Daun M. koenigii sangat umum digunakan sebagai rempah dalam berbagai masakan di Aceh, namun pemahaman masyarakat terhadap pentingnya daun M. koenigii perlu ditingkatkan agar pemanfaatannya dapat ditingkatkan. Dalam penelitian ini dilakukan uji aktivitas antibakterial daun M. koenigii terhadap bakteri E. coli dan S. aureus serta uji aktivitas antioksidan terhadap radikal bebas DPPH (1,1-Diphenyl-2-pycrilhydrazil). Uji antibakterial dilakukan dengan metode difusi cakram, dan hasilnya ekstrak etil asetat menghasilkan zona hambat yang lebih besar dibandingkan ekstrak metanol dan n-heksana. Zona hambat yang dihasilkan pada konsentrasi 1, 5 dan 10 % secara berturut-turut adalah 8,7; 7,7 dan 6,7 mm terhadap E. coli, dan 12,8 ;10,7 dan 8,0 mm terhadap S. aureus. Ekstrak etil asetat memiliki aktivitas antioksidan paling kuat dibandingkan dengan ekstrak metanol dan nheksana secara berturut-turut berdasarkan nilai IC50 yaitu 23; 50,54; dan 64,70 ppm. Berdasarkan kromatografi kolom diperoleh 8 (A-H) subfraksi dan subfraksi G menunjukan aktifitas antioksidan yang paling kuat sebesar 14,41 ppm   Murraya koenigii (M. koenigi) leaves are very commonly used as spices in various cuisines in Aceh, but people's understanding of the importance of M. koenigi leaves needs to be improved, so it can be utilized better. In this study, the antibacterial activity of M. koenigii leaves was carried out against E. coli and S. aureus and antioxidant activity against DPPH free radicals (1,1- Diphenyl-2-pycrilhydrazil). Antibacterial tests were carried out using the disc diffusion method, and as a result, the ethyl acetate extract produced a larger inhibition zone than the methanol and n-hexane extracts. The inhibition zones produced at concentrations of 1, 5 and 10% were 8.7; 7.7 and 6.7 mm against E. coli, and 12.8; 10.7 and 8.0 mm against S. aureus. Ethyl acetate extract had the strongest antioxidant activity compared to methanol and nexexane extracts based on IC50 values, 23; 50; 54; and 64.70 ppm. Based on column chromatography obtained, 8 (A-H) subfraction and subfraction G showed the strongest antioxidant activity of 14.41 ppm.

Abstract Cucumis ficifolius (family Cucurbitaceae) is traditionally used for the treatment of diarrhea, teeth-ach, wound, burns, bleeding from external cuts and against different infectious diseases. With the absence of previous scientific report on the roots of the plant, this study was focused on isolation of chemical constituents and evaluation of their anti-bacterial activities of the roots of C. ficifolius. The n-hexane, ethyl acetate and methanol extracts of the roots of C. ficifolius were investigated for their phytochemical constituents and the results revealed that C. ficifolius is rich in many secondary metabolites such as saponins, terpenoids, glycosides, and steroids. But tannins and alkaloids were not observed. The ethyl acetate extract was subjected to column chromatography over silica gel and furnished three compounds namely compound-1, compound- 2 and compound-3. The structures of these compounds were characterized using FTIR, 1H-NMR, 13C-NMR and DEPT-135. The crude extracts and the isolated compounds were tested against four bacteria strains (Gram negative, E. coli and p. aeruginosa; Gram positive, S. aureus and B. subtilis) using Agar well diffusion method, the results showed that the methanol extracts and compound-2 were active against all the tested bacteria. Accordingly, 12.34, 18.25 and 23.75 mm zone of inhibition against B. subtilis were found at the concentration of 20, 30 and 40 μg/mL MeOH extract respectively. Similarly 9.5, 13.25 and 17.25 mm zone of inhibition were found for the same bacteria strains and concentrations of compound-2. These results showed the increase of inhibition zone with concentration. However, the n-hexane crude extract was found not to have anti-bacterial activity towards the tested bacteria strains.

Abstract Endosymbiont fungi from marine sponges are a rich source of medicinally active compounds. Indonesia has a huge number of marine sponges. This research was conducted to determine the antibacterial potential of endosymbiont fungi isolated from marine sponges collected from Kotok Kecil Island, Seribu Islands. The screening was conducted with two methods, the modified GIBEX and disc diffusion. Species of marine sponges were isolated, namely Petrosia sp., Stylissa carteri, Cinachyrella australiensis, Callyspongia sp., Petrosia nigrians, and Stylissa massa, and obtained 9 isolates of endosymbiont fungi. The GIBEX test against Escherichia coli and Streptococcus mutans showed that ethyl acetate extract had strongest antibacterial activity. Disc diffusion test of ethyl acetate extract, five isolates had antibacterial activity against S. mutans and four against E. coli. The isolate that showed the strongest antibacterial activity was from Petrosia sp. Which has an inhibition zone of 8.4 mm against E. coli and 7.45 mm against S. mutans. The main active compounds from the isolate of Petrosia sp. are butylhydroxytoluene and phthalic acid di-(2-propylpentyl) esters. Based on this study, we concluded that the endosymbiont fungi of marine sponges are potential to be developed for further development as source of antibacterial agents.

