During enzymatic process of cheese manufacturing, rennin cleaves κ-casein releasing two fractions: para-κ-casein and glycomacropeptide (GMP), which remains soluble in milk whey. GMP is a peptide with structural particularities such as chain carbohydrates linked to specific threonine residues, to which a great variety of biological activities is attributed. Worldwide cheese production has increased generating high volumes of milk whey that could be efficiently used as an alternative source of high quality peptide or protein in foodstuff formulations. In order to evaluate isolation and recovery on whey GMP by means of thermal treatment (90 °C), 18 samples (2 L each) of sweet whey, resuspended commercial whey (positive control) and acid whey (negative control) were processed. Indirect presence of GMP was verified using chemical tests and PAGE-SDS 15%. At 90 °C treated sweet whey, 14, 20 and 41 kDa bands were observed. These bands may correspond to olygomers of GMP. Peptide recovery showed an average of 1.5 g/L (34.08%). The results indicate that industrial scale GMP production is feasible; however, further research must be carried out for the biological and nutritional evaluation of GMP's incorporation to foodstuff as a supplement.

Objective: The current study was carried out to evaluate the anthelmintic activity of the rhizome extract of Curcuma longa as an alternative source of effective remedies for nematodiasis.Methods: The anthelmintic activity of the C. longa was assessed in vitro against Haemonchus spp., a gastrointestinal (abomasum) parasite of goats. Different concentrations of the extracts (1 mg/mL, 2.5 mg/mL, 5 mg/mL, and 10 mg/mL) in phosphate-buffered saline (PBS) were tested, and the results expressed in terms of time of paralysis (minute) and time of death (minute) of the worms. Albendazole (1 mg/mL) was used as a reference (positive control) and PBS as a control group (negative control).Results: The qualitative phytochemical analysis of the methanolic extract (ME) of the plant disclosed the presence of alkaloids, glycosides, terpenoids, flavonoids, tannins, saponins, phenol, anthraquinone, and carbohydrates; whereas, the aqueous extract (AE) showed the presence of alkaloids, carbohydrate, flavonoids, and saponins. Both ME and AE of the C. longa (rhizome) expressed significant efficacy (p≤0.05) in causing paralysis as well as the death of the worms within 12 h of exposure at all tested concentrations, as compared to the negative control. The rhizome extracts of C. longa showed dose-dependent efficacy in causing paralysis of the worm motility and the final progression to death. The results showed that the ME at 10 mg/mL was significantly more potent (p≤0.05) over the AE.Conclusions: This study concluded that the rhizome extract of C. longa exhibited potent anthelmintic efficacy against the nematode parasite, Haemonchus spp.

Microalgal oils (AOs) emerged recently as an alternative to fish oil and to nutritionally poorer vegetable oils for fish species. In this trial, two experimental diets containing fish oil (negative control: 2.1%; positive control: 13.8%) and two diets incorporating AO at 3.5 and 0.7% were fed to grow out gilthead seabream (Sparus aurata) of 64.5 g initial body weight. After 110 days of experimental feeding, performance (final body weight mean = 147 g) and survival (>99%) were similar across treatments. The highest eicosapentaenoic acid (EPA) + docosahexaenoic acid (DHA) content in positive control (PC) and 3.5 AO feeds (3.11 and 2.18% of diet, respectively) resulted in the highest EPA + DHA deposition in the fillets (18.40 and 12.36 g/100 g fatty acid, respectively), which entirely reflected the dietary fatty acid profile. Feed and fillets from fish fed the AO diets had lower levels of dioxins and polychlorinated biphenyls (PCBs). Moreover, sensory quality of AO fillets scored equally to the PC fish. Collectively, these findings offer a more resilient means for sustaining the future growth of seabream aquaculture, whilst maintaining the nutritional value of the resulting seafood. The data supports the addition of seabream to the list of aquaculture species where microalgal oil can be used as an ingredient to fulfil their challenging nutritional demands.

