Background: Traditional Siddha medicine literature claims that the Amaranthus spinosus Linn. Whole plant possesses diuretic property. The aim of the present study is to investigate the diuretic activity of aqueous extract of amaranthus spinosus linn on rats.Methods: The rats were randomly divided into four groups of six animals each and deprived of food for 18 hours before the experiments. Control (Group I) rats were treated with Normal saline (5ml/Kg,) and Standard (Group II) rats were administered furosemide 15mg/Kg, I.P. Group III and IV were treated with Aqueous extract of Amaranthus spinosus Linn (AEAS) 250 and 500 mg/kg in normal saline respectively, The urine was collected after 5 and 24 hours. The urine volume and the total concentration of Na+, K+, and Cl- in the urine were estimated. The data were presented as mean ± standard deviation and analyzed using one-way analysis of variance and Student's "t"-test. P <0.05 was considered statistically significant. Conventional windows software was used for statistical analysis.Results: Urine analysis showed that Na+, K+, and Cl- levels were significantly (p<0.05) increased with AEAS 250 and 500 mg/kg as rats treated with furosemide 15mg/kg. Urine volume is significantly (p<0.05) increased in rats treated with drugs (AEAS 500 mg/kg and furosemide 15mg/kg) compared to normal saline 25ml/Kg, treated rats at 5 and 24 hours.Conclusions: These data suggest that AEAS has produced notable diuretic effect which appeared to be comparable to that produced by the standard diuretic furosemide.

The present study was undertaken to establish the diuretic activity of alcoholic and aqueous extract of dried rhizomes of Curcuma amada Roxb in rats. Alcoholic and aqueous extracts of rhizomes were administered to experimental male Wistar rats orally at doses of 250 and 500 mg/kg and compared with furosemide (10 mg/kg) as the reference standard. The parameters measured for diuretic activity were total urine volume, urine electrolyte concentration such as sodium, potassium and chloride have been evaluated. The rats treated with alcoholic and aqueous extract of Curcuma amada in a dose of 250 and 500 mg/kg showed higher urine output when compared to the respective control. Both alcoholic and aqueous extracts have showed a significant dose-dependent increase in the excretion of electrolytes when compared to the control group. The result indicates that alcoholic and aqueous extract is an effective natriuretic and kaliuretic diuretic, which supports the traditional claim about the Curcuma amada Roxb being used as diuretics.

Background: Sphaeranthus indicus Linn (S. indicus Linn) is a medicinal plant widely used in Indian traditional system of medicine for treating various ailments. The aim of the study was to evaluate diuretic activity by Lipschitz method in albino rats.Methods: Thirty albino rats were grouped into five groups and each contained six rats. Group I (control group) received 25ml/kg each of 0.9% sodium chloride orally. Group II (standard) received Hydrochlorothiazide 25 mg/kg orally dissolved in isotonic normal saline. Groups III, IV and V received ethanolic extracts of S. indicus Linn in doses 100mg/kg, 200mg/kg and 300mg/kg respectively dissolved in isotonic saline orally. The mice were put in metabolic cages and urine samples were collected for all the groups up to 24 hours after dosing. Urine was analysed for volume, urinary excretion ratio, diuretic activity, sodium and potassium composition electrolytes. Data was analysed using SPSS version 16 for windows.Results: Single dose administration of S. indicus in doses of 100, 200 and 300 mg/Kg as compared to Hydrochlorothiazide (25 mg/kg)have significantly (p<0.001, ANOVA) increased total urine output along with an increase in concentration of sodium and potassium. S. indicus Linn 300 mg/Kg produced greater diuretic activity, which is comparable to the effect of standard.Conclusions: The extract treated rats showed a dose dependent increase in urine volume. Maximum diuretic and kaliuretic activity were observed at the dose of 300 mg/kg of the extract. The present study supports and confirms the basis for folklore use of S. indicus Linn as a diuretic agent.

Aim: To Compare frequency of sore throat in early post operative period among patients undergoing general anaesthesia and endotracheal intubation for abdominal surgeries who are given dexamethasone and normal saline. Study Design: Randomized controlled study Setting: Department of Anesthesia/ ICU, Sheikh Zayed Hospital, Lahore Duration of study: Six months i.e. 25-09-2009 to 25-03-2010. Methodology: 120 patients undergoing elective general surgery on abdomen were selected. They were divided into two groups. Group I received dexamethasone 8mg (2ml) I/V pre-operatively and group II received 2ml normal saline I/V pre-operatively. Chi square test was used. Visual analogue (VAS) scale was used for recording sore throat. The VAS score ≤4 was considered as no sore throat and VAS scores>4 were considered as the sore throat. Results: Frequency of post-operative sore throat after the first 24 hours following GA and endotracheal intubation was lower in group (I) as compared to the control group (II). Eleven (20%) patients with dexamethasone had post-operative sore throat compared to thirty one (56.3%) patients in control group. (p<0.01). Conclusion: Pre-operative use of dexamethasone was associated with decreased incidence of post-operative sore throat. Abstract Nephrolithiasis appear to be a major health crisis among the population with serious medical related consequences throughout the lifetime of patient. The aim of the study was to evaluate the preventive effect of the hydro-alcoholic extract of A. lanata roots on Urolithiasis rats. Thirty adults male wistar Albino rats weighing 200 – 250 g were divided into five groups comprising 6 rats in each. Group I Served as positive control with water ad libitum. Group II as negative control which is disease treated group receiving 0.75% ethylene glycol mixed with drinking water for 28 days. Group III chosen as standard group receiving ethylene glycol for first 14 days and Cystone 750 mg/Kg from day 15 till day 28. Group IV and V received ethylene glycol for first 14 days and treatment regimen of LD (400 mg/Kg) and HD 800 mg/Kg orally from day 15 till day 28. Invitro studies like Nucleation, Aggregation and Growth assays were performed. Urine volume and pH was collected and observed for change in appearance, pH, odour and turbidity. Extract was given by preparing suspension and stability was observed by measuring its parameters. On Day 29, the kidneys were dissected and histopathology was done to check tubular injury. There was Increase in urine volume, enhanced excretion of urinary constituents like citrate, oxalate etc. and improving clearance rate. Improvement in pH and antioxidant activity was observed in treated groups. The extract showed that it has prominent effect on nephrolithiasis and has better safety profile in the dose given.