Zanthoxylum alatum (Z. alatum), an important medicinal plant is used for various ailments including chest infection, cough, cholera, fever, stomach disorders, gas problems, indigestion, piles, toothache, gum problems, dyspepsia and stomachic worldwide. Keeping in view the medicinal potential of this plant, fruit and leaves methanolic (MeOH) extracts were prepared, evaluated for antioxidant activity by 1,1-diphenyl-2-picrylhydrazyl (DPPH) and for antibacterial potential by well diffusion and macrodilution methods in-vitro. Our results confirmed that Z. alatum fruit and leaves extracts have significant antioxidant activity with IC50 values 0.28 ± 0.07 mg/ml and 0.34 ± 0.05 mg/ml, respectively. The inhibitory trend at highest tested concentration (120 mg/ml) at 24 hr incubation in well diffusion method was recorded as 0.39, 0.30, 0.28, 0.27 and 0.18 cm against S. pyogenes, B. cereus, E. coli, S. aureus and S. enterica for leaf extract, respectively. In case of fruit extract inhibitory trend at highest tested concentration was observed as 0.33, 0.32, 0.31, 0.30 and 0.28 cm against S. aureus, S. enterica, S. pyogenes, B. cereus and E. coli, respectively. The fruit extract showed higher zones of inhibition than leaves extracts against all the test bacteria except S. pyogenes. Moreover, highest zones of inhibition were observed at lowest incubation (24 hr) and lowest zones were observed at highest incubation period (72 hr) for all tested concentrations. Macrodilution method showed antibacterial susceptibility in liquid medium with different levels of IC50 values ranging from 1.6±0.13 mg/ml to 10.3±0.7 5mg/ml. Interestingly, none of the tested bacteria showed resistance against any of the test extract in well diffusion or macrodilution method expressing the Z. alatum as potent candidates to kill bacteria in semisolid or in liquid medium to fulfill medical needs in future.

Caulerpa racemosa merupakan rumput laut hijau yang mengandung senyawa aktif yang berpotensi sebagai antibakteri dan antioksidan. Perbedaan profil fitokimia dan bioaktivitas suatu ekstrak dipengaruhi oleh kepolaran pelarut ekstraksi. Penelitian ini bertujuan untuk mempelajari profil fitokimia, aktivitas antibakteri dan antioksidan ekstrak C. racemosa. Ekstrak diperoleh dari sampel segar C. racemosa yang dimaserasi menggunakan pelarut n-heksana, etil asetat, dan metanol. Uji antibakteri dilakukan dengan metode Resazurin Microtitter Assay (REMA) terhadap bakteri Escherichia coli dan Staphylococcus aureus. Aktivitas antioksidan diuji menggunakan metode 1,1-diphenyl-2-picrylhydrazyl (DPPH) dan Ferric Reducing Antioxidant Power (FRAP). Hasil penapisan fitokimia memperlihatkan bahwa ekstrak n-heksana mengandung alkaloid dan triterpenoid, sedangkan ekstrak etil asetat dan metanol mengandung alkaloid, flavonoid, tanin, fenol, dan steroid. Aktivitas antibakteri terbaik ditunjukkan oleh ekstrak etil asetat dengan nilai Minimum Inhibitory Concentration (MIC) terhadap E. coli sebesar 250 mg/mL. Ekstrak etil asetat C. racemosa juga memperlihatkan aktivitas antioksidan terbaik dengan nilai IC50 DPPH sebesar 110,7 mg/mL dan nilai FRAP sebesar 96,68 mmol Fe (II)/g. ABSTRACTCaulerpa racemosa is a green seaweed that contains active compounds that are potential for antibacterial and antioxidant. The phytochemical constituents and bioactivity of an extract depend on the extraction solvent polarity. This study aimed to determine the phytochemical profiles, antibacterial and antioxidant activity of C. racemosa extracts. Extracts were obtained from the fresh C. racemosa samples that were macerated with n-hexane, ethyl acetate, and methanol. Antibacterial assay was tested by Resazurin Microtiter Assay (REMA) method against Escherichia coli and Staphylococcus aureus. Antioxidant activity was determined using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and Ferric Reducing Antioxidant Power (FRAP) method. Phytochemical screening showed that n-hexane extract contained alkaloid and triterpenoid, while ethyl acetate and methanol extracts contained alkaloids, flavonoids, tannins, phenols, and steroids. Dumortiera hirsuta is one of the liverworts that can be used as a medicinal to prevent infection by pathogenic bacteria. The content of secondary metabolites of D. hirsuta has potential as antibacterial properties includes flavonoids, alkaloids and steroids. This research is to analyze the antibacterial activity of moss D. hirsuta against pathogenic bacteria that will be beneficial to humans. Liverworts of D. hirsuta were extracted using ethyl acetate solvent and tested against three types of pathogenic bacteria using the agar well-diffusion method. The results of this study indicated that the ethyl acetate extract of D. hirsuta at 100% concentration could inhibit the growth of Escherichia coli, Staphylococcus aureus, and Salmonella typhi bacteria. The range of antibacterial activity categories of the ethyl acetate extract of D. hirsuta to E. coli, S. aureus, and S. typhi between weak to moderate.

