AbstractRubus hyrcanus Juz. (Rosaceae), known as Caspian blackberry, is wildly distributed around the Caspian Sea. This study focused on antioxidant, cytotoxic, and antibacterial activities of total extracts and different fractions from the roots and leaves of this species. The total phenolics and flavonoid contents were also evaluated. Finally, the phenolic profiles of selected fractions were determined using HPLC–DAD and LC–MS/MS. The results indicated that the total phenolics content (TPC) of root total extract (RTE) was 3.5 times that of leaves (340.4 and 102.7 mg GAE/g, respectively). The TPC of three root fractions ranged from 226.6 to 392.9 mg GAE/g, while in leaves fractions, it ranged between 68.3 and 101.8 mg GAE/g. The total extract of leaves had higher contents of total flavonoids than roots (70.5 and 8.9 mg QE/g, respectively). The methanol fractions of both parts had the highest amounts of flavonoids. The root methanol fraction (RMF) had the best antioxidant effect in both DPPH radical scavenging assay (IC50: 9.16 μg ml−1) and total antioxidant capacity test (1010.5 mg ɑTE/g). The RMF and RTE had potent antibacterial activities against Bacillus subtilis and Staphylococcus aureus (MIC 1.5 mg ml−1). In the MTT assay, ethyl acetate fractions of roots and leaves exhibited the best cytotoxicity (IC50 247 and 227 μg ml−1, respectively) and the highest selectivity indexes (4.73 and 5.31, respectively). Phytochemical analysis revealed the presence of gallic acid, p-coumaric acid, and chlorogenic acid in leaves ethyl acetate fraction, chlorogenic acid in leaves methanol fraction, and gallic acid in the root ethyl acetate fraction.

The antioxidant capacity of the crude extract and fractions ofTabernaemontana catharinensis fruits and branches, was evaluated by the 2,2-diphenyl-1-picrylhydrazyl (DPPH) method and the content of polyphenols, flavonoids, alkaloids and condensed tannins were determined by the spectrophotometric method. The ethyl acetate fraction of the fruits and the n-butanol fraction of the branches showed IC50 of 181.82 µg/mL and 78.19 µg/mL, respectively. All fractions were analyzed by high performance liquid chromatography (HPLC), in the branches were quantified chlorogenic acid in the chloroform (8.96 mg/g), ethyl acetate (4.31 mg/g) and n-butanol (3.33 mg/g) fractions; caffeic acid in the ethyl acetate (5.24 mg/g) and n-butanol (1.81 mg/g); gallic acid (0.52 mg/g) in the n-butanol. In the fruits, chlorogenic acid in the chloroform (1.67 mg/g); rutin in the ethyl acetate (3.45 mg/g) and n-butanol (8.98 mg/g) fractions. The present study showed that these quantified compounds can contribute to antioxidant capacity which was higher in the branches than in the fruits.

Abstract Background Dryopteris ramosa has numerous potentials uses in the treatment of different maladies as old traditional medication. The fronds of D. ramose are edible and orally administered for producing antibiotic effect. They are also used as astringent and febrifuge, and as a pesticide. Methods Extraction of fronds of D. ramosa using solvents of increasing polarity, namely, ethyl acetate, methanol and water were tested for phytochemical (qualitative tests, GC-MS), antimicrobial (well method), antioxidant (DPPH), antifungal (tube dilution), cytotoxic activity (brine shrimps lethality assay) and LOX and COX inhibitory activities were performed using standard methods. Results The phytochemical analysis of the crude methanolic extract revealed that the fronds are rich in flavonoids, alkaloids, saponins, tannins, glycosides and triterpenoids. The total flavonoid content of the ethyl acetate fraction was 46.28 μg QE/mg extract. The GC-MS analysis revealed nine major compounds that constituted the crude drug and potentially had a role in reported activities. The crude extract was the most active amongst all the fractions against the bacterial and fungal strains used such that it inhibited the growth of P. aeruginosa with a zone of 13 mm and a MIC value of 16 μg/ml as compared to the standard cefixime, which inhibited the zone by only 10 mm and a MIC value of 32 μg/ml. The highest antioxidant potential in DPPH assay was shown by the crude extract with 91.948% free radical scavenging activity. The bring shrimps lethality potential of the crude extract was the highest, with a LD50 value of 47.635 μg/ml. The ethyl acetate fraction inhibits 91.36% of alpha glucosidase enzyme at a concentration of 0.5 mg/ml. In case of acetylcholine esterase inhibition assay, the methanol fraction inhibits 58.26% of the enzyme activity. Similarly, for butyrylcholine esterase inhibition, the maximum inhibitory effect was seen in the methanol fraction, with a percentage inhibition of 47.32%. Conclusion These test results support traditional medicinal uses of the plant. Dryopteris ramosa could be imperative for being used as a therapeutic agent and the medicinal importance of this plant should be further investigated.