To evaluate the effect of Aqueous extract of Terminalia belliricafruit pulp (AETB) on learning by Hebb William maze model in mice with acute alcohol consumption.Swiss albino mice (n=48) of either sex weighing 20-30g will be divided into eight groups of six mice each. Drugs were given orally after 12 hours of fasting. Group I mice received 10ml/kg of Normal Saline, Group II mice received Piracetam 200mg/kg, Group III received AETB 36mg/kg, Group IV received ethanol 1.5g/kg orally, Group V received ethanol(1.5g/kg )+ piracetam (200mg/kg), Group VI mice received ethanol(1.5g/kg) +AETB(9mg/kg), Group VII mice received ethanol(1.5g/kg) +AETB (18mg/kg), Group VIII mice received ethanol(1.5g/kg) +AETB(36mg/kg). Time taken by the animal to reach the reward chamber from the start chamber (TRC) in Hebb-William maze was used as a parameterto evaluate the learning.Acute alcohol administration showed increase in TRC. Whereas, acute administration of Aqueous extracts of Terminalia belliricafruit pulp showed a decrease in TRC when compared to the control group. The TRC values for the groups that were administered AETB along with acute alcohol administration showed decrease in TRC values compared to the negative control.Current study showed acute alcohol administration caused impairment of thelearning ability in mice. Whereas, acute administration of Aqueous extracts of Terminalia belliricafruit pulp (AETB)caused enhancement of learning. Pre-treatment with AETB before acute alcohol administration indicated protective action of AETB on alcohol affected learning in mice.

Objective: Present study was undertaken to investigate the nephroprotective activity of whole plant ethanol extract of Biophytum sensitivum Linn. DC (B. sensitivum; EEBS) on gentamicin induced-nephrotoxicity in Wistar albino rats.Methods: Animals were divided into five groups, containing six animals in each. Gentamicin (GM) 100 mg/kg/d; i. p., was given to all groups except normal control to induce nephrotoxicity for a period of 8 d in rats. Animals in Group I served as control and Group II as GM-treated or nephrotoxic control. Group III received standard quercetin (50 mg/kg; p. o.); group IV received EEBS (250 mg/kg; p. o.) and group V received EEBS (500 mg/kg; p. o.), for 8 d. Several renal functional tests and injury markers such as a change in body weight, water intake, urine volume and pH, urinary levels of total protein, albumin, sodium, potassium, calcium and magnesium were measured. Different hematological parameters including, red blood cells (RBC), hemoglobin (Hb), packed cell volume (PCV), mean corpuscular haemoglobin (MCH), white blood corpuscles (WBC), lymphocyte, monocyte, polymorphs and eosinophil were also analyzed.Results: The results revealed that co-administration of EEBS at 250 and 500 mg/kg significantly reduced the urinary excretion of total protein (4.05±0.04, 3.53±0.05 g/dl), albumin (0.81±0.08, 0.71±0.00 g/dl), calcium (8.05±0.05, 7.81±0.06 mg/dl) and magnesium (0.79±0.05, 0.77±0.06 mg/dl) respectively. EEBS treatment in group IV and V animals produced a significant increase in body weight (2.58±0.17, 2.52±0.22 g), water intake (17.35±0.82, 17.87±0.86 ml/24 h) urine output (7.24±0.15, 7.88±0.08 ml/24 h), compared to animals in group II. Co-therapy with both doses of extracts also showed significant increase in RBC (6.85±0.31, 7.10±0.43 million/mm3) Hb (13.96±0.21, 13.83±0.31 g/dl), MCH (20.45±0.49, 20.67±0.51 pg) and PCV (38.29±0.42, 38.53±0.20%) levels, whereas WBC (8.24±0.21, 8.15±0.10 1X103/mm3) and polymorphs (14.19±0.20, 14.05±0.11%) levels were found to be significantly reduced respectively in animals than those in toxic control group.Conclusion: This study substantiated and confirmed the ethnomedical usefulness of B. sensitivum as a nephroprotective and antioxidant agent.

