The objective of this study was to evaluate the reuse of an intravaginal progesterone (P4) device during diestrus of Nelore cow embryo recipients in order to improve conception rate, accelerate estrus cycle re-synchronization of non-pregnant animals, and reduce the amount of waste generated by animal breeding biotechnologies. Two experiments were performed using 268 multiparous Nelore cows. In experiment 1, all animals were subjected to a timed embryo transfer (TET) procedure, but at the time of embryo transfer, two treatment groups were established: T1 - the control treatment (N = 132) and T2 - animals receiving a second-use CIDR® device for 12 days (N = 136). Experiment 2 was performed on cows that had not remained pregnant after experiment 1 using two groups: G1 - a control group (N = 69) and G2 -re-synchronized cows that received a P4 device for 12 days for the first TET (N = 74). In experiment 1, no significant effect of the P4 treatment was observed on conception rate (T1 = 37.9%, T2 = 39.7%; P = 0.50) and corpus luteum (CL) diameter (T1 = 17.5 ± 3.4 mm, T2 = 18.1 ± 3.4 mm; P = 0.61). In experiment 2, no significant effect of the treatment was observed on conception rate (G1 = 22.2%, G2 = 35.7%; P = 0.24), recipients utilization rate (G1 = 75.4%, G2 = 70.3%; P = 0.86), and CL diameter (G1 = 17.4 ± 3 mm, G2 = 18.1 ± 3.2 mm; P = 0.27). However, the P4 treatment (for re-synchronization) significantly increased the conception rate (G1 = 22.2%, G2 = 35.7%; P = 0.04), which was similar to that in a conventional TET protocol performed with a new device (38.8%). We conclude that reusable intravaginal P4 devices can help accelerate TET protocols, suggesting an alternative application method; furthermore, this protocol may help reduce waste production in assisted animal breeding.

Background: The activation of Nrf2/HO-1 pathway has been shown to protect against cisplatin- induced nephrotoxicity by reducing oxidative stress. Berberine (Ber), an isoquinoline alkaloid, has demonstrated antioxidant, anti-inflammatory and anti-apoptotic activities in various experimental models. Aim: To check the effect of Ber on cisplatin-induced nephrotoxicity and to explore the involved mechanism. Methods: Adult male Wistar rats were divided into 6 groups: Normal, cisplatin-control, treatment groups and per se group. Normal saline and Ber (20, 40 and 80 mg/kg; p.o.) was administered to rats for 10 days. A single intraperitoneal injection of cisplatin (8 mg/kg) was injected on 7th day to induced nephrotoxicity. On 10th day, rats were sacrificed, the kidney was removed and stored for the estimation of various parameters. Results: As compared to cisplatin-control group, Ber pretreatment improved renal function system and preserved renal architecture. It also diminished oxidative stress by upregulating the expression of Nrf2/HO-1 proteins. In addition, Ber attenuated the cisplatin mediated inflammation and apoptosis. Furthermore, it also reduced the phosphorylation of p38/JNK and PARP/Beclin-1 expression in the kidney. Conclusion: Ber attenuated renal injury by activating Nrf2/HO-1 and inhibiting JNK/p38MAPKs/ PARP/Beclin-1 expression which prevented oxidative stress, inflammation, apoptosis and autophagy in renal tissue.

The present study was conducted to investigate the efficacy of Ovsynch and Ovsynch plus protocol in postpartum (day 60) Sahiwal cows (n=18). Animals were randomly divided into three equal groups, viz., Ovsynch group Ovsynch plus group and Control group. Animals of group I (n = 6) were treated with traditional Ovsynch protocol. The animals (n = 6) of group II were treated with Ovsynch plus protocol which consisted of an initial intramuscular injection of eCG (Folligon) @ 250 IU on day 60 postpartum followed 3 days later by GPG (Ovsynch) protocol. In group-III Control, no treatment was given to animals (n = 6). Treated animals were inseminated at a fixed time between 14 and 20 hrs after second GnRH injection, irrespective of estrus detection. Blood samples were collected from each animal on days 50 and 60 postpartum to determine the status of cyclicity in animals based on serum concentrations of progesterone (P4). A third blood sample was collected on the day of prostaglandin treatment to determine the response of first GnRH injection. Four animals each were cyclic, and two were acyclic in both treatment groups. Four animals each responded to first GnRH treatment in both treatment groups. Similarly, two animals each got conceived giving conception rate of 50% (2/4) in each treatment. In the control group, one out of 6 animals got conceived yielding 16.66 % conception rate (1/6) during the study period. It may be thus concluded that Ovsynch and Ovsynch plus protocol may be used during the early postpartum period to improve the reproductive efficiency in postpartum Sahiwal cows.

