AbstractGlobally,An experimental laboratory-based study was done following standard methods and procedures. Nine experimental groups (I, II, III, IV, V, VI, VII, VIII, IX) each comprising of six animals were used. Group I received normal saline. Groups II to IX received 200 mg/kg bwt cyclophosphamide at the beginning of the study. Group III received 50 mg/kg bwt of an immunostimulatory drug levamisole. Groups IV to IX were dosed daily for 14 days with extract at doses of 250, 500, and 1000 mg/kg bwt, respectively, using an intragastric tube. Complete blood count (CBC), delayed-type hypersensitivity reaction (DTH), neutrophil adhesion test, and hemagglutination antibody titer were determined using standard methods and procedures. Statistical analysis was performed using GraphPad prism 5.0a Software.There was an increment in WBC, lymphocyte, and neutrophil counts at a dose of 1000 mg/kg bwt similar to the levamisole-positive control group. The neutrophil adhesion was statistically significant (p≤0.05) for treatment groups that received 1000 mg/kg bwt (29.94%) and 500 mg/kg bwt at 17.28%. The mean percentage increment in footpad thickness was highest (26.9%) after 8 h of injection of antigen in the footpad of rats dosed 500 mg/kg bwt and this later reduced to 25.6% after 24 h. There was a dose-dependent increment in the mean hemagglutination antibody titer to sheep red blood cells (SRBC) from 10.73±0.57 HA units/μL for the 250 mg/kg bwt to 26.22±1.70 HA units/μL for the 1000 mg/kg bwt.Methanolic leaf extract of

Diclofenac is used to treat rheumatism disorders, which are associated with the damages of renal, gastric and hepatic organs. Diclofenac is a pharmaceutical drug that is known to induce toxicity on its overdosage and long-term usage. Madhuca longifolia is known to have antioxidant, anti-inflammatory and anti-ulcer activity. It is an evergreen tree that is reported to have many ethnomedicinal uses. The other properties of Madhuca longifolia include anti-diabetic, analgesic and anti-microbial activities. Our study aims to evaluate the pre-treatment activity against the diclofenac-induced toxicity by the Madhuca longifolia aqueous leaf extract in Wistar albino rats for 10 and 15 days. Rats were divided as Group-I: Normal control, Group-II: Diclofenac on the last two days, Group-III and group-IIIa: Diclofenac + Aqueous Leaf Extract of Madhuca longifolia, Group-IV and group-IVb: Diclofenac + Silymarin, Group -V and group-Va: Aqueous Madhuca longifolia leaf extract. After the sacrifice, the rats were studied for antioxidant assay, renal enzyme markers, liver enzyme markers, and histopathological analysis of the kidney, stomach, intestine, and liver. As a result, we could identify that Madhuca longifolia has reduced the toxic changes in rats caused by diclofenac.

Background: Mashaparni is a plant belonging to the family Fabaceae and used in ayurvedic system of medicine and is being used in diet by various people of India and it also forms a part of traditional Indian cuisine. Teramnus labialis Spreng as accepted botanical source of Mashaparni. Mashaparni is widely used as Balya drug as mentioned by Charaka acharya in Balyamahakashaya gana. Hence this study is aimed to evaluate the Immunomodulatory activity of Mashaparni. Methodology: Experimental studies to determine the Immunomodulatory activity of Mashaparni Teramnus labialis Spreng by antigen induced rat paw oedema in Wister albino rats and the drug in the Aqueous and Ethyl extract form respectively. The studies were carried out on 4 groups i.e. Control group, Standard group, Aqueous extract and Ethyl extract form. Standard drug taken for the study is Guduchi Tinospora cordifolia in Ethyl extract form. Immunomodulatory activity determined by Delayed Type Hypersensitivity method with the help of Digital Plethysmograph method for duration of 15 days. The rat paw volume was the assessment parameter and the results were recorded at 2nd, 4th, 6th, and 24th hours after induction of antigen. Results: The Aqueous extract of Mashaparni shows a significant Immunomodulatory action compared to Ethyl extract of Mashaparni.

