Phytochemical Investigation and Antibacterial Activity of Bergenia ligulata

In the course of screening of plant extracts for antimicrobial activity, the methanolic extract of Bergenia ligulata (Saxifragaceae) exhibited antibacterial activity against gram-positive bacteria. Fractionation of ethylacetate soluble phase of the methanol extract led to the isolation of bergenin (1), 11-O-p-hydroxybenzoylbergenin (2), 6-O-galloylarbutin (3) and epiafzelechin (4). Except bergenin, other compounds were isolated for the first time from B. ligulata. Euphorbia antiquorum is a large shrub or small tree belonging to the largest and the most diverse family in the plant kingdom, Euphorbiaceae. E. antiquram latex was used as a classical binding agent for the kshara sutra in the management of the anal fistula. In the present study, an attempt was made to evaluate the antimicrobial properties of latex of E. antiquorum on certain microbes. The methanolic extract of the latex was prepared. The fractions of methanol extract were tested for their antibacterial activity against the Gram positive bacteria, Streptococus agalactiae and Staphylococcus aureus and Gram negative bacteria Escherichia coli and P. aeruginosa. The results revealed that the latex showed minimum inhibition only to E. coli and S. aureus. Latex did not posses antibacterial activity against S. agalactiae and P. aeruginosa. The results suggest that the E. antiquorum latex extract account for the antibacterial properties and has a potential for use as an antimicrobial agent.

Bioassay-directed fractionation for the determination of antimicrobial activity of Uncaria tomentosa, has led to the isolation of isopteropodine (0.3%), a known Uncaria pentacyclic oxindol alkaloid that exhibited antibacterial activity against Gram positive bacteria.

Polyphenolic enriched extracts from two species of Cyperus, Cyperus glomeratus and Cyperus thunbergii, possess mammalian arginase inhibitory capacities, with the percentage inhibition ranging from 80% to 95% at 100 µg/mL and 40% to 64% at 10 µg/mL. Phytochemical investigation of these species led to the isolation and identification of two new natural stilbene oligomers named thunbergin A-B (1–2), together with three other stilbenes, trans-resveratrol (3), trans-scirpusin A (4), trans-cyperusphenol A (6), and two flavonoids, aureusidin (5) and luteolin (7), which were isolated for the first time from C.thunbergii and C. glomeratus. Structures were established on the basis of the spectroscopic data from MS and NMR experiments. The arginase inhibitory activity of compounds 1–7 was evaluated through an in vitro arginase inhibitory assay using purified liver bovine arginase. As a result, five compounds (1, 4–7) showed significant inhibition of arginase, with IC50 values between 17.6 and 60.6 µM, in the range of those of the natural arginase inhibitor piceatannol (12.6 µM). In addition, methanolic extract from Cyperus thunbergii exhibited an endothelium and NO-dependent vasorelaxant effect on thoracic aortic rings from rats and improved endothelial dysfunction in an adjuvant-induced arthritis rat model.

Ayurvedic medicines are being used increasingly to fight or prevent common diseases. Udhara vayuhara churna is a polyherbal formulation containing Piper longum (Piperceae) Zingiber officinalis (Zingiberaceae), Elettaria cardamom (Zingiberaceae), Plumbago zeylanica (Plumbaginaceae), Carum carvi (Umbelliferae), Acorus calamus (Araceae) and Embilica officinalis (Euphorbiaceae). Methanolic and aqueous extracts of Udhara vayuhara churna were screened for their antimicrobial activity against Bacillus subtilis, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Aspergillus niger and Candida albicans. The results of antimicrobial activity of aqueous and methanolic extracts of the Udhara vayuhara churna indicated that methanolic extract inhibited the growth of one or more test pathogens than aqueous extract. Udhara vayuhara churna formulation extracts showed a broad spectrum of antimicrobial activity. Phytochemical investigation revealed the presence of tannins, saponins, alkaloids, glycosides, flavonoids and essential oils. This may be due to the multifunctional effect of all the seven plant ingredients of Udhara Vayuhara churn.