 Objective: The current study was designed to estimate the potential protective role of the aqueous extract of rosemary (AER) (Rosmarinus officinalis) against trichloroacetic acid (TCA)-created hepatotoxicity in male albino rats.Methods: Forty male albino rats were separated into four groups of ten: Group I served as control; Group II was given AER (200 mg/kg/day) by gavage; Group III received TCA at the dose 50 mg/kg/day, and Group V was treated with AER (200 mg/kg/day) and received TCA (50 mg/kg/day). The experiment was carried out for 2 months.Results: The toxicity of TCA for rats was revealed by an elevation in liver marker enzymes activities (gamma-glutamyl transferase [GGT], alkaline phosphatase [ALP], aspartate transaminase [AST], alanine aminotransferase [ALT]) and conjugated bilirubin (CB) level, and a decrease in albumin and total protein (TP) levels. The TCA administration also caused a significant increase in the activities of catalase (CAT), glutathione peroxidase (GPx) and superoxide dismutase (SOD), and also malondialdehyde (MDA) level in liver tissues. These biochemical effects were accompanied by histological indicators of liver damage. Treatment with ARE recovered the liver damage instigated by TCA, as showed by perfection of liver enzyme markers (GGT, ALT, AST, ALP), CB, TP and albumin; as well as antioxidant parameters (CAT, SOD, GPx) and lipid peroxidation (MDA) and amelioration of histopathology changes in the liver tissues.Conclusion: It could be concluded that AER supplementation for 2 months in TCA-induced toxicity in rats benefited hepatic antioxidant status and improved liver injury and damage in male albino rats exposed to TCA.

Doxorubicin (DOX) is one of the most potent and widely used chemotherapeutic agents to treat several malignancies. However, the clinical use of DOX is seriously restricted due to its acute and chronic cardiotoxic side effects This study investigated the protective effect of (Ajwa) date aqueous extract (AJDAE) against doxorubicin-induced cardiotoxicity in rats. Sixty Wister albino male rats (150-200 gms.) were comprised in our study and divided into six equal groups: group I (untreated control), group II, group III, rats were orally received AJDAE (0.75 & 1.5 gm/ kg.bw) respectively, for 4 weeks, rats of groups IV, V and VI were intraperitoneally injected with one dose of doxorubicin (5 mg/kg.bw) at the end of the 4th week of the study to induce cardiotoxicity, rats of groups V & VI were orally received AJDAE (0.75 & 1.5 gm/ kg.bw) respectively. Cardiac enzymes, lipid profile, SOD, GR, GST, GPx, CAT and MDA in rats’ hearts homogenate, urinary 8OHdG as well as DNA integrity and histopathological changes were investigated in all studied rats.Oral administration of AJDAE (0.75 & 1.5 gm/ kg.bw) attenuated the cardiotoxicity of DOX, improved the cardiac enzymes, lipid profile, reduced the urinary 8OHdG and prohibited the depletion of endogenous antioxidants and suppressed lipid peroxidation (MDA). Moreover, AJDAE enhanced DNA integrity. Histological findings showed that AJDAE (0.75 & 1.5 gm/ kg.bw) administration reduced cardiomyocytes alterations, congestion, edema and the intense cellular stress exerted on myocardial fibers as well as restored the cardiomyocytes architecture. Our data showed that AJDAE obviously resulted in protective effects against DOX-induced cardiotoxicity in rat’s heart. It can be concluded that Ajwa date offers a considerable protection against DOX-induced cardiotoxicity.

INTRODUCTION Diabetes mellitus is the most common metabolic disease worldwide. Nigella sativa also known as black cumin, is an emerging miracle herb with a rich historical and religious background. In the present study, we have attempted to study the hypoglycemic effect of aqueous extract of Nigella sativa on diabetes-induced rats. MATERIAL AND METHODS This preclinical study was conducted in Department of Pharmacology from August to October 2019. In this study, hyperglycemia was induced by a single intraperitoneal dose of 60 mg/kg streptozotocin in 0.1 M citrate buffer (pH 4.5). Animals were divided into five groups containing normal control, negative control, standard, test-1, and test-2. The aqueous extract of Nigella sativa was administered 400 and 800 mg/kg per oral in both the test groups for 18 days. Blood glucose level was measured at three-days interval by glucometer. Oral glucose tolerance test was done by administrating 2 gm/kg per oral glucose and the blood glucose was recorded every two-hours interval for eight hours. RESULTS An oral glucose tolerance test showed a decrease in blood glucose level at the dose of 400 and 800 mg/kg compared to the negative control group. The administration of aqueous extract of Nigella sativa for 18 days showed significant decrease in blood glucose level (p=0.001) compared to negative control group. The blood glucose level in test 1 (400 mg/kg) was significantly reduced compared to the standard group (p***<0.001). The blood glucose level in test 2 at 18th day was less significant (p*< 0.05) compared with standard group and test 1 (p**<0.01, p***<0.001). CONCLUSION The findings of the present study indicate that the aqueous extract of Nigella sativa has significant hypoglycemic effect in Wistar rats.
