Abstract Older adults are viewed as being vulnerable to COVID-19. Previous research revealed that individuals would internalize or dissociate with attitudes toward aging when they aged. In this study, data collected before the COVID-19 pandemic were compared with those collected during the pandemic to assess whether the pandemic might make older adults internalize or dissociate with attitudes toward aging to a greater extent.123 Hong Kong participants (50.4% females, M=60.59±13.28 years old) were recruited in a two-wave survey (wave 1 in 2018 and wave 2 in 2020) on attitudes toward aging and future self-views. After comparing the correlations between attitudes toward aging and future self-views in the two waves, we found stronger positive correlations between these 2 variables in wave 2 than in wave 1 in the personality and finance domains, but not in the family, independence, or health domains. These findings suggest that internalization of attitudes toward aging might be domain-specific. The pandemic might make older adults more likely to internalize positive personality attitudes toward aging and negative finance attitudes toward aging into their future self-views. Professionals may consider utilizing the internalization process to promote a positive attitude toward aging during the pandemic.

Background: Osteoarthritis is the most common chronic disease among elders, and it crucially requires self-management. This study aimed to qualitatively examine exercise experiences of elderly Medicaid beneficiaries with osteoarthritis receiving exercise intervention based on self-determination theory (SDT).Methods: Eight elders who received a community-based exercise intervention were interviewed three times. Using the interview transcriptions and field notes, researchers conducted an interpretative phenomenological analysis based on SDT to explore the motivational process of exercise compliance. Results: Three meaningful profiles (non-compliant, intermitter, and compliant) were identified along with satisfaction with needs (autonomy, competence, and relatedness) that motivate exercise compliance based on SDT. Compliance of exercise is primarily initiated by external regulation and followed by the internalization process of recognizing and accepting the value of the behavior. Conclusions: The concepts of SDT could be used to explore factors associated with motivation and gradual behavior change among elderly Medicaid beneficiaries with osteoarthritis. Autonomy, competence, relatedness, external regulation, and identified regulation control may be applied to establish a strategy that promotes behavior change by satisfying the psychological needs and internalizing the behavior of these elders.

The phenomenon of sexual violence is illustrated as the iceberg phenomenon. Every year the number of cases of sexual violence increases. Among the victims of sexual violence, women are the most common ones. This study aimed to examine how the social construction of sexual violence from the side of women as female politicians. The research method was qualitative. Primary data were obtained through observation and in-depth interviews. The theory employed was the social construction theory by Peter L. Berger and Thomas Luckmann. The results showed that in the externalization process, sexual violence is a reality that still has weaknesses in its handling in Indonesia due to the existence of patriarchal values in society. The objectification process resulted in a debate between the use of the KUHP and the Elimination of Sexual Violence Bill (RUU PKS) as the legal basis for sexual violence. The internalization process is a reinterpretation of sexual violence after externalization and objectivation. This study concludes, in this process, female politicians interpret sexual violence as an act of attacking the sexuality of the victim which damages the physical and psychological aspects.

This in vitro study aims to evaluate the magnetic hyperthermia (MHT) technique and the best strategy for internalization of magnetic nanoparticles coated with aminosilane (SPIONAmine) in glioblastoma tumor cells. SPIONAmine of 50 and 100 nm were used for specific absorption rate (SAR) analysis, performing the MHT with intensities of 50, 150, and 300 Gauss and frequencies varying between 305 and 557 kHz. The internalization strategy was performed using 100, 200, and 300 µgFe/mL of SPIONAmine, with or without Poly-L-Lysine (PLL) and filter, and with or without static or dynamic magnet field. The cell viability was evaluated after determination of MHT best condition of SPIONAmine internalization. The maximum SAR values of SPIONAmine (50 nm) and SPIONAmine (100 nm) identified were 184.41 W/g and 337.83 W/g, respectively, using a frequency of 557 kHz and intensity of 300 Gauss (≈23.93 kA/m). The best internalization strategy was 100 µgFe/mL of SPIONAmine (100 nm) using PLL with filter and dynamic magnet field, submitted to MHT for 40 min at 44 °C. This condition displayed 70.0% decreased in cell viability by flow cytometry and 68.1% by BLI. We can conclude that our study is promising as an antitumor treatment, based on intra- and extracellular MHT effects. The optimization of the nanoparticles internalization process associated with their magnetic characteristics potentiates the extracellular acute and late intracellular effect of MHT achieving greater efficiency in the therapeutic process.

