Binding and Peroxidative Action of Oxyfluorfen in Sensitive and Tolerant Algal Species

Peroxidative activity of oxyfluorfen and binding of this nitrodiphenyl ether to cell fractions was investigated with the susceptible alga Scenedesrnus aeutus and the resistant alga Bumilleriopsis filiformis. Although a 10-fold higher concentration of oxyfluorfen was applied to Bumilleriopsis, the lag phase for initiation of peroxidative evolution of short-chain hydrocarbons from fatty acids was much longer than found with Seenedesmus. Oxyfluorfen was predominantly recovered after homogenization from the pellet which was separated into a lipid and a chloroform/methanol insoluble fraction. Parts of the oxyfluorfen which is present in the insoluble pellet fraction during the lag phase before the onset of peroxidation can be found in the lipid fraction when measurable peroxidative activities have started. This was observed with Seenedesmus as well as with Bumilleriopsis. During peroxidation initiated by oxyfluorfen acyl lipids are degradated as monitored by the disappearance of the plastidic sulfolipid. Analysis of bound fatty acids showed that they are targets for peroxidative reactions in acyl lipids. Destruction of polyunsaturated fatty acids was higher than for saturated ones.

Abstract The marine diatom Ditylum brightwellii (West) Grunow isolated from the Baltic Sea could be synchronized by a light/dark rhythm of 6.5:17.5 h (white light intensity 8 W m-2) at 18 °C and 0.035 vol.% CO2. Content of protein, DNA and RNA increased linearly up to the end of the cell cycle. Pigments (chlorophyll a, chlorophyll c1 + c2, carotenoids) and galactolipids were synthesized in the light period only. A lag phase of 2 h was observed in the biosynthesis of sulphoquinovosyl diacylglycerol and phosphatidylglycerol. Formation of phosphatidylglycerol and phosphatidylcholin continued in the dark period (30% and 28%, respectively). The pattern of major fatty acids (C14:0, C16:1, C16:0, C18:1 and C20:5) varied during the cell cycle of Ditylum.Biosynthesis of acyl lipids was reduced in dependence on the UV-B dose. The most sensitive lipid was digalactosyl diacylglycerol (total inhibition at 585 J m-2), whereas phosphatidylcholin was less affected (20% reduction). UV-B radiation during the dark period had no effect on the lipid and pigment content. Strongest inhibitory effect of UV-B on cell division, synthesis of protein, pigments, sulphoquinovosyl diacylglycerol and phosphatidylglycerol was found after UV-B radiation at the beginning of the cell cycle (0.-2. h). An exposure time at the end of the light period (4.-6. h) led to a marked damage on the synthesis of monogalactosyl diacylglycerol and phosphatidylglycerol. These findings indicate a stage-dependent response of Ditylum to UV-B irradiance. The impact of UV-B resulted in an increase of unsaturated long chained fatty acids (C18, C20) and in a diminution of short chained fatty acids (C14, C16). Content of ATP was not affected by UV-B radiation under the used conditions. The inhibitory effect of UV-B on synthesis of DNA, RNA, protein and acyl lipids was mainly reversible. Results were discussed with reference to UV-B damage on the enzymes involved in the biosynthesis of acyl lipids and by a reduction of available metabolites.

The livers of male chicks which had been fed a soybean and corn type ration for 8 weeks were exhaustively extracted with chloroform–methanol. The total (4.47%) lipid extract was separated into neutral and phospholipid fractions on a Unisil column and the mixed fatty acids in each fraction determined with the aid of gas chromatography. The neutral lipid fraction contained 3.16% hydrocarbons, 4.73% cholesterol esters, 74.99% triglycerides, 0.28% free fatty acids, 11.57% cholesterol, 3.37% diglycerides, and 1.54% monoglycerides. The phospholipid fraction contained 5.7% cardiolipin, 32.3% phosphatidyl ethanolamine, 7.3% phosphatidyl inositol, 3.0% phosphatidyl serine, 47.9% phosphatidyl choline, and 3.8% sphingomyelin and lysophosphatidyl choline. The cholesterol ester and diglyceride fractions contained twice as much linoleic acid as the other neutral lipid fractions. Phospholipids were characterized by a high content of polyunsaturated fatty acids.

