Biotelemetry transmitter implantation in rodents: impact on growth and circadian rhythms

The implantation of a biotelemetry transmitter for core body temperature (Tc) and motor activity (MA) measurements is hypothesized to have effects on growth and circadian rhythmicity depending on animal body-to-transmitter (B:T) size ratio. This study examined the impact of transmitter implantation (TM) on body weight, food intake (FI), water intake (WI), and circadian Tc and MA rhythms in mice (23.8 ± 0.04 g) and rats (311.5 ± 5.1 g) receiving no treatment (NT), anesthesia, laparotomy (LAP), and TM. The B:T size ratio was 6:1 and 84:1 for mice and rats, respectively. In mice, body weight required 14 days to recover to presurgical levels and never attained the level of the other groups. FI recovered in 3 days, whereas WI never reached presurgical levels. Rat body weight did not decrease below presurgical levels. FI and WI recovered to presurgical levels in rats by day 2 postsurgery. Anesthesia decreased mouse body weight for 1 wk, but was without effect in rats. LAP significantly decreased body weight for 5 days in mice and 1 day in rats, showing a significant effect of the surgical procedure in the absence of TM in both species. Circadian Tc and MA rhythms were evident within the first week in both species, indicating dissociation between circadian rhythmicity and recovery of growth variables. Cosinor analysis showed a TM effect on Tc min, Tc max, mesor, amplitude, and period of mice, whereas only the amplitude of the rhythm was affected in rats. These data indicate that a large B:T size ratio is associated with minimization of the adverse effects of surgical implantation. We recommend that B:T size ratio, recovery of presurgical body weight, and display of a robust circadian Tc and MA rhythm be established before collection of biotelemetry data collection under an experimental paradigm.

Abstract Background Heatstroke is associated with exposure to high ambient temperature (AT) and relative humidity (RH), and an increased risk of organ damage or death. Previously proposed animal models of heatstroke disregard the impact of RH. Therefore, we aimed to establish and validate an animal model of heatstroke considering RH. To validate our model, we also examined the effect of hydration and investigated gene expression of cotransporter proteins in the intestinal membranes after heat exposure. Methods Mildly dehydrated adult male C57/BL6J mice were subjected to three AT conditions (37 °C, 41 °C, or 43 °C) at RH > 99% and monitored with WetBulb globe temperature (WBGT) for 1 h. The survival rate, body weight, core body temperature, blood parameters, and histologically confirmed tissue damage were evaluated to establish a mouse heatstroke model. Then, the mice received no treatment, water, or oral rehydration solution (ORS) before and after heat exposure; subsequent organ damage was compared using our model. Thereafter, we investigated cotransporter protein gene expressions in the intestinal membranes of mice that received no treatment, water, or ORS. Results The survival rates of mice exposed to ATs of 37 °C, 41 °C, and 43 °C were 100%, 83.3%, and 0%, respectively. From this result, we excluded AT43. Mice in the AT 41 °C group appeared to be more dehydrated than those in the AT 37 °C group. WBGT in the AT 41 °C group was > 44 °C; core body temperature in this group reached 41.3 ± 0.08 °C during heat exposure and decreased to 34.0 ± 0.18 °C, returning to baseline after 8 h which showed a biphasic thermal dysregulation response. The AT 41 °C group presented with greater hepatic, renal, and musculoskeletal damage than did the other groups. The impact of ORS on recovery was greater than that of water or no treatment. The administration of ORS with heat exposure increased cotransporter gene expression in the intestines and reduced heatstroke-related damage. Conclusions We developed a novel mouse heatstroke model that considered AT and RH. We found that ORS administration improved inadequate circulation and reduced tissue injury by increasing cotransporter gene expression in the intestines.

Purpose:Exercise increases core body temperature (TC) due to metabolic heat production. However, the exercise-induced release of inflammatory cytokines including interleukin-6 (IL-6) may also contribute to the rise in TC by increasing the hypothalamic temperature set point. This study investigated whether the exercise-induced increase in TC is partly caused by an altered hypothalamic temperature set point.Methods:Fifteen healthy, active men age 36 ± 14 y were recruited. Subjects performed submaximal treadmill exercise in 3 randomized test conditions: (1) 400 mg ibuprofen and 1000 mg acetaminophen (IBU/APAP), (2) 1000 mg acetaminophen (APAP), and (3) a control condition (CTRL). Acetaminophen and ibuprofen were used to block the effect of IL-6 at a central and peripheral level, respectively. TC, skin temperature, and heart rate were measured continuously during the submaximal exercise tests.Results:Baseline values of TC, skin temperature, and heart rate did not differ across conditions. Serum IL-6 concentrations increased in all 3 conditions. A significantly lower peak TC was observed in IBU/APAP (38.8°C ± 0.4°C) vs CTRL (39.2°C ± 0.5°C, P = .02) but not in APAP (38.9°C ± 0.4°C) vs CTRL. Similarly, a lower ΔTC was observed in IBU/APAP (1.7°C ± 0.3°C) vs CTRL (2.0°C ± 0.5°C, P < .02) but not in APAP (1.7°C ± 0.5°C) vs CTRL. No differences were observed in skin temperature and heart-rate responses across conditions.Conclusions:The combined administration of acetaminophen and ibuprofen resulted in an attenuated increase in TC during exercise compared with a CTRL. This observation suggests that a prostaglandin-E2-induced elevated hypothalamic temperature set point may contribute to the exercise-induced rise in TC.

