Objective: The objective of the study was to investigate the effect of dietary flaxseed-derived lignan supplement as anti-hyperlipidemic (fatty liver changes) on rabbit liver.Methods: Rabbits were assigned randomly into three groups (five rabbits for each group): group-1, normal diet (negative control); group-2, 1.6 % cholesterol diet for 30 d (positive control) and group-3, 1.6 % cholesterol for 30 d then gave 40 mg/kg/day of pure flax lignan for 14 d.Results: Marked proliferation of fibrous connective tissue infiltrated with mononuclear cells was recorded; congested dilated of sinusoids and fibrosis in the portal area around the proliferation of bile ducts and congested portal blood vessels with the proteinous material in the lumen of bile ducts were also recorded. In other sections, the liver showed large necrotic area replacement with RBCs as well as enlarged foamy hepatocytes with mononuclear cells in narrowing sinusoids were seen.Conclusion: From the results of this study we can conclude that lignan has been used successfully in decreasing the inflammatory cells in congested blood vessels and sinusoids and has decreased the inflammatory cells infiltration in moderate fibrosis in the portal area around the bile ducts of rabbit liver tissue.

Diazinon is an insecticide that has a higher toxicity than other insecticides. Normally, insecticides are detoxified by liver, but this process produced free radicals which causes cell damage. Free radicals also increase lipid peroxidation which directly increase malondialdehyde levels. This research was aimed to determine the influence of diazinon toxicity to the liver histopathology and levels of malondialdehyde (MDA) in the serum of rats (Rattus norvegicus). The rats were divided into 4 groups which consist of negative control group and three experimental groups which were given diazinon 20 mg/kgBW (P1), 40 mg/ kgBW (P2), and 60 mg/ kgBW (P3). The rats were given diazinon for 8 weeks orally. The parameters used in this research was liver histopathology with hematoxilin eosin stain and Malondialdehyde levels with Thiobarbituric test. Liver histopathology was analyzed in descriptive qualitative and the level of MDA was analyzed quantitatively using ANOVA and Tukey's exact test with α = 0.05. The result of this research showed the influence of organophosphate (diazinon) on liver histolopathology was shown by inflammatory cells infiltration in hepatic parenchyma, sinusoidal congestion, and cytoplasmic vacuolation of the hepatocytes. Statistical analysis proved that the diazinon was able to increase the level of MDA serum significantly (p<0,05) up to 71% in experimental group P1 (dose 20 mg/kgBW), which the increase level of MDA serum depend on dose of diazinon. This research concluded that diazinon is one of the organophosphate pesticide that toxic based on liver histopathology and MDA serum levels.

Introduction: Some studies indicated that antlions (Myrmeleon sp.) is traditionally used by many people in Indonesia as a diabetes treatmenter. However. reseach has not been done to find the effect of antlion extract in pancreas, an important organ in diabetes mellitus pathophysiology. Objective: To prove the effect of antlion extract towards histopathological feature of pancreas in diabetic mice.Methods: In the post test only control group design, 25 Swiss mice were divided into five groups. Normal group (Nor-G), Negative control group (Neg-G), and the other 3 groups were intraperitoneally injected with Streptozotocine 200 mg/kg to induce diabetic state. In Neg-G had no antlion treatment, whereas in At-14, At-28, and At-56 were treated with antlion extract at the dose of 14, 28, and 56 mg/kg BW respectively for 24 days. Pancreas then collected and prepared to be examined under microscope with 400x magnification. The score of pancreatic islet injury was determined by assessing structural damage and inflammatory cells infiltration. Kruskal-Wallis and Mann-Whitney were adopted as statistical analysis.Results: Mann-Whitney analysis indicated that the score of pancreatic islets injury in At-56 was significant lower compared to that of Neg-G, p<0.05. The score of pancreatic islets injury in At-56 also significant lower compared to that of At-14 and At-28, p<0.05. In contrary, when compared to that of Nor-G, the pancreatic islets injury in At-56 was not signicantly lower, p>0.05.Conclusion: Antlion extract treatment at the dose of 56 mg/kg BW was capable of reducing pancreatic islet injury in diabetic mice induced by streptozotocine.