To advance the use of embryo vitrification technology in veterinary practice, we developed a system in which embryo vitrification, warming, and dilution can be performed within a straw. An in-straw embryo cryopreservation method reduces the need for equipment and technical skills and can facilitate direct embryo transfer to the uterus. This study proposes the use of a new device named “Sarah” that is designed to permit all in-straw embryo cryopreservation procedures. Ovine in vitro-produced (IVP) embryos were vitrified at either early blastocyst stage (EB, n = 65, 6 days post-IVF) or fully expanded blastocyst stage (FB, n = 168, 7 days post-IVF). The vitrification procedure using Sarah constituted a 0.25-mL straw with a capsule having 50-µm pores inserted at one end. Embryos at each stage (EB and FB) were divided into 2 subgroups and vitrified by 1 of 2 methods: (1) multi-step (MS) group-a straw containing 2 embryos was sequentially loaded vertically into 1.5-mL tubes containing 6 different vitrification solutions: 10, 20, 40, 60, 80, or 100% ES (with 100% ES being 7.5% DMSO +7.5% EG + 20% FCS in TCM-199; 90 s each step) followed by 30 s each in 75 and 100% VS (100% VS being 18% DMSO +18% EG + 0.5 M trehalose + BSA in TCM-199); and (2) two-step (TS) group-the straw (2 embryos/straw) was loaded with 100% of ES (5 min), followed by 100% VS solution for 30 s. For both methods, at the end of the preparation steps, the straws were plunged directly into liquid N2. Non-vitrified embryos were maintained in in vitro culture as a control group (n = 102). The warming procedure consisted of placing the straws directly into 5-mL tubes containing 100, 50, 25% WS (WS = 1 M sucrose in TCM-199+ 20% FCS) at 38.6°C (for first solution) and at room temperature for all the rest (5 min each), before being placed into the holding medium. Embryos were recovered from the straws, incubated at 38.6 C in 5% CO2 in air in TCM 199 + 5% FCS, and evaluated for blastocoel re-expansion, embryo survival, and hatching rate at 2, 14, 48 h post-warming. Blastocyst re-expansion (2 h) after warming increased as the developmental stage progressed and was not affected by the vitrification method. In fact, it was significantly (P < 0.05) higher for FB vitrified in the MS and TS methods (77.90% and 71.25%, respectively) compared with the EB method (62.5% and 48.50%, respectively). At 24 h, survival rate of vitrified FB was significantly higher (P < 0.05) in the MS system (95.35%) compared with those in TS (86.25%). Survival rates of FB embryos for both methods (MS and TS) were significantly higher (P < 0.001) than EB embryos vitrified in MS (56.25%) and TS (56.55) methods. After 48 h of culture, the hatching rate for FB vitrified in the MS system (87.21%) was comparable with TS (77.5%) and control (85.3%) groups but significantly higher (P < 0.001) than vitrified EB in MS (43.75%) and TS (36.36%). In conclusion, we showed that a high survival rate of IVP embryos can be achieved by this new in-straw vitrification and warming device (“Sarah”), with hatching rates in vitro comparable with that of control fresh embryos. This method has the potential for use in direct embryo transfer in field conditions.

Background: The god’s crown fruits have properties as antioxidants and anti-inflammatory. Toxic doses of paracetamol can injure the liver through toxic metabolite bonds with cytoplasmic proteins that cause free radicals to form. The aim of this research is to know the effect of the crown of gods extracts on paracetamol hepatotoxicity. Method: A total of 30 of Webster swiss mice with a weight of ± 20 grams, age 3 months were randomly assigned to five groups, negative control, positive control, treatment 1,2 and 3. A 70% ethanol extract of god’s crown fruit given in doses 60mg, 120mg and 240mg per kgBB mice. The extract was administered for 14 days in the treatment groups, then on day 15 paracetamol ware administered in a given dose 300mg/kgBB for 1 day for the positive control group and treatment groups. On day 16, the serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels were examined from the orbital sinuses and animals terminated liver tissues taken and immediately fixed in 10% buffer formalin for histological examination. Results and Discussion: The 70% ethanol extract of the god’s crown fruits decreased blood serum levels of AST and ALT, and these results were supported by histopathologic scores of the liver in which histopathologic scores were improved with the increasing doses (p < 0.05). The secondary metabolite contents of the god’s crown fruit extract served as an antioxidant and anti-inflammatory, protecting hepatic injury from the toxic metabolite of paracetamol. Conclusion: A 70% ethanol extract of god’s crown fruit (Phaleria macrocarpa) have hepatoprotective properties that effectively prevent hepatic injury due to paracetamol toxic dose.

