Effects of Selected Antioxidants on Atherosclerosis in Hyperlipidemic Wistar Rats

The interplay of hyperlipidemia and oxidative stress in atherosclerosis has been fairly established by previous studies. There remains however, paucity of data in this environment on the direct effects of antioxidants on atherosclerosis. This study therefore aimed at determining the protective effects of EDTA, vitamin C and Vitamin E on atherosclerosis in diet induced heperlipidemic wister rats. Thirty Wister rats were investigated in this study. The rats were randomly divided into five groups (n=6). The control group was fed with growers mash and water only while group II-V were induced with hyperlipidemic diet for ten weeks. In addition to the hyperlipidemic diet; group III received 1 g/kg body weight of EDTA, group IV received 1 g/kg body weight of vitamin C, group V received 1 g/kg body weight of vitamin E, and group VI received EDTA, vitamin C and E. The group’s treatments were orally for two weeks. C-reactive protein, Total cholesterol (TC), Triglyceride (TG), HDL-cholesterol, LDL-cholesterol, Total calcium and Total antioxidant status were analyzed using standard methods after the treatments. The study showed significant effect in the use of EDTA, Vitamin C and Vitamin E in the treatment of atherosclerosis in rats which could be due to their antioxidant and anti-hyperlipidemic properties. Therefore the combinations EDTA, vitamin C and vitamin E appear greatly protective against atherosclerosis.

The current study was designed to determine the antioxidant effects of vitamin C and vitamin E against oxidative stress induced by vancomycin in some antioxidants changes in the male rats. The study was conducted in the animal house of the Faculty of Science/University of Kufa for the period from April, 2018 to May, 2018 on 119 animals of male rats aged 2.5–3 months and the weight of 150-200 gm. Two experiments designed in this study addressed the first and two experiments to study the oxidative effect of vancomycin in addition to the protective effects of vitamin C and vitamin E to reduce these effects in the treatment of animals for one week and three weeks with vancomycin and vancomycin plus vitamins. The results indicated a significant increase (p less than 0.05) in the MDA, CAT, and significant decrease (p less than 0.05) in SOD, and GPX. In the animals treated with vancomycin 40,60 mg/kg only compared to the control group for the two periods of administration at the same time occur a significant decrease(p less than 0.05) in the MDA, CAT and a significant increase (p less than 0.05) in the SOD and GPX after treated animals with vancomycin 40,60 mg/kg with vitamin C and vitamin E for a period of one and three weeks compared with vancomycin group.

Abstract Background Circadian disruption is often associated with aggravation of atherosclerosis; however, the pathophysiological mechanisms underlying atherogenic initiation in normolipidemic diet remains unclear. Most of the studies done for understanding circadian disruption induced atherosclerosis have been carried out in murine model of hyperlipidemia induced atherosclerosis. The present study investigates pro-atherogenic events in response to chronic photoperiodic manipulation induced chronodisruption (PMCD) in C57BL/6J mice fed with laboratory chow diet. Results The results were compared with atherogenic initiation induced by high fat high fructose (HFHF) diet. The combined effects of HFHF and PMCD on atherogenic initiation were also investigated for possible synergy of both variants. The HFHF and HFHF+PMCD groups recorded increments in body weight gains and serum lipid parameters (TC, TG, LDL-cholesterol, VLDL) and a decrement in HDL-cholesterol as compared to the control group. However, PMCD group recorded body weight gain similar to that of the control group, but the serum lipid parameters (TG and VLDL) were significantly elevated and the HDL levels were lowered. However, prominent hypertrophic remodeling, higher collagen deposition, and elastin derangement, along with endothelial dysfunction, its activation, and macrophage infiltration, were observed in thoracic aorta of all the three experimental groups. But the mRNA and immunoblots of heat shock protein 60 (HSP60) in thoracic aorta was found to be maximum in PMCD followed by HFHF and HFHF+PMCD groups. Conclusion Laboratory chow feeding coupled with photoperiodic manipulation mediated chronodisruption overexpress HSP60 that in turn plays a central role in PMCD mediated pro-atherogenic remodeling in thoracic aorta of C57BL/6J mice.