Inhibitor of DNA binding 2 (ID2) is a helix-loop-helix transcriptional repressor rhythmically expressed in many adult tissues. Our previous studies have demonstrated thatId2null mice have sex-specific elevated glucose uptake in brown adipose tissue (BAT). Here we further explored the role ofId2in the regulation of core body temperature over the circadian cycle and the impact ofId2deficiency on genes involved in insulin signaling and adipogenesis in BAT. We discovered a reduced core body temperature inId2−/− mice. Moreover, inId2−/− BAT, 30 genes includingIrs1,PPARs, andPGC-1s were identified as differentially expressed in a sex-specific pattern. These data provide valuable insights into the impact ofId2deficiency on energy homeostasis of mice in a sex-specific manner.

The study aimed to investigate the impact of supplementary feeding on the young camels calve growth rate and daily gain. A total of 16 heads of young male camel calves from the Arabic camel breeds were selected and div ided into four equal groups (1, 2,3,4). Four heads of camel calves in each group, their body weight ranged from 176 - 220 kg. Experimental animal in group one was control supplemented nothings jus t depend on the natural range land, group two was feed on watermelon seeds, group three was allowanced concentrated diet composed of ground nut cake and the animals in group four intake mixture diet consist of watermelon seeds and ground nut cake 50% for e ach. The body weight of the experimental animal was determined biweekly using table balance. The random complete experimental design was used and the Analysis of variance and Tukey test were applied for data analysis and means separation. The results indic ated that the average Body weights of the camel calves were 174.75±3.3, 275.93± 9.0, 241.50± 8.5 and 236.50±8.2 Kg, in group one, two, three and four respectively. The higher body weight was recorded in group two (275.93±9 Kg) in which the animals feed on watermelon seeds and followed by group three and four (241.50± 8.5 and 236.50±8.2) respectively. On the other hand the less camel calves body weight was obtained in group one (control) that experimental animals not intake supplementary feeding. Highly diff erences (P ≤ 0.01) were obtained in the body weight between the groups. The daily gain of the first group, second, third and fourth were 214±12g, 624±28g, 542±19g and 528±12g, respectively, while no significant differences (P ≥ 0.01) between diets groundnut cake 242 kg and mixture diet 237 kg. The protein content was higher in ground nut cake 41% when compared to the protein content in watermelon seeds 36%, on the other hand the percentage of fat was highest in watermelon seeds 27.8% compared to ground nut ca ke 13.4%.

The aim of this study was to assess the impact of the leptin concentration and body weight of mink females on the birth parameters of their kits. Blood samples were collected 3 times during preparation for reproduction, 7±2 days before mating. Total leptin concentration in serum was measured using commercial RIA kits (Millipore, St. Charles, Missouri USA). Research was conducted on 20 clinically healthy scanbrown female minks (Neovison Vison) aged 2-3 years. To evaluate their body condition, the females were divided into two groups based on the BCS scoring system. In the first group, referred to as BCS 2, lean females were included, and the second group (BCS 4) consisted of obese females. In mink dams belonging to BCS 2, the average leptin values were one-fourth of those in the BCS 4 group. The results obtained indicate the influence of the body mass of minks during preparation for reproduction on serum leptin concentration. The results of the Mann-Whitney U test showed a statistically significant difference between weights and leptin concentrations in groups BCS 2 and BCS 4. Among the other traits, such differences were not found. The analysis of results obtained in the two mink groups did not reveal a clear relationship between leptin in the serum of mothers before mating and the birth parameters of their kits.

Proper functioning of the human circadian timing system is crucial to physical and mental health. Much of what we know about this system is based on experimental protocols that induce the desynchronization of behavioral and physiological rhythms within individual subjects, but the neural (or extraneural) substrates for such desynchronization are unknown. We have developed an animal model of human internal desynchrony in which rats are exposed to artificially short (22-h) light–dark cycles. Under these conditions, locomotor activity, sleep–wake, and slow-wave sleep (SWS) exhibit two rhythms within individual animals, one entrained to the 22-h light–dark cycle and the other free-running with a period >24 h (τ>24 h). Whereas core body temperature showed two rhythms as well, further analysis indicates this variable oscillates more according to the τ>24 h rhythm than to the 22-h rhythm, and that this oscillation is due to an activity-independent circadian regulation. Paradoxical sleep (PS), on the other hand, shows only one free-running rhythm. Our results show that, similarly to humans, (i) circadian rhythms can be internally dissociated in a controlled and predictable manner in the rat and (ii) the circadian rhythms of sleep–wake and SWS can be desynchronized from the rhythms of PS and core body temperature within individual animals. This model now allows for a deeper understanding of the human timekeeping mechanism, for testing potential therapies for circadian dysrhythmias, and for studying the biology of PS and SWS states in a neurologically intact model.