Recently emerged severe acute respiratory syndrome coronavirus (SARS-CoV)-1 and -2 initiate virus infection by binding of their spike glycoprotein with the cell-surface receptor angiotensin-converting enzyme 2 (ACE2) and enter into the host cells mainly via the clathrin-mediated endocytosis pathway. However, the internalization process post attachment with the receptor is not clear for both SARS-CoV-1 and -2. Understanding the cellular factor/s or pathways used by these CoVs for internalization might provide insights into viral pathogenesis, transmission, and development of novel therapeutics. Here, we demonstrated that the cytoplasmic tail of ACE2 is not essential for the entry of SARS-CoV-1 and -2 by using bioinformatics, mutational, confocal imaging, and pseudotyped SARS-CoVs infection studies. ACE2 cytoplasmic domain (cytACE2) contains a conserved internalization motif and eight putative phosphorylation sites. Complete cytoplasmic domain deleted ACE2 (∆cytACE2) was properly synthesized and presented on the surface of HEK293T and BHK21 cells like wtACE2. The SARS-CoVs S1 or RBD of spike protein binds and colocalizes with the receptors followed by internalization into the host cells. Moreover, pseudotyped SARS-CoVs entered into wtACE2- and ∆cytACE2-transfected cells but not into dipeptidyl peptidase 4 (DPP4)-expressing cells. Their entry was significantly inhibited by treatment with dynasore, a dynamin inhibitor, and NH4Cl, an endosomal acidification inhibitor. Furthermore, SARS-CoV antibodies and the soluble form of ACE2-treated pseudotyped SARS-CoVs were unable to enter the wtACE2 and ∆cytACE2-expressing cells. Altogether, our data show that ACE2 cytoplasmic domain signaling is not essential for the entry of SARS-CoV-1 and -2 and that SARS-CoVs entry might be mediated via known/unknown host factor/s.

Character education in Indonesia needs to be instilled from an early age, given the decline in morality of students in the era of globalization. Morality is an element that shows the quality of people's lives. Religious people assume that religion always teaches its followers to do good and prohibits them from doing wrong. Meanwhile, humanists assume that a person's morality is seen from how someone obeys the values and norms in society. Cultivating character education through three stages, namely internalization and externalization. The three processes of planting can show positive results, of course, the character of the students is also good. However, if the three processes are not optimal, the results obtained by students are also not optimal. The internalization process becomes the basic process or foundation for the character education of students, where the family forms the basis of a character in the form of character recognition to students. And externalization is the next process of internalization where this process is filled by education staff, peers, social media.

Abstract The internalization of G protein-coupled receptors (GPCRs) is an important mechanism regulating the signal strength and limiting the opportunity of receptor activation. Based on the importance of GPCRs, the detailed knowledge about the regulation of signal transduction is crucial. Here, current knowledge about the agonist-induced, arrestin-dependent internalization process of rhodopsin-like GPCRs is reviewed. Arrestins are conserved molecules that act as key players within the internalization process of many GPCRs. Based on highly conserved structural characteristics within the rhodopsin-like GPCRs, the identification of arrestin interaction sites in model systems can be compared and used for the investigation of internalization processes of other receptors. The increasing understanding of this essential regulation mechanism of receptors can be used for drug development targeting rhodopsin-like GPCRs. Here, we focus on the neuropeptide Y receptor family, as these receptors transmit various physiological processes such as food intake, energy homeostasis, and regulation of emotional behavior, and are further involved in pathophysiological processes like cancer, obesity and mood disorders. Hence, this receptor family represents an interesting target for the development of novel therapeutics requiring the understanding of the regulatory mechanisms influencing receptor mediated signaling.
