Background: Calcium bentonite is a type of clay produced by a devitrification of volcanic ash which is often used as a traditional medicine to absorb toxins and waste products of metabolism. The aim of this study was to determine the lethal dose of calcium bentonite in Wistar rats to explore its toxicity level and safe use.Methods: Fifty male and female Wistar rats were randomly divided into five groups for each sex. The control group was given 5 cc aquadest whereas the other four groups received calcium bentonite solution of 50 mg/kgBW, 300 mg/kgBW, 2,000 mg/kgBW, and 5,000 mg/kgBW, respectively, at a single dose. The weight was observed for up to 7 days and analyzed using the unpaired t-test and Mann-Whitney test. The death rate was calculated using the probit analysis. Median lethal dose results were then classified according to Loomis Criteria. Results: No deaths occurred at the highest dose, suggesting that the median lethal dose value of calcium bentonite was >5,000 mg/kgBW. No weight loss occurred due to the administration of calcium bentonite and a significant increase in the body weight was even observed in the male rat group. Interestingly, a significant decrease was found in the female rats group when compared to the control group.Conclusions: Calcium bentonite is classified as a practically non-toxic material with a median lethal dose of over 5,000 mg/kgBW.

Cadmium (Cd) present in the air, drinking water and food has the potential to affect the health of people, mainly those who live in highly industrialized regions. Cd affects placental function, may cross the placental barrier and directly modify fetal development. It is also excreted into milk. The body is particularly susceptible to Cd exposure during perinatal period. The effect on rat oral epithelium (floor of the mouth) after continuous exposure to drinking water containing low levels of Cd during lactation was studied. Female rats were supplied with ad libitum drinking water containing 300 mg/l of CdCl2 throughout the whole lactation period. Control animals received a similar volume of water without Cd. Lactating rats (21 day-old) were killed by lethal dose of anesthetic. The heads were retrieved, fixed in "alfac" solution (alcohol, acetic acid and formaldehyde) for 24 h, serially sectioned in frontal plane, at the level of the first molars. The 6 µm sections were then stained with hematoxylin and eosin. Nuclear epithelium parameters were estimated, as well as cytoplasm and cell volume, nucleus/cytoplasm ratio, numeric and surface densities, and epithelial thickness. Mean body weight was 34.86 g for the control group and 18.56 g for the Cd-treated group. Histologically, the floor of the mouth epithelium was thinner in the treated group, with smaller and more numerous cells. In this experiment, Cd induced epithelial hypotrophy, indicating a direct action in oral mucosa cells, besides retarded development of the pups.

The present study was aimed at investigating the safety of Lacidipine (LCDP) loaded nanostructured lipid carriers (NLCs) in Wistar rats. NLCs were formulated using ultrasound dispersion technique. Animals were orally treated once daily with NLCs containing 0.140 mg, 0.350 mg, and 0.875 mg of LCDP as low, medium, and high dose per kg body weight, respectively, during 28 days along with blank formulation and pure LCDP. Control rats were fed with water. Animals were observed throughout experiment period and their body weight was recorded once weekly. Overnight fasted rats were sacrificed on the 29th day. Study revealed no signs or symptoms of toxicity or morbidity. No significant changes in the body weight were observed between treated and control group. Significant increase in left testis weight and liver weight was observed in male and female rats, respectively. Haematological estimation revealed significant decrease in haemoglobin count in male rats while female rats showed significant increase in granulocyte count. All the serum clinical parameters were within the normal range and no gross histopathological changes were observed. No delayed effect was noted in satellite group. The results indicate that developed LCDP loaded NLCs are safe when administered orally in rats.

Background: Nicotine has been widely reported to generate free radicals in several organs of the body. Therefore, this study investigated the ameliorative potentials of Quercetin on selected reproductive biomarkers in female Wistar rats.Methods: Forty female rats were randomly divided into five groups (n=8) which were treated as follows: Group I (CN) received normal saline (1 ml/kg bwt); Group II (LN) received nicotine (0.5 mg/kg bwt) only; Group III (HN) received nicotine (1.0 mg/kg bwt) only; however, Groups IV (LNA) and V (HNA) received 0.5 and 1.0 mg/kg of nicotine with 20 mg/kg of quercetin respectively. All administration were done orally and lasted 28 days. Results: Nicotine significant reduction in the bioavailability of estrogen and progesterone in the treated rats when compared to control. Furthermore, relative to the control group, nicotine administration resulted in significant elevation in MDAand consequently caused significant reduction in the antioxidant enzymes (SOD and CAT) activities. However, quercetin treatment ameliorated these alterations. Conclusion: Quercetin dampens the reproductive distruptive effects exerted by nicotine through anti-oxidative mechanism in female Wistar rats.