Abstract Probiotic attributes of lactic acid bacteria isolated from goat and sheep milk samples were analysed by culturing them on MRS (de Man, Rogosa and Sharpe) agar media. The most potential isolates, GMB24 and SMB16, were identified by biochemical tests, which were further identified as Enterococcus faecium GMB24 and Enterococcus hirae SMB16 by 16S rRNA gene sequencing approach. The probiotic potential of the GMB24 and SMB16 were possessed for probiotics attributes including antimicrobial activity against five pathogenic bacteria viz., Escherichia coli (MTCC118), Staphylococcus aureus (MTCC7443), Pseudomonas aeruginosa (MTCC424), Listeria monocytogens (MTCC657) and Salmonella typhimurium (MTCC733), and antibiotic susceptibility test. These bacterial isolates had ability to tolerate different concentrations of acid, bile and phenol resistance. Further, immunomodulating activities of potential probiotic bacteria were tested on neutrophil adhesion test, haemagglutinating antibody titer and delayed type hypersensitivity. The data from these experiments were used for the principal component analysis (PCA) for assessing survivability of isolates at different factors. The probiotic bacterial isolates showed good response for the enhanced antibody production and delayed type hypersensitivity (DTH) activity. Probiotic isolates E. faecium GMB24 and E. hirae SMB16, at 109 cells/ml doses per day, increased the neutrophil adhesion, haemagglutinating antibody titer and DTH in comparison to the untreated control group. These differences were statistically significant (P < 0.05). The isolates showed negative test for hemolytic and gelatinase activities and hence were considered safe. E. faecium GMB24 and E. hirae SMB16 possessed an excellent probiotic potential and had a strong potential immune-stimulant activity.

Eriobotrya japonica is traditionally used as an antipyretic, digestive, and diuretic agent. Its flowers possess free radical–scavenging, antioxidative, and hepatoprotective effects. We investigated the hepatoprotective potential of E. japonica leaf extract and its various fractions against hepatotoxicity in rats. Liver injury was stimulated by the oral administration of carbon tetrachloride (CCl4; 2.5 mL/kg b.wt.). Male albino rats (n = 55) were distributed arbitrarily into 11 groups: Group I, normal control group; Group II, CCl4 (positive control group); Group III, CCl4 + silymarin; Groups IV and V, CCl4 + two doses of 250 and 500 mg/kg of the 80% methanolic extract of E. japonica leaves, respectively; Groups VI and VII, CCl4 + 250 mg/kg and 500 mg/kg of the ethyl acetate fraction, respectively; Groups VIII and IX, CCl4 + 250 and 500 mg/kg of the butanol fraction, respectively; and Groups X and XI, CCl4 + 250 and 500 mg/kg of the aqueous fraction of E. Japonica leaves, respectively. CCl4-treated rats that were given 250 or 500 mg/kg of the methanol extract of E. Japonica leaves, or its ethyl acetate, butanol, or aqueous fractions, had significantly lower levels of biochemical parameters such as alanine aminotransferase, aspartate transaminase, alkaline phosphate, total protein, gamma-glutamyl transferase, and bilirubin levels than those of the CCl4 positive group. However, the extract and fractions did not significantly affect lipid profiles. Thus, we conclude that Eriobotrya leaf extract and its fractions have a hepatoprotective effect against CCl4-induced hepatotoxicity in rats.

<p class="abstract"><strong>Background:</strong> Acrylamide (ACR) is a widely used chemical in industry and it accounts for major health problems as it has been detected in highly consumed food items, carbohydrate-rich food items cooked at high temperature. Accordingly the population is highly exposed to ACR. The aim of the study was to assess the effect of vitamin E on biochemical and ultrastructure changes in acrylamide-induced renal toxicity in wistar albino rats.</p><p class="abstract"><strong>Methods:</strong> Three groups of adult albino wistar rats weighing about (200-250 gm) were used in this study to investigate the effect of vitamin E on acrylamide induced renal toxicity; 10 rats in each group; Group I: Control group, Group II: Acrylamide treated and Group III: Acrylamide-Vit E treated group. Blood samples were collected for estimation of serum creatinine, blood urea nitrogen (BUN), lactic dehydrogenase (LDH) and albumin. Kidneys specimens were processed for light and electron microscopic studies. Kidney sections were stained with H&amp;E, MT, PAS and immuno-histochemical stains for detection of NF-kβp65 and Bcl-2. Morphometric study was done followed by biochemical and morphometric statistical analysiss.</p><p class="abstract"><strong>Results:</strong> Acrylamide treated rats showed degeneration of cells lining PCT and DCT, atrophy of glomeruli and fibrosis. Ultrastructurally; tubules lining cells showed loss of microvilli, basal membrane in foldings and mitochondrial changes. Podocytic changes include: Vacuolation, irregularity and disorientation of their processes and thinning of glomerular basement membrane. Significant increase in the mean number of NF-kβp65 positive cells and decrease in the mean area% of Bcl-2 immuno-expression, increase in the mean area % of collagen fibers and urinary space diameter and decrease in thickness of epithelial cells of PCT and DCT were also observed. Serum creatinine, BUN anLDH were increased whereas serum albumin was decreased. Vitamin E co-administration with ACR improve all these histological, ultrastructural and biochemical changes.</p><strong>Conclusions:</strong> Acrylamide induced renal toxicity could be ameliorated by vitamin E co-administration.

