Abstract Objective: The rabbit VX2 bone tumor model is an ideal experimental animal model for studying malignant bone tumors, but few studies on cytokines and tumor proliferation. This paper is aimed to study the effect of interferon-γ (IFN-γ) and interleukin-4 (IL-4) on the expression of proliferating cell nuclear antigen (PCNA) in tumor tissue. Methods: The experiment included 30 Japanese white rabbits divided into an experimental group (n=15) and a control group (n=15) according to the random number method. Peripheral blood and tumor tissue were collected at 1 and 2 weeks after tumor implantation. We used enzyme-linked immunosorbent assay to detect the expression levels of IFN-γ and IL-4 in peripheral blood and PCNA in tumor tissue. After the rabbits were euthanasia, the tissues were taken for pathological examination. Results: After tumor implantation, the expression level of IFN-γ in the experimental group in the 2nd week was significantly lower than that in the 1st week and 2nd week in the control group (p-value<0.005). In contrast, the expression level of IL-4 in the experimental group in the 2nd week was significantly higher than that in the 1st week and 2nd week in the control group (p-value<0.005). The expression level of PCNA in the experimental group in the 2nd week was significantly higher than that in the 1st week (P<0.005), and the PCNA expression in the 1st and 2nd week in the experimental group was higher than that in the 1st and 2nd week in the control group (p-value<0.005). In the 1st and 2nd week of tumor implantation, the correlation between IFN-γ and PCNA was negatively correlated(r=-0.566, p-value=0.028; r=-0.604, p-value=0.017), while the correlation between IL-4 and PCNA was positively correlated(r=0.583, p-value=0.023; r=0.884, p-value<0.005). Conclusion: In the VX2 bone tumor model of rabbit, with the extension of time after tumor implantation, there was a negative correlation between IFN-γ and PCNA and a positive correlation between IL-4 and PCNA.

Tuberculosis (TB) is still one of the major health problems in the community. Micronutrients, including iron, are crucial in the body's defense mechanism. Iron modulates the activation of IFN-γ, which in turn will activate the macrophages. This study explores the effect of iron supplementation given with anti-tuberculosis drug therapy on the IFN-γ level in tuberculosis spondylitis patients. This was a single-blind randomized control trial study at Dr. Hasan Sadikin General Hospital Bandung, from December 2018 to March 2019, using the consecutive sampling approach. Thirty-four respondents with category 1 TB spondylitis and an sTfR level of> 21.7 pg /µL were recruited and randomized into control and experimental groups. The control group (n=17) was treated with 2 RHZE for 2 months while the experimental group (n=17) received 2 RHZE and iron tablet for 2 months. An increase in the IFN-γ level of the experimental group was observed from an average of 21.7 pg/ml before therapy to an average of 72.39 pg/mL after therapy. Meanwhile, the IFN-γ level in the control group increases from an average of 22.5 pg/mL to an average of 35.35 pg/mL. This difference was statistically significant based on the analysis using the independent t-test (p-value <0.05). Therefore, the administration of anti-tuberculosis drugs with the addition of iron can increase the body's immune response, which is indicated by the increasing level of IFN-γ.

Anxiety disorder on of the most common illnesses in the context of psychiatry. Potential causes include genetic and environmental factors, as well as the parental attachment of the individuals. The purpose of this study is to examine the relationship between parental attachment style and anxiety disorders for a group of children and their parents. Study data were collected from the mothers (N = 40) of children with an anxiety disorder who visited a child psychiatry outpatient clinic at a city hospital and a private institution in Kayseri (Turkey) in 2018. For the control group, 40 mothers of children without any mental illness were also included in the study. The purposive sampling method was used in the selection of the participants for both groups (experimental and control). Sociodemographic data sheet and parental bonding instrument (PBI) were utilized as the data collection instruments. Then, data were analyzed based on the descriptive analysis methodology that included mean scores, standard deviation, p-value, t-experimental, two-way ANOVA and Pearson correlation experiments by using SPSS v.22. The findings revealed that the mothers of the participants with a college degree in the experimental group had fewer perceptions of protection (t = 2.38, p < 0.01), but more perception of care from their mothers than fathers (t =−2.28, p < 0.05). In addition, although the perceived care of parents was found lower than the participants in the control group, the participants in both groups evaluated their parents analogously for overprotection. Findings showed that the mothers in the experimental group predominantly described their parents as neglecting.

Asthma is a chronic disease that affects the airways in the lungs and is common in many countries. Studies have shown that vitamin D levels are associated with the onset and exacerbation of asthma symptoms. Given the role of vitamin D in regulating immune responses and the importance of the two major cytokines IL-4 (an inflammatory cytokine) and IL-10 (an anti-inflammatory cytokine) in immunological processes, in this study, the relationship between vitamin D intake and the level of expression of these ILs in asthmatic mice was investigated. In the case-control study performed in three groups of mice, 10 experimental asthma mice, 10 asthmatic vitamin D-treated mice, and 10 healthy mice as the control group were studied. For analyses, after RNA extraction and cDNA synthesis, the expression level of two IL-4 and IL-10 genes was evaluated by real-time PCR. Finally, the results were analyzed by SPSS software. The results showed that in the control group, the expression level of IL-4 and IL-10 genes was 0.01 and 0.02, respectively. Accordingly, the expression level of the IL-4 gene in asthmatic mice and asthmatic mice treated with vitamin D was 0.6 and 0.2, respectively, while the expression level of the IL-10 in these groups was 1.2 and 2.8, respectively, which showed significant changes (p-value < 0.05) in treated mice compared to the asthmatic mice without treatment. Based on results, in asthmatic mice treated with vitamin D, a significant increase in IL-10 expression was observed (1.2 to 2.8), while IL-4 expression was decreased from 0.6 to 0.2. It appears that vitamin D supplementation in asthmatic patients by affecting T-cell maturation and increasing anti-inflammatory IL-10 and suppressing IL-4 may play a role in reducing the symptoms of asthma.