Introduction. Knowledge about the effects of medications, vitamins, and various supplements on orthodontic tooth movement (OTM) is imperative for orthodontists. This study aimed to assess the effect of methotrexate (MTX) injection on OTM in rats. Materials and Methods. Twenty-eight male Wistar rats were randomized into four groups (n = 7). The first molar and central incisor were connected using a nickel-titanium (NiTi) coil spring with a 50 g load in each rat. The two experimental groups received 0.75 mg/kg and 1.5 mg/kg MTX, respectively, intraperitoneally for 21 days. The negative control group did not receive any injection and did not undergo orthodontic treatment. The positive control group underwent orthodontic treatment and received 0.9% saline (NaCl) injections for 21 days. All rats were sacrificed with chloroform inhalation after 21 days; their maxilla was resected, and the mean number of Howship’s lacunae, blood vessels, osteoclasts, and resorption lacunae was counted. The reduction in bone volume (bone volume to total volume ratio (BV/TV)) at the site of the maxillary molar was quantified by microcomputed tomography (micro-CT). Results. OTM, the number of osteoclasts, and the number of blood vessels significantly increased in rats treated with MTX ( P < 0.05 ). However, the increase in the number of Howship’s lacunae and resorption lacunae was not significant ( P > 0.05 ). Lower BV/TV in the MTX groups was in agreement with the increased number of osteoclasts. Conclusion. Injection of MTX can significantly increase OTM and decrease root resorption in rats.

Abstract: Aspirin is an anti-inflammatory drug which can cause side effect such as damage of the gastric mucous. Soursop leaf is usually used for medical treatment because it contains flavonoid compound which has the antioxidant and anti-inflamatory activity and may protect gastric mucous from the side effects of aspirin. This study aimed to reveal the histopathological features of the gaster of wistar rats administered with soursop leaf extract after induced with aspirin. This was an experimental study using 20 Wistar rats. Rats were divided into negative control group (A) and treatment groups. Treatment groups were divided into; rats induced with aspirin 30mg for 10 days (B); rats administered with soursop leaf extract 80mg before induced with aspirin 30mg for 10 days (C); rats induced with aspirin 30mg for 10 days and administered with soursop leaf extract 80mg for the next 3 days (D); and rats induced with aspirin 30mg for 10 days and not treated for the next 3 days (E). Groups A, B and C were terminated on 11th day, meanwhile groups D and E were terminated on 14th day. The results showed normal histological features in group A. Group B showed acute gastritic features such as many PMN inflammatory cells in the mucous to serous layers, submucous edema, and capillary dilatation. Groups C and D showed many PMN inflammatory cells in the mucous to submucous layers. Group E showed decreased PMN inflammatory cells in mucous to submucous layers. Conclusion: Administration of soursop leaf extract could not decrease the acute gastritic signs such as inflammatory cells, edema and capillary dilatation in the gaster of Wistar rats induced with aspirin.Keywords: aspirin, soursop leaves, gaster. Abstrak: Aspirin merupakan obat anti inflamasi yang bisa menyebabkan efek samping gangguan mukosa lambung. Daun sirsak sering digunakan sebagai obat tradisional yang berkhasiat karena mengandung senyawa flavonoid yang berkhasiat sebagai antioksidan dan antiinflamasi yang mungkin dapat melindungi lambung dari efek samping aspirin. Penelitian ini bertujuan untuk mengetahui gambaran histopatologik lambung tikus Wistar yang diberikan ekstrak daun sirsak setelah induksi aspirin. Jenis penelitian ini ialah eksperimental yang menggunakan 20 ekor tikus Wistar. Hewan uji dibagi dalam kelompok kontrol negatif (A) dan kelompok perlakuan. Kelompok perlakuan dibagi atas kelompok tikus yang diberi aspirin 30mg selama 10 hari (B); tikus yang diberi ekstrak daun sirsak 80mg sebelum induksi aspirin 30mg selama 10 hari (C), tikus yang diberi aspirin 30mg selama 10 hari dan diberikan ekstrak daun sirsak 80mg selama 3 hari berikutnya (D); dan tikus yang diberi aspirin 30mg selama 10 hari dan tidak diberi perlakuan selama 3 hari berikutnya (E). Kelompok A, B dan C diterminasi pada hari ke-11, kelompok D dan E diterminasi pada hari ke-14. Hasil penelitian menunjukkan gambaran histologik lambung normal pada kelompok A. Kelompok B menunjukkan gambaran histopatologik gastritis akut yakni baanyak sel-sel radang PMN pada lapisan mukosa sampai serosa, edema submukosa, dan pelebaran pembuluh darah kapiler. Kelompok C dan D menunjukkan banyak sel-sel radang PMN pada lapisan mukosa sampai submukosa. Kelompok perlakuan E menunjukkan sel-sel radang PMN yang lebih sedikit pada lapisan mukosa sampai submukosa. Simpulan: Pemberian ekstak daun sirsak tidak dapat mengurangi tanda-tanda gastritis akut berupa sel-sel radang, edema, dan pelebaran pembuluh darah kapiler pada lambung tikus wistar yang diinduksi aspirin. Tukhme Karafs is the seed of Apium graveolens L., which is an important aromatic plant grown globally as a fresh herb and widely used as salad crop. This seed possesses potential pharmacological activity like hepatoprotective, diuretic and lithotriptic. The present study was designed to evaluate nephroprotective effect of Tukhme Karafs against gentamicin (80 mg/kg BW) induced nephrotoxicity in male Wistar rats. The effect was tested at two dose levels i.e. 500 mg/kg and 1000 mg/kg BW (calculated based on clinical dose mentioned in Unani literature). The study was divided into curative and protective models. The efficacy was determined based on estimation of biochemical parameters (creatinine, uric acid, BUN and serum electrolytes) along with quantification of kidney injury molecule-1 (KIM-1) using ELISA method in kidney tissue homogenate and histopathological study of kidney tissue. The findings of the study showed that Tukhme Karafs at both tested doses showed prominent reduction of biochemical parameters as compared to negative control group in both models. KIM-1 reduction was observed but was not statistically significant in both the models. Histopathological changes showed that drugs have inhibited gentamicin induced injury in both models. These results confirm the nephroprotective effect of Tukhme Karafs.

