Abstract Determination of ileal endogenous amino acids (IEAAs) is necessary for the calculation of standardized ileal amino acid digestibility. This experiment was conducted to compare the response of amino acids composition of IEAAs of broilers, and digestive physiology fed the nitrogen-free diet (NFD) formulated with different ratios of dextrose to corn starch (D/CS). 28d-old broiler chickens (n = 210) with similar body weight were allocated to 5 treatment groups, including a control group (CT, basal diet, normal level of protein) and four NFD groups for a 3-days trial, designated as A (D/CS = 1.00), B (D/CS = 0.60), C (D/CS = 0.33), and D (D/CS = 0.14). The results showed that NFD significantly reduced serum IGF-1, albumin and uric acid levels when compared with the control (P < 0.05). A higher ratio of D/CS (1.00 and 0.60) increased Asp, Thr, Ser, Glu, Gly, Ala, Val, Ile, Leu, His, Tyr, Arg, and Pro contents of IEAAs when compared with the ratio of 0.33 and 0.14 (P < 0.05). Moreover, ileal DM digestibility and digestive enzyme increased with an increasing ratio of dextrose to corn starch (P < 0.001). The number of ileal goblet cells and the gene expression of Mucin 2 were higher in group A (D/CS=1.00) than in group C (D/CS = 0.33) and the control (P < 0.05). It was further observed that NFD indeed reshaped the gut microbiota, characterized by lower Bacteroidetes, a significantly increased proportion of Proteobacteria, and decreased microbial diversity (P < 0.05).Our results indicate that the chicken fed NFD were accompanied by huge digestive physiological alterations, presenting with malnutrition and accumulation of Proteobacteria in the gut. Different proportions of dextrose and starch directly affect the basal IEAAs of broiler chickens. A higher proportion of dextrose (D/CS = 1 and 0.6) in NFD increase IEAAs by promoting secretion of digestive enzyme and mucin. But the excessive proportion of starch is unsuitable for the chicken to digest NFD (D/CS = 0.14). Therefore, we suggest the ratio of dextrose to corn starch in NFD at 0.33 might be more appropriate to detect IEAAs of broiler chickens.

Скрябина и ФГБНУ ФНЦ ВИЭВ РАН, аминокислотный состав мяса перепелов в Государственном бюджетном учреждении Краснодарского края Кропоткинская краевая ветеринарная лаборатория . Из цыплят в суточном возрасте кросса Кобб500 было сформировано три группы по 10 голов в каждой две опытные и контрольная опытным группам выпаивали кормовые добавки из расчета 1 мл/кг живой массы птицы до 50 суток выращивания, убой птицы проводили на 56 сутки. Тушки птицы после 24х часов созревания в холодильной камере при температуре 4 С подвергали исследованиям по общепринятым методикам: ГОСТ Р 519442002. Мясо птицы. Методы органолептических показателей, температуры и массы ГОСТ 314702012. Мясо птицы, субпродукты и полуфабрикаты из мяса птицы. Методы органолептических и физикохимических исследований аминокислотный состав мяса исследовали согласно М 0438 2009. Корма, комбикорма и сырье для их производства. Методика измерений массовой доли аминокислот методом капиллярного электрофореза с использованием системы капиллярного электрофореза Капель. Применение кормовых добавок в дозе 1 мл/кг живой массы способствовало увеличению живой массы птицы при применении Абиотоника на 54,23, а при применении Чиктоника на 37,70 по отношению к контролю. Во всех исследуемых пробах количество ЛЖК находится в пределах нормы и составило: в 1й опытной 1,390,03 мг КОН, во 2й опытной 1,420,04 мг КОН и в контрольной группе 1,810,06 мг КОН, что говорит о свежести и доброкачественности мяса. Значение рН мяса цыплятбройлеров находилось в трех группах в пределах нормы и не превышало 6,0. Abstract A digestibility experiment was conducted to evaluate the effects of dietary exogenous monocomponent protease on the coefficient of apparent total tract digestibility (ATTD) and coefficient apparent ileal digestibility (AID) of amino acids in meat and bones meal (MBM), poultry byproducts meal (PBPM), and feather meal (FM). A total of 512 Cobb-500 male broiler chickens (aged 14 d) were randomly placed into 64 metabolism cages (8 birds per pen) and were allocated to eight treatments with eight replicates in a semi-controlled environmental room. The experimental diets consisted of the basal diet (corn/SBM) and the replacement of 300 g/kg on a weight basis with MBM, PBPM, or FM. The excreta were collected during 3 d (19 to 21 d), and the ileal digesta (using Celite as an indigestible marker) only 1 d (21 d). The protease contained 75,000 PROT units/g. The use of the enzyme increased (P &lt; 0.05) ATTD for alanine, cysteine, glycine, and threonine in the basal diet and AID for the amino acids alanine, cysteine, glycine, lysine, threonine, and valine for the basal diet (vegetable). Regarding meals, there was an increase (P &lt; 0.05) in the amino acid digestibility in large part due to the amino acids of MBM (14) and PBPM (9), with only five amino acids for FM. The amino acids glycine and threonine showed increases (P &lt; 0.05) in both total and ileal digestibility for all animal meals and for the diet based on corn and soybean meal, which indicates a high specificity of the enzyme for these amino acids. The overall results obtained in this study found satisfactory effectiveness of this exogenous protease. The total collection method was lower by 1.83% of amino acids digestibility than the ileal method.