Pycnarrhena cauliflora (Miers.) Diels., local name sengkubak, is one of indigenous plants from West Kalimantan that has been used as natural flavor. Pycnorrhena cauliflora is one of species of Menispermaceae family which is rich in bisbenzylisoquinoline alkaloids. This alkaloids are known to have various biological activities including antiprotozoal, antiplasmodial, antifungal and antibacterial activities. This study aimed to investigate antimicrobial activity of  the P. cauliflora (Miers.) Diels. methanolic extracts against gram-positive and gram-negative bacteria. The methanolic extract of P. cauliflora (Miers.) Diels., root, leaf and stem were prepared by maceration. The disk-diffusion method was then used to determine the antimicrobial activity of the extracts against Streptococcus pyogenes, S. mutants, Staphylococcus aureus, S. epidermidis, Salmonella typhi, Shigella flexneri, Pseudomonas aeruginosa and Escherichia coli after 18-24 h incubation at 37 oC. Amoxicillin was used as positive control for gram-positive bacteria and ciprofloxacin was used as gram-negative bacteria. The inhibition zones were then measured in mm. Analysis were conducted in duplicates. The results showed in general the methanolic extracts of P. cauliflora (Miers.) Diels. root (inhibition zone diameter= 10-23 mm) were more active than that leaf (0-15 mm) and stem (0-17 mm) extracts against gram-positive bacteria. The zone inhibition diameter of amoxicillin as positive control was 8-42 mm. In addition, the methanolic extracts of P. cauliflora (Miers.) Diels. root (12-17 mm) were also more active than that leaf (0-12 mm) and stem (0-12 mm) extracts against gram-negative bacteria. The zone inhibition diameter of ciprofloxacin as positive control was 33-36 mm. In conclusion, the methanolic extract of P. caulifloria (Miers.) Diels. root is the most extract active against both gram-positive and gram-negative bacteria. Further study will be focused to isolate active compounds in the methanolic extract of the root.

Comparative study of plant extracts crude and aqueous, methanolic and ethanolic with antibiotics, provide evidence that calotropis procera extracts has the similar antibacterial activity as these antibiotics against test pathogens i.e. Salmonella typhi and E.coli. The analysis of antimicrobial activity of aqueous, methanolic and ethanolic extract of leaves and flower of Calotropis procera was carried out in disc method and also determined MIC value at 600nm through optical density using spectrophotometer. The zone of inhibition produced by extracts was examined and compares it with zone produced by antibiotics. The effect exhibited by ethanolic extract of leaves and flower was significantly greater than the aqueous and methanolic extract of leaves and flower. Crude extracts i.e. latex, leaves, fruit and flower crude extracts. Among them, flower crude extracts shows similar zone of inhibition to test anitibiotics.while in MIC value, we made different concentration of extracts and antibiotics.i.e for crude we made 25%, 50%.75% and 100% concentration of crude juice and for the aqueous, methanolic and ethanolic we made 0.5mg/mL,1mg/mL,2mg/mL,3mg/mL,4mg/mL, 5mg/mL and 6mg/mL in Dimethyl salfoxide, and same antibiotics concentration. Aqueous leaves extracts show MIC at 0.5mg/ml against E.coli while against Salmonella it shows MIC at 1mg/ml... We also determine phytochemical analysis for presences of different compounds in crude extracts. The obtained results provide a support for the use of Calotropis procera, in traditional medicine and suggest its further advance investigation.