Cisplatin-induced nephrotoxicity is associated with an increase in lipid peroxidation and oxygen free radicals in rat kidneys. In this study, the effects of desferrioxamine were compared to vitamin C and E on cisplatin-induced lipid peroxidation and antioxidant enzyme activities in rat kidneys. Rats were divided into five groups, with 15 Wistar rats in each group. In the control group, rats received 1 mL/100 g isotonic saline solution intraperitoneally (i.p.). In Group II, 10 mg/kg cisplatin i.p. was injected to rats. Thirty minutes before the same dosage of cisplatin administration, 100 mg/kg i.p. vitamin C or E was given to rats in groups III and IV, respectively. Rats in Group V received 250 mg/kg desferrioxamine i.p., before the same dose of cisplatin administration. All rats were killed by cervical dislocation after 72 hours. The kidneys were immediately removed and washed in cold saline. Spectrophotometric method was used for all analyses. While catalase, glutathione reductase (GR), and super oxide dismutase (SOD) levels were found to be significantly decreased (P B < 0.001), malondialdehyde (MDA) (P < 0.05) and hydrogen peroxide (H2O2) (P < 0.001) levels were significantly increased in the cisplatin group when compared to the controls. MDA levels were decreased by desferrioxamine (P < 0.005) as well as vitamin C and E (P < 0.05 and P < 0.001, respectively). These three compounds induced a significant increase in SOD levels (P B < 0.05), but only in the vitamin C group, were SOD levels not significantly different than the levels of the controls (P > 0.05). In the desferrioxamine (P < 0.05), vitamin C and E groups (P < 0.001 for both), the cisplatin elevated H2O2 levels were decreased. None of these drugs had any effect on GR and catalase levels (P > 0.05). Desferrioxamine is useful to prevent cisplatin-induced lipid peroxidation, however, vitamin C and E are more effective on antioxidant enzymes than desferrioxamine.

The present study was undertaken to find out the efficacy of vitamin E and/or vitamin C against mercury (Hg) induced toxicity in mice. Sixty mice were randomly divided into 5 equal groups (n=12). One group of mice (Group A) was kept as control and each of rest four groups (B, C, D and E) were fed with mercuric chloride (HgCl2) in drinking water @ 65 mg/L. In addition to HgCl2 alpha-tocopherol (vitamin E) @ 100 mg/L, ascorbic acid (vitamin C) @ 250 mg/L and combination of vitamin E and vitamin C at same dose were given to the mice of groups C, D and E respectively. All treatments were continued for 28 consecutive days. Four mice of each group were sacrificed on day 1, 14 and 28 and efficacy of vitamin E and vitamin C against Hg induced toxicity were evaluated by observing toxic signs, body weight, hemato-biochemical parameters and postmortem lesions. Mild (++) toxic signs as evident by reduced feed and water intake, salivation, vomiting, excitement, muscle tremor, ataxia, restlessness, incordination and ruffled hair coat were observed from 2nd week (group B) and from 3rd week (group C and D) by intoxication with HgCl2. Significant (P<0.01) reduction of body weight (18.38%) and hematological parameters i.e. TEC (19.88%), TLC (27.89%), Hb content (34.09%) and PCV (9.15%) were observed at day 28 in HgCl2 induced intoxicated mice (group B). At identical period in same group biochemical parameters i.e. AST (46.99%) and ALT (58.72%) increased significantly (p<0.01). Pinpoint hemorrhages throughout the liver and highly (++++) congested kidney was also observed at post mortem (group B). All the parameters i.e. toxic signs, body weight, hemato-biochemical and post mortem lesions were found to be slight (+) or mild (++) and/or improved in rest three groups of mice following treatment with vitamin E, vitamin C and combination of vitamin E and vitamin C. The present study reveals that vitamin E and C have a protective role against Hg poisoning. AbstractThe aim of the study was to assess the gut microbiota and selected haematological and biochemical blood parameters of weaned piglets following dietary supplementation with a probiotic and vitamin C. the piglets were divided into a control group (group C) and an experimental group (group E), with 30 piglets in each group. All animals received the same feed ad libitum. The animals in the control group (group C) received feed with no added probiotic and vitamin C. the piglets in the experimental group (group E) were given a supplement containing a probiotic (Bacillus cereus 1×109 CFU/kg) in the amount of 1.5 g/piglet/day and vitamin c in the amount of 300 mg/piglet/day. The supplement was administered for 28 days. The total numbers of bacteria of the family Enterobacteriaceae and of the genus Lactobacillus were determined in faeces. The erythrocyte count, haemoglobin level, haematocrit, leukocyte count, and percentages of neutrophils and lymphocytes were determined in the blood. The biochemical analysis concerned the concentration of triacylglycerols, total cholesterol, and LDL and HDL cholesterol. In group E a significant decrease (P<0.01) in the total number of Enterobacteriaceae bacteria was observed in the faeces of the piglets, accompanied by an increase in the number of lactobacilli relative to group C. E. coli was found to predominate over other microorganisms. Salmonella choleraesuis bacteria were present in the faeces of both groups before administration of the supplement, but were not found after its use. The supplement with probiotic and vitamin C caused a significant increase in the erythrocyte, haemoglobin and haematocrit levels in the blood of the piglets and a significant decrease in the concentration of triacylglycerols, total cholesterol and LDL cholesterol.