Abstract This study evaluated the effectiveness of a combined cognitive training (CT) and physical activity (PA) intervention in improving cognitive function for institutionalized older residents with cognitive impairment. An experimental design with pre/post-test evaluations in a double-blind assessments at three points (baseline, T0; post-treatment, T1; 8-weeks-follow-up, T2), conducted an 8-week-CTPA intervention. Participants (N=134) were recruited from 12 institutions. Centers were randomly assigned into wait-list control, treatment I or treatment II groups. Treatment I group (low frequency) underwent combined 30-minutes sessions of individual-non-computer-based multi-domain CT (twice a week) with 30-minutes-group-chair-based PA (3 times a week). Treatment II group underwent the same protocol as Treatment I group, but with high frequency, 5 days per week for both CT and PA. The primary outcome, Cognitive Assessment Screening Instrument(CASI) total scores showed significant improvement in the treatment I and treatment II groups at T1-T0 and T2-T0, compared to the wait-list control group(10.55±9.60, 12.75±11.64, -8.01±6.61, p=0.000; 8.32±7.81, 11.75±10.19, -7.11±5.78, p=0.000), however there were no significant differences between two treatment groups. In CASI’s nine sub-domains, all the mean difference between groups were also significant at T1-T0 and T2-T0 (all p&lt;0.05). The two treatment groups only significantly differ on CASI-ORIENT domain at T2-T0 (p=0.02). The findings revealed that a combined CT-and-PA intervention have positive immediate (T1-T0) and delayed (T2-T0) effects in cognitive function for older institutional residents with cognition-impairment. The two treatment groups did not show dose-response relationship. Even more, the low frequency intervention was more effective on several domains than high frequency intervention did.

The purposes of this investigation were to determine the effects of different drugs such as GnRH analogue, vitamin AD3E and combination of vitamins and minerals on induction of cyclicity followed by conception in crossbred anoestrus heifers. The effects of body condition score (BCS) and age at anoestrus heifers during treatment on induction of cyclicity and conception were also determined. Sixty crossbred anoestrus heifers having > 2.5 BCS at >24 months age at Potiya Upazilla of Chittagong district were used. The cyclicity of the cows was confirmed by using real time ultrasonography on the basis of presence or absence of corpus luteum. The anoestrus heifers were allotted for 1 to 4 different treatment groups with GnRH analogue, vitamin AD3E and vitamin mineral solution including one no-treatment control group. AI was done in observed oestrus heifers and the conception was determined transrectally by using B-mode real time ultrasonography between 28-35 days post-Al. Heifers showed oestrus at higher (46.67%) proportion in treatment groups than that in no-treatment counterpart (6.67%) (P<0.05). The conception rate was 35.56% in treatment received heifers. The GnRH treated heifers showed the highest (60.00%) proportion of cyclicity and conception rate (46.67%) whereas vitamin mineral solution treated heifers showed the lowest cyclicity (33.33%) and conception rate (26.67%) (P<0.05). Heifers with BCS 3.5 had the highest (57.14%) rate of oestrus induction and (57.14%) rate of conception (P<0.05) whereas heifers with BCS 2.5 had the lowest (41.67%) rate of oestrus induction and (20.83%) rate of conception (P>0.05).  Heifer showed highest rate (50.00%) of cyclicity and (38.89%) rate of conception when their age ranges from 36 to 42 months where as heifer showed lowest (37.50%) rate cyclicity and (31.58%) rate conception when their age ranges from 24 to 29 months (P>0.05). In conclusion, treatment of crossbred anoestrus heifers with drugs has positive effect on induction of cyclicity followed by conception. GnRH may be the drug of choice among the drugs tested in this study. The BCS of heifers during treatment influences the treatment outcome with respect to induction of cyclicity followed by conception.