Abstract: Monosodium glutamate (MSG) is one of the ingredients used as flavour additive. However, excessive consumption of MSG could damage the liver. This study was aimed to obtain the impact of MSG exposure on histopathological findings of the liver of Wistar rats administered with tomato juice. This was an experimental study with the post-test only control group design. Subjects were Wistar rats divided into three groups, as follows: the negative control group, treatment group I, and treatment group II. The negative control group was given AD2 pellets; the treatment group I was given AD2 pellets and MSG, while the treated group II was given MSG and tomato juice. This study was conducted for 14 days. The histopathological examination of the treatment group I showed fatty degeneration of hepatocytes and infiltration of inflammatory cells meanwhile of the treatment group II, there was less fatty degeneration of hepatocytes compared to the treatment group I and no inflammatory cells. Moreover, there was no fatty degeneration and inflammatory cells in the negative control group. Conclusion: Wistar rats treated with MSG showed fatty degeneration of hepatocytes and infiltration of inflammatory cells which could be reduced by administration of tomato juice along with MSG.Keywords: tomato juice, MSG Abstrak: Monosodium glutamat (MSG) merupakan salah satu bahan makanan yang sering digunakan sebagai penyedap rasa namun konsumsi MSG berlebihan dapat merusak hati. Jenis penelitian ialah eksperimental dengan post-test only control group design. Tikus dibagi menjadi 3 kelompok, yaitu kelompok kontrol negatif, kelompok perlakuan I, dan kelompok perlakuan II. Kelompok kontrol negatif hanya diberikan pelet AD2; kelompok perlakuan 1 diberikan pelet AD2 dan MSG; kelompok perlakuan 2 diberikan MSG dan jus tomat. Penelitian ini dilakukan selama 14 hari. Hasil pengamatan gambaran histologik hati tikus Wistar pada kelompok perlakuan I mendapatkan adanya degenerasi lemak pada hepatosit dan sel-sel radang PMN. Pada kelompok perlakuan 2 masih ditemukan adanya degenerasi lemak pad hepatosit tetapi dengan jumlah yang lebih kurang daripada kelompok perlakuan I, serta tidak ditemukan sel-sel radang PMN. Pada kelompok kontrol negatif tidak ditemukan adanya degenerasi lemak pada hepatosit dan sel radang. Simpulan: Pada tikus wistar dengan pemberian MSG terdapat degenerasi lemak pada hepatosit dan adanya sel-sel radang. Pemberian jus tomat bersamaan dengan pemberian MSG berefek menurunkan terjadinya degenerasi lemak pada hepatosit dan tidak disertai sel-sel radang.Kata kunci: jus tomat, MSG

