The Influence of Various Factors and Changes in the Concentration of the Substrate on the Action Enzymes of Microbial Origin

Использование ферментных препаратов микробного происхождения в промышленности имеет устойчивую тенденцию к увеличению, при этом 2/3 текущего объема составляют ферменты для пищевой промышленности, а их основная доля приходится на спиртовую отрасль. При этом потребность спиртовой отрасли России в комплексных ферментных препаратах составляет около 7 тыс. т, доля отечественных препаратов составляет менее 15%. Объектами исследований служили ферменты микробного происхождения, плесневые грибы, затор спиртового производства. Осахаривание проводили при температуре 57…58 °С, длительность брожения при этом составляла 62 ч. Установлено, что для достижения максимального действия фермента, при других постоянных условиях, требуется сравнительно большая концентрация субстрата. У большинства ферментов вне клетки она выше, чем концентрация данного вещества в организме, то есть фермент действует менее эффективно, чем в искусственно созданных условиях, где он насыщается субстратом. В зависимости от активности расход грибной культуры может колебаться в определенных пределах. Скорость и глубина гидролиза углеводов сырья определяются активностью глубинной культуры плесневого гриба. Таким образом, свойства ферментов определяются главным образом особыми свойствами белков. При этом в молекуле белка изменяется укладка пептидных цепей, что приводит к потере характерных свойств белка. Способность ферментов в благоприятных условиях пережить материнские клетки и проявлять свое действие вне клетки позволяет широко использовать биологические катализаторы в различных отраслях пищевой промышленности. The use of enzyme preparations of microbial origin in industry has a steady tendency to increase, with 2/3 of the current volume being enzymes for the food industry, and their main share is in the alcohol industry. At the same time, the need of the alcohol industry in Russia for complex enzyme preparations is about 7 thousand tons, the share of domestic preparations is less than 15%. The objects of research were enzymes of microbial origin, mold fungi, mash of alcohol production. Saccharification was carried out at a temperature of 57…58 °C, the duration of fermentation was 62 hours. It was found that to achieve the maximum effect of the enzyme, under other constant conditions, a relatively high concentration of the substrate is required. For most enzymes outside the cell, it is higher than the concentration of a given substance in the body, so the enzyme acts less efficiently than in artificially created conditions, where it is saturated with a substrate. Depending on the activity, the consumption of the mushroom culture can fluctuate within certain limits. The rate and depth of hydrolysis of raw carbohydrates are determined by the activity of the deep culture of the mold. Thus, the properties of enzymes are determined mainly by the special properties of proteins. In this case, the folding of peptide chains in the protein molecule changes, which leads to the loss of the characteristic properties of the protein. The ability of enzymes to survive mother cells under favorable conditions and to exert their action outside the cell makes it possible to widely use biological catalysts in various branches of the food industry.

Asparaginase, derived from microbial origin hydrolyses L-asparagine to L-aspartic acid. The enzyme finds principal use in the treatment of Acute Lymphoblastic Leukemia during childhood that primarily occurs between two to ten years of age. L-Asparaginase finds its use in management of haemopoietic disorders especially in pediatrics that is caused due to proliferation and enlargement of lymphoblast in bone marrow and in blood as well as other part of the body. L- Asparaginase from bacterial sources exhibit quaternary and tertiary structural forms. However for using it in therapeutic and clinical application it should not generate any fatal allergic reaction to the patient. Such effects can occur due to the enzyme associated L-Glutaminase activity and also due to the endotoxins from bacteria in enzyme preparations. Therefore, with the recent development in biotechnology with respect to production and purification techniques it is possible to get pure L- asparaginase from microbial origin. The present article provides an insight into the mechanism of action of L-Asparaginase as an anticancer agent and its industrial applications.

At the present stage of development, the entire world industry has faced the problem of rational use of renewable natural resources, in particular the most efficient ways of wastewater treatment and the use of accumulated waste in the production process as a secondary raw material. In particular, the alcohol industry, as one of the components of food, medical, chemical and various industries,leads to the formation of huge amounts of waste, including wastewater. The food industry, like any other industry, has a negative impact on the environment. Water bodies are the most affected by the food industry. Almost the first place in terms of water consumption per unit of production is the production of alcohol. Consumption of large amounts of water leads to the formation of wastewater, which is highly polluted and adversely affects the environment. Due to the high chemical and biological consumption of oxygen, specific color and odor, suspended solids, low pH value, the purification of such waste in the filtration fields and discharge into water bodies is not possible. The purpose of our work is: 1) conducting the analysis of the alcohol industry potential in Ukraine in recent years, and methods of waste disposal as a potential source for the development of bioenergy. 2) environmental aspects of the alcohol industry modernization at present stage of development and implementation of modern wastewater treatment technologies.