The transition from experimentally induced poor nutritional conditions to feeding was studied with 69 eight-month-old female reindeer (Rangifer tarandus tarandus). During a pre-experimental period, all reindeer were fed a simulated winter diet with 80% lichens Cladina spp. and 20% Vaccinum myrtillus shrubs and Salix spp. leaves (lichen diet) ad lib. The reindeer were divided into five groups. A control group (group C) was fed the lichen diet ad lib. throughout the experiment. Four groups were fed half of that ration for eight days and were then totally deprived of feed for one day (restriction period). During the following 34 days (feeding period) the groups were re-fed the lichen diet (group L), fed pelleted reindeer feed combined with either lichen (group PL) or grass silage (group PS), or fed silage with a gradually increasing addition of pellets (group SP). Weekly measurements of blood samples and body weighr showed that the control group remained clinically healthy and had stable blood plasma concentrations of protein, urea, glucose and insulin throughout the experiment, but they lost weight. At slaughter, before and after the restriction period, all animals had lost rumen-free body weight, but the reindeer fed a restricted amount of feed lost more than the control group. Also the plasma metabolites were affected by the restricted feeding, with increased concentrations of urea and decreased concentrations of glucose. Group L responded immediately to the ad lib. feeding with blood metabolite levels rapidly approaching those of group C. The body weight developments were similar in groups L and C. Although the feed rations were increased gradually, diarrhoea occurred in some animals belonging to groups PL and PS within the first week of the feeding period. All reindeer recovered, after antibiotic treatment of the worst affected animals. The PL and PS groups, which had high contents of metabolisable energy and crude protein in their diets, showed increased con-centtations of plasma protein, urea and insulin. At the end of the feeding period, these groups had increased their body and carcass weights and gained fat, whereas reindeer fed the lichen diet had lost weight. Severe health problems (malnutrition and so-called wet belly) occurred in group SP during the first weeks of feeding and led to loss of animals, and consequently the SP group was excluded from the remainder of rhe experiment. The general conclusion is that the lichen diet did not cause any digestive problems, but resulted in a continuous decline in body weight and small or deficient fat reserves. After the initial diarrhoea, feeding with diets comprising pellets from the start resulted in improved condition, expressed as increased body weight, fat gain and higher concentrations of plasma protein, urea and insulin in relation to the control group. The diet initially based on grass in the form of silage of the given quality seemed insufficient as feed to reindeer calves in a poor nutritional state.

Free radical that are released in cigarette smoke, believed to play an important role in male infertility. The free radicalproduces Reactive Oxygen Species (ROS) that is found increased in male smokers. ROS is one of the free radicalscomponent can be neutralized by vitamin C and E as an antioxidant. This study using those agents to the fifteen minuteshalf piece cigarette to expose mice and count their sperm as the fertilility parameter. The research design used was posttest-only control group with twenty mice were divided into five groups. A group of no smoke exposed was categorizedas a baseline control, the exposed groups were consisted of one group without antioxidant, one group with vitamin Ctreatment, a group of vitamin E treatment and at last the combination of vitamin C and E. All groups were treatedfor fourteen days. The results were analyzed using oneway ANOVA with p<0.05. It is obviously proven that thecombination of vitamin C and vitamin E treatment on the usual dose gave the highest sperm count compared to othergroups.