The influence of method (direct vs difference method) used to determine the apparent ileal digestibility coefficient (AIDC) of amino acids in two cereals (maize and wheat) and two grain legumes (Australian sweet lupins and peas) was investigated. For the direct method, the test ingredients were incorporated as the sole source of protein in assay diets. The assay diets used in the difference method were formulated by substituting the cereals and legumes for 50 and 25% (w/w), respectively, of a maize-soy basal diet. Each diet contained 3 g/kg titanium dioxide as an indigestible marker and were offered ad libitum to four replicate cages of broilers (four birds/cage) from d 28 to d 35 post-hatching. On d 35, digesta were collected from the terminal ileum and the AIDC of amino acids were calculated using marker ratios in the diet and digesta. Except for histidine, methionine, cysteine and tyrosine, the main effect of methods was found to be significant (P<0.05 to 0.001) for the AIDC of all amino acids. The main effect of the ingredients was significant (P<0.05 to 0.001) for all amino acids, except for histidine, aspartic acid and serine. Interactions (P<0.05) between ingredients and method were observed only for leucine, lysine, proline and serine. The AIDC of amino acids, determined by the difference method, was found to be distinctly higher than those determined by the direct method. This suggested that the direct method underestimated amino acid digestibility in low and medium protein ingredients.

Amino acids are known to play a key role in gene expression regulation. Amino acid signaling is mediated via two pathways: the mammalian target of rapamycin complex 1 (mTORC1) and the amino acid responsive (AAR) pathways. Cationic amino acid transporters (CATs) are crucial in these pathways due to their sensing, signaling and transport functions. The availability of certain amino acids plays a key role in the intake of other amino acids, hence affecting growth in young birds. However, the specific mechanism for regulating lysine transport for growth is not clear. In this study, we analyze the transcriptome profiles and mRNA expression of selected cationic amino acid transporters in the livers of broilers fed low and high lysine diets. Birds consumed high-lysine (1.42% lysine) or low-lysine (0.85% lysine) diets while the control group consumed 1.14% lysine diet. These concentrations of lysine represent 125% (high lysine), 75% (low lysine) and 100% (control), respectively, of the National Research Council’s (NRC) recommendation for broiler chickens. After comparing the two groups, 210 differentially expressed genes (DEGs) were identified (fold change >1 and false discovery rate (FDR) <0.05). When comparing the high lysine and the low lysine treatments, there were 67 upregulated genes and 143 downregulated genes among these DEGs. Analysis of Kyoto Encyclopedia of Genes and Genomes (KEGG) and the Gene Ontology (GO) enrichment analysis show that cellular growth, lipid metabolism and lysine metabolism pathways were among the significantly enriched pathways. This study contributes to a better understanding of the potential molecular mechanisms underlying the correlation between lysine intake, body weight gain (BWG) and feed intake (FI) in broiler chickens. Moreover, the DEGs obtained in this study may be used as potential candidate genes for further investigation of broiler growth customized responses to individualized nutrients such as amino acids.