The aim of this study was to investigate the clinical efficacy of vanadium complexes on triglycerides (TG), total cholesterol (Chol), uric acid (UA), urea (U), and antioxidant parameters: nonenzymatic (FRAP—ferric reducing ability of plasma, and reduced glutathione—GSH) and enzymatic (glutathione peroxidase—GPx, catalase—CAT, and GPx/CAT ratio) activity in the plasma of healthy male Wistar rats. Three vanadium complexes: [VO(bpy)2]SO4⋅2H2O, [VO(4,4′Me2bpy)2]SO4⋅2H2O, and Na[VO(O2)2(bpy)]⋅8H2O are administered by gavage during 5 weeks in two different diets such as control (C) and high fatty (F) diets. Changes of biochemical and antioxidants parameters are measured in plasma. All three vanadium complexes statistically decrease the body mass growth in comparison to the control and fatty diet. In plasma GSH was statistically increased in all vanadium complexes-treated rats from control and fatty group in comparison to only control group. Calculated GPX/CAT ratio was the highest in the control group in comparison to others.

The aim of this study was to evaluate the influence of ligature-induced periodontal disease in pregnant rats on their newborn's health parameters. Twenty-four female adult Wistar rats were divided into two groups: the control group (G1) and the group that was submitted to dental ligatures around second upper molars (G2). After the four week period of development of periodontitis, the female animals were mated with male adult Wistar rats. There were no differences in the body weight of females between the two groups during mating and pregnancy. No differences were observed among the groups in relation to the viable newborn index. However, there were differences in newborn birth weight, explained by the diverse size of the litters. In this study, ligature-induced periodontal disease did not promote changes during pregnancy that resulted in low birth weight in newborn Wistar rats.

The aim of this research was to investigate the effects of extract ethanol of pegagan (Centella asiatica) after treated with restraint stress by measuring the eight arms radial maze performance of rats. Pre-test and post-test controlled groups design was applied in this research. Male Wistar rats (Rattus novergicus) with the mean age of 8 weeks and the body weight of 150-200 grams which used for sample were divided randomly into 4 groups, each group consisted of 6 rats, i.e. (i) control group (without any treatment), (ii) stress group (it was given by restraint stress for 2 hours/day for each rat), (iii) pegagan group (it was given by 0.3 mg/g BW/day/rat extract ethanol of pegagan), and (iv) pegagan and stress group (it was given by 0.3 mg/g BW/day/rat extract ethanol of pegagan and restraint stress for 2 hours/day for each rat). The treatments were given for 21 days. Within 12 days for each pre-treatment and post-treatment, a test on the eight arm radial maze was conducted on individual rat to observe its performance. The assessment of rat performance in the eight arms radial maze test was conducted based on error type B. Kruskal-Wallis and Mann-Whitney tests with SPSS for Windows 16 version were applied to analyze statistically the difference between four groups. Kruskal-Wallis test was used to show the significant performance level difference between four groups of rats with p=0.001, while Mann-Whitney test was used to determine the significant difference between stress group and pegagan group (p=0.001), also stress group and pegagan and stress group (p=0.001). The result of research showed that there was no significant difference between control group and stress group (p=0.051), control group and pegagan group (p=0.143), control group and pegagan and stress group (p=0.143), also pegagan group and pegagan and stress group (p=0.952). It was concluded that extract ethanol of pegagan improved the performances of rats on the eight arms radial maze after treated with restraint stress.

