Chitinase (EC.3.2.2.14) is an enzyme which can degradatechitin became N-acetilglucosamin. Chitinase has many benefits made the demand of it increases. High demands spur its availability in large quantities, cheap, fast production, resistant to any physical factor and chemical environment. Rapid and resistant enzyme production to environment factor can be obtained using chitinolitic bacteria of Geothermal Dieng. The utilization of chitin as bacterial growth substrate from waste of shell crab can be done considering high prices of commercial chitin on the market. The purpose of the research is to get the isolate of termoleranchitinolitic of watery mud in Geothermal Dieng and to know the character of the chosen isolate producing highest chinitase activity type of chitin source treatment and pH of media production. The research is done by growing the chitinolitic in the room temperature for 14 days. The experimental design used in this study is a complete randomized design of factorial pattern (two factors). The first factor is the type of chitin source that includes commercial chitin and chitin crab kits. The second factor is the pH of liquid chitin media for the production of enzymes, ie pH 6, 7 and 8.Chitinase activity is tested by measuring the result of sugar reduction. Obtained data is analyzed with Analysis of Variance (ANOVA). Result of isolation and selection is obtained one potential isolate, KSR 121. The isolate produce 1,4 cm of chitinolitic index after 96 hour incubation. Result of statistical test show both citin source type, pH of media production treatment and interaction were not significantly different (P˃0,05). KSR 121 isolate experience the highest growth of crab chitin treatment pH 8 (K2P3) with 6 hour incubation, whereas highest kinitase activity happen on crab chitin treatment pH 7 (K2P2) with 24 incubation, in amount of 0,125 (U/mL). Key words: N-acetil glucosamin, chtinase activity, chitinase, chitin, chitinolitic bacteria, isolation

The plum is a fruit prized for its nutraceutical properties because of its high content of fiber and sorbitol, which aid in digestion; furthermore, it is characterized as containing antioxidant pigments and an antiseptic action with anthocyanins. These fruits are classified as climacteric and continue the process of respiration and ripening after harvesting; because of this, it is necessary to harvest early so they can withstand transport, which often causes the fruits to not reach the consumption maturity required by consumers. The effect of ethylene on the ripening of plum fruits was evaluated, for which a completely randomized design with 10 treatments was used, which resulted from the combination of two factors plus a control without applications. The first factor was the ethylene dose (100; 1,000 and 2,000 mg L-1) and the second factor was the length of exposure to the ethylene (5, 10 and 15 minutes). The fruits were stored at room temperature for 9 days. In the fruits treated with ethylene, a significant increase in the respiration index and total soluble solids was observed. Additionally, greater total titratable acids and firmness values were obtained with the control treatment than with the ethylene; similarly, the ethylene application increased the fresh mass loss in the plum fruits. Overall, differences between the fruits treated with ethylene and the control were observed, but not between the doses, indicating that the lowest ethylene dose (100 mg L-1) can obtain the desired uniformity in plum fruits.

Lemon is a fruit that is rich in vitamin C. The processing of lemon as an infused water is one way to get optimal benefits from lemon. Lemon infused water is treated by soaking it in water for several hours. The temperature and soaking time are thought to influence the physical and chemical characteristics of lemon infused water. This study aims to determine the level of turbidity, vitamin C and antioxidant activity of lemon infused water. This study used a completely randomized design of two factors, with two differences in temperature (refrigerator temperature and room temperature) and five differences in soaking time (1, 2, 3, 4, 5 hours) and then analyzed turbidity levels, vitamin C levels and antioxidant activity. Observation data were analyzed using Two Way Anova. The results showed turbidity, vitamin C, and antioxidant activity at room temperature were higher than the refrigerator temperature and the longer soaking the turbidity level, vitamin C, and antioxidant activity were also as high as possible. The results of the analysis of variance showed that there were effects of temperature and soaking time on the level of turbidity and vitamin C of lemon infused water but there was no effect of soaking time on antioxidant activity. Soaking time at room temperature for 1 hour or at refrigerator temperature for 4 hours is the best time to get optimum vitamin C and antioxidant activity.

Nut grass is Cyperaceae group and underestimated its existence. Nut grass has many benefits so  potential to be developed. This study aims to determine the effect of planting media and the addition of NPK fertilizer to the growth of the nut grass and find out the nutrient content of chips made from nut grass tuber. The design of this study was factorial complete randomized design (RAL) which consisted of two factors. First factor is Planting Media, consist of M1 = soil, M2 = Soil: Sand (2: 1), M3 = Soil: Compost Fertilizer (2: 1) and M4 = Soil: Cage Fertilizer (2: 1) and second factor is fertilizer dosage NPK 15: 15: 15 with the level P1 = Without NPK fertilizer, P2 = 5 g NPK fertilizer, P3 = 10 g NPK fertilizer. Research result showed that the media significantly affected the number of clumps, root weight, clump weight, overall weight and number of flowers but did not significantly affect the height, number of tubers and tuber weight. Fertilizer significantly affected the number of clumps, root weight, clump weight, overall weight, number of flowers, number of tubers and tuber weight but did not significantly affect the height at P≤0.05 level. Interaction between planting media and NPK fertilizer did not significantly affect all parameters. Keywords: Chips, NPK Fertilizer, Nut Grass, Planting Media