The present studies aimed to analyse and assess the health-promoting properties of the musculus semimembranosus (MS) of roe and red deer harvested in the wild. The experimental materials comprising the carcasses of roe deer (15 does and 15 bucks) and red deer (15 hinds and 15 stags) were selected for analysis based on the following criteria: age of animals at harvest—3–4 years; time that passed from the harvest of animals to carcass cutting—48–54 h. After chilling the carcasses for 24 h at +2 °C, the haunches were cut from the carcasses and transported to the laboratory in isothermal ice-packed containers. Samples of the musculus semimembranosus were analysed to determine the chemical composition (proximate components, energy value, Fe, Pb, Cd, fatty acids). More (p ≤ 0.05) dry matter and total protein and less (p ≤ 0.05) crude fat was found in the musculus semimembranosus of roe deer in comparison to red deer. No significant influence of the animal’s sex was observed in the content of the evaluated nutrients, apart from crude fat content, which was increased in the haunch of females from both species. The energy content in the muscle of roe deer was 10% higher than the energy value of MS in red deer (p ≤ 0.05). The content of iron was significantly (p ≤ 0.05) higher (6.64 mg/kg) in the meat of red deer compared to the meat of roe deer (31.68 mg/kg). Roe deer haunch contained more lead but less cadmium than red deer haunch. Irrespective of sex, the lipid fraction of red deer muscle contained more saturated fatty acids (SFA). Intramuscular fat (IMF) in roe deer meat contained a higher percentage of polyunsaturated fatty acids (PUFA). The atherogenicity (AI) and thrombogenicity (TI) index values were significantly lower in roe deer haunch, and the hypocholesterolaemic-to-hypercholesterolaemic fatty acids ratio (HH) was lower (p ≤ 0.05) in red deer meat. To sum up, the evaluated roe deer and red deer haunch featured high dietary value as it contained a high protein and low fat content and had an advantageous fatty acids composition. As regards the content of cadmium and lead, roe deer and red deer haunch can be considered safe for consumers.

Various biomolecules induce neutrophil extracellular trap (NET) formation or NETosis. However, the effect of fatty acids on NETosis has not been clearly established. In this study, we focused on the NETosis-inducing ability of several lipid molecules. We extracted the lipid molecules present in Arabian Gulf catfish (Arius bilineatus, Val) skin gel, which has multiple therapeutic activities. Gas chromatography–mass spectrometry (GC-MS) analysis of the lipid fraction-3 from the gel with NETosis-inducing activity contained fatty acids including a furanoid F-acid (F6; 12,15-epoxy-13,14-dimethyleicosa-12,14-dienoic acid) and common long-chain fatty acids such as palmitic acid (PA; C16:0), palmitoleic acid (PO; C16:1), stearic acid (SA; C18:0), and oleic acid (OA; C18:1). Using pure molecules, we show that all of these fatty acids induce NETosis to different degrees in a dose-dependent fashion. Notably, F6 induces a unique form of NETosis that is rapid and induces reactive oxygen species (ROS) production by both NADPH oxidase (NOX) and mitochondria. F6 also induces citrullination of histone. By contrast, the common fatty acids (PA, PO, SA, and OA) only induce NOX-dependent NETosis. The activation of the kinases such as ERK (extracellular signal-regulated kinase) and JNK (c-Jun N-terminal kinase) is important for long-chain fatty acid-induced NETosis, whereas, in F-acid-induced NETosis, Akt is additionally needed. Nevertheless, NETosis induced by all of these compounds requires the final chromatin decondensation step of transcriptional firing. These findings are useful for understanding F-acid- and other fatty acid-induced NETosis and to establish the active ingredients with therapeutic potential for regulating diseases involving NET formation.

The effects of two synthetic diets and four algal diets on the survival and fecundity of Moinodaphnia macleayi for use in life-cycle toxicity tests were determined. The synthetic foods were a suspension of blended cichlid fish pellets with powdered alfalfa, either prepared fresh or fermented in natural water for 3 days. The algae, isolated from the habitat of M. rnacleayi in the Alligator Rivers Region, were Selenastrum sp., Ankistrodesmus (falcatus?), Scenedesmus quadricauda and a Chlamydomonas sp. Each alga was tested alone or as a supplement to the fermented synthetic food. The diet that supported the greatest number of young and the highest survival rate over a 5-day period was a combination of the fermented synthetic food and Chlamydomonas sp. Analysis of the fatty acid composition of the lipid fraction of the four algal species showed that Chlamydomonas sp., Selenastrum sp. and Scenedesmus quadricauda had significant concentrations of the highly unsaturated fatty acids 18 : 2(n-6), 18 : 3(n-6) and 18: 3(n-3). In terms of size and dry weight, Chlamydomonas sp. was the smallest of the algal cells studied. The toxicity of uranium to M. macleayi survival, determined under standard conditions, was a lowest-observed-effect concentration (LOEC) of 200 �g L-1 after 48 h and 25 �g L-1 after 5 days.

