass regrowth) was greater for tetraploid than diploid annual ryegrasses. Yields of fall pasture (cuts 3 and 4) were similar among ryegrass cultivars. Cuts 2 and 3 (ryegrass only) of Italian ryegrasses had superior quality (higher IVDDM and protein; and lower ADF, lignin, and NDF) to Westerwolds ryegrassses, but all cultivars had similar quality in the late fall (Cut 4). It was concluded that a range of barley and annual ryegrass cultivars can be successfully intercropped to produce forage in south central B.C.Key words: Intercropping, barley annual ryegrass, forage yield and quality

Most of the environmental organism possess endotoxin which presence indicates the organisms are much lethal and for the purposes bacteriological quality of pharmaceutical raw products is much important. In these current study, 2 raw materials out of 10 were found to containing pathogenic bacteria Escherichia coli and Pseudomonas species in the culture medium, that indicates the raw materials were contaminated with the deadly organism. These two raw materials checked for the presence of Endotoxin and both provided positive gel clot by Limulas amoebocyte lysate (LAL) assay. Quality maintenance and assurance is the essential need of Drug preparation in pharmaceutical sector. The result ensure that Pharmaceutical industry should need to follow GMP and HACCP to minimize the contamination for improving the biological safety of the product in a cost-effective manner. Background and Objective: With the significant increase in the prevalence of infectious diseases and the development of drug resistance by human pathogenic bacteria, there is a continuous need to discover new antimicrobial compounds from plants. Methods: Four extracts of wild Myrtus communis L. berries (myrtle berries) were prepared with the addition of Cinnamomum verum and Eugenia caryophyllata. The extracts were screened in vitro for their antimicrobial activities using agar-well diffusion method against Escherichia coli, Staphylococcus aureus, Enterobacter cloacae, Listeria monocytogenes, Pseudomonas aeruginosa and Proteus mirabilis cultures. Results: The inhibition zones ranged from 12 to 22 mm. The MICs values of extracts lies between the ranges of 30 to 100 mg/ml. Of the extracts studied, the most active ones were those obtained from the myrtle berries:cloves, myrtle berries:cinnamon:cloves with the highest inhibition zones 22 mm and 17mm against S. aureus and L. monocytogenes at 50 mg/ml and 80 mg/ml, respectively. None of the extracts was active against E. coli and P. mirabilis. Conclusion: The present investigations have exposed that the myrtle berries:cloves, myrtle berries: cinnamon:cloves extracts could be used in traditional medicine as natural antimicrobial agents in treatment the bacterial infections.

The objectives of this investigation were a) to isolate bacteria from different foods (dairy products, fruits, and vegetables) and evaluate their probiotic potential and b) to select, identify, and characterize the strain with the highest probiotic potential. From 14 food samples, a total of 117 strains were isolated; however, only 42 (T1 to T42) showed the morphology (gram-positive, coco, and bacillar form) and were catalase- and oxidase-negative to be considered as a presumptive lactic acid bacteria (LAB). The antagonistic activity of the 42 strains was evaluated on Escherichia coli (O157:H7E09), Listeria monocytogenes (ATCC 19115), Staphylococcus aureus (ATCC 25923), and Salmonella enterica serotype Typhimurium (ATCC 14028). The strains with the highest antagonistic activity were nine isolates from the following: pulque (T1), sprouted beans (T26), ranchero cheese (T30, T31, T32, T33, T35, T36), and tenate cheese (T40) with inhibition zones from 17.0 ± 1.2 to 19.3 ± 2.8 mm. Based on the antagonistic activity against pathogenic bacteria and resistance to low pH and bile salts, strain T40 exhibited the highest probiotic potential. Using the 16S rRNA technique, strain T40 was identified as Lacticaseibacillus paracasei (the previous taxonomic nomenclature was Lactobacillus paracasei , prior to the nomenclature change in April 2020); this strain presented no resistance to ampicillin, gentamicin, erythromycin, and tetracycline. The antagonistic activity was evaluated in situ (fresh cheese) against pathogenic bacteria, evidencing the probiotic potential of L. paracasei . Finally, Lacticaseibacillus paracasei isolated from tenate cheese showed characteristics as a probiotic microorganism and high potential in food technology.