Pure dense silicon carbide (SiC) ceramics were obtained via the high-temperature physical vapor transport (HTPVT) method using graphite paper as the growth substrate. The phase composition, the evolution of microstructure, the thermal diffusivity and thermal conductivity at RT to 200∘C were investigated. The obtained samples had a relative density of higher than 98.7% and a large grain size of 1[Formula: see text]mm, the samples also had a room-temperature thermal conductivity of [Formula: see text] and with the temperature increased to 200∘C, the thermal conductivity still maintained at [Formula: see text].

The areas of tropical forests in Indonesia always decrease every year, so that efforts are needed to cultivate plants efficiently for the supply of seedlings. Sungkai (Peronema canescens) has good quality wood that can be used for various purposes, either for construction, furniture, plywood or the leaves can be used for medicine. The purpose of this study was to analyze the growth of stem cuttings of sungkai based on the number of latent buds by calculating the root dry weight, bud dry weight, and total dry weight. This study used a factorial complete randomized design. There were two factors studied, namely the number of buds consisting of 2 levels: 2 buds and 1 bud, and the stem split factor. Each treatment was replicated 3 times and each experiment unit used 5 cuttings. The growth of sungkai cuttings in the treatment of two buds had a total dry weight of 170.04 grams, while the treatment of one bud had a total dry weight of 98.30 grams. The results of this study indicated that the more the number of latent buds, the more root and bud growth of cuttings.Keywords: Coppice; Latent buds; Cuttings; Sungkai

Efforts to improve the production performance of clown loach in recirculate aquaculture system can be done by increasing stocking density and water discharge. This study aimed to analyze the effect of increasing stocking density and water discharge on production performance, stress response and water quality in order to obtain the best stocking density and water discharge to obtain maximum profit in a short time. A factorial completely randomized design with two factors, i.e. the stocking density of 1, 2, and 3 fish L-1 and the water discharge of 0,05; 0,10; 0,15 L s-1 was performed. The volume of water used in each aquarium was 48 L and using a ½ inch faucet stop to regulate the water discharge from the inlet pipe. Clown fish fed Tubifex sp. with a feeding frequency of two times a day according to the treatment for 60 days. The result indicates that there was no interaction between the two factors on production performance, payback period, and R/C ratio. Stocking density has a significant effect on specific growth rate, absolute growth rate of individual weight, feeding consumption rate and R/C ratio of clown loach. Stocking density and water discharge had an interaction on the visual color of pectoral and caudal fins of clown loach. Different stocking densities with the combination of water discharge in this study resulted that the water quality were within tolerable range for clown loach so that they did not experience stress, as well as high production and business performance. It is recommended to intensify clown loach with a stocking density of 3 fish L-1 and 0,15 L s-1‑ of water discharge.

(Chemical and Organoleptic Characteristics of Mung Bean Flakes with The Addition of Fillers and Water) ABSTRACT. The objective of this research was to determine the chemical and organoleptic characteristics of mung bean flakes with the addition of fillers and water. The study was conducted using completely randomized design with two factors namely the addition of fillers and water. The results showed that the addition of fillers and water has significant (P≤0,01) effect on the water absorption ratio and protein levels of mung bean flakes. The best treatment was obtained by the mung bean flour without the addition of fillers with 150% of addition water with water absorption ratio of 204%, water content of 5,7%, protein of 22,01%, starch levels of 61,76%, organoleptic values of aroma of 2,61, color of 2,42, crispness of 2,72 and flavor of 2,83.

Fusarium oxysporum is one of the most limiting fungal pathogens of lulo crop. To determine its pathogenicity, this work morphologically and pathogenically characterized F. oxysporum isolates from different lulo-growing municipalities of the Department of Nariño. Twenty isolates were evaluated through a completely randomized design with two factors and three replicates per treatment, including a control. The first factor corresponded to 20 isolates of F. oxysporum and the second to 10 lulo genotypes. The morphological characterization involved determining growth rate (GR), color (CO), mycelial type (MT), medium coloration (Mc), shape (Sh), size (S), number (N) of macroconidial (Ma) and microconidial (Mi) septa, presence of chlamydospores (PC), and chlamydospore shape (CS). Moreover, the pathogenic characterization was based on the incubation period (IP), absolute growth rate (AGR), disease severity (DS), disease incidence (I), and vascular discoloration (VD). The morphological characterization demonstrated that all isolates corresponded to F. oxysporum. For IP, genotype G1 showed the lowest average at 18 days. For AGR, genotype G2 had the lowest height increase at 0.05 cm.day-1. For DS, genotype G1 reached the highest severity level (level 9) and a disease incidence of 100%. This study provides the first report of the special form of F. oxysporum f. sp. quitoense in Nariño. Solanum hirtum, Solanum sessiliflorum, and Solanum estramonifolium were resistant to the isolates evaluated, demonstrating that wild species should be considered as sources of resistance for breeding programs aiming to obtain resistant commercial genotypes.