Abstract This study aimed to compare antioxidant, antibacterial and toxicity tests of various types of kesambi tree bark extract (Schleichera oleosa (Lour) Oken). The research stages included extraction of kesambi stem skin from Mancak Serang sub-district with multilevel maceration techniques using 3 solvents: Methanol, n-hexane, and ethyl acetate. The extract was then tested for antioxidant activity, antibacterial, and toxicity. The results of antioxidant testing using the DPPH method, antibacterial with agar diffusion method, and toxicity test with BSLT method, the results showed that ethyl acetate extract had better antioxidant activity (IC50 7,723 ppm.) compared to extract of methanol (IC50 7,801 ppm), and n-hexane extract (IC50 8,568 ppm). Antibacterial activity showed the ability to inhibit the growth of Streptococcus aureus compared to Escherichia coli at a concentration of 10000 ppm, and the results of the toxicity test showed that ethyl acetate extract had better activity (LC50 305,17 ppm) than n-hexane extract (LC50 374, 96 ppm) and methanol extract (LC50 431,26 ppm).Keywords: DPPH, BSLT, Agar Diffusion, Schleichera oleosa (Lour) Oken

Antibacterial Extract of Mangosteen (Garcinia mangostana L.)Fruit Shell           The skin of the mangosteen fruit is extracted with n-hexane and ethyl acetate to determine the Minimum Inhibitory Concentration (MIC) against Staphylococcus aureus ATCC 25923 and Pseudomonas aeruginosa ATCC 27 853. Results showed that n-hexane extract gave inhibition zone larger than the ethyl acetate extract on all concentrations . Extract n-hexane has a value of MIC against S. aureus ATCC bacterial test 25923 62.5 mg / ml while the ethyl acetate extract of 125 mg / ml . N- hexane extracts had MIC values of the test bacteria P.aeuroginosa ATCC 27 853 was 125 mg / ml , and while the ethyl acetate extract had a MIC value of 500 mg / ml . Treatment of solvent, concentration and interaction between the solvent and concentration significantly affected the test bacteria ATCC 25923 S. aureus at the level of 5 %, the highest interaction N-Hexane solvent with a concentration of 15,625 mg / ml and was not significantly different interactions with the concentration of 31.25 mg/ml and 125 mg/ml. Treatment solvent and concentration significantly while the interaction between the solvent and the concentration has no effect on the test bacteria P.aeuroginosa ATCC 27 853 at 5% level .Keywords: Garcinia mangostana L., Staphylococcus aureus and Pseudomonas aeruginosa ABSTRAK           Kulit buah manggis diekstrak dengan n-heksan dan etil asetat   untuk mengetahui Konsentrasi Hambat Minimum (KHTM) terhadap bakteri Staphylococcus aureus ATCC 25923 dan Pseudomonas aeruginosa ATCC 27853. Hasil penelitian menunjukkan bahwa ekstrak n-heksana  memberikan zona hambatan lebih besar dibandingkan dengan ekstrak etil asetat pada semua konsentrasi. Ekstrak n-heksana  memiliki nilai kadar hambat minimal (KHM) terhadap bakteri uji S. aureus ATCC 25923  62,5 mg/ml sedangkan ekstrak etil asetat 125 mg/ml.   Ekstrak n-heksana memiliki nilai KHM   terhadap bakteri uji P.aeuroginosa  ATCC 27853 adalah 125 mg/ml dan  sedangkan ekstrak etil asetat memiliki nilai KHM  500 mg/ml. Perlakuan pelarut, konsentrasi dan interaksi antara pelarut dan konsentrasi  berpengaruh nyata terhadap bakteri uji S. aureus ATCC 25923 pada taraf 5%, interaksi tertinggi yaitu pelarut N-Heksan dengan konsentrasi 15,625mg/ml dan interaksi ini tidak berbeda nyata dengan konsentrasi 31,25 mg/ml dan 125 mg/ml. Perlakuan pelarut dan konsentrasi  berpengaruh nyata sedangkan interaksi antara pelarut dan konsentrasi  tidak berpengaruh terhadap bakteri uji P.aeuroginosa  ATCC 27853 pada taraf 5%. Kata kunci:  Garcinia mangostana L., Staphylococcus aureus dan Pseudomonas aeruginosa