The purpose of this study was to develop a mouse model of asthma (MMA) using house dust mite Dermatophagoides pteronyssinus (Der p) extract. Methods. BALB/c mice were i.p. immunized with different doses of Der p lyophilized extract three times in three week interval in the mixture with Al(OH)3. 8 weeks after the final immunization mice were challenged with Der p during five consecutive days by intranasal applications (INA) or aerosol administration (AA). All mice were divided into 5 experimental groups: group 1 was immunized with 50 ^g/mouse of Der p (in protein equivalent) in the mixture with 2 mg/mouse Al(OH)3 and challenged by INA; group 2 was immunized in the same way and challenged by AA; group 3 was immunized with 100 ^g/mouse Der p in the mixture with 2 mg/mouse of Al(OH)3 and challenged by INA; group was immunized in the same way and challenged by AA; group 5 (negative control) was immunized and challenged with saline. 24 hours after the last challenge airway hyperresponsiveness (AHR) to different concentrations of metha-choline was evaluated in all groups by whole-body plethysmography. 48 hours after the last challenge in all groups blood was collected for differential cell count, brochoalveolar lavage fluid (BALF) was sampled for the determination of inflammatory cells and lungs were removed for histological analysis. Histopathological changes in lungs (allergic inflammation) were graded according to semi-quantitative scoring system. Anti-Der p IgE, IgG1 and IgG2a antibodies in individual sera samples were detected by ELISA seven days after the last immunization and 48 hours after the challenge. Results. The levels of anti-Der p IgE antibodies in groups 1-4 before as well as after the challenge were substantially higher than that of in the group 5 (negative control). The highest level of serum Der p-specific IgE antibodies was observed in the group 2. The levels of anti-Der p IgG1 and IgG2a antibodies in the groups 1-4 during all periods of observation were higher than that of group 5 (negative control). At the same time the maximal levels of anti-Der p IgG1 and IgG2a antibodies were observed in group 3 both after immunization and after challenge. The maximum of AHR was observed in the groups 1 and 3 challenged by INA. Analysis of cell composition in BALF demonstrated significant elevated number of eosinophils in group 3 in comparison with group 5 (negative control) and other experimental groups. Regarding peripheral blood leukocyte count we observed decreasing of band neutrophils in group 4 and increasing of segmented neutrophils in groups 1 and 3 in compare to group 5 (negative control). In group 1 we found statistically significant decreasing of lymphocytes and increasing of eosinophils in compare to negative control group 5. Histological picture of general allergic inflammation in lungs as well as peribronchial and perivascular infiltration with inflammatory cells were the most noticeable (according to score system) in group 3 in comparison with negative control group and other experimental groups. Conclusion. Data obtained indicate that immunization (sensitization) of mice with Der p in a dose 100 μg/mouse together with Al(OH)3 and challenge with Der p by mean of intranasal applications is a suitable approach for modeling of mouse allergic asthma.