Carboxylated poly-l-lysine (CPLL) is an ampholytic polymer compound, and it is obtained by converting 65% amino groups to carboxyl groups after synthesising ε-poly-l-lysine aqueous solution and succinic anhydride. CPLL has cryoprotective property similar to antifreeze protein, and addition of CPLL into cryopreservation medium improves the post-thaw survival rate of cells and embryos. In this research, we examined the effectiveness of CPLL as a bovine germ cell cryoprotective material. In experiment 1 (in sperm), the conventional cryopreservation medium used for control group was consisted of 6.5% (vol/vol) glycerin, and the cryopreservation medium used for CPLL group was consisted of 3.25% (vol/vol) glycerin and 0.5% CPLL (wt/vol). The post-thaw survival and motility were assessed by using Sperm Motility Analysis System (DITECT Corp., Tokyo, Japan). There was no significant difference for post-thaw survival rate and motility (control v. CPLL; 98.8% v. 96.6% and 69.7% v. 62.2%, respectively). Artificial insemination was carried out in 65 cows (control v. CPLL; 34 v. 31), and the conception rate of the CPLL group was higher than that of the control group (80.6% v. 67.6%; P = 0.23). In experiment 2 (embryos), the conventional cryopreservation medium used for control group was consisted of 5% (vol/vol) ethylene glycol and 6% (vol/vol) propylene glycol in PBS. In the CPLL group, 7% (wt/vol) CPLL was added to the conventional medium. In vitro fertilization embryos were cryopreserved at Day 7 and Day 8. There was no significant difference in survival rate at 0, 24, and 48 h and hatched rate until 72 h after thawing (control v. CPLL: 93.6% v. 93.2%, 69.0% v. 64.7%, 56.1% v. 56.3%, 12.9% v. 10.2%, respectively). Embryos obtained by superovulation treatment and in vivo fertilization at Day 7 were cryopreserved using above 2 media, and transferred non-surgically into synchronized recipient cows (1 embryo per animal). Embryo transfer (ET) was carried out in 81 cows (control v. CPLL: 31 v. 50), and recipients were diagnosed for pregnancy ultrasonically 50 days after embryo transfer. Conception rate of CPLL group was higher than control group (50.0% v. 29.0%; P = 0.063). In both experiments, the significant differences between control group and CPLL group were determined by chi-squared test. The effectiveness of CPLL in cells and embryos has been reported; however, there is no report using CPLL in bovine germ cells. In this research, CPLL improved the conception rate of AI and ET, probably due to its low toxicity and protection of the cell membrane. These results suggest that CPLL is available as a new cryoprotective material for bovine sperm and embryo in slow freezing methods.

Objective: The effect of environmental enrichment on the behaviour of piglets in the nursery phase was evaluated.Methods: A total of 450 hybrid pigs (21 day old), including both females and uncastrated males, weighing approximately 6 kg, were distributed in a completely randomised design with 3 treatments and 3 replicates of 50 animals each. The treatments were: i) pen without environmental enrichment (control), ii) treatment consisting of continuous environmental enrichment (CEE) with rubber balls throughout the experimental period, and iii) treatment consisting of environmental enrichment with washed balls (EEWB) during the whole experimental period which were removed daily for washing. For the behavioural evaluation, 10 animals were randomly selected per replicate. The behavioural assessments were performed once a week, from 8 am to 6 pm, using images captured with a video camera. The data were submitted to non-parametric analyses, the means were compared using the Bonferroni test, and Person’s correlations were also calculated.Results: A statistical difference (p = 0.001) was observed in the B5 (playful) behaviour; the animals in the EEWB treatment group had a higher frequency of this behaviour than animals in the control treatment group. The animals in the control group showed a higher frequency of B7 behaviour (lying down) (p = 0.026) than those in the EEWB and CEE treatment groups. The animals in the control group had a higher frequency of the B9 (belly nosing) behaviour than those in the EEWB group (p = 0.015). There was a tendency towards a higher frequency of behaviour B3 (walking in the pen) (p = 0.067) when the animals received the control treatment than treatments EEWB or CEE.Conclusion: The use of an enriching object improved the animal welfare and altered the correlation between the evaluated behaviours compared to the animals that did not have environmental enrichment.