Abstract   Background: Purple sweet potato leaves (Ipomoea batatas L.) which has been known as immunomodulatory activity, which contains chemical compounds, such as vitamins, minerals, flavonoids (anthocyanin), especially iron found in a higher proportion in the leaves. Therefore, it is necessary to study the immunomodulatory activity of the leaf extract of purple sweet potato (Ipomoea batatas L.) on the immune response in Balb/c mice who induced by hepatitis B vaccines. Method: Inducing performed at day 0 and day 15. Mice with weights ± 20 g, which has been divided into normal control group, I, II, III, IV and V (each 5 mice), respectively were given orally by the leaf extract of purple sweet potato (Ipomoea batatas L.) (doses 100 mg / kg, 200 mg / kg, 300 mg / kg, 400 mg / kg and 500 mg / kg), normal controls (CMC Na 0,5%). With the volume of giving 1% of body weight in mice, performed during the study (20th days). Blood sampling is done via the tail vein (laeralis vein), to calculate the number of leukocytes (21st days) and lymph decision to count lymphocytes (21st days). Results: The results that the leaf extract of purple sweet potato (ipomoea batatas L.), especially a dose of 500 mg / kg as immunomodulatory activity can increase the number of lymphocytes proliferation cell. Conclusion: Purple sweet potato leaf extract has activity against the increase in the number of lymphocytes cell especially on dose 500 mg/kg BW who induced by hepatitis B vaccine that can stimulated an immune response.

Pyrethroid cyhalothrin (PC) is an insecticide that is used worldwide for pest control in agriculture and household use. Samoa extract (SE) is a potent antioxidant protecting cells from oxidative stress. The present study investigates the protective and therapeutic effect of SE on PC-induced changes in sperm quality in male rats. Fifty adult male albino rats were divided into five groups: group I: served as control; group II: received PC i.p. only (6.2 mg/kg b.wt.); group III: received SE only (100 mg/kg b.wt., p.o.) for eight weeks; group IV: received SE as a protective agent daily for eight weeks, then followed by the administration of PC (i.p.) three times a week for two weeks; group V: exposed to PC (i.p.) three times a week for two weeks, then treated with the SE daily for 8 weeks. Results showed that PC caused markedly impaired sperm quality (a count, viability, motility, and abnormality). Compared to PC-treated animals, SE in the protective group markedly restored the alteration of sperm indices. However, SE in the curative group was found to be less effective in restoring PC-induced alterations. In conclusion, the data of this study revealed that the SE as a protective agent is more effective than as a therapeutic agent. Keywords: Samoa; Pyrethroid; Sperm quality; Rat

Abstract Background Repairing urinary bladder (UB) defect by enterocystoplasty remains the gold standard surgical bladder reconstruction procedure to increase the capacity and compliance of dysfunctional bladders. However, many complications were recorded. Aim of the work This work aimed to compare the consequences of reconstruction of urinary bladder defect using untreated small intestinal submucosal (SIS) matrix versus seeded and unseeded decellularized SIS matrix. Material and Methods Fifty female albino rats were used in this study. The animals were divided into three groups: Group I (Control) included ten adult rats from which ileal tissue was obtained. Group II included ten adult rats in which their UB defect was repaired by untreated cellular SIS. Group III included twenty adult rats that were subdivided into two subgroups, 10 rats each; Subgroup IIIA where rats had their UB defect repaired by acellular SIS and subgroup IIIb where rats had their UB defect repaired by acellular SIS seeded with adipose mesenchymal stem cells (AMSCs).Ten young rats were used for preparation of AMSCs. Morphometric and statistical analysis were also performed. Results In rats where UB defect was repaired by untreated cellular SIS, the graft area showed loss of epithelial polarity, presence of intraepithelial cysts and occasional extension of urothelium to the outer surface forming fistula. There were areas of metaplasia with the appearance PAS positive cells. In the lamina propria, there was areas of lymphocytic infiltration together with significant increase in the collagen fiber deposition (p &lt; 0.05). There was a significant decrease thickness of muscle layer as compared to control (p &lt; 0.05). In rats where UB defect was repaired by acellular SIS, urothelium in the graft area showed occasional squamous metaplasia and often the urothelium extended to the deeper layers forming Brunn's nest. There was minimal muscle regeneration in the graft area. However, in rats where UB defect was repaired by acellular SIS seeded with AMSCs, the urothelium in the graft area was nearly similar to control group with uniform urothelium thickness, minimal collagen fibers deposition and thick muscle layer that showed no significant difference from the control (p &gt; 0.05). Conclusion Acellular SIS seeded with AMSCs showed better results compared to non-seeded and cellular SIS in reconstructing urinary bladder defects.