Abstract Considering that the involvement of reactive oxygen species (ROS) has been implicated in the toxicity of various pesticides, this study was designed to study the ameliorative effect of Vitamin E (100 mg/kg body weight) on mice (25 - 30 mg) treated with diazinon (32.5 or 16.25 mg/kg body weight) organophosphate insecticide for 14 days. Subchronic DZN exposure and the protective effects of vitamins E (vitE) were evaluated for their effects on haematological indices, the enzymes concerning liver damage [plasma alanine aminotransferase (ALT), aspartate aminotaransferase (AST), alkaline phosphatise (AIP), and some parameters of kidney function (urea and creatinine) in mice. Additionally, the histopathological changes in liver and kidney tissue were examined. The high dose of diazinon (DZNH) decreased the body weight significantly at the end of experiment. Additionally, the liver and kidney were examines for histopathological changes. The high dose of diazinon decreased body weight significantly. Moreover, there was a statistically significant decrease in haemoglobin (Hb), red blood cell (RBC) and hematocrit (Hct) in diazinon-treated mice compared to controls. This decrease was partially remedied in the diazinon-treated group that also received vitE. Damage in the liver and kidney tissues was also evident as elevated plasma ALT, AST, ALP, urea and creatinine. VitE partially counteracts the toxic effect of DZN and repairs tissue damage in the liver and kidney, especially when supplemented to 1/4 LD50 intoxicated animals. Histopathological changes in liver and kidney were observed only in 32.5 mg/kg DZN given group. These results suggest that the effects of DZN are dose dependent. No pathological findings were observed in vitE + DZN treated groups. According to the present study, we conclude that vitE can reduce the detrimental impacts of diazinon on haematological indicies, as well as liver and kidney function.

Obesity increases risk of Alzheimer’s Disease (AD). A high fat diet (HFD) can lead to amyloidosis and amyloid beta (Aβ) accumulation, which are hallmarks of AD. In this study, protective effects of the ethyl acetate fraction of Acer okamotoanum (EAO) and isoquercitrin were evaluated on obesity and amyloidosis in the HFD- and Aβ-induced mouse model. To induce obesity and AD by HFD and Aβ, mice were provided with HFD for 10 weeks and were intracerebroventricularly injected with Aβ25–35. For four weeks, 100 and 10 mg/kg/day of EAO and isoquercitrin, respectively, were administered orally. Administration of EAO and isoquercitrin significantly decreased body weight in HFD and Aβ-injected mice. Additionally, EAO- and isoquercitrin-administered groups attenuated abnormal adipokines release via a decrease in leptin and an increase in adiponectin levels compared with the control group. Furthermore, HFD and Aβ-injected mice had damaged liver tissues, but EAO- and isoquercitrin-administered groups attenuated liver damage. Moreover, administration of EAO and isoquercitrin groups down-regulated amyloidosis-related proteins in the brain such as β-secretase, presenilin (PS)-1 and PS-2 compared with HFD and Aβ-injected mice. This study indicated that EAO and isoquercitrin attenuated HFD and Aβ-induced obesity and amyloidosis, suggesting that they could be effective in preventing and treating both obesity and AD.

Rats were divided into two groups such that mean weight and hemoglobin and hematocrit levels were not significantly different. One group (controls) was then fed a normal chow ad libitum. The other group was fed 6 g daily (30% of normal intake) for 2 weeks. The hemoglobin levels of rats fed the restricted diet rose 1.4–3.5 g/100 ml and hematocrit level rose 2–6%. At the end of 2 weeks total red cell mass (Cr51) was 5.5–6.0 ml in the underfed groups compared with 6.8 ml in the control group. Body weight fell proportionally more than did red cell mass, elevating the calculated red cell mass per unit body weight. Serum osmolality and K+ were not significantly different from control values, and there was a slightly higher serum Na+ and Cl– in the restricted diet group. It is concluded that restriction of food intake produced a relative polycythemia. At the end of 2 weeks of restriction an isosmotic reduction in plasma volume was present.
