<p>Background. The change in metabolism of the connective tissue elements of heart is the central chain in<br />pathogenesis of diffuse ischemic necrotic cardiosclerosis (DINC), which occurs after repeated epinephrine injury<br />of myocardial tissues.<br />Objective. This study proves that trimetazidine (TM) metabolic therapy has a protective effect on the<br />development of DINC in rats with different rates of hypoxia resistance.<br />Methods. Male white rats were divided into three groups due to the different rates of hypoxia resistance by<br />means of the method of hypobaric hypoxia: rats with low, middle and high rates of hypoxia resistance. Each<br />group was divided into equal subgroups: a control group, a DINC group (injections of epinephrine hydrotartrate<br />(0,5 mg/kg of body weight) and calcium gluconate (5 mg/kg of body weight) two times), a control group administrated<br />with trimetazidine dihydrochloride (10 mg/kg of body weight), a DINC group treated with TM every day<br />(10 mg/kg of body weight) for all period of observation. Concentration of protein-bound oxyproline in blood<br />serum was evaluated on the 7th, 14th and 30th days after the pathology simulation. Histological examination of<br />Masson trichrome staining of myocardium was performed on the 30th days after the pathology simulation.<br />Results. DINC increased the concentration of protein-bound oxyproline in blood serum on the 7th, 14th and<br />30th days after the pathology simulation, and followed by metabolic imbalances in diffuse connective tissue elements,<br />which are rich in collagens. DINC+TM increased the concentration of protein-bound oxyproline in blood<br />serum less intensively.<br />Conclusions. The intensity of metabolic imbalances in diffuse connective tissue elements is the highest in<br />the low resistant animals to hypoxia. Those results are confirmed by histological examination of the myocardium<br />of rats with different resistance to hypoxia. Fibrotic regions in myocardium are rich in collagens. It has been<br />revealed that the most pronounced therapeutic effect of TM is observed in animals with low resistance to hypoxia,<br />slight – in animals with medium resistance to hypoxia, and the lowest – in animals with high resistance to<br />hypoxia.<br />KEY WORDS: hypoxia, heart, diffuse cardiosclerosis, trimetazidine, oxyproline.</p>

Introduction: The prevalence and incidence of cardiovascular diseases have been attracting considerable attention in recent decades. This is partly due to the fact that myocardial fibrosis is the major consequence of the most nosological units of cardiovascular diseases. We believe that early pathogenic therapy of myocardial fibrosis should be taken into consideration as a solution to this issue. The change of the connective tissue metabolism in myocardium is the central chain in pathogenesis of diffuse ischemic necrotic cardiosclerosis (DINC) occurs after repeated epinephrine injury of myocardial tissues. The aim: The present study establishes that use of metabolic therapy by trimetazidine (TM) has a protective effect on myocardium repeatedly damaged by epinephrine in hight concentration during the development of DINC in rats with different resistance to hypoxia. Materials and methods: Using the method of hypobaric hypoxia, male albino rats were divided into three groups due to their different resistance to hypoxia. Each group was divided into four equal subgroups: control group, DINC group (2 times repeated injections of epinephrine hydrotartrate (0,5 mg/kg body weight) and calcium gluconate (5 mg/ kg body weight), control group introdused with trimetazidine dihydrochloride (10 mg/kg body weight), DINC treated with TM group (2 times repeated injections of epinephrine hydrotartrate (0,5 mg/kg body weight) and calcium gluconate (5 mg/kg body weight) group introduced with TM (10 mg/kg body weight) for all period of observation. The concentration of protein-bound oxyproline in homogenate of myocardium was determined at 7, 14 and 30 days after the modelling pathology and the histological examination of Masson trichrome staining of myocardium was performed. Results: Experimental modeling of DINC increased the concentration of protein-bound oxyproline in homogenate of myocardium at 7, 14 and 30 days after the modelling pathology, as well as accompanied by metabolic imbalances in diffuse connective tissue elements, which are rich in collagens. Experimental modeling of DINC+TM increased the concentration of protein-bound oxyproline in blood serum significantly less intensive. Conclusions: The intensity of metabolic imbalances in diffuse connective tissue elements of myocardium is the highest in the low resistant animals to hypoxia. Those results are confirmed by histological examination of the myocardium of rats with different resistance to hypoxia. Fibrotic regions in myocardium are rich in collagens. It has been revealed that the most pronounced therapeutic effect of TM is observed in animals with low resistance to hypoxia, slightly less – in animals with medium resistance to hypoxia, and the lowest – in animals with high resistance to hypoxia.