Aim. To investigate the composition and properties of the samples of cheese and acid milk whey, obtained in industrial conditions using a combination of nanofi ltration and electrodialysis methods. Methods. Determi- nation of physical-chemical indices using standard methods, study of functional-technological properties of demineralized whey by common methods. Results. It was established that there was high effi ciency of apply- ing membrane methods for processing of secondary resources in current conditions of raw materials source, which are presented by different kinds of milk whey, formed during cheese production. It was determined that processing of different kinds of whey using the combination of nanofi ltration and electrodialysis methods led to a considerable decrease in the content of ash compared to the initial whey. The level of demineraliza- tion of cheese whey may amount to 90 %, that of acid whey – 75 %. In addition to dry kinds of whey, liquid demineralized whey is of some interest for practical application, which may be used during the production of sour-milk and milk-containing drinks due to a high content of dry substances. It was found that the increase in protein content in dry demineralized whey, obtained using the complex of membrane methods of processing, led to a considerable increase in its foam-forming, moisture-retaining, fat-retaining and emulsifying abilities compared to milk whey, obtained by a traditional technology. Conclusions. It was established that dry demin- eralized whey, obtained by a combination of nanofi ltration and electrodialysis methods, had better organoleptic and physical-chemical indices compared to dry whey. The investigated industrial samples were remarkable for improved functional and technological properties which allows using them in the formulations of other food products.

Marine organisms have been known produce certain compounds those could lead for medicine purposes. Sponges are one and the most studied for this aim. oneof the important biological activities which expected from sponge are larvacide activity. The aims this research was to test the larvacide activity aagainst Aedes aegypti larvae from some of sponge extract. Sponge samples were taken from Malalayang Waters, (N 01 ° 27'37 "E 124 ° 47'30") on November 2014 with the depth varies from 2-15 m  with SCUBA. The extraction, preparation of the larvae and activity testing was performed on Biomolecular and Marine Pharmacy Laboratory Faculty Fisheries and Marine Science. The sponge samples were cutted and soaked in 95% Ethanol for over night with 3 repetitions to obtain ethanolic extracts. The extract were filtered and evaporated using freeze dryer then tested onto 10 instars 3 instars m larvae that had been previously maintained. the test was made in triplowith 24 hours observation. abate was used as positive control while negative control clean water was used.The test results showed that of 11 Sponge tested, 10 species showed larvacidal activity and marine sponge extract Tedania sp. has the highest activity compared to 9 extracts. As a suggestion of this research the further purification of Tedania sp. extract is needed to know the structure of active compound.Keywords: Aedes aegypti, Larvacide, Sponge extractABSTRAKOrganisme laut yang dapat dikembangkan menjadi bahan sediaan obat antara lain sponge, dan merupakan salah satu organisme laut yang banyak diteliti. Beberapa aktivitas biologis penting yang diharapkan dari ekstrak sponge salah satunya adalah aktivitas larvasida. Penelitian ini bertujuan untuk menguji aktivitas larvasida nyamuk Aedes aegypti dari beberapa ekstrak Sponge. Sampel sponge diambil di perairan Malalayang, tepatnya di koordinat N 01°27’37” E 124°47’30” pada bulan November 2014 di kedalaman 2-15 m. Sedangkan untuk tahap ekstraksi, penyiapan larva uji dan pengujian aktivitas larvasida di lakukan di laboratorium Biomolekular dan Farmasitika Laut program studi Ilmu Kelautan, FPIK UNSRAT. Dalam penelitian yang dilakukan, sampel diambil di perairan menggunakan peralatan SCUBA. Setelah itu diekstrak dengan larutan etanol 95% dan direndam selama 24 jam dan dilakukan 3 kali pengulagan untuk mendapatkan ekstrak etanolik. Sampel dikeringkan dengan menggunakan alat freeze dryer kemudian diujikan ke 10 ekor larva nyamuk fase instar 3 yang telah dipelihara sebelumnya. Pengujian dilakukan sebanyak 3 kali pengulangan dengan lama pengamatan 24 jam pengamatan. Sebagai kontrol positif digunakan bubuk abate yang biasa dijual di pasaran sedangkan kontrol negative atau tanpa perlakuan digunakan air bersih. Data hasil pengamatan diolah menggunakan Microsoft excel.Hasil pengujian menunjukkan bahwa Dari 11 Sponge yang diuji, 10 jenis menunjukan aktivitas larvasida dengan persentase mortalitas yang bervariasi dan ekstrak sponge laut Tedania sp. memiliki aktivitas tertinggi dibandingkan dengan 10 jenis ekstrak sponge lain dalam pengujian. Sebagai saran dalam penelitian ini yaitu Perlu dilakukan pemurnian lanjut ekstrak lebih lanjut dari ekstrak sponge Tedania sp. yaitu ke tahap partisi dan Perlu adanya variasi konsentrasi dalam pengujian.Kata Kunci : Aedes aegypti, Larvasida, ekstrak Spong