The plant of castor is widely spread in the Iraqi land, and characterized with containing ricin toxin, which has a very serious effects, and because the seeds of this plant scattered in the agricultural soil and rivers water, which increases the exposure of humans and animals to these beans. Objective: This experiment was designed to study the effect of high concentration of castor bean powder in some physiological and biochemical parameters and changes in some tissues of the body, as well as trying to use doxycycline to reduce the effects of ingestion of these seeds. Materials and Methods: In the experiment, 24 local rabbits were raised and fed in the Animal House of the Faculty of Medicine / Al-Muthanna University, then divided into four groups and treated for three weeks (21 days), Control group: treated with normal saline solution (0.9) orally throughout the experiment, G1: was treated orally with a concentration of 25 mg / kg of castor bean powder daily during the experiment, G2 : orally treated 25 mg / kg of castor bean and 25 mg / kg of doxycycline, G3: orally treated 25 mg / kg of castor powder with 50 mg / kg of doxycycline daily throughout the trial period. Results: The results of the experiment showed significant changes (P less than 0.05) in all physiological and biochemical blood tests when compared with control group. There was a significant decrease in PCV, Hb, RBC, T.protein and body weights, while demonstrated a significant increase in WBC, Urea, Creatinine, ALT, AST and ALP, with distortions in liver and kidney of animals that treated with Castor beans. In contrast, the treatment with doxycycline and caster beans showed significant improvement reflected by a normal proportion in physiological tests and biochemical tests with improvement in the tissues when compared to control group. Conclusions: It can be concluded from this study that castor bean has high toxic and pathogenic effects that may be dangerous to the life of the organism. Therefore, it is advisable to be cautious of these pills and avoid exposure to them, also recommended to take high concentrations of doxycycline treatment when infected with castor bean poisoning.

Abstract Background Diabetes, a global cause of mortality in developing countries is a chronic disorder affecting the metabolism of macromolecules and has been attributed to the defective production and action of insulin characterized by persistent hyperglycemic properties. This global disorder harms organs of the body such as the liver, kidney and spleen. Medicinal plants such as Hunteria umbellate have been shown to possess hypoglycemic, antioxidative and anti-diabetic properties owing to the high concentration of active phytochemical constituents like flavonoids and alkaloids. The present study seeks to evaluate the hypoglycemic activities of ethanolic seed extract of Hunteria umbellate on streptozotocin-induced diabetes rats. Methods Thirty (30) female experimental rats were randomly divided into five groups with six rats per group and were administered streptozotocin (STZ) and Hunteria umbellate as follows. Group 1 served as control and was given only distilled water, group 2 rats were administered 60 mg/kg STZ; Group 3 was administered 60 mg/kg STZ and 100 mg/kg metformin; group 4 rats were administered 60 mg/kg STZ and 800 mg/kg Hunteria umbellate, group 5 rats 60 mg/kg STZ and 400 mg/kg Hunteria umbellate. The fasting blood glucose level of each rat was measured before sacrifice. Rats were then sacrificed 24 h after the last dose of treatment. Results The results showed that Hunteria umbellate significantly reversed STZ-induced increase in fasting blood glucose and increase in body and organs weight of rats. Hunteria umbellate significantly reversed STZ-induced decrease in antioxidant enzyme in liver, kidney and spleen of rats. Hunteria umbellate significantly reversed STZ-induced increase in oxidative stress markers in liver, kidney and spleen of rats. Conclusion Collectively, our results provide convincing information that inhibition of oxidative stress and regulation of blood glucose level are major mechanisms through which Hunteria umbellate protects against streptozotocin-induced diabketes rats.

Carrots were grown on soils polluted by heavy metal salts. Each particular microelement reached a high concentration [molybdenum (Mo) 39.00, cadmium (Cd) 2.30, lead (Pb) 4.01, mercury (Hg) 30.00, and selenium (Se) 36.20 mg/kg dry matter] in the carrot. In a metabolic balance trial conducted with 15 male and 15 female New Zealand White rabbits, the control animals (n = 5) were fed ad libitum with concentrate as basal diet, while the other rabbits received the basal diet and carrots containing the particular microelement. Blood samples were taken to determine the activity of serum enzymes. To investigate the metabolism of Mo, Cd, Pb, Hg and Se, samples were taken from the heart, liver, lungs, kidneys, spleen, ovaries/testicles, entire digestive tract, adipose tissue, femur, hair, faeces and urine. Carrot had significantly higher digestibility for all nutrients than the rabbit concentrate. Carrot samples of high Pb content had the lowest digestibility of crude protein. The microelements differed in their rate of accumulation in the organs examined: Mo and Cd accumulated in the kidneys, Pb in the kidneys, liver, bones and lungs, Hg in the kidneys and liver, while Se in the liver, kidneys and heart. The proportions of microelements eliminated from the body either via the faeces and urine (Mo 80.18% and Se 47.41%) or via the faeces (Cd 37.86%, Pb 66.39%, Hg 64.65%) were determined. Pathohistological examination revealed that the rate of spermatogenesis was reduced in the Mo, Cd, Pb and Hg groups compared to the control. Lead, Cd and Hg intake resulted in a considerable decrease in gamma-glutamyltransferase (GGT) and in an increase of alkaline phosphatase (ALP) activity because of damages to the kidneys and bones. All experimental treatments decreased the activity of cholinesterase (CHE) because of lesions in the liver.

