ABSTRACT Objective To evaluate the impact of low to moderate aerobic exercise and ovariectomy on body composition and food consumption in female rats. Methods Forty adult Wistar female rats (age: 23 weeks; body weight: 275.2±3.6g; mean±SEM) were divided into 4 groups (n=10): laparotomy-sedentary; laparotomy-exercised; ovariectomy-sedentary; and ovariectomy-exercised. The exercised groups were submitted to a treadmill running program (16m/min; 30min/day, 5 days/week), for 8 weeks. Body weight and food consumption were monitored during the experiment. Visceral fat and carcass water, protein, ash, fat and carbohydrate fractions were analyzed. Two-way ANOVA plus the Tukey’s post hoc test was used for comparisons and p<0.05 was considered significant. Results The ovariectomized (ovariectomy-sedentary+ovariectomy-exercised) and sedentary (laparotomy-sedentary+ovariectomy-sedentary) animals showed higher (p<0.05) weight gain, food consumption, food efficiency ratio and weight gain/body weight ratio than laparotomy animals (laparotomy-sedentary+laparotomy-exercised) and exercised (exercised laparotomy+exercised ovariectomy), respectively. The ovariectomized and sedentary animals showed higher (p<0.05) carcass weight, fat percentage and visceral fat than laparotomy and exercised rats, respectively. Conclusion Ovariectomy and physical inactivity increase obesogenic indicators, whereas regular aerobic exercise of low to moderate intensity attenuates these unfavorable effects in female rats.

Epidemiological and animal studies suggest that the alteration of hormonal and metabolic environment during fetal and neonatal development can contribute to development of metabolic syndrome in adulthood. In this paper, we investigated the impact of maternal high-fat (HF) diet on hypothalamic leptin sensitivity and body weight gain of offspring. Adult Wistar female rats received a HF or a control normal-fat (C) diet for 6 wk before gestation until the end of the suckling period. After weaning, pups received either C or HF diet during 6 wk. Body weight gain and metabolic and endocrine parameters were measured in the eight groups of rats formed according to a postweaning diet, maternal diet, and gender. To evaluate hypothalamic leptin sensitivity in each group, STAT-3 phosphorylation was measured in response to leptin or saline intraperitoneal bolus. Pups exhibited similar body weights at birth, but at weaning, those born to HF dams weighed significantly less (−12%) than those born to C dams. When given the HF diet, males and females born to HF dams exhibited smaller body weight and feed efficiency than those born to C dams, suggesting increased energy expenditure programmed by the maternal HF diet. Thus, maternal HF feeding could be protective against adverse effects of the HF diet as observed in male offspring of control dams: overweight (+17%) with hyperleptinemia and hyperinsulinemia. Furthermore, offspring of HF dams fed either C or HF diet exhibited an alteration in hypothalamic leptin-dependent STAT-3 phosphorylation. We conclude that maternal high-fat diet programs a hypothalamic leptin resistance in offspring, which, however, fails to increase the body weight gain until adulthood.

Purpose. Transitioning from high school to college generally results in reduced physical activity and weight gain at a rate that is higher than the general population. The purpose of this study was to examine the effects of three progressively higher step recommendations over 24 weeks on changes in body weight and body composition. Methods. Ninety-two freshmen college women wore a multifunction pedometer for 24 weeks after being randomly assigned to a daily step level: 10,000, 12,500, or 15,000. Pedometer data were downloaded every two weeks and participants were counseled on meeting their step recommendation. Body weight and body composition were assessed at baseline and 24 weeks. Body composition was assessed by dual X-ray absorptiometry. Results. On average, women took 10,786 ± 1501, 12,650 ± 2001, and 13,762 ± 2098 steps per day for the 10,000-, 12,500-, and 15,000-step groups, respectively (F = 15.48, P<0.0001). Participants gained 1.4 ± 2.6, 1.8 ± 2.1, and 1.4 ± 2.1 kg for the 10,000-, 12,500-, and 15,000-step groups, respectively (F = 37.74, P<0.0001). Weight gain was not significantly different between groups (F = 0.18, P=0.8385). There was also no difference in fat weight gain (F = 0.41, P=0.7954). Discussion. A step recommendation beyond 10,000 does not prevent weight or fat gain over the first year of college. Future research should focus on either intensity of physical activity or the addition of dietary interventions to prevent weight gain during the first year of college.