Aphis gossypii Glover hama penting tanaman cabai, dapat dikendalian dengan menggunakan pestisida nabati. Penelitian bertujuan untuk mengevaluasi waktu aplikasi dan konsentrasi ekstrak kasar biji bintaro (Cerbera odollam G.) dalam mengendalikan A. gossypii Glover serta pengaruhnya terhadap pertumbuhan tanaman cabai. Penelitian menggunakan Rancangan Acak Lengkap, dua faktor dan tiga ulangan. Faktor pertama adalah waktu aplikasi ekstrak (waktu sebelum dan waktu setelah infestasi A. gossypii Glover). Faktor kedua adalah konsentrasi ekstrak bintaro (0%, 1%, 2%, 3%, 4% dan 5%). Hasil penelitian menunjukkan konsentrasi 1% ekstrak biji bintaro menyebabkan mortalitas kutu daunnya 68% sedangkan pada konsentrasi 3% mortalitasnya mencapai 90%. Ekstrak biji bintaro yang diberikan sebelum hama diinfestasikan menyebabkan mortalitas kutu daun 59,5% sedangkan jika diberikan setelah hama diinfestasikan mortalitasnya meningkat menjadi 77,6%. Nilai LC50 dan LC90 ekstrak biji bintaro diaplikasikan sebelum dan setelah hama diinfestasikan secara berurutan adalah 1,8%; 4,4%; 0,57% dan 2,8%. A. gossypii Glover yang diinfestasikan pada tanaman berumur 4 minggu dan disemprot dengan ekstrak biji bintaro pada konsentrasi 1% dapat menurunkan intensitas kerusakan pada hari ke tujuh setelah infestasi dan tidak berpengaruh terhadap jumlah daun, bobot segar maupun bobot kering tanaman cabai. Ekstrak biji bintaro dapat dikembangkan sebagai pestisida nabati untuk mengendalikan hama A. gossypii Glover pada tanaman cabai. Aphis gossypii Glover is an important sucking insect pest of the pepper (Capsicum annum L.), Can be controlled by using natural pesticides. The study aimed to evaluate the application time and concentration of Cerbera odollam G. seed extract in controlling A. gossypii and their effects on pepper, C. annum. Complete randomized design was used in this experiment, with two factors, and was repeated three times. The first factor was the time of application of the extract, consisting of before and after A. gossypii infestation. The second factor was the concentration of C. odollam extract (0%, 1%, 2%, 3%, 4% and 5%). The results indicated the C. odollam seed extract affected the mortality of A. gossypii. At a concentration of 1% of C. odollam seed extract, the mortality of A. gossypii was 68% while at a concentration of 3% the mortality reached up to 90%. C. odollam seed extract applied before infestation, the mortality of A. gossypii was 59.5% whereas, after the infestation, the mortality increased up to 77.6%. LC50 and LC90 extract of C. odollam seeds applied before and after infestation were 1.8%; 4.4%; 0.57% and 2.8% respectively. A. gossypii infested at 8 weeks old pepper and sprayed with C. odollam seed extract at a concentration of 1% lowered the crop damage on the seventh day after infestation. On the other hand, the application of C. odollam seed extract did not affect the number of leaves, fresh weight, and dry weight of pepper. This study indicate that C. odollam seed extract can be developed as a natural pesticide to control A. gossypii on pepper..

This study aims to determine the dose of Trichoderma koningii which can induce soursop germination; know the best soaking time that can affect the induction of soursop germination; to know the interaction between soaking time and Trichoderma koningii dose on soursop germination. This research was conducted in April - June 2019 at the Green House of Plant Sciences, Faculty of Agriculture, Sam Ratulangi University, Manado. The material used in this study was soursop seed obtained from Lota Village, Pineleng Tomohon Subdistrict, Trichoderma koningii with 106 solids obtained from the North Sulawesi Province Plant Protection Center Collection, Aquadest, 70% Alcohol, 80% Acetone, sandalready sifter and sterilized. This research was arranged in factorial with a Completely Randomized Design (CRD) consisting of two factors, namely 5 treatments of Trichoderma koningii dose and 4 immersion treatments repeated three times (3x) so that a total of 60 treatment units. Each treatment unituses 10 seeds. The results of this study indicated that the dose of Trichoderma koninggi 200 g without soaking can increase the germination power by 86.67% and the vigor index by 32.96%; Immersion with Koninggi Trichoderma suspension has not been able to induce soursop seed germination rate; Theinteraction between Trichoderma koningii dose and soaking time did not affect the speed of soursop seed germination.*eprm*