Fetal Alcohol Syndrome (FAS) is a condition associated with drinking alcohol during pregnancy and affect embryo development. FAS is the most common congenital disabilities in the world. The effects of FAS are mental retardation, cranial abnormalities, and heart defects. Moringa oleifera is a highly valued plant. It has an impressive range of biological activities, including antioxidant, antiproliferation, and anti-inflammatory. This study aims to assess the effect of Moringa oleifera leaf extracts in inhibiting growth retardation of chicken embryo growth induced by alcohol. The study was a true experimental design with posttest controlled group design. There were five groups with 5 of chicken embryo each group. The treatments contained 15% alcohol and aquadest as positive control (P+), without alcohol as negative control (P-), P0.5 (0,5g/dl), P5 (5g/dl), P50 (50g/dl) by adding Moringa oleifera leaf extracts, respectively. Moringa oleifera leaf extracts showed the improvement of length and head diameter as a benchmark for chicken embryo development with P5 concentration showed the best results to overcome growth retardation in alcohol-induced chicken embryos. This finding implies against possibility Moringa oleifera leaf extracts can be alternative medicine to prevent growth retardation.

Comprehensive genetic testing has become integral in the evaluation of children with deafness, but the amount of blood required to obtain DNA can be prohibitive in newborns. Dried blood spots (DBSs) are routinely collected and would provide an alternative source of DNA. Our objective was to evaluate the use of DBSs for comprehensive genetic testing for deafness. DNA derived from fresh and archived DBS samples was compared with DNA from whole blood. We performed next-generation sequencing of all known deafness genes in 4 DBS samples: 2 positive controls, an unknown sample, and a negative control. The DBS-derived DNA was of sufficient quantity and quality for clinical testing. In the 2 positive control samples, pathogenic variants were identified; in the negative control, no pathogenic variants were found; and in the unknown sample, homozygous deletion of the OTOA gene was identified as the cause of deafness. This pilot study shows that comprehensive genetic testing for deafness is feasible with fresh and/or archived DBSs.

Inappropriate use of antibiotics can cause bacteria to become resistant and have side effects, therefore an alternative source of antibacterial is needed. One species of plant that has antibacterial properties is calincing (Oxalis corniculata L.). Antibacterial activities of the juice and decoction of calincing leaves against Streptococcus mutans have not reported yet. This study aims to determine the antibacterial activity of the juice and decoction of calincing leaves against Streptococcus mutans. This study uses a well diffusion method with eight treatment groups, namely juice and decoction at the concentrations of 20%, 40%, 60%; listerin 100% as a positive control, and distilled water as a negative control. The diameter of the inhibitory zone of the juice and decoction of calincing leaf at a concentration of 20% are 19 mm and 24.33 mm respectively, and classified as strong growth inhibition, while the concentrations of 40% and 60% are classified as very strong growth inhibition. Growth inhibition of the juice and decoction of calincing leaf at concentration of 20% was significantly higher than the positive control. The best antibacterial activity against Streptococcus mutans is produced by the juice and decoction of calincing leaf at concentration of 60%. Increasing in the concentration of the juice or decoction of calincing leaf also increases the diameter of the inhibitory zone.