Given the medicinal importance of the avocado plant in alternative medicine,the present study aimed to study the effect of the phytochemical content of P. americana leaf extract on the hematological system of albino rats. Albino rats were obtained andclustered into sixgroups,with five rats per groupsimultaneously were assigned experimental and control. The rats in the experimental group were administered with varying doses of the prepared aqueous extract of P. americanawithin the period of the study. The rats in the control group were administered with other substances such as water and feed for the study period.The hematological system of the experimental animal was assessed after the administration of the extract, and it was observed that the aqueous extract of P. americana leaf recorded no significant adverse effect on the hematology system of the experimental study animal.

Abstract: Papaya leaves contain several antioxidants (flavonoid, tanin, and vitamin C) which have antiatherogenic effects that may inhibit the progression of an atherosclerotic lesion. One of the risk factors causing atherosclerosis is the consumption of food containing highly saturated fatty acids e.g. lard. Low density lipoproteins (LDL) accumulate within the intima and then are oxidized (LDL-ox). This LDL-ox is ingested by macrophages, resulting in foam-cell formation (early lesion of atherosclerosis). This study aimed to find out the histological features of the aorta of wistar rats having lard diets without the addition of the papaya leaf extract; having lard diets along with the papaya leaf extract; and having lard diets followed by papaya leaf extract. This was an experimental study on 16 wistar rats divided into 4 groups: group I without treatment (negative control group), group II lard diet for 14 days (positive control group), group III lard diet with papaya leaf extract for 14 days (treatment group I), and group IV lard diet for 14 days, and then followed by papaya leaf extract for 14 days (treatment group II). It was found that the aorta of group I showed adipose cells in the intima and media layers; group II and III showed foam cells in both layers; and group IV showed foam cells in fewer numbers than group II. Conclusion: The aorta histological features of wistar rats given lard diets for 14 days, with or without papaya leaf extract, showed foam cells in the intima and media layers. Papaya leaf extraxt added to lard diets had no effect on decreasing foam cells (no protective effect), meanwhile papaya leaf extract following lard diets showed a reduction of foam cells (therapeutic effect). Keywords: papaya leaf, lard dietary, foam cells, wistar rat.   Abstrak: Daun pepaya mengandung antioksidan (flavonoid, vitamin C) yang berefek anti-aterogenik, sehingga diharapkan dapat menghambat perkembangan lesi aterosklerosis. Salah satu faktor risiko penyebab aterosklerosis yaitu makanan yang berkandungan tinggi asam lemak jenuh, antara lain lemak babi. Konsumsi lemak jenuh berlebihan dapat mengganggu fungsi sel endotel, sehingga lipoprotein berdensitas rendah (LDL) dapat masuk dan menjadi LDL teroksidasi (LDL-oks). Makrofag menangkap LDL-oks dan menjadi sel busa (lesi dini aterosklerosis). Penelitian ini bertujuan untuk mengetahui gambaran histologik aorta tikus wistar dengan diet lemak babi tanpa pemberian ekstrak daun pepaya, diet lemak babi bersamaan pemberian ekstrak daun pepaya, dan setelah diet lemak babi dilanjutkan pemberian ekstrak daun pepaya. Penelitian ini bersifat eksperimental. Subyek penelitian terdiri dari 16 ekor tikus wistar yang dibagi menjadi empat kelompok: kelompok I tanpa perlakuan (kelompok kontrol negatif); kelompok II dengan diet lemak babi selama 14 hari (kelompok kontrol positif); kelompok III dengan diet lemak babi serta pemberian ekstrak daun pepaya selama 14 hari (kelompok perlakuan I); dan kelompok IV dengan diet lemak babi selama 14 hari, dilanjutkan pemberian ekstrak daun pepaya selama 14 hari (kelompok perlakuan II). Hasil penelitian memperlihatkan gambaran histologi aorta kelompok I tampak perlemakan; pada kelompok II terdapat sel-sel busa; pada kelompok III masih terdapat sel-sel busa; dan pada kelompok IV terdapat sel-sel busa, namun dalam jumlah yang lebih sedikit dibandingkan pada gambaran kelompok II. Simpulan: Tikus wistar dengan diet lemak babi selama 14 hari, baik dengan maupun tanpa ekstrak daun pepaya, memperlihatkan gambaran histologik adanya sel-sel busa pada tunika intima dan tunika media aorta. Pemberian ekstrak daun pepaya bersamaan dengan diet lemak babi tidak berefek menurunkan jumlah sel busa (tidak ada efek protektif) sedangkan pemberian ekstrak daun pepaya setelah diet lemak babi berefek mengurangi jumlah sel-sel busa yang terbentuk (efek terapi). Kata kunci: daun pepaya, diet lemak babi, sel-sel busa, tikus wistar.