ABSTRAKAedes aegypti merupakan serangga vektor dari virus Dengue penyebab Demam Berdarah Dengue (DBD). Kandungan senyawa dalam tumbuhan lakum berpotensi sebagai larvasida. Buah lakum dimaserasi dengan pelarut methanol kemudian difraksinasi menggunakan etil asetat. Fraksi diteliti kandungan senyawanya menggunakan analisis fitokimia. Uji larvasida diamati selama 48 jam terdiri dari tujuh konsentrasi berseri yaitu kontrol; 0,04; 0,12; 0,2; 0,6; 1; 3% (b/v). Setiap konsentrasi diulang 3 kali dan setiap ulangan terdiri dari 10 ekor larva nyamuk Ae. aegypti. Data mortalitas dianalisis menggunakan analisis probit untuk menentukan nilai LC50. Fraksi metanol buah lakum  berpotensi sebagai larvasida dengan nilai LC50 6 jam sebesar 1,125%. Fraksi etil asetat  buah lakum  berpotensi sebagai larvasida dengan nilai LC50 6 jam sebesar 1,102%. Domin; larvasida; LC50  ABSTRACTLarvacidal activity of methanol and ethyl acetate fraction of lakum fruits (Cayratia trifolia (L.) Domin.) against Aedes aegypti larvae. Aedes aegypti is a vector from the Dengue virus which causes Dengue Hemorrhagic Fever (DHF). The compounds of lakum plants has the potential larvicide. The fruit of C. trifolia was macerated with methanol then fractionated using ethyl acetate as a solvent. The fraction was examined for its compound using phytochemical analysis. Larvacide test was observed for 48 hours consisting of seven concentration of 0 (control); 0.04; 0.12; 0.2; 0.6; 1; 3% (w/v). Each concentration was repeated 3 times and each repetition consisted of 10 larvae of Ae. aegypti. Mortality data were analyzed using probit analysis to determine the LC50 of the fraction of methanol and ethyl acetate from lakum fruit. The methanol fraction has the potential larvicide with a six hour LC50 value 1.125%. The ethyl acetate fraction has the potential larvicide with a six hour LC50 value 1.102%. Phytochemical analysis, antimicrobial and antioxidant properties of the leaf extract and fractions of Sabicea brevipes were studied. The leaves were defatted and the marc extracted with methanol. The extract was further purified by solvent-solvent partitioning using n-hexane, ethyl acetate and n-butanol to obtain the three solvent fractions. They were screened for phenolics, flavonoids, tannins, saponins, terpenoids, glycosides, and steroids. Total phenolics, flavonoids and tannins were determined quantitatively. The antimicrobial test was screened in vitro by agar diffusion method. Analysis of variance (ANOVA) was used to test for significant difference at p ≤ 0.05 in all study groups. The methanol extract exhibited the most significant amount of phenolics (110.78 ± 1.06 mg GAE/g) while ethyl acetate fraction had the least total phenolics content (50.55 ± 2.91 mg GAE/g). The same trend was observed for the total flavonoids content whereas the methanol extract measured (418.40 ± 14.03 mg QE/g) while ethyl acetate fraction had 192.40 ± 3.06 mg QE/g. Total tannins contents were: methanol extract (102.22 ± 7.58 mg GAE/g) and ethyl acetate (27.33 ± 0.77 mg GAE/g). The antioxidant results showed that the methanol extract had the highest DPPH free radical scavenging ability (93.69%) with half maximal inhibitory concentration (IC50) of 0.601± 0.02 and also highest ferric ion reducing power (50.381 ± 1.56 µmol Fe2+/g). Also, the methanol extract showed high total antioxidant capacity (96.79 ± 0.31 mg AAE) and IC50 of 0.798± 0.01. The antimicrobial results revealed that the methanol extract showed better activity against Escherichia coli, Bacillus subtilis, Pseudomonas aeruginosa, Staphylococcus aureus, and Candida albicans than the solvent fractions at concentrations of 200, 100, 50, 25, 12.5, mg/mL having various inhibition zone diameters (IZDs). The methanol extract and fractions of S. brevipes compared favourably in terms of zone of inhibition and minimum inhibition concentration (MIC) with the standard drug disc (Gentamycin and Ketoconazole) against the tested microorganisms. The MIC of the extract and solvent fractions ranged from 6.31 mg/mL to 50.12 mg/mL. The continual use of the extract of Sabicea brevipes in preventing oxidative stress and in the treatment of common infection is justified by these results.