The aim of this paper is the isolation of individual compounds, which are caused the antibacterial activity of the leaves of the bearberry [Arctostaphylos uva-ursi (L.) Spreng.]. The leaves of Arctostaphylos uva-ursi, collected in Perm region, there were extracted with 70% ethanol, the obtained water-alcoholic infusion there was evaporated in vacuum.By means of the chromatographic methods with the using of silica gel 40/100 and eluent systems (chloroform and ethanol in several ratio) from the evaporated water-alcoholic extract of the leaves of Arctostaphylos uva-ursi, a substance with antibacterial activity, ethyl ester of p-digallic acid, which is a new natural compound, was isolated along with arbutin ((1-О-b-D-glucopyranoside of hydroquinone) from the leaves of this plant. The chemical structures of the ethyl ester of p-digallic acid and arbutin were established with the using of data of 1H-NMR-spectroscopy, UV-spectroscopy and mass-spectrometry..The antibacterial activity of ethyl ester of p-digallic acid against test cultures of gram-positive bacteria Bacillus cereus and Staphylococcus aureus, gram-negative bacteria Escherichia coli and Pseudomonas aeruginosa for the first time was determined. By antibacterial activity, arbutin was inferior not only to ethyl ester of p-digallic acid, but also to decoction from the leaves of the bearberry. Consequently, the ethyl ester of p-digallic acid is one in main component, which is take the contribution in the antibacterial activity of the decoction and other preparations of the leaves of Arctostaphylos uva-ursi.

Genus Mentha presents group of plants which are the most studied in family Lamiaceae. Aboveground parts are used for different purposes in pharmacy, food industry or confectionery. Most important is natural product extracted from leaves - essential oil (EO). The aim of presented experiment was to demonstrate different chemotype and compare antibacterial activity of two Mentha species EO. Plant samples were obtained from various environments – from Slovakia and from Italy. Dominant compounds were determined by GC/MS. The results showed high amount of menthol and menthone in tested Slovak peppermint EO. On the other hand, carvone and 1,8-cineole were determinate as dominant compounds in Italian spearmint EO. The antimicrobial activity of the EO was investigated by disc diffusion and broth micro dilution methods. EO was evaluated for their antibacterial activity against 7 microorganisms: Enterobacter cloacae, Salmonella spp., Klebsiella pneumoniae, Escherichia coli, Staphylococcus aureus, Streptococcus pyogenes. The determination results of antibacterial activity by agar disk diffusion method ranged from 7 to 14 mm of the growth inhibition zone. MIC of tested mint EO varied from 0.625 to 2.5 μg/mL. In addition, both EO showed relatively the same antibacterial activity against the selected Gram-negative bacteria. However, there is a variation in the antibacterial activity against Gram-positive bacteria.

In the present study, Coccinea cordifolia root was extracted with various solvents according to their increasing order of polarity and the extracts were subjected to preliminary phytochemical investigation. The extracts were then subjected to antioxidant assay using FTC, TBA, DPPH and Reducing Power assay as models and Antimicrobial assay. The Methanolic extract of the plant root has shown good antioxidant and antimicrobial activity among the extracts under study. The findings of the study here justify the traditional claim against Coccinea cordifolia root.

A phytochemical investigation of whole <em>Aspidistra letreae</em> plants led to the isolation of 2<em>H</em>-chromen-2-one (<strong>1</strong>), <em>α</em>-tocopherol (<strong>2</strong>), (<em>E</em>)-phytol (<strong>3</strong>), asparenydiol (<strong>4</strong>) and (25<em>S</em>)-spirost-1<em>β</em>,3<em>α</em>,5<em>β</em>-triol (<strong>5</strong>). Their structures were determined on the basis of NMR spectral evidences and in comparison with the reported data. Of these, asparenydiol (<strong>4</strong>) was isolated from the genus <em>Aspidistra</em> for the first time. This is also the first report on the separation and structural determination of (25<em>S</em>)-spirost-1<em>β</em>,3<em>α</em>,5<em>β</em>-triol (<strong>5</strong>) as a pure compound. The methanol extract exhibited a moderate cytotoxicity against the LU-1, HeLa, MDA-MB-231, Hep-G2, and MKN-7 human cancer cell lines with <em>IC</em><sub>50</sub> values ranging from 52.58 ± 3.65 to 64.78 ± 4.89 μg/mL.
