Microwave Assisted Synthesis and Antibacterial Evaluation of 1, 3, 4-Thiadiazole Derivatives

1,3,4-Thiadiazole is an important heterocyclic moiety, forms an integral core structural component of different categories of drugs such as antimicrobial, antitubercular, anti-inflammatory, antiepileptic, antiviral, antineoplastics, and analgesic agents. It is a key moiety in current discovery and designing of new drugs. The compounds were synthesised by both conventional method and microwave method. The targeted derivatives can be synthesised in a shorter time under microwave condition than under conventional reaction condition. Their structures were confirmed by FT-IR and NMR Spectroscopy. Antibacterial property of two synthesised analogs were evaluated by Agar well diffusion method against Escherichia coli and Staphylococcus aureus. The results of antibacterial activity showed that both the compounds were active against Staphylococcus aureus and inactive against Escherichia coli. Results of invitro studies showed that modifications in SB-2-PHB and SB-8-PHB will make it as a promising lead molecule for further research.

Modern medicines that are developing now come from active ingredients isolated from plants that require large amounts of plants. The development of new drugs from endophytic fungi found obstacles in the amount of pure compounds produced. Therefore further research is needed by using endophytic fungi as a new antimicrobial producer. This study aims to see the ability or activity of pure compounds produced by Sporothrix sp endophytic fungi from Dahlia tuber (Dahlia variabilis). Test the activity of pure compounds produced by Sporothrix sp. Endophytic fungi on E. coli and Staphylococcus aureus determined by disc diffusion method. With doses of 10, 30 and 50μg/disk. In Escherichia coli bacteria doses 10 and 50μg/disk gave significant inhibition of pure compounds from isolation compared to the positive control of ciprofloxacin, which was marked by a statistically significant test result (p <0.05). In contrast to Staphylococcus aureus there was no significant difference in doses of both doses of 10.30 and 50μg/disk. Determination of pure compounds was carried out by HPLC and Infra Red Spectrophotometry.

Antirrhinum Orontium is belonging to the family Plantaginacea commonly known as lesser snapdragon which is a flowering plant belong to genus Antirrhinum. It was traditionally used in treatment of Tumor, Eye inflammation and Ulcer. Antirrhinum Orontium is contain mostly tertiary alkaloids bases one of them is identified as 4-methyl-2, 6-naphthyridine and Choline is identified as quaternary base. The recent study is to investigate the antibacterial activity of Ethyl acetate, Dichloromethane, Hexane and Water extracts of Antirrhinum Orontium, against the pathogen like Escherichia coli and Staphylococcus aureus. Both the Soxhlet and Maceration water extract were found most active from the other extract against Escherichia coli and Staphylococcus aureus by using agar well diffusion method. The measured zone of inhibition for Soxhlet water extract is 21±0.05mm and Maceration water extract is 20±0.05mm against gram positive Staphylococcus aureus, while for gram negative Escherichia coli 23±0.05mm is measured for Soxlet and 21±0.05mm measured for Maceration water extracts

ABSTRACT  Akway (Drimys piperita) is a woody, evergreen and aromatic plant that belongs to family winteraceae. This plant is used by Sougb tribe lived in Sururey village, District of Anggi, to enhance the vitality of body. The objectives of the research were to determine antimicrobial stability of akway bark extracts influenced by heating time of 100OC, levels of acidity (pH) and salt contents.. Antimicrobial assays were done by using agar well diffusion method against four species of bacteria, i.e.  Escherichia coli ATCC25922, Bacillus cereus ATCC10876, Pseudomonas aeruginosa ATCC27853 and Staphylococcus aureus ATCC25923. The results showed that ethanolic extracts of akway bark only inhibited growth of B. cereus and S. aureus with minimum inhibitory concentration 0,99% and 0,89% . The levels of concentration and acidity of ethanol extracts  influenced the antimicrobial capacity of extracts.. Whereas heating time on 100OC during 25 minutes and salt contents  up to 5% of extract solution did not influence the antimicrobial stability of  akway bark extracts. Key words : akway, extracts, antimicrobe, pH, Heating, salt ABSTRAK Akway (Drimys piperita) adalah tumbuhan berkayu, aromatik dan hijau sepanjang tahun dan tergolong dalam suku winteraceae. Tumbuhan ini digunakan oleh Suku Sougb yang bermukim di desa Sururey Distrik Anggi, untuk mengobati malaria dan meningkatkan vitalitas tubuh. Tujuan dari penelitian ini adalah untuk menentukan stabilitas antimikroba ekstrak kulit kayu akway pada waktu pemanasan ekstrak pada 100OC, tingkat keasaman (pH) dan kandungan garam. Pengujian antimikroba dilakukan dengan menggunakan metode difusi agar terhadap empat spesies bakteri yaitu Escherichia coli ATCC25922, Bacillus cereus ATCC10876, Pseudomonas aeruginosa ATCC27853 and Staphylococcus aureus ATCC25923. Hasil menunjukkan bahwa ekstrak etanol kulit kayu akway hanya dapat menghambat bakteri Bacillus cereus dan Staphylococcus aureus dengan konsentrasi hambat minimum masing-masing adalah 0,99% dan 0,89%. Tingkat konsentrasi dan keasaman (pH) mempengaruhi kapasitas antimikroba ekstrak etanol kulit kayu akway. Sedangkan perlakuan pemanasan pada suhu 100OC dengan lama pemanasan sampai dengan 25 menit dan penambahan garam NaCl sampai konsentrasi 5%  tidak berpengaruh pada stabilitas antimikroba ekstrak etanol kulit kayu akway.