Samama (Anthocephalus macrophyllus) is one of a pioneer indigenous fast growing species in Indonesia which have been used as traditional medicine for various diseases. The objectives were to determine the yield extract, to analyze their antidiabetic activity by inhibition assay for α-glucosidase enzyme activity and chemical analysis with GCMS. Extraction of leaves, bark and wood samples were done by using ethanol 95%. Fractionation the most active  ethanol extract was conducted by using n-hexane and ethyl acetate. The yield of ethanol extracts in leaves, bark and wood were 13.90%, 12.87%, and 2.18% respectively. Based on antidiabetic activity assay, the bark ethanol extract was the most active extract by the IC50 value of 5.86 μg mL-1. Phytochemical analysis on bark ethanol extracts showed that they contained flavonoid, quinon, triterpenoid, saponin and tannin which were assumed have high contribution in antidiabetic activities. The result of fractionation ethanol extract bark showed that the ethyl acetate fraction was the most active fraction (IC50 6.82 μg mL-1). GCMS analysis indicated the presence of dominant phenolic compounds such as pyrocatechol, antiarol, isopropyl myristate and phenol in which were suspected have antidiabetic activity. These results strongly suggested that ethyl acetate fraction of Samama bark was a potential natural source for antidiabetic agents.

Mangosteen leaves have flavonoid compounds, tannins, and saponins that can be efficacious as antibacterial. The aim of this study was to determine the fraction of mangosteen leaves having an antibacterial effect and knowing the class of compounds identified as having antibacterial activity after TLC- Bioautography testing was carried out. The samples were extracted using 96% maceratarion method and fractioned using liquid-liquid fractionation method with methanol, n-hexane and ethyl acetate solvents, antibacterial activity using agar diffusion method (Kirby and Bauer) with 3 concetrations namely 10%, 20% and 30%. Thin Layer Chromatography (TLC) uses n-hexane and chloroform solvens. TLC-Bioautography uses contact bioautography methods. The resultd showed that mangosteen leaves in methanol fraction with a concentration of 30% had a very large inhibitory activity again Staphylococcus aureus and ethyl acetate fraction with a concentration of 30% had the gratest antibacterial activity against  Escherichia coli. The results of the TLC- Bioautography study showed that the flavonoids compounds after spraying with AlCl3 and the mangosteen leaf Biosutography test had inhibitory zone activity against the bacteria Staphylococcus aureus dan Escherichia coli. Keywords: Mangosteen Leaves. Penelitian ini bertujuan untuk mengetahui fraksi daun manggis memiliki efek antibakteri dan mengetahui golongan senyawa yang teridentifikasi memiliki aktivitas antibakteri setelah dilakukan pengujian KLT Bioautografi. Sampel diektrak dengan metode maserasi dengan pelarut 96% dan difraksinasi dengan metode  fraksinasi cair-cair dengan pelarut metanol, n-heksan dan etil asetat, aktivitas antibakteri menggunakan metode difusi agar (Kirby and Bauer) dengan 3 kosentrasi yaitu 10%, 20% dan 30%. Kromatografi Lapis Tipis (KLT) menggunakan pelarut n-heksan dan klorofom. KLT-Bioautografi menggunakan metode bioautografi kontak. Hasil penelitian menunjukan daun manggis pada fraksi metanol  dengan kosentrasi 30% memiliki aktivitas zona hambat ppaling besar terhadapat bakteri Staphylococcus aureus dan fraksi etil asetat dengan kosentrasi 30% memiliki aktivitas antibakteri paling besar terhadap  Escherichia coli. Hasil penelitian KLT-Bioautografi menunjukan golongan senyawa flavonoid setelah disemprotkan dengan AlCl3 dan uji Bioautografi daun manggis memiliki aktivitas zona hambat terhadap bakteri Staphylococcus aureus dan Escherichia coli. Kata kunci : Daun Manggis, Antibakteri, KLT- Bioautografi.