Forty male and 40 female Crl:SD® CD® IGS rats were fed diets containing 0, 40 000, 80 000, or 120 000 ppm tamarind seed polysaccharide (equivalent to 3450.8, 6738.9, or 10 597.1 mg/kg bw/day and 3602.1, 7190.1, or 10 690.7 mg/kg bw/day for males and females, respectively) for 28 days. Animals were observed for adverse clinical signs, body weight, feed consumption, hematology and clinical chemistry parameters, urinalysis values were recorded, and at the end of the study the rats underwent a full necropsy. Functional Observational Battery (FOB) and Motor Activity (MA) tests were performed on all animals. There were no mortalities, no clinical or ophthalmologic signs, body weight, body weight gain, food consumption and food efficiency, FOB or MA findings associated with the administration of tamarind seed polysaccharide. Initial statistically significant decreases in body weight gain and food consumption resolved after the first week and were considered the result of reduced palatability. There were no adverse changes in hematology, coagulation, clinical chemistry or urinalysis parameters in male or female rats considered the result of test substance administration. At necropsy, there were no macroscopic, histopathological findings, estrus cycle, or organ weight changes deemed related to administration of the test substance. Under the conditions of this study and based on the toxicological endpoints evaluated, the no-observed-adverse-effect level (NOAEL) for tamarind seed polysaccharide in the diet was the highest concentration tested of 120 000 ppm (equivalent to 10 597 mg/kg bw/day and 10 691 mg/kg bw/day for male and female rats, respectively).

Objective The purpose of this study was to investigate the effects of aerobic exercise on PKC/CaV1.2 pathway in mesenteric arterial smooth muscle from spontaneously hypertensive rats (SHRs) Methods Twelve-week-old male normotensive Wistar–Kyoto (WKY) rats and SHRs were randomly assigned to sedentary groups (SHR-SED, WKY-SED) and exercise training groups (SHR-EX, WKY-EX). Exercise groups were performed an 8-week moderate-intensity treadmill running. After 8 weeks, vascular contractility of mesenteric arteries was measured. Vascular smooth muscle cells (VSMCs) were obtained with an enzymatic isolation method. CaV1.2 channel currents were examined by using whole-cell patch clamp recording technique. Results  1) Body weight and systolic blood pressure (SBP) in both WKY-EX and SHR-EX were significantly lower than those of their sedentary counterparts (both P<0.05). Body weight in SHR-SED was remarkably lower than WKY-SED (P<0.05), while SBP was much higher than WKY-SED (P<0.05). 2) PDBu (PKC activator) elicited a tension increase, and Gö6976 (PKC inhibitor) induced vasodilation. Both the responses of PDBu and Gö6976 in SHR-SED were notably increased compared with WKY-SED (both P<0.05), however, exercise training significantly suppressed these increases (both P<0.05). 3) Nifedipine (CaV1.2 inhibitor) induced vasodilation. Response to nifedipine in SHR-SED was more sensitive than both SHR-EX and WKY-SED (both P<0.05). 4). The current density of SHR-SED and WKY-EX exhibited an increase compared to the WKY-SED (both P<0.05), and the current density of the SHR-EX decreased obviously in contrast with SHR-SED (P<0.05). Besides, PDBu enlarged current density of all the groups, while Gö6976 decreased current density. The increase or decrease amplitude in SHR-SED was significantly higher than WKY-SED (both P<0.05), whereas exercise training markedly inhibited those responses (both P<0.05). Conclusions Aerobic exercise efficiently prevents the upregulation of PKC/CaV1.2 pathway in hypertension, and enhances the function of vascular smooth muscle.