This study aims to examine the antibacterial activity of n-hexane, ethyl acetate, and n-butanol extract of tenggulun leaves (Protium javanicum Burm. F.) in inhibiting the growth of S. aureus bacteria and determine the minimum inhibitory concentration of the most active extracts and identify its active compounds. A total of 1000 g of tenggulun leaf powder was macerated with methanol and 91.78 g of crude methanol extract was obtained which was further partitioned into n-hexane, ethyl acetate, and n-butanol extracts. Antibacterial activity test was carried out using the diffusion well method and the result showed that the ethyl acetate extract at a concentration of 20% was strongly able to inhibit the growth of S. aureus bacteria with inhibition zones of 20.08 mm, followed by n-butanol 16.66 mm, and n-hexane 13.33 mm. The minimum concentration of ethyl acetate extract was 0.2% in inhibiting the growth of S. aureus with inhibition zone diameter of 6.20 mm. Separation of the active ethyl acetate extract was conducted by silica gel column chromatography with mobile phase of n-hexane: ethyl acetate (7.5: 2.5) gave 6 combined fractions. Fractions B, G, H and I showed antibacterial activity at concentrations of 50% with a diameter of inhibition zone of 5.62 mm, 6.87 mm, 8.50 mm, and 6.75 mm respectively. The results of mass spectra analysis from the chromatogram peaks of LC-MS / MS suggested that the G and I fractions were tentatively identified as crotarin, benzophenone-2, and medroxyprogesterone acetate which may contributed to the antibacterial activity by acting synergisticly.    Keywords: antibacterial, tenggulun, Staphylococcus aureus, LC-MS/MS.

Background : The treatment of some diseases caused by free radicals and pathogenic bacteria usually by using antioxidants and antibiotics. Due to excessive use of antibiotics and other environmental cues, some bacteria are now resistant to certain antibiotics or even to multiple antibiotics. Some Vibrio cholerae bacterial strains are multiresistant to many antibiotics.Objective : The antioxidant and antibacterial activities of Brugueira gymnorrhiza stem extracts against pathogenic bacteria V. cholerae.Method : The B. gymnorrhiza stem was extracted by gradient maceration method. The DPPH method was used to determine the antioxidant activity and the disc diffusion method was used to determine the antibacterial activities. The column chromatography method was used to fractionate the selective extract with the best activity. The LC-MS/MS method was used to identify the compound obtained from the fraction with the best antioxidant and antibacterial activity.Result : Ethyl acetate extract of B. gymnorrhiza stem had the best antibacterial activity with MIC and MBC values of 62.50 mg/L. Ethyl acetate extract also showed the best value of antioxidant activity as indicated by an IC50 value of 255.03 mg/L. The results of fractions test showed that fraction 3 had the best antibacterial and the best antioxidant activities with both the MIC and MBC values of 7.90 mg/L and IC50 value of 348.91 mg/L, respectively.Conclusion : Ethyl acetate extract of B. gymnorrhiza stem has good potential as antioxidant and antibacterial. The compound which is thought as antioxidant and antibacterial from Ethyl acetate extract is 2-Ethyl-4-methyl-1H-imidazole.

Generally, Gluta renghas L. is known because of its very toxic latex which can cause hard irritations to skin. Although, rengas latex has efficacy as an antibacterial agent. Related to previous research, it has been reported that there are urisol, rengol, glutarengol, laccol, and thitsiol in rengas latex. The woody trunk was reported to contain flavonoid, benzenoid, lipid, and steroid compounds. The leaves, were reported shows that the leaves of G. renghas contain an anticholinesterase substance and can be reduced the rate of hydrolysis of acetylcholine. Extraction of rengas leaves has been carried out. It has been shown phenols, steroids, and coumarins compounds in methanol extract, flavonoids, phenols, saponins and alkaloids compounds in ethyl acetate extract, steroids and alkaloids compounds in hexane extract. In the present, cytotoxicity and antibacterial activity have been tested. The results show that the great cytotoxicity activity by ethyl acetate extracts which have an LC50 value is 123,718 µg/mL (R2 0.9822), while the great antibacterial activity shown by methanol extract 1,000 µg/mL with a diameter of inhibition zone 19.02 mm (S. aureus) and 16.06 mm (E. coli).