Objective: Arginase inhibition could be a potential therapeutic approach for endothelial dysfunction. Syzygium cumini (L.) Skeels leaves containphenolic acids and flavonoids, which have been predicted to exhibit arginase inhibitory activity. Moreover, these leaves contain tannins, which canform complexes with enzymes and lead to false-positive results during biological testing. Therefore, this study was conducted to evaluate the arginaseinhibitory activity of S. cumini leaf extract and fractions as well as to elucidate the effects of tannins on this activity.Methods: S. cumini leaves were fractionated using n-hexane, ethyl acetate, and methanol. A colorimetric method was employed to evaluate arginaseinhibitory activity. Tannin elimination was performed through the gelatin precipitation method. Total phenolic and flavonoid contents of the fractionswere calculated using the Folin–Ciocalteu and aluminum chloride methods, respectively.Results: Ethyl acetate and methanol fractions showed arginase inhibitory activity with half-maximal inhibitory concentrations (IC50) of 46.96 and15.35 μg/mL, respectively. The methanol fraction was positive for tannins. After tannin elimination, this fraction exhibited less potent arginaseinhibitory activity, with an IC50 value of 53.03 μg/mL. The ethyl acetate fraction showed higher total phenolic and flavonoid contents than the methanolfraction.Conclusion: Tannins affected the arginase inhibitory activity of the methanol fraction of S. cumini leaves; however, the ethyl acetate fraction did notcontain tannins and could inhibit arginase activity.

The species Inga laurina is native to the Brazilian Cerrado. There are no studies about the chemical composition and biological activities of extracts of this endangered species. The ethanolic extract and its successive fractions are rich in phenolic compounds and presented good antifungal activities. HPLC/MS-MS/MS and H1/C13 analysis led to the identification of seventeen compounds, most of which are gallic acid derivatives, myricetin and quercetin glycosides. The ethyl acetate fraction (EAF) contained high levels of total phenolics, expressed in milligrams of gallic acid equivalents per gram of extract (475.3 ± 1.9 mg GAE gextract-1) and flavonoids expressed in milligrams of quercetin equivalents per gram of extract (359.3 ± 10.6 mg QE gextract-1). This fraction was active against fungi of the Candida genus. The EAF showed MIC value 11.7 μg mL−1 against C. glabrata and a selectivity index of 1.6 against Vero cells. The flavonol glycoside myricetin-3-O-rhamnoside was isolated for the first time from the Inga laurina. These results make I. laurina a promising plant as a source of pharmaceutical and biological active antifungal compounds.

Fourteen compounds belonging to different chemical classes were characterized in the roots and fruits extracts from Berberis libanotica, using the same HPLC-DAD-MS method. Thirteen were reported, for the first time, from the fruits whereas the roots contained mostly alkaloids of which 3 out of 5 are reported for the first time. Their structures were established on the basis of MS data as gallic acid (1), chlorogenic acid (2), delphinidin (3), oxyacanthine (4), rutin (5), hyperoside (6), berbamine (7), isoquercitrin (8), quercitrin (9), jatrorrhizine (10), palmatine (11), berberine (12), quercetin (13) and luteolin (14). Extracts containing compounds 4 and 7 showed significant cytotoxicity against the HT29 cell line with an IC50 of 12.2–26.1 μg/mL. Fruits extracts, due mostly to compounds 1 and 2, showed potent antioxidant activities with an EC50 of 0.0025-0.019 mg/mL.

Oxidative stress and aldose reductase activity have been implicated in the development of diabetic complications. In this study, the antioxidant and aldose reductase (AR) inhibitory effects ofMaackia amurensis(MA) were investigated. The ethyl acetate fraction of the MA extract showed the highest inhibitory activity in antioxidant and rat lens AR (RLAR). To identify and isolate the active components in the ethyl acetate fraction of the MA extract, high-speed countercurrent chromatography and Sephadex LH-20 column chromatography were performed and guided by an offline HPLC-ABTS assay and HPLC microfractionation AR assay. Four antioxidants, namely, piceatannol (IC50= 6.73 μM), resveratrol (IC50= 11.05 μM),trans-ferulic acid (IC50= 13.51 μM), and chlorogenic acid (IC50= 27.23 μM), and six AR inhibitors, namely, chlorogenic acid (IC50= 4.2 μM), tectoridin (IC50= 50.4 μM), genistein (IC50= 57.1 μM), formononetin (IC50= 69.2 μM), resveratrol (IC50= 117.6 μM), and daidzein (IC50= 151.9 μM), were isolated and identified. The screening results of the offline HPLC-ABTS assay and HPLC microfractionation AR assay matched the activity of isolated compounds. Thus, MA is potentially valuable for antioxidant and AR inhibitor discovery and efficient drug design for the prevention and treatment of diabetic complications.
