The purpose of this research was to determine the antagonistic bacteria Pseudomonas spp. of paddy roots endophytic against disease blast (Pycularia oryzae) in vitro. This research was conducted at the Laboratory of Agroecology, Faculty of Agriculture, University of Sultan Ageng Tirtayasa on April to June 2015. This research implemented experimental one factor that arranged in Completely Randomized Design with three replications. The treatment consisted of 9 isolates of Pseudomonas sp. which was screened from endophytic roots of paddy plants.  The test  results  showed  that  antagonistic in  vitro  testing could inhibit the growth P. oryzae. Inhibition of 9 isolates to the growth Pseudomonas spp. had the significant clear zone. Isolate Ps 17 (58.89%), Ps 22 (54.44%), Ps 37 (52.06%) and Ps 39 (61.42%) have the largest inhibition. The results showed that the bacteria Pseudomonas sp. on this research is potential on controlling disease blast in paddy plant.

This research aims to determine the effectivity of seaweeds extract (Ulva lactuca) as an antifungal in the control of kerling (Tor sp) infected with saprolegnia and achlya fungal. The research was conducted in May 2018 at the MIPA Laboratory, Syiah Kuala University and the Hatchery of the Faculty of Fisheries and Marine Sciences, Teuku Umar University. The research method was experimental with a completely randomized design (RAL), 3 treatments and 3 replications. The treatment given was immersion of kerling fish seeds infected with fungi in a solution of sea lettuce extract with the concentration of treatment namely: K (control) = 0 ppm, P1=5 ppm, P2=10 ppm, dan P3=15 ppm. Phytochemical test results showed that the content of bioactive compounds contained in ethanol extract of seaweed were alkaloids, steroids and phenolic/tannins. Ethanol extract of sea lettuce can inhibit the growth of Saprolegnia and Achlya in vitro by forming a clear zone of 5.7 mm (medium). The highest percentage of fish survival rate was in the treatment (P3) concentration of 15 ppm.

Has been done research that aims to test the inhibitory power of pronojiwo (Euchresta horsfieldii) leaf extract to the fungus P.oryzae. the experimental study was conducted using a completely randomized design with 9 concentrations of liquid extract of pronojiwo leaf and 1 control. Ie the concentration of each pepeated 4 times and in duplo. Application is done by the method of well by means of liquid extract dripped on the diffusion well, dilusion method by means of liquid extract mixed on the media PDA, method of calculating the amount of fungus conidia by means of liquid extract mixed with media PDB, to determine the inhibition zone of the fungus, the growth of the diameter of the fungus, the number of living conidia. Observations were made after incubation for 7 days. The results showed that liquid extract pronojiwo leaves can inhibit and reduce the growth of fungus P.oryzae, at a concentration of 6,25% with a resistor zone result of 0,9 mm, a growth diameter percentage of 71,2%, and may reduce by 2,9 conidia/ml.   Keywords : pronojiwo leaves, growth, reduce , fungus P.oryzae

Efficacy wood vinegar produced from wood laban (Vitex pubescens) against Ophiostoma piliferum fungus was evaluated. The objectives of this research to perform in vitro antifungal of wood vinegar from laban wood against O. piliferum. The source of lignocelluloses biomass was carbonization process used three temperature i.e. 350, 400 and 450 °C.  Efficacy of wood vinegar to the fungus carried on PDA (potato dextrose agar) in a Petri dish that has been mixed with the wood vinegar with a concentration of 0, 0.5, 1.0, 1.5, and 2.0 % (v/v). Antifungal test was a factorial 3 by 5 in a completely randomized design. The means were seperated using using Tukey’s test at p = 0.05. All data were analyzed using the SAS softwere (version 8.2, SAS Institute Inc., NC. USA). The test results showed that the pyrolysis suhue effect and concentration of wood vinegar on the inhibition of fungal growth. The higher pyrolysis temperature and concentration the inhibition of fungal growth was increased.Efikasi asap cair dari kayu laban (Vitex pubescens)terhadap Ophiostoma piliferum telah dilakukan.Penelitian ini bertujuan mengevaluasi kemampuan asap cair sebagai antijamur (O. piliferum) secara in vitro. Asap cair yang digunakan diperoleh dengan cara pirolisis pada suhu produksi asap cair dan konsentrasi asap cair yang berbeda. Efikasi asap cair terhadap jamur dilakukan pada media PDA (potato dextrose agar) dalam cawan Petri dengan campuran asap cair pada konsentrasi 0, 0,5, 1,5, dan 2,0 (v/v).Pengujian aktivitas antijamur dilakukan menggunakan rancangan acak lengkap tipe faktorial.Data yang diperoleh dianalisis dengan sidik ragam.Pengaruh  antarperlakuan dilakukan uji BNJ pada taraf nyata 5%. Analisis data menggunakan SAS versi 9.13. Hasil pengujian menunjukkan bahwa suhu pirolisis produksi asap cair dan konsentrasi asap cair berpengaruh terhadap daya penghambatan pertumbuhan jamur. Makin tinggi suhu pirolisis produksi asap cair dan konsentrasinya dan makin tinggi daya penghambatan terhadap pertumbuhan jamur, O. piliferum.