Guanidinoacetic acid (GA) is a natural precursor of creatine in the body and is usually used to improve the feed conversion and cellular energy metabolism of broiler chickens. The objective was to elucidate the effect of dietary supplementation of GA on carcass yield, muscle amino acid profile, and concentrations of brain neurotransmitters in laying hens. In total, 128 72-week-old ISA Brown laying hens were assigned to four equal groups (32 birds, eight replicates per group). The control group (T1) was fed a basal diet with no supplements, while the other experimental groups were fed a basal diet supplemented with 0.5 (T2), 1.0 (T3), and 1.5 (T4) g GA kg−1 diet. The T3 and T4 groups showed higher hen-day egg production and carcass yield compared to the control group (p = 0.016 and 0.039, respectively). The serum creatine level increased linearly with the increased level of dietary GA (p = 0.007). Among the essential amino acids of breast muscle, a GA-supplemented diet linearly increased the levels of leucine, isoleucine, phenylalanine, methionine, and threonine in the breast (p = 0.003, 0.047, 0.001, 0.001, and 0.015, respectively) and thigh (p = 0.026, 0.001, 0.020, 0.009, and 0.028, respectively) muscles. GA supplementation linearly reduced the level of brain serotonin compared to the control group (p = 0.010). Furthermore, supplementation of GA in the diet of laying hens linearly increased the level of brain dopamine (p = 0.011), but reduced the level of brain Gamma-aminobutyric acid (p = 0.027). Meanwhile, the concentration of brain nitric oxide did not differ between the experimental groups (p = 0.080). In conclusion, the dietary supplementation of GA may improve the carcass yield and levels of essential amino acids in the breast muscles, as well as the brain neurotransmitters in aged laying hens.

Aim and Objective:: To determine the mechanisms present in the etiopathogenesis of nasal polyposis. It is not clear whether amino acids contribute in a causal way to the development of the disease. Therefore, the aim of this study was to determine the plasma-free amino acid profile in patients with nasal polyposis and to compare the results with a healthy control group. Materials and Methods:: This was a prospective controlled study that took place in the Otolaryngology Department at the Harran University Faculty of Medicine between April 2017 and April 2018. Plasmafree amino acid profile levels were studied in serum samples taken from a patient group and a healthy control group. Patients who were diagnosed with bilateral diffuse nasal polyposis and were scheduled for surgical interventions were included in this study. Individuals whose age, gender, and body mass index values were compatible with that of the patient group and who did not have any health problems were included in the control group. All the participants whose levels of plasma-free amino acid were thought to be affected by one or more of the following factors were excluded from the study: smoking and alcohol use, allergic rhinitis presence, the presence of acute or chronic sinusitis, a history of endoscopic sinus surgery, unilateral nasal masses, a history of chronic drug use, systemic or topical steroid use in the last three months for any reason, and liver, kidney, hematological, cardiovascular, metabolic, neurological, or psychiatric disorders or malignancies. Conclusion: In this study, plasma levels of 15 free amino acids were significantly higher in the nasal polyposis group than in the healthy control group. A plasma level of 1 free amino acid was found to be significantly lower in the nasal polyposis group compared to the healthy control group. Therefore, it is important to determine the possibility of using the information obtained to prevent the recurrence of the condition and to develop effective treatment strategies. This study may be a milestone for studies of this subject. However, this study needs to be confirmed by further studies conducted in a larger series.

AbstractThe prime purpose of this review is to explore the pathways whereby progress towards reduced-crude protein (CP) diets and sustainable chicken-meat production may be best achieved. Reduced-CP broiler diets have the potential to attenuate environmental pollution from nitrogen and ammonia emissions; moreover, they have the capacity to diminish the global chicken-meat industry’s dependence on soybean meal to tangible extents. The variable impacts of reduced-CP broiler diets on apparent amino acid digestibility coefficients are addressed. The more accurate identification of amino acid requirements for broiler chickens offered reduced-CP diets is essential as this would diminish amino acid imbalances and the deamination of surplus amino acids. Deamination of amino acids increases the synthesis and excretion of uric acid for which there is a requirement for glycine, this emphasises the value of so-called “non-essential” amino acids. Starch digestive dynamics and their possible impact of glucose on pancreatic secretions of insulin are discussed, although the functions of insulin in avian species require clarification. Maize is probably a superior feed grain to wheat as the basis of reduced-CP diets; if so, the identification of the underlying reasons for this difference should be instructive. Moderating increases in starch concentrations and condensing dietary starch:protein ratios in reduced-CP diets may prove to be advantageous as expanding ratios appear to be aligned to inferior broiler performance. Threonine is specifically examined because elevated free threonine plasma concentrations in birds offered reduced-CP diets may be indicative of compromised performance. If progress in these directions can be realised, then the prospects of reduced-CP diets contributing to sustainable chicken-meat production are promising.