An experiment was designed to evaluate alternative treatment protocols for fixed-time embryo transfer (FTET) in recipients in order to reduce cattle handling. Non-lactating, cycling Bos taurus � Bos indicus beef cows (n = 918), with a body condition score (BCS) of 2.5 to 3.5 out of 5, received a progesterone-releasing device (DIB; Syntex, Buenos Aires, Argentina) and 2 mg estradiol benzoate (EB; Syntex) IM on random days of the cycle (Day 0); DIB were removed on Day 8. Cows were placed randomly into 6 treatment groups in a 3 � 2 factorial design. The factors evaluated were time of administration of PGF and eCG [control treatment: PGF Day 5 and eCG Day 5 (Bo et al. 2002 Theriogenology 57, 53–72); PGF Days 0 and 8 and eCG Day 5; PGF Days 0 and 8 and eCG Day 8] and the time of administration of the second injection of EB (at DIB removal or 24 h later). Therefore, cows in the control treatment received 150 �g of d-Cloprostenol (PGF, Ciclase; Syntex) and 400 IU eCG (Novormon; Syntex) on Day 5, whereas cows in the other 2 treatments received 75 �g of PGF on Days 0 and 8 and 400 IU eCG on either Day 5 or Day 8. Each treatment group was further subdivided to receive 1 mg EB IM on either Day 8 (EB Day 8) or Day 9 (EB Day 9). All recipients, whether observed in estrus or not and with more than one CL with luteal area (total CL area minus cavity area) &gt;76 mm2 on the day before embryo transfer, received frozen-thawed embryos (IETS Grade 1) on Day 16 (EB Day 8) or Day 17 (EB Day 9) by direct transfer. Pregnancy was determined by ultrasonography 30 days after FTET. Continuous data were analyzed by ANOVA and pregnancy rates were analyzed by logistic regression. No effects of body condition score or technician were detected (P &gt; 0.1). Although there were no differences in conception rates between recipients treated with EB on Day 8 or Day 9 (168/321, 52.3% vs. 203/350, 58.0%, respectively; P &gt; 0.1), treatment with EB on Day 8 resulted in fewer recipients selected/treated (321/460, 69.7% vs. 350/458, 76.4%; P &lt; 0.05) and pregnant/treated (168/460, 36.5% vs. 203/458, 44.3%; P &lt; 0.05) than 24 h later (EB Day 9), respectively. The mean interval from DIB removal to estrus was shorter (P &lt; 0.05) for recipients in the EB Day 8 (28.1 � 7.2 h) than in the EB Day 9 (43.5 � 9.4 h) group, and the mean (� SEM) CL area was also less in the EB Day 8 group than in the EB Day 9 group (261.7 � 96.7 vs. 293.1 � 114. mm2; P &lt; 0.05). The number of recipients selected/treated was higher in the group receiving PGF on Days 0 and 8 and eCG on Day 8 (243/305, 79.6%) than in the group receiving PGF on Days 0 and 8 and eCG on Day 5 (208/301, 69.1%) or the control group (220/312, 70.5%). Although the luteal area was greater (P &lt; 0.05) in the PGF on Days 0 and 8 and eCG on Day 5 group (304.4 � 127.1 mm2) than in the groups receiving PGF and eCG on Day 5 (274.6 � 98.6 mm2) or PGF on Days 0 and 8 and eCG Day 8 (258.4 � 90.2 mm2), pregnancy rates did not differ between PGF and eCG treatment groups (P &gt; 0.05). It was concluded that it may be possible to reduce animal handling by treating recipients with a half dose of PGF at the time of DIB insertion and removal, when eCG is also administered, without compromising pregnancy rates.

The aim of study was to determine the impact of different selenium sources on blood biochemical picture of Aseel. Three-weeks-old 240 Aseel chickens (120 and 120) from Lakha, Mushki, Peshaweri and Mianwali varieties were randomly distributed into four groups; 60 birds/variety (30 and 30). The birds of each group of either sex were further subdivided into A, B (experimental) and C (control) treatment groups with 10 birds each. Group A and B were fed the diet containing 0.3 mg/kg Se-enriched yeast and sodium selenite, respectively, while group C was fed diet without selenium (control diet). Birds were maintained individually in cages from 4 to 21 weeks. In blood serum, glutathione peroxidase (GPx) levels increased (P£0.05) while, total cholesterol and thyroxin (T4) levels decreased (P£0.05) in Se-yeast supplemented group than sodium selenite supplemented or control group. The study concluded that Se-enriched yeast gave the superior performance to improve blood biochemical profile of Aseel.