Saluang Belum Root (Lavanga sarmentosa) and Yellow Root ((Arcangelisia flava) Merr) plants that have antibacterial effects need to be proven, for this research this program uses Saluang Belum Root (Lavanga sarmentosa) with Yellow Root (Arcangelisia flava) Merr) against bacterial inhibition. Aeromonas hydrophila by In Vitro. The method used in this study was a Completely Randomized Design (CRD) with 6 (six) settings and 3 (three) replications related to the aid of A = 25 gr / 150 ml distilled water, treatment B = 35 gr / 150 ml distilled water, C treatment = 45 gr / 150 ml distilled water, treatment D = 55 gr / 150 ml distilled water, handling E = 500 mg chloromphenicol / 150 ml distilled water, and treatment F = negative control only using the use of 150 ml distilled water. The results of this study indicate a combination of Saluang Belum Root Root (Lavanga sarmentosa) with Yellow Root (Arcangelisia flava Merr) which is able to inhibit the growth of My Aeromonas hydrophila bacteria because it has an antibacterial effect, this can be seen from the results of the zone average (inhibitory power) on each The treatment is the highest clear zone according to the D 55 gr / 150 ml treatment with an average clear zone of 15 mm and the lowest clear zone yield is A 25 gr / 150 ml approval with an average clear zone value of 11 mm.  Keywords: Aeromonas hydrophila; Root of Saluang Not yet; Yellow Root; Antibacterial.

Blood Disease Bacteria is one obstacle in cultivation of bananas. BDB can be controlled using biological agents. Biological agents that can be used to control of blood diseases are Pseudomonad fluorescent and microorganisms in EM4. Pseudomonad fluorescent used as biological agents because of its ability to produce antimicrobial compounds such as siderophores, antibiotics, volatile and cyanide. other than that, the EM4 microorganisms can produce antimicrobial against several bacterial pathogens. This study aims to determine the effect of combination Pseudomonad fluorescent and EM4 to inhibit the growth of BDB in vitro. This research was conducted in January 2016 in the Laboratory of Microbiology Department of Biology FMIPA UNP. Research using a completely randomized design (RAL) with 6 treatments and 3 replications. The study treatment is a combination of Pseudomonads fluorescent and EM4. The observation was done by measuring the clear zone formed around the paper disc. Data were analyzed using analysis of variance (ANOVA) and the test of Duncan New Multiple Range Test (DNMRT) at 5% level. The research proves that the combination resulted in inhibition zone around the paper disc. Inhibition zone shows that the combination inhibits the growth of BDB in vitro. Inhibition zone is formed as fluorescent Pseudomonads and microbes on EM4 produce secondary metabolites that can inhibit the growth of BDB.Key Word: Pseudomonad flourecent, BDB, EM4

Shallot (Allium ascalonicum L.) has been known as traditional medicine. Shallot which has same genus with garlic(Allium sativum L.) contains allicin that is also found in garlic and has been suspected has fungicidal activity toCandida albicans. It is supported by several researches. Therefore, shallot is suspected has antifungal activity too.The aim of this research was to know antifungal activity of shallot’s water extortion againsts Candida albicans invitro. This was a laboratory experimental research which used completely randomized design, with diffusion method.Shallot’s water extortion was devided into three concentrations, there were 50%, 100% and 200%. Ketoconazole 2%was positive control and aquadest was negative control. The result of this research based on analysis of varians(Anova), there was significant difference between several treatments and was confirmed with Duncan New MultipleRange Test (DNMRT) p<0,05, there was significant difference between 100% shallot’s water extortion with othertreatments, but there was no significant difference between 50% shallot’s water extortion with 200% shallot’s. Theconclusion was shallot’s water extortion had antifungal activity againsts Candida albicans with the best concentration100%, but it was lower than ketoconazole 2%.