Immunosuppressant and steroid therapy for SLE have not shown satisfactory results. Another method of therapy that is being developed is vaccines and escalating dose immunotherapy using self-antigen. The aim of this study was to assess the balance of immune cells through the ratio of pro-inflammatory and anti-inflammatory cells and cytokines in SLE using self-antigen dsDNA therapy. Methods: Female Balb/c mice 6-8 weeks old separated randomly to negative control group and pristane induced lupus (PIL) mice group. PIL mice groups were injected pristane intraperitoneally. Twelve weeks after the injection, the mice were evaluated for clinical and serological manifestations (anti-dsDNA levels). Mice with lupus signs were divided into four groups; positive control group: PIL mice without EDI dsDNA therapy, treatment A: PIL mice with EDI dsDNA therapy dose I (0.01μg/ml, 0.1μg/ml, 1μg/ml), treatment B: PIL mice with EDI dsDNA therapy dose II (0.1μg/ml, 1μg/ml, 10μg/ml), and treatment C: PIL mice with EDI dsDNA therapy dose III (1μg/ml, 10μg/ml, 100μg/ml). dsDNA were injected once a week and the dose was increased every week. Samples were analyzed for active/inactive dendritic cells ratio, Th1/Th2 cells ratio, Th17/Treg cells ratio and IL-17/TGF-β levels ratio. Results: Escalating dose antigen specific therapy with dsDNA injection of third dose reduced active/inactive dendritic cells ratio (p=0.000), Th1/Th2 cells ratio (p=0.010), Th17/Treg ratio (p=0.004) and decrease IL-17/TGF- β levels ratio (p=0.004) significantly compared to positive control. Conclusion: Escalating dose antigen specific therapy with dsDNA injection of dose III was able to regulate balance ratio of inflammatory cells and cytokines in PIL mice thus the immune tolerance may improve compared to control groups.

The aim of this study to conduct the effects of fimbrial and lipopolysacchride (LPS) immunization is on the pathohistological changes in rabbits, Fifteen rabbits of both sexes (Weight 1500-2000 gm) divided into three groups (5 animals of each group). The first group was immunized by 1ml (200µg /animal) of fimbrial subcutaneously the second group gave 1 ml ( 200 µg /animal) LPS while the third group was left as negative control group that injected 1 ml phosphate buffer control subcutaneously. First and second groups recived the same dose after two weeks give as booster dose. All animals challenged after 5 weeks of immunization by5X107CFU/ml Proteus vulgaris intra peritoneally .After 7 days from challenge all the animals, sacrificed for histopathological examination . The results showed that the fimbrial group had a severe infiltrations of mononucleart cells in liver and kidney ,but there was no clear histopathological changes observed in the spleen compared with lipopolysaccharide and control group . Also this group showed a slight mononuclear cells infiltration in lamina properia of intestine ;while the lipopolysaccharide group showed hypertrophy of epithelial cells with a mild mononuclear cells infiltration in lamina properia as compared with the control group that showed presence of mononuclear cells aggregation in the lamina properia .Our conclusion That the defense of animals against Proteus vulgaris more efficient in fimbrial antigen than lipopolysaccharide antigen by decrease the pathological effects of this bacteria.

The aim of the study is to determine the effectiveness of betamethasone in the treatment of local post-tattoo complications, depending on the mode of administration.Materials and methods. The work was carried out on 90 male rats which had been tattooed (n = 30 – a negative control group; n = 30 – a comparison group: a subcutaneous administration of 1 ml of a betamethasone solution; n = 30 – an experimental group: an administration of 1 ml of a betamethasone solution using a tattoo machine), 15 – intact rats. Withdrawal from the experiment took place on the 3rd, 10 th and 21st days. The skin samples were fixed in 10% formalin, followed by histological posting and manufacturing of micropreparations, then staining with hematoxylin and eosin, according to Van Gieson. A morphometric study included determination of the volume fraction (VF) of the epidermis; dermal fibers; pigment; inflammatory cells; macrophages (%), as well as the pigment depth (μm) and the severity of edema.Results. The study found out that in the process of the betamethasone administration using a tattoo machine, the drug was uniformly administrated over the entire area of the tattoo; hereby, the phenomena of edema and inflammatory infiltration were insignificant. The dermal fibers were located in each layer with no signs of edema and with single cells of inflammation, respectively. The data of the histological processing were completely consistent with the results of morphometry: it was found out that in the experimental group, edema significantly decreased, the volume fraction of the pigment and macrophages decreased, and the volume fraction of the dermal fibers increased. The estimation of the inflammatory reaction was carried out according to the morphometric parameters of the volume fraction of inflammatory cells and had significant differences in all the experimental groups, decreasing in the following series: the negative control group> the comparison group> the experimental group> the group of intact animals (p <0.05).Conclusion. Based on the data obtained, the effectiveness of betamethasone in the treatment of local post-tattoo complications has been proved. In this case, the treatment of choice is the administration of this drug not traditionally subcutaneously, but using a tattoo machine that enables the targeted delivery of the substance to the area of the pathological process.Abbreviations: NC – negative control, VF – volume fraction