Phytochemical composition and antioxidant activity of flowers decoction at post-flowering stage (F3D) of Opuntia dejecta were determined. The obtained findings demonstrate that F3D has a marked antioxidant activity in all tested assays. Furthermore, the present study was designed to test the protective activity of F3D against induced Diabetes type 2 (DT2) in male rats. Those metabolic syndromes were induced by a high-fructose diet (HFD) (10% fructose solution) for a period of 20 weeks. F3D was administered orally (100 and 300 mg/kg body weight) daily for the last 4 weeks. Metformin (150 mg/kg body weight) was used as a standard drug and administrated orally for the last 4 weeks. The results showed a significant increase in blood glucose, triglycerides and hepatic markers (ALAT, ASAT and ALK-P) in HFD group. A significant increase in hepatic TBARS and a significant decrease in SOD, CAT and GPX were observed in fructose fed rats compared to control group. Administration of F3D showed a protective effect in biochemical and oxidative stress parameters measured in this study. Also, oral administration of F3D restored the histological architecture of rat liver in comparison with rats fed HFD. In conclusion, F3D attenuated hepatic oxidative stress in fructose-fed rats.

This study aimed to evaluate the efficacy of ivermectin, levamisole HCl and albendazole against gastrointestinal nematodes in naturally infected goats of government goat development farm, Sylhet, Bangladesh. The study included 52 black Bengal breed of which 30 were naturally infected and randomly selected 20 on the basis of their weight and egg count. Twenty black Bengal goats of 12-13 month old irrespective of sex infested with gastrointestinal nematodes were selected for this experiment and randomly divided into four equal groups (group A, B, C and D) where each group consisted of 5 goats and goats of group D were kept as control group. One injectable ivermectin (200?gkg-1 body weight, S/C) preparations (Techno Drugs Ltd. Bangladesh) and two solid levamisole HCl, albendazole (7.5 mgkg-1 body weight, orally) preparations (The ACME Laboratories Ltd. and Square Pharmaceuticals Ltd. Bangladesh) were used for positive control of gastrointestinal nematodes as group A, B and C. Goats of group D was kept as control without giving any treatment. Before trials (day 0), total egg count, blood samples and initial body weight were recorded. During the study period the fecal and blood samples were collected directly from rectum and examined on 7th, 14th, 21st and 28th day using McMaster fecal egg counting method. Body weight was recorded on day 28 following the treatments. The results of the comparative efficacies of different anthelmintic of ivermectin was 86.75%, followed by levamisole HCl 85.07% and albendazole 92.81%. McMaster fecal egg counting method discloses the percentage of Haemonchus spp. (15.38%), with Trichostrongylus spp, Strongyloides spp., and Cooperia spp. also present. After treatment with ivermectin, levamisole HCl and albendazole, Total Erythrocyte Count (TEC), Hemoglobin (Hb) content and Packed Cell Volume (PCV) were increased significantly (p<0.01 and p>0.05) in goats but Erythrocyte Sedimentation Rate (ESR) and Total Leukocyte Count (TLC) were decreased significantly (p<0.05 and p>0.01) in all treated goat and body weight was increased significantly (p<0.01) on day 28.International Journal of Natural Sciences (2015), 5(1) 26-34