Dendrobium anosmum is one of natural orchids in Indonesia. Optimization of medium composition for orchid propagation through in vitro culture is necessary to enhance propagule multiplication capabilities and quality. This study was aimed to study the influence of concentration of coconut water in culture medium on in vitro growth and development of D. anosmum orchid species and to determine the optimal coconut water concentration in culture media.  The experiment were arranged in a Completely Randomized Design with four treatments and eight replications. The treatments consisted of the addition of coconut water with concentrations: 0 ml•l -1 (control), 50 ml•l-1, 100 ml•l-1 and 150 ml•l-1. The results showed that addition of coconut water in culture medium gave different effect on shoot growth and multiplication of D. anosmum orchids.  Coconut water concentration of 100 ml•l-1 was the best concentration for growth and multiplication of D. anosmum orchids, based on both shoots and roots growth, plantlet height and wet weight.

<em>Inhibition of Pseudomonas fluorescens isolates the origin of tomato, soybean and corn against Ralstonia solanacearum tested using a completely randomized design (CRD).        P. fluorescens growth was measured at King's B medium by way of suspension antagonist put on filter paper of 0.5 cm in diameter. Inhibition of P. fluorescens is done by placing the antagonist suspension of 0.5 cm diameter filter paper on NA media that has been deployed R. solanacearum. The results showed that: (a) the growth of P. fluorescens origin of tomato, soybean and corn on King's B media were not significantly different, (b) the inhibition of P. fluorescens isolates against R. solanacearum not significantly different and, (c) the inhibition of isolates P. fluorescens origin of tomato, soybean and corn against R. solanacearum in vitro relatively strong</em>

The objective of this study was to evaluate the efficacy of the active ingredient abamectin, either in isolation or in combination with fungicides and insecticides formulated for the industrial seed treatment, on the population of Meloidogyne javanica, tested under greenhouse and in vitro conditions. In both tests, the combination of the following treatments was assessed: abamectin; thiamethoxam; fludioxonil + metalaxyl-M + thiabendazole. Water was used as control for the in vitro assay, whereas under greenhouse conditions, controls were inoculated and not inoculated with M. javanica. The tests were conducted in a completely randomized design with six (in vitro) and ten (greenhouse) replicates. For in vitro studies the effect on hatching, motility, and mortality of juveniles of M. javanica was evaluated. Under greenhouse conditions, the soybean ‘BRS 133’ seeds were treated, and at 15, 30, and 60 days after inoculation (DAI) with M. javanica, plant measurements were recorded. The penetration of second stage juveniles (J2) was also evaluated at 15 DAI. At 30 DAI, galls, egg masses, nematodes/g of root, and final population were evaluated. At 60 DAI, the final population of nematodes in the roots was quantified. The treatments containing abamectin were the most effective in diminishing the hatching of juveniles. All treatments had an effect on nematode motility when compared to the control, and in the treatment containing only abamectin, total juvenile mortality was observed. In greenhouse conditions, at 15 DAI, the treated soybean plants had the highest root mass and shoot length, differing statistically from the inoculated control. All treatments reduced the number of nematodes per gram of root, differing from the control. At 30 DAI, treatment efficiency was observed in reducing the final population of M. javanica, particularly the treatments using abamectin, and abamectin + thiamethoxam + fludioxonil + metalaxyl-M + thiabendazole. However, at 60 DAI, the effect of the treatments on the population of M. javanica did not persist.

Pseudomonas species are metabolically robust, with capacity to produce secondary metabolites including cyclic lipopeptides (CLPs). Herein we conducted a chemical analysis of a crude CLP extract from the cocoyam rhizosphere-derived biocontrol strain Pseudomonas sp. COW3. We performed in silico analyses on its whole genome, and conducted in vitro antagonistic assay using the strain and purified CLPs. Via LC-MS and NMR, we elucidated the structures of four novel members of the bananamide group, named bananamides D-G. Besides variability in fatty acid length, bananamides D-G differ from previously described bananamides A-C and MD-0066 by the presence of a serine and aspartic acid at position 6 and 2, respectively. In addition, bananamide G has valine instead of isoleucine at position 8. Kendrick mass defect (KMD) allowed the assignment of molecular formulae to bananamides D and E. We unraveled a non-ribosomal peptide synthetase cluster banA, banB and banC which encodes the novel bananamide derivatives. Furthermore, COW3 displayed antagonistic activity and mycophagy against Pythium myriotylum, while it mainly showed mycophagy on Pyricularia oryzae. Purified bananamides D-G inhibited the growth of P. myriotylum and P. oryzae and caused hyphal distortion. Our study shows the complementarity of chemical analyses and genome mining in the discovery and elucidation of novel CLPs. In addition, structurally diverse bananamides differ in their antimicrobial activity.