A thorough analysis and comparison of the fatty acid profiles of stipe and blade from Laminaria hyperborea, a kelp species found in the northern Atlantic, is presented. Lipids were extracted and fractionated into neutral lipids, free fatty acids and polar lipids, then derivatized to fatty acid methyl esters prior to GC-MS analysis. A total of 42 fatty acids were identified and quantified, including the n-3 fatty acids α-linolenic acid, stearidonic acid and eicosapentaenoic acid. An n-6/n-3 ratio of 0.8:1 was found in blade and 3.5:1 in stipe, respectively. The ratios vary between the lipid fractions within stipe and blade, with the lowest ratio in the polar lipid fraction of blade. The fatty acid amounts are higher in blade than in stipe, and the highest amounts of n-3 fatty acids are found within the neutral lipid fractions. The amounts of polyunsaturated fatty acids are 3.4 times higher in blade than stipe. This study highlights the compositional differences between the lipid fractions of stipe and blade from L. hyperborea. The amount of polyunsaturated fatty acids, compared to saturated- and monounsaturated fatty acids, as well as the n-6/n-3-ratio, is known to influence human health. In the pharmaceutical, food, and feed industries this can be of importance for production and sale of different health products. Additionally, lipids are today among the unused by products of alginate production, exploiting this material for commercial interest should give both economical and environmental benefits.

This article discusses the effect of the disintegration of the cell wall of the microalgae Chlorella sorokiniana on the output of the lipid fraction. The biomass of the microalgae Chlorella sorokiniana was grown under laboratory conditions in special photobioreactors at a temperature of 25 °C, with a constant aeration of a mixture of carbon dioxide and air at a rate of 1.5 liters/min, illumination 2200-2800 Lx. Nutrient medium for cultivation contained macro – and micronutrients for high-speed growth of microalgae. Selection of optimal cultivation parameters allows obtaining biomass with desired properties. Disintegration was carried out with the homogenization of biomass and under the influence of microwave radiation. Extraction of lipids was carried out on a semi-automatic extractor according to the Randall method, using organic solvents. The output of the lipid fraction without treatment was 10.18% after the destruction of the cell wall 14.45% with the homogenization of biomass and 13.85% under the influence of microwave radiation. A qualitative analysis of the lipid fraction, carried out under gas chromatography, obtained under various conditions showed that there was no significant difference in composition from the disintegration method. Lipid fractions (more than 50%) in both cases consist mainly of unsaturated fatty acids, of which irreplaceable unsaturated fatty acids constitute more than 18% for both samples. The residual biomass formed after the extraction of the lipid fraction can be used as fertilizer in the plant, for the manufacture of sorption materials for the purification of industrial water and as a biofuel. The purpose of this study was to study the effect of cell wall disintegration on the output of the lipid fraction and qualitative composition.

Background: Chromatography is one of the important laboratory technique in which the components of a mixture are separated on an adsorbent in order to analyze, identify, purify and quantify a mixture. Thin Layer Chromatography (TLC)is used to support the identity of a compound in a mixture when the Rf of a compound is compared with the Rf of a known compound. High Performance Thin Layer Chromatography is a sophisticated and automated form of Thin Layer Chromatography (TLC). The procedure simultaneously processes the sample and standard that results in better analytical precision and accuracy at a faster pace. Pharmacological/ Toxicological action of Nux Vomica is because of its active principles present in the seeds namely strychnine, brucine etc. This research paper aims to corelate the active principles present in Nux Vomica with the toxicological action of the same. Materials and Methods: 1. Standard Nux Vomica mother tincture was tested for its alkaloid markers and its correlation with the toxicological action was studied. 2. Analysis of the mother tincture was done using High Performance Thin Layer Chromatography. 3. Stationary phase consisted of TLC Aluminium sheets with silica gel 60 F253 pre-coated layer (20cm x 10cm), thickness-0.2mm, no. of tracks-18, band length-6mm. 4. Mobile Phase consisted of Chloroform: Methanol (9.5:0.5). 5. The plate was developed in developing chamber and observed under U.V. Light. Results: Colours seen on the HPTLC Plates of samples are greenwhich corresponds to strychnine, dark blue which corresponds to brucine, orange to alkaloids fluorescent green to sterols and pink to fatty acids which are evident on the chromatogram. Conclusion: Therapeutic action of Nux Vomica as noted in Homoeopathic Materia Medica is because of the active principles like strychnine, brucine, alkaloids, sterols, fatty acids present in it which is evident from the chromatogram.

The objective of this study was to determine the quality and composition of fatty acid in the lipid fraction of silages obtained from the residue of tilapia processing. Stratification of the lipid layer of the silages occurred at different times among the two types of silage (acid and fermented) and the greatest volume of oil was observed in acid silage (8.67% p/p). Although acid silage was more oxidized, it showed lower contents of free fatty acids probably because the degree of hydrolysis of its components is lower than that of fermented silage. Fatty acid composition did not differ among processes inasmuch as level of ϖ-3 was slightly higher in fermented silage. According to the degree of saturation, monounsaturated fatty acids stood out as the predominant category in acid and fermented silages with values of 39.69% and 33.39%, respectively. The use of antioxidants in the silage is needed because the process of production is carried out at temperatures higher than room temperature. The oil in the silages has excellent nutritional value and contains fatty acids essential for animal feeding.