Background: The extract of Corchorus olitorius has a reasonable content of vitamins A and C, calcium, iron and fibre, and therefore enjoys a universal application in the treatment of some disease conditions, even as the whole leaf is a very important component of food in so many cultures. Purpose: The study was designed to investigate the effect of Corchorus olitorius extracts on the testis of adult Wistar rats. Method: A total number of thirty two adult Wistar rats weighing between 180 and 200 grammes separated into four groups labeled A,B,C and D respectively were used for the study. Animals in group A which served as the control group were fed with the normal rat chow and water only. The group B rats were administered 100mg/kg body weight of aqueous extract of Corchorus olitorus; while those in group C were administered 500mg/kg body weight of the extract. The group D rats received 1000mg/kg body weight of the extract. For a period of four weeks, the different experimental animal groups received the respective aforementioned treatments once daily, around nine- o’clock in the morning through oral intubation. At the end of the 28 day treatment, the animals were sacrificed and the testes harvested for histological, investigation, and through cardiac puncture, blood samples for some hormonal studies was also collected and investigated using standard laboratory standards. Results: Observations made showed that the extract produced no histological distortions, degenerative or defective effects on the testicular tissues. The using of plant-based medicine is a popular approach to health care. A study presented by American Hearth Association showing that brown rice can lower cholesterol level in bood serum. To know the benefi ts of Angkak’s steeping water to decrease serum cholesterol levels in mice and determine the effects of Angkak’s steeping water to decrease serum cholesterol levels in rats compared with simvastatin. This study used an experimental research design, and using pre and post test control group design, performed in the laboratory of biomedical III FK UMS on November 1, 2012 to date of January 1, 2013. The purposive sampling technique, sampel determinaton using formula Federer by the number of sample of 30 male white rats wistar strain. After the data is collected, analyzed using the Shapiro-Wilk test, ANOVA, LSD, and the unpaired t test. The result of this study demnstrate hypothesis testing using ANOVA test with signifi cant level 95% of the negative control group, the treatment group a dose of 1, 2, and 3, it was found p<0,05 were signifi cantly different meaning. In the test dose of LSD between treatment groups 2 and 3 the value of p=0,839 (p>0,05) wich means no signifi cant difference. In the unpaired T test between the treatment group a dose of 1, 2, and 3 with positive control group were signifi cant difference. The unpaired T test between the treatment group a dose of 1, 2, and 3 with white rice also found signifi cant difference. Conclusion: Angkak’s steeping water can lower serum cholesterol levels of white rat as well as the water steepimg Angkak has the same effectiveness with simvastatin to decrease serum cholesterol levels of mice.Keyword: Cholesterol, Angkak

Objectives: A study on the hepatoprotective effect of Vernonia amygdalina leaves ethanolic extract on white rats induced by paracetamol was aimed to obtaining scientific data and evidence the hepatoprotective ability of the extract.Methods: The research was a complete randomized design, using 36 white rats which were divided into 6 groups. The normal control group was given 1% carboxy methyl cellulose for 7 days and followed by distilled water 8 hours after treatment. The positive control group was given 100 mg curcumin per kg body weight of rats for 7 days and followed by 2.5 g paracetamol per kg body weight after treatment. The negative control group was given 1% carboxy methyl cellulose for 7 days and followed by 2.5 g paracetamol per kg body weight after treatment. The test group was given 50, 100 and 200 mg extract per kg body weight of rats for 7 days and followed by 2.5 g paracetamol per kg body weight after treatment. Blood sampling was carried out 3 times (before treatment, after treatment and after induction).Results: With doses of 50, 100, and 200 mg extract per kg body weight of rats give significant difference on the decrease of Serum Glutamic Oxaloacetic Transaminase (SGOT) and Serum Glutamic Pyruvic Transaminase (SGPT) levels after paracetamol administration.Conclusions: It can be concluded that Vernonia amygdalina leaves ethanolic extract were potentially hepatoprotective.