Background: Early life stress (ELS) models such as maternal deprivation (MD) are used to in¬vestigate behavioral changes in rodents under stressful situations. MD is a situation in which rat pups are separated from the dam; MD has different paradigms. The purpose of this research is to evaluate the effects of maternal deprivation on anxiety, depression, and empathy in adult Wistar rats. Materials and Methods: MD was applied to pups as per specifically designed protocol to compare rats of the control group with maternal deprivation rats and also the group, which faced novel objects. Each group consisted of eight rats. In this study, separation started from postnatal day (PND) 14 for various periods up to PND 60. EPM test was undertaken to measure anxiety; moreover, FST was used to indicate levels of depression. Also, changes in the empathy ratio were also demonstrated. One-way analysis of variance (ANOVA), Tukey’s post hoc analysis, and t-test were applied to analyze the results. Results: MD-treated rats showed a significant decrease in anxiety and empathy indexes compared with those in the control group (P<0.05). However, MD significantly increased depression in both male and female rats (P<0.05). Final¬ly, exposure to novel objects decreased depression but did not have any effect on anxiety and empathy levels in MD rats (P<0.05). Conclusion: ELS may lead to various states of mood and behavior in adulthood. According to the findings of this study, depression increases due to MD, though both anxiety and empathy decrease in both male and female Wistar rats. Moreover, ex¬posure to novel objects decreases depression, while anxiety and empathy do not change signifi¬cantly with exposure to novel objects. [GMJ.2019;8:e1093]

Ficus vogelii is a medicinal plant mainly found in tropical Africa and reported to treat inflammatory complaints. This study aims to evaluate the acute and sub-chronic toxicity of the aqueous extract of Ficus vogelii stem bark in wistar rats. For acute study, aqueous extract at a single dose of 5000 mg/kg body weight was administered to female rats and observed for 14 days. In the sub-chronic study, the extract was administered daily to both sex rats at the doses of 100, 200, 400, and 600 mg/kg body weight for 28 consecutive days. Body weight was measured weekly, while hematological, biochemical, and histopathological parameters were analyzed after euthanize. Aqueous extract of Ficus vogelii at all tested doses didn’t produced any mortality or significant change on the body weight and relative weight of rats on acute and sub-chronic studies. The lethal dose 50 was estimated greater than 5000 mg/kg (DL50˃5000 mg/kg). Hematological parameters were recorded non-significant in all treated rats. Aqueous extract at 600 mg/kg significantly changed transaminases and alkaline phosphatase activities, these changes were reversible in satellites. The concentrations of bilirubin was increased at 200 and 600 mg/kg in male rats, at 100, 400 mg/kg in female rats. The levels of lipids markers didn’t changed, except the significant decrease of LDL-cholesterol. Histological examination didn’t showed any change in the architecture of the liver and kidney of rats treated compared to control. Thus aqueous extract of Ficus vogelii stem bark didn’t produced adverse effects in rats after oral acute and sub-chronic treatment.

Objective: Potash known as potassium carbonate (K2CO3) is a mixture of salt with other components, including impurities which coexist in mineral and salt is highly consumed in various forms by pregnant women. The aim of this research is to determine the effect of potash on the weight index of pregnant Wistar rats. Methods: A total of 25 albino Wistar rat with weights ranging from 180 to 300 g were used and allocated into five groups of five animals each (four females and one male) designated as Groups A, B, C, D, and E. The experimental Groups B, C, D, and E were administered through oral route different doses of potash of 300 mg/kg, 600 mg/kg, 900 mg/kg, and 1200 mg/kg, respectively, after pregnancy was detected by checking for mucus plug in the vagina. Group A served as the control group and was administered distilled water only. The animals were allowed for 1 week for acclimatization under normal temperature (270–300°C), which they were being fed with normal feed (grower’s mash) and water ad libitum for 1 week. Results: The result showed a significant (p˂0.05) reduction in weight with the highest level seen with the 1200 mg/kg group when compared to the control. Conclusion: Therefore, the effect of potash alters the physical activity and decreases weight, by implication may induce growth retardation of the Wistar rats which is not healthy for a pregnant animal.

PURPOSE: Thermal injury causes catabolic processes as the body attempts to repair the damaged area. This study evaluated the effects of a scald injury on the morphology of muscle fibers belonging to a muscle distant from the lesion. METHODS: Thirty Wistar rats were divided into control (C) and scalded (S) groups. Group S was scalded over 45% of the body surface, standardized by body weight. Rats in both groups were euthanized at four, seven and 14 days following the injury. The middle portions of the medial gastrocnemius muscles were sectioned, stained with hematoxylin and eosin and Picrosirius, and submitted to histological analysis. RESULTS: Control group sections exhibited equidistantly distributed polygonal muscle fibers with peripheral nuclei, characteristic of normal muscle. The injured group sections did not consistently show these characteristics; many fibers in these sections exhibited a rounded contour, variable stain intensities, and greater interfiber distances. A substantially increased amount of connective tissue was also observed on the injured group sections. CONCLUSION: This experimental model found a morphological change in muscle distant from the site of thermal injury covering 45% of the body surface.