Abstract Like galanin, the 60-amino-acid peptide, galanin-like peptide (GALP), has orexigenic actions, demonstrated by an acute increase in feeding after central injection in rodents. However, in contrast to galanin, GALP causes a prolonged rise in core body temperature and a reduction in body weight over 24 h. In an attempt to identify potential explanations for the observed differences between GALP and galanin, this study examined which brain areas were activated by these peptides. Intracerebroventricular injection of GALP into conscious rats significantly stimulated feeding over 0–1 h, increased core body temperature, but reduced body weight gain over 24 h. Immunohistochemistry to detect c-fos demonstrated that intracerebroventricular injection of GALP or galanin activated several brain regions in common, including the dorsomedial nucleus of the hypothalamus, lateral hypothalamus, and nucleus tractus solitarius of the brainstem. However, GALP also induced c-fos expression in the periventricular hypothalamic region and supraoptic hypothalamic nucleus. Cell activation induced by GALP in the supraoptic hypothalamic nucleus and nucleus tractus solitarius was dependent on food intake but independent of food consumption in all other brain regions. Double immunohistochemistry indicated that small cells expressing c-fos in the periventricular hypothalamic region after GALP were astrocytes and not microglia.

Abstract The research was conducted on a group of 20 3-weeks-old chicken from the Cobb breed that were divided into two batches. Both batches received a 21/1 ratio feed diet for broiler chicken in their finishing period. The experimental group feed was supplemented with 0.5 ppm selenium, with 0.25 ALKOSEL R397 g/kg mixed fodder. 30 days into this feeding regime, the following determinations were made weekly: body weight, body temperature and computerized thermography of the comb and wattle. The obtained results were tabled and bio statistically analyzed revealing that individuals in the experimental batch showed statistically significant increases of the values for body weight (average body weight was 14.78% larger than that of the control batch) and of the temperature measured in the combs (average temperature was 12.97% higher) and wattles (average temperature was 4.12% higher). The experimental group also registered a higher core body temperature, but not statistically significant in comparison with the control group. Association of the higher values of core and peripheral body temperature with increased body weight in the individuals from the experimental group, correlated with an increased number of red blood cells and lower cholesterol levels suggest an improved basal metabolism and a better feed conversion.

Judokas commonly train the <em>seoi-otoshi</em> technique (aka, drop-knee <em>seoi-nage</em>). A controversy exists about the convenience of its use by the younger judokas due to the risk of high loads produced by the impacts on their growing structures. The aim of the present paper was to measure the impacts against the tatami when executing the knee <em>seoi-otoshi</em> technique and its relationship with the years of practice and the degree or level (color of the belt). Thirty-three judokas from different years and degree volunteered to participate. Two force plates covered by standard tatami, registered the ground reaction forces while five consecutive repetitions were executed. We measured the mean and maximum peaks of force relative to their own body weight (BW). The results show peaks of more than 10 BW, which can be considered a potential risk of injury in the younger judokas, especially when repeated in time. In addition, a tendency to decrease the impact as the years of practice increase is observed (potential function; R<sup>2</sup>= 0.41, p&lt;0.000 in the force peak), and the force in the expert group has been significantly lower than in the other groups (p&lt;0.001). On the other hand, the degree (belt color) shows a quadratic relationship (R<sup>2</sup>= 0.45, p&lt;0.000 in the force peak). The lack of agreement between the years of practice and the degree shows that the promotion criteria does not appear to be a valid one from a preventive viewpoint, especially in the lower degrees which would correspond to younger practitioners whose locomotor structures are still not fully developed.

Rumen temperature boluses are becoming increasingly used as a means of monitoring core body temperature for the detection of ill health. However, the effect of behavior on rumen temperature is largely unknown. This research investigates the impact of behaviour and diet on the rumen temperature of Holstein bulls, both at grass, and in a housed environment. Rumen temperature was recorded at five-minute intervals using a bolus. Direct observations were conducted on young bulls in two studies (i) at grass (n = 30) and (ii) while housed (n = 32). In addition, activity monitors were attached to bulls at grass (n = 24). Within each study, diet differed by the level of concentrate supplementation. There was no effect of diet on rumen temperature. Significant differences in rumen temperature were observed between behaviour groups for bulls at grass (p < 0.001) and housed (p < 0.001). Furthermore, drinking resulted in the lowest rumen temperature (grass 35.97 °C; housed 36.70 °C). Therefore, rumen temperature is affected by behavior; however, the temperatures recorded were not outside the normal temperature range for healthy cattle.