AbstractThe present study was conducted to investigate effects of Bacillus subtilis on growth performance, serum parameters, digestive enzymes, intestinal morphology, and colonic microbiota in piglets. A total of 72 piglets were weighed and randomly allotted into three treatments (four replication pens per treatment with six piglets/pen) for a 28-day experiment. The dietary treatments were as follows: basal diet (control group, CTR), basal diet supplementation with antibiotic (antibiotic group, ABT), and basal diet supplementation with 0.1% Bacillus subtilis (probiotic group, PBT). The average daily gain of body weight increased in both the ABT and PBT groups, and dietary antibiotics decreased the feed:gain ratio (F:G), as compared to the CTR group (P < 0.05). Both ABT and PBT piglets had increased serum triglycerides and lipase, amylase, maltase activities and villus height:crypt depth ratio (V/C) in ileum (P < 0.05). The PBT group also showed an increase in serum glucose and villus height in the ileum (P < 0.05). Dietary antibiotics increased Lactobacillus johnsonii, as compared to the CTR group, but decreased bacterial diversity and increased Escherichia coli, as compared to the PBT group (P < 0.05). Piglets dietary with B. subtilis modulated the microbiota by increasing the abundance of Firmicutes (L. johnsonii, L. reuteri) and decreasing the abundance of E. coli, as compared to the control group (P < 0.05). These results indicate that dietary of B. subtilis improves growth performance and intestinal health and can be a promising alternative to antibiotics in piglets diet.

AbstractCa2+-sensing receptor (CaSR) represents a potential therapeutic target for inflammatory bowel diseases and strongly prefers aromatic amino acid ligands. We investigated the regulatory effects of dietary supplementation with aromatic amino acids – tryptophan, phenylalanine and tyrosine (TPT) – on the CaSR signalling pathway and intestinal inflammatory response. The in vivo study was conducted with weanling piglets using a 2 × 2 factorial arrangement in a randomised complete block design. Piglets were fed a basal diet or a basal diet supplemented with TPT and with or without inflammatory challenge. The in vitro study was performed in porcine intestinal epithelial cell line to investigate the effects of TPT on inflammatory response using NPS-2143 to inhibit CaSR. Dietary supplementation of TPT alleviated histopathological injury and decreased myeloperoxidase activity in intestine challenged with lipopolysaccharide. Dietary supplementation of TPT decreased serum concentration of pro-inflammatory cytokines (IL-1β, IL-6, IL-8, IL-12, granulocyte-macrophage colony-stimulating factor, TNF-α), as well as the mRNA abundances of pro-inflammatory cytokines in intestine but enhanced anti-inflammatory cytokines IL-4 and transforming growth factor-β mRNA levels compared with pigs fed control diet and infected by lipopolysaccharide. Supplementation of TPT increased CaSR and phospholipase Cβ2 protein levels, but decreased inhibitor of NF-κB kinase α/β and inhibitor of NF-κB (IκB) protein levels in the lipopolysaccharide-challenged piglets. When the CaSR signalling pathway was blocked by NPS-2143, supplementation of TPT decreased the CaSR protein level, but enhanced phosphorylated NF-κB and IκB levels in IPEC-J2 cells. To conclude, supplementation of aromatic amino acids alleviated intestinal inflammation as mediated through the CaSR signalling pathway.

Studies were carried out with 12 pigs (Yorkshire × Landrace) to determine the effect of dietary fibre level on amino acid digestibility. The pigs were weaned at 21 d of age and fitted with a simple T-cannula at the distal ileum at 23 or 24 d of age. The pigs were fed four isonitrogenous corn starch-based soybean meal diets (22.5 % crude protein) containing 4.3, 7.3, 10.3 and 13.3% Solkafloc according to a balanced two-period change-over design. The pigs were fed four times daily, equal amounts, at 6-h intervals. The diets were supplied at a rate of 5% of the average body weight which was determined at the initiation of the first (8.8 kg) and second (11.7 kg) experimental period. Each experimental period comprised 10 d. Feces were collected for 48 h on days 6 and 7 and ileal digesta for 24 h during days 8, 9 and 10. Chromic oxide was used as digestibility marker. The dietary inclusion of Solkafloc, up to 13.3 %, did not affect (P > 0.05) the apparent ileal as well as the fecal digestibilities of crude protein and amino acids. In conclusion, starter pigs can tolerate up to 13.3% Solkafloc, equivalent to 16.8% neutral-detergent fibre, in corn starch-based soybean meal diets without a depression in amino acid digestibility.