The dairy industry seems to have convinced the food industry that whey is a miracle product. The list of supposed benefits it gives to food is as long as your arm. Some of the benefits may be real. Whey is the liquid remaining after milk has been curdled and strained. It is a by-product of the manufacture of cheese or casein and has several commercial uses. To produce cheese, rennet or an edible acid is added to heated milk. This makes the milk coagulate or curdle, separating the milk solids (curds) from the liquid whey. Sweet whey is the byproduct of rennet-coagulated cheese and acid whey (also called sour whey) is the byproduct of acid-coagulated cheese. Sweet whey has a pH greater than or equal to 5.6, acid whey has a pH less than or equal to 5.1. Whey is also a great way to add sweetness to a product without having to list sugar as an ingredient as whey contains up to 75% lactose. And it sounds healthy. This study is done to research the examinations for the production of mozzarella cheese from Cow’s milk, after research and analyses of a physical-chemical peculiar feature of whey from coagulum. We have followed the processes from the drying of whey from the coagulum analyzer's physical-chemical peculiar feature. We carried out three experiments. For every experiment, we took three patterns and analyzed the physical-chemical. The calculation was appraised statistically. This paper deals with the research of% of whey fat during the process of milk production from standardized to non-standardized milk. Where% of whey fat should be an economic indicator for standardizing milk for dairy production.

Shallot (Allium ascalonicum L.) has been known as traditional medicine. Shallot which has same genus with garlic(Allium sativum L.) contains allicin that is also found in garlic and has been suspected has fungicidal activity toCandida albicans. It is supported by several researches. Therefore, shallot is suspected has antifungal activity too.The aim of this research was to know antifungal activity of shallot’s water extortion againsts Candida albicans invitro. This was a laboratory experimental research which used completely randomized design, with diffusion method.Shallot’s water extortion was devided into three concentrations, there were 50%, 100% and 200%. Ketoconazole 2%was positive control and aquadest was negative control. The result of this research based on analysis of varians(Anova), there was significant difference between several treatments and was confirmed with Duncan New MultipleRange Test (DNMRT) p<0,05, there was significant difference between 100% shallot’s water extortion with othertreatments, but there was no significant difference between 50% shallot’s water extortion with 200% shallot’s. Theconclusion was shallot’s water extortion had antifungal activity againsts Candida albicans with the best concentration100%, but it was lower than ketoconazole 2%.

<p><strong><em>Background:</em></strong><em> Orthodontic tooth movement is a </em><em>interaction prosess</em><em> of resorption and deposition of bone remodeling. Orthodontic tooth movement by mechanical strength causes changes in alveolar bone. Osteocyte is an essential cell to respond bone remodelling. Hyperbaric Oxygen Therapy affects production of osteocyte because it can release Reactive Oxygen Species (ROS) and Nitrid Oxide (NO).  <strong>Purpose: </strong>To determine the difference number  of osteocyte in pressure and tension area during tooth movement by adjuvant of Hyperbaric Oxygen 2,4 ATA during 7 days starting on day 8 to day 14. <strong>Materials and Methods</strong>: This research used Completery Randomized Control Group Post Test Only Design. 36 cavia cobaya (male)  were divided into 3 groups randomly : the negative control groups, positive control group, and treatment group. Preparat staining used Hematoxylin Eosin (HE) and calculated on microscop 1000x with 20 field of view. Data analyses used one way ANOVA and LSD test then compared each area by using paired T test. <strong>Result:</strong> The data showed that the treatment group (P=10,67) tension area has the highest number of osteocyte than  negative control group (K-=3,67), positive control (K+=7,42).