The efficacy of tulsi (Ocimum sactum) and neem (Azadirachta indica) leaves extract as a growth promoter were studied in broiler. A total of 40 day-old broiler chicks were purchased from Kazi hatchery and after three days of acclimatization the chicks were randomly divided into four groups (n=10). No vaccination schedule was practiced and no antibiotic was added in ration of group A, B, C, and D respectively. Group A served control without any supplements while group B, C and D were supplemented with combination of tulsi and neem extract @ 1 ml, 2ml and 3 ml/liter of drinking water. Live body weight gain was recorded weekly up to 6th weeks and hematological studies were performed at 21st and 42nd day of experiments. At the end of 42nd day of experiment final body weight of group A, B, C and D were 1561± 12.10 g, 1698± 12.87 g, 1608± 12.04 g and 1763± 13.28 g, respectively. The birds of group D utilized their feed more efficiently among the treatment groups (p<0.05). The net body weight gain were, 1533± 11.98 g, 1443± 11.32 g and 1588± 12.10 g in treated groups compared to control group (1393± 11.07 g ) and total net profit per broiler was 19.08, 40.10, 20.68 and 45.07 Taka , respectively. Hematological parameterss (TEC, PCV, Hb and ESR) were not significantly changed among the treated and control group suggesting no side effects of herbal extracts in broiler. This experiment was conducted to determine the effect of papaya leaf (Carica papaya) and kalo jeera (Nigella sativa) seeds powdered supplementation in drinking water as a growth promoter in broiler chickens. A total of 20 Cobb-500 broiler chicks (day-old) were purchased from local hatchery (Nourish Poultry and Hatchery Ltd.) and after seven days of acclimatization chicks were randomly divided into two groups, A (n=10) and B (n=10). The group A was kept as a control and not treated. The group B was supplemented with papaya leaf and kalo jeera powder with feed and water. Weekly observations were recorded for live body weight gain up to 5th weeks and hematological tests were performed at 35th day’s age of broiler to search for hematological changes between control (A) and treatment (B) groups. The initial body weight of groups A and B on 1st were 41.00±0.56 gm and 41.50±0.35 gm, respectively and after 35th day of experiment final body weight were 1470±57.35 gm and 1720±58.56 gm, respectively and economics of production were analyzed and found that net profit per broiler was Tk. 8.91 and Tk. 20.69, respectively. The treatment group B was recorded statistically significant (at 1% level) increased (17.00%) for live body weight than that of control group A. The hematological parameters total erythrocyte count (TEC), erythrocyte sedimentation rate (ESR) and hemoglobin (Hb) estimation value of treatment group shows significant difference, while hemoglobin estimation does not show significant difference from control group. The results suggest that better growth performance could be achieved in broilers supplemented with papaya leaf and kalo jeera seeds.

Objectives: Drugs can cause undesired effects on the fetus during pregnancy, especially embryonic/organogenesis which could lead to defects in the fetus because some types of drugs can penetrate the placenta and will undergo biotransformation into a highly reactive compound that has the potential to become a teratogenic compound. The aim of this research was to examine the teratogenic effect of bintangur leaves (Calophyllum soulattri Burm. F) ethanol extract to Sprague Dawley strain white rats. Methods: The white rats are divided into four treatment groups: Control group was given carboxymethyl cellulose Na 1%, comparison group was given trimethoprim 360 mg/kg BW, C. soulattri leaves ethanol extract (CLE) 100 mg/kg BW, and CLE 500 mg/kg BW. The treatment was administrated since organogenesis period. Cesarian section was performed to pregnant rat at the 20th day to separate the fetuses. Observation covered body weight of pregnant rats, fetal biometric, morphological malformation, and skeletal formation. Results: CLE 100 mg/kg BW and 500 mg/kg BW did not cause any change in the number of a living fetus, body weight, and length of fetuses like the comparison group. Both doses of CLE shown have a normal skeletal formation. Resorption was found in the comparison group and CLE 100 mg/kg BW with the percentage was 65.21% and 6.67%. It was found that there is no significant difference (p<0.05) between both doses of CLE compared to control group. Conclusion: From the results, it is concluded that CLE did not have the teratogenic effect.

Chloroacetonitrile (CAN) is a by-product of chlorination of drinking water. Epidemiological studies indicate that it might present a hazard to human health. The present study was designed to investigate the potential adverse effects of intrauterine exposure to CAN on fetal body weight and development of the musculoskeletal system in mice. At gestation day 6, pregnant mice were given CAN (12.5, 25, or 50 mg/kg/day) till gestation day 18. Uteri were then examined and live fetuses were collected, weighed, and evaluated for any malformations. High doses of CAN (50 mg/kg) significantly elevated fetal anomalies and reduced fetal viability. Chloroacetonitrile at a dose of 25 mg/kg did not affect fetal viability and significantly reduced fetal body weight. Subsequent experimentation was performed using this dose level. Histological examination of fetal axial skeleton indicated that CAN resulted in delayed appearance of endochondral ossification centers, widening of the vertebrae, and destruction of the calcified zone. In addition, the skeletal muscle fibers were markedly distorted, were small in size, and were widely separated by connective tissue. Both connective tissue perimysium and endomysium were less cellular compared with control sections. The histological findings were further confirmed by assessing the morphometric changes. Ratios of calcified cartilage to non-calcified cartilage areas in both control and CAN-exposed groups were determined. Also, skeletal muscle fiber diameter of CAN-exposed fetuses was significantly decreased compared with control group. In conclusion, intrauterine exposure to low levels of CAN decreases fetal body weight and induces malformations in the musculoskeletal system in mice.