To analyse the change on leptin, body composition, blood pressure (BP), cardiorespiratory fitness (CRF) and some biochemical parameters in physically inactive women and men with primary hypertension (HTN) and obesity, and to evaluate the potential sex differences in the change after intervention. METHODS. Participants (n = 37 women, n= 40 men, 52.9 ± 6.9 yrs) from the EXERDIET-HTA study were randomized into attention control group (physical activity recommendations) or one of three supervised aerobic exercise groups [two days/week: high-volume (HV) with 45 min of moderate-intensity continuous training, HV and high-intensity interval training (HV-HIIT), and low volume-HIIT (LV-HIIT, 20 min)]. All participants received the same hypocaloric diet. All variables were assessed pre and post intervention (16 weeks). 24 h ambulatory BP monitoring was used to analyze systolic and diastolic BP. A cardiopulmonary exercise test was performed to determine peak oxygen uptake (VO2peak). RESULTS. Following the intervention, there were increments (P &lt; 0.01) in CRF by VO2peak (Women, W = 21.1 ± 3.7 vs. 24.6 ± 4.4 mL·kg-1·min-1, Men, M = 26.3 ± 6.0 vs. 33.1 ± 10.2 mL·kg-1·min-1) and decreases (P &lt; 0.05) in leptin (W = 49.5 ± 23.0 vs. 41.8 ± 19.9 ng/mL, M = 20.5 ± 14.8 vs. 12.9 ± 18.6 ng/mL), body mass (W = 84.7 ± 12.1 vs. 80.3 ± 11.5 kg, M = 97.9 ± 14.4 vs. 91.5 ± 13.3 kg), waist perimeter (W = 97.3 ± 10.7 vs. 94.3 ± 10.9 cm, M = 107.9 ± 8.7 vs. 101.5 ± 7.9 cm), fat mass (W = 42.3 ± 5.1 vs. 38.6 ± 8.4 %, M = 31.2 ± 5.0 vs. 28.0 ± 4.4 %), systolic BP (M = 136.5 ± 12.1 vs. 129.3 ± 12.5 mmHg), diastolic BP (W = 76.2 ± 8.9 vs. 74.1 ± 8.7 mmHg, M = 79.3 ± 7.2 vs. 75.0 ± 8.2 mmHg), total cholesterol (M = 216.1 ± 44.5 vs. 196.1 ± 35.0 mg/dL), insulin (W = 13.4 ± 7.9 vs. 9.4 ± 4.2 mU/L) values. There were significant between-sex differences in body mass (W=-5.2%, M=-6.5%, effect size, ES = 0.073), waist circumference (W=-3.1%, M=-5.9%, ES = 0.114), and VO2peak (W = 14.2%, M = 20.5%, ES = 0.059). CONCLUSIONS. Aerobic exercise along with hypocaloric diet is an effective non-pharmacological tool to induce beneficial changes in women and men in BP and leptin as a mediator of obesity-induced HTN, and other regulatory mechanisms such as body composition, CRF and biochemical profile. The found sex-related differences could confirm the need of individual non-pharmacological strategies.

Doxorubicin (DOX), an anticancer drug, causes a dose-dependent cardiotoxicity. Some evidence suggests that female children have an increased risk for DOX-mediated cardiac damage. To determine whether the iron chelator dexrazoxane (DXR) could reduce DOX-induced cardiotoxicity in the young, we injected day 10 neonate female and male rat pups with a single dose of saline or DOX, DXR, or DXR + DOX (20:1). We followed body weight gain with growth, measured cardiac hypertrophy after a 2-wk swim exercise program, markers of apoptosis (Bcl-2, BAX, BNIP1, caspase 3 activation), oxidative stress (heme oxygenase 1, protein carbonyl levels), the chaperone protein clusterin, and the transcriptional activator early growth response gene-1 (Egr-1) in hearts of nonexercised and exercised rats on neonate day 38. All DOX-alone and DXR + DOX-treated rats showed decreased weight gain, with female rats affected earlier than male rats. DXR-alone, DOX-alone, and DXR + DOX-treated rats had an increased heart weight-to-body weight (heart wt/body wt) ratio after the exercise program with female rats showing the largest increase in heart wt/body wt. Drug-treated females also showed increased cardiac apoptosis, as measured by the increased expression of the proapoptotic proteins BAX and BNIP1 and the appearance of caspase 3 activation products, and oxidative stress, as measured by increased heme oxygenase 1 expression, and reduced Egr-1 and clusterin expression when compared with the similarly treated male rats. We conclude that DXR preinjection did not reduce DOX-induced noncardiac and cardiac damage and that young female rats were more susceptible to DXR and DOX toxicities than age-matched male rats.

Abstract Background The COVID-19 pandemic has forced governments around the world to impose strict hygiene and national lockdown measures, which in turn has changed the dietary and lifestyle habits of the world’s population. Thus, the aim of this study is to evaluate whether dietary and physical activity behaviors of Saudi Arabia’s adult population changed during the COVID-19 quarantine. Methods An electronic questionnaire which assessed changes in body weight, dietary habits, and physical activity of Saudi Arabia’s adult population (n = 2255) during the COVID-19 quarantine was distributed on social media between June and July 2020. To test the differences between changes in dietary and physical activity behaviors in relation to changes in body weight a Chi-square test was used. Results Over 40 and 45% of participants reported eating and snacking more, respectively, which led to weight gain in around 28%. Most participants reported that they consumed home-cooked (73%) and healthy meals (47%), while only 7% reported that they consumed foods from restaurants. Feelings of boredom and emptiness (44%) and the availability of time for preparing meals (40%) were the main reasons for changing dietary habits. Honey (43%) and vitamin C (50%) were the most consumed immune-boosting food and dietary supplement, respectively. COVID-19 also had a negative impact on physical activity, lowering the practice in 52% subjects, which was associated with significant weight gain (p < 0.001). Conclusion Assessing the changes to the population’s dietary habits and physical activity during the lockdown will help predict the outcome of the population’s future health and wellbeing after the pandemic.