Na-Y Zeolite synthesized from fly ash was exchanged with Ag+ and Cu2+ ions by ion exchange process. The antibacterial action and efficacy of silver and copper exchanged Na-Y zeolite on Escherichia coli and Staphylococcus aureus were investigated. Escherichia coli and Staphylococcus aureus was suspended in broth media and exposed to silver and copper exchanged zeolite Y (ranging from 25-1000 μg) for a period of 24 h. The minimum bactericidal concentration (MBC) of silver and copper loaded zeolite-Y powder was 50 and 250 μg for Escherichia coli. Staphylococcus aureus was destroyed at a powder concentration of 100 μg for both Ag-Y and Cu-Y zeolite. Further, the present work also focuses to incorporate 5% silver and copper exchanged zeolite pigment in coating and study the antibacterial effect on the coated 304 stainless coupons. Paint containing copper exchanged zeolite shows antibacterial activity compared to paint with silver containing zeolite.

Bacteria of the genus Thermoactinomyces have the ability to produce antibacterial bioactive compounds. This bioactive compound can be used for combating diarrheal agents such as Escherichia coli and Staphylococcus aureus. This study aims to determine the antibacterial activity of the metabolite extract from Thermoactinomyces sp. (H24) against Escherichia coli and Staphylococcus aureus. Methanol was used as a solvent for the extraction of bacterial bioactive compounds. Antibacterial activity was analyzed by the diffusion method with several extract concentrations (0.75 mL, 1.5 mL, 2.25 mL, and 3 mL), 10% DMSO as the negative control, and ciprofloxacin as the positive control. Our result shows that Termoactinomyces sp. (H24) extract has an inhibitory effect on the growth of  E. coli and S. aureus with an effective concentration of 2.25 mL (inhibition strength: very strong).

Background: Bovine mastitis, a global disease that is responsible for large economic losses each year due to lower milk yield and reduced milk quality. In some countries, especially in China, Streptococcus agalactiae has become one of the most frequently detected pathogen. Antibiotic treatment and vaccine immunization are important strategies for the control of infectious diseases. The main objective of the present study was to evaluate distribution of bovine mastitis pathogens and antimicrobial resistance of S. agalactiae, and contribute to the treatment of bovine mastitis.Materials, Methods & Results:Clinical mastitis samples (n= 1,122) were collected from 27 dairy farms located in 15 different provinces of China during 2012-2018. Thepathogens were identified by 16S rDNA method. Antimicrobial susceptibility was assessed by disc diffusion method. Molecular characteristics was distinguished based on PCR. The results showed that the main pathogens were Streptococcus agalactiae (n= 324, 26.2%), Escherichia coli (n= 287, 23.2%), and Staphylococcus aureus (n= 131, 10.6%). The serotypes of Streptococcus agalactiae were serotype II (53.6%), Ia (44 %) and VII (1.2%), respectively. Streptococcus agalactiae were resistant to kanamycin (93.8%), gentamicin (49.4%), vancomycin (49.4%), tetracycline (35.8%), clindamycin (34.6%) and erythromycin (32.1%). The main resistance genes were ermA (53.1%) and ermB (85.2%). Resistance to erythromycin was attributed to the genes ermA (P < 0.05) and resistance to tetracycline was attributed to the genes tetK, tetM, tetO (P < 0.01). The virulence genes scpB (81.4%), cyl (100%), glnA (76.6%), cfb (98.8%), hylB (98.8%), scaA (69.1%) were detected in almost all isolates.Discussion: In the present study, Streptococcus agalactiae, Escherichia coli and Staphylococcus aureus were the pathogens isolated most frequently from clinical mastitis. In the case of S. agalactiae, we performed capsular serotyping of isolates. As a result, serotype II (53.6%), Ia (44 %) and VII (1.2%) were detected whichrevealed variation in the distinct geographical areas. We found that serotypes (Ia and II) and β-hemolytic have significant correlation (P < 0.01) in all isolated strains. We made an assumption that either in processes of capsular and haemolytic appearance effected the expression of another. The unclear mechanism remains to be resolved in the future. Penicillin was recommended as a preferred antibiotic for the treatment of both human and bovine S. agalactiae infection. In the present study, resistance to erythromycin and clindamycin were observed in 32% and 34.6% of our strains, respectively. The results indicated that the ermB gene was most frequent among the erythromycin-resistant S. agalactiae. However, we found that the susceptibility to erythromycin and gene ermA have a significant interaction, while susceptibility to erythromycin and gene ermB have a not significant interaction by analyzing the relationship of phenotypic and genotypic resistance. The severity of S. agalactiae infections may be determined by various virulence factors. Surface enzyme ScpB, a C5a peptidase, encode by scpB gene, could promote bacterial invasion of epithelial cells by attenuating recruitment of polymorphonuclear leukocytes to the site of infection. In the present study, the scpB gene was found in 81.4% of all strains. The results suggested the cyl, cfb, hylB and scpB genes may play an important role in the virulence of Streptococcus agalactiae pathogens.