Denture-associated stomatitis is a common candidal infection that may give rise to painful oral symptoms, as well as be a reservoir for infection at other sites of the body. As poly (methyl methacrylate) (PMMA) remains the main material employed in the fabrication of dentures, the aim of this research was to evaluate the adhesion of Candida albicans cells onto PMMA surfaces by employing an atomic force microscopy (AFM) single-cell force spectroscopy (SCFS) technique. For experiments, tipless AFM cantilevers were functionalized with PMMA microspheres and probed against C. albicans cells immobilized onto biopolymer-coated substrates. Both a laboratory strain and a clinical isolate of C. albicans were used for SCFS experiments. Scanning electron microscopy (SEM) and AFM imaging of C. albicans confirmed the polymorphic behavior of both strains, which was dependent on growth culture conditions. AFM force-spectroscopy results showed that the adhesion of C. albicans to PMMA is morphology dependent, as hyphal tubes had increased adhesion compared with yeast cells ( P < 0.05). C. albicans budding mother cells were found to be nonadherent, which contrasts with the increased adhesion observed in the tube region. Comparison between strains demonstrated increased adhesion forces for a clinical isolate compared with the lab strain. The clinical isolate also had increased survival in blood and reduced sensitivity to complement opsonization, providing additional evidence of strain-dependent differences in Candida-host interactions that may affect virulence. In conclusion, PMMA-modified AFM probes have shown to be a reliable technique to characterize the adhesion of C. albicans to acrylic surfaces.

Magnesium/chlorine and sulphur/chlorine ratios have been measured in the body fluids of some planktonic organisms by x-ray emission analysis of frozen hydrated specimens in a scanning electron microscope. Homarus vulgaris (Anthropoda: Decapoda) larvae excluded Mg2+ and SO42-ions from the haemolymph, but to a lesser extent than does the adult lobster. Bipinnaria larvae of Asterias (Echinodermata) excluded Mg2+ and SO42-ions from the coelomic fluid. Obelia medusae excluded Mg2+ and SO42-ions from the mesogloea but concentrate them in the gastrovascular system. The high concentration of sulphate in the gastrovascular fluid of medusae has been confirmed by rhodizonate titration in Cyanea and Rhizostoma jellyfish. Some implications of magnesium and sulphate regulation are discussed.

Purpose The purpose of this paper is to divulge the current knowledge about the nutritional and functional characteristics of Cañihua (Chenopodium pallidicaule Aellen), in addition to its potential applications in the food industry since research studies related to it are still limited compared to other cereals of greater diffusion. Design/methodology/approach The scientific information was collected from Web of Science, Scopus and Google Scholar databases, using keywords such as nutrition value of Chenopodium pallidicaule, amaranth and pseudocereals. Consistent information was selected according to its relevance, year of publication and accuracy with the topic. A total of 49 research papers were selected. Findings Cañihua is a grain with high nutritional potential, considered a superfood because it has a high protein quality, a balanced composition of essential amino acids and unsaturated fatty acids, with a high concentration of linoleic and oleic acid. Besides, it has a good level of bioactive compounds with high antioxidant capacity. However, its production and consumption are limited outside its area of origin, although its cultivation is possible under harsh conditions. Originality/value This paper, through a systematic bibliographic review, highlights the potential of cañihua to be considered in the development of food products with high nutritional and functional value. The information compiled will help researchers and professionals become aware of the importance of this grain and join forces in its processing and enhancement of its attributes.

The relationship between the distribution of the lateral line canals and their functionality has not been well examined in elasmobranchs, especially among Neotropical freshwater stingrays of the family Potamotrygonidae. The spatial distribution of the canals and their tubules and the quantification of the neuromasts were analyzed in preserved specimens of Potamotrygon motoro, P. orbignyi, Potamotrygon sp. "cururu", and Paratrygon aiereba from the middle Negro River, Amazonas, Brazil. The hyomandibular, infraorbital, posterior lateral line, mandibular, nasal and supraorbital canals were characterized and their pores and neuromasts quantified. The ventral canals are known to facilitate the accurate localization of prey items under the body, and our results indicate that the dorsal canals may be employed in identifying the presence of predators or potential prey positioned above the stingray's body. The presence of non-pored canals in the ventral region may be compensated by the high concentration of neuromasts found in the same area, which possibly allow the accurate detection of mechanical stimuli. The concentration of non-pored canals near the mouth indicates their importance in locating and capturing prey buried in the bottom substrate, possibly aided by the presence of vesicles of Savi.

The aim of present study was to compare the changes in thyroid gland with the reproductive cycle of S. heathi. Thyroid showed marked seasonal variation in weight, quantity of colloid and follicular epithelial height, suggesting the thyroid gland to be inactive during quiescence and winter dormancy and active during the time of recrudescence and breeding similarly to the testicular cycle. Plasma thyroxin (T4) concentration showed a significant seasonal change with high concentration during breeding and post-breeding and low concentration during quiescence. However, the T4 concentration increased from breeding to post-breeding phase, when the testes weight was declining. It is suggested that in S. heathi the positive correlation between thyroid and testicular cycles occurs only during the phases of the reproductive cycle when the body weight and testicular activity are also closely correlated.