Diseases are the worst enemy to man currently. This study was aimed at isolating pathogenic bacteria from water obtained from shallow wells in Dundori Kenya. Also, the study aimed at testing the isolates for sensitivity to antibiotic metabolites previously extracted from Actinomycetes isolates from soils of Egerton University. Water samples were collected from shallow wells randomly selected from Dundori and abbreviated as A, B, C, D, and E. Bacterial pathogens were isolated from the water samples using the membrane filtration technique. The isolates were characterized using biochemical means. Antimicrobial sensitivity testing was carried out using Kirby Bauer disk diffusion method. Data analysis was carried out using the Statistical Package for Social Sciences (SPSS). Comparison of means was carried out using one way ANOVA. Shallow wells B, D and E were highly contaminated with pathogenic bacteria. Biochemical characterization of the isolates indicated that the most common isolates were Vibrio cholera, Klebsiella pneumoniae, Proteus sp, Escherichia coli, and Staphylococcus aureus. There was no significant difference between the zones of inhibition produced by the antibiotic metabolites (F=2.149 P=1340) when tested against the test isolates. There were no significant differences between the MIC's of the antibiotic metabolites on the bacterial pathogens (F=2.01 P=0.15). Water from some shallow wells in Dundori is highly contaminated with Klebsiella pneumoniae, Escherichia coli, Proteus sp., Vibrio cholerae and Staphylococcus aureus. The pathogens can effectively be controlled using antibiotics from the Actinomycetes. There is a need to sensitize the residents of the study area on ways of preventing seepage of contaminants into the shallow wells.

This study aimed to determine the phytochemical and antimicrobial activity of jasmine flower. Extraction was done by maceration, such as using water solvent, methanol, ethyl acetate, and hexane as the solvent. The sample bacteria used were Escherichia coli and Staphylococcus aureus. This study used disc diffusion method, with a complete 2-factor random design and 4 replications. Processing of the data is using ANOVA. The results showed that the jasmine extracts with a concentration of 25%, 50%, 75% and 100% had an effect to inhibit of the growth of the bacteria. Jasmine extract with ethyl acetate solvent was the most effective to inhibit the growth of bacteria. It was known that jasmine flower extract is one of the good natural antimicrobials gave an inhibitory effect to the growth of Escherichia coli and Staphylococcus aureus bacteria. Kedua bakteri ini memiliki sifat yang berbeda tetapi memilki kesamaan dari penyakityang disebabkannya. Kualitas dan jenis madu juga sesuai dengan tempat dan lokasi lebah berkembang biak.Tujuan penelitian ini adalah untuk melihat perbandingan efek antibakteri madu asli Sikabu dan Lubuk Minturunterhadap Escherichia coli dan Staphylococcus aureus secara in vitro. Metode Penelitian: Madu yang diuji untukpenelitian ini adalah madu yang berasal dari Sikabu dan Lubuk Minturun. Penelitian ini dilaksanakan pada bulanDesember 2011 sampai Mei 2012 di laboratorium Mikrobiologi Fakultas Kedokteran Universitas Andalas. Jenispenelitian yang digunakan adalah eksperimental dengan menggunakan metode difusi (metode cakram) dananalitik dengan menghubungkan perbedaan yang dimiliki kedua madu sebagai efek antibakteri. Both of these bacteriahave different properties but have the common of the diseases it causes. The quality and type of honey is also inaccordance with the breeding places and their location. The purpose of this study was to see a comparison of theantibacterial effect of honey Sikabu and Lubuk Minturun against Escherichia coli and Staphylococcus aureus by invitro. Methods: Honey to be tested for this study were derived from Sikabu and Lubuk Minturun. This study wasconducted in December 2011 to May 2012 in the laboratory of Microbiology, Faculty of Medicine, AndalasUniversity. Type of research is experimental diffusion method (disc method) and analytical distinction owned bylinking the two of honey as an antibacterial effect. Results: These results indicate that the honey Sikabu and LubukMinturun have no antibacterial effect against Escherichia coli, both of honey have differences antibacterial effectagainst Staphylococcus aureus.
