The present research study was designed to evaluate the effect of zinc supplementation on body weight, serum triglyceride, cholesterol, glucose homeostasis, oxidative stress, and hepatic function in mice. Mice were treated with zinc sulphate at an equivalent weight of 6.5 mg/kg-body weight elemental zinc for four weeks. Bodyweight, serum glucose, triglyceride, cholesterol, serum MDA, nitric oxide, vitamin C, and hepatic enzymes level were determined at the end of the study period. Data from this study showed that supplementation with zinc in mice maintained a balanced blood glucose homeostasis throughout the experimental period. Moreover, treatment with zinc showed a significant (p <0.05) decrease in serum triglyceride and cholesterol level along with a decrease in the body weight compared to control. Treatment with zinc significantly attenuated the rate of lipid peroxidation whereas increased the level of vitamin C and NO level. The protective effect of zinc on liver activity was observed. Treatment with zinc showed a strong negative association with serum total cholesterol (r= -0.934, p = 0.02), triglycerides level (r= -0.709, p = 0.05), and body weight (r= -0.899, p = 0.01). The present findings demonstrate that zinc supplementation can be helpful to maintain a glucose homeostasis, ameliorate hyperlipidemia, oxidative stress, and liver dysfunction. Therefore, zinc supplementation can be suggested to alleviate diseases associated with metabolic syndrome and oxidative stress. Dhaka Univ. J. Pharm. Sci. 20(1): 59-66, 2021 (June)

The present study was designed to investigate the effect of Vitamin C (450 mg/kg diet) and sodium selenite (0.5 mg/kg diet) in adult white Leghorn male chickens (30 weeks of age), whose concomitantly exposed to oxidative stress induced by hydrogen peroxide (0.5%) supplemented with drinking water for 6 weeks on reproductive performance. Semen were collected at 0, 2, 4 and 6weeks, The study results showed that hydrogen peroxide treatment caused a significant decrease in the body weight, sperm concentration, mass motility and individual motility during experimental period. Also a decrease in testosterone and testis glutathione concentration at the 6th week of the treatment, accompanied with a significant increase in dead and abnormal sperm percentage, testis malondialdehyde level compared with the control group. Histopathological changes revealed presence of necrosis and sloughing in the epithelial lining of the semineferous tubules and vacuolar degeneration of the supporting cells that fall in the lumen of the semineferous tubules and necrosis of interstitial cells. Vitamin C and sodium selenite with hydrogen peroxide caused a significant increase in body weight, sperm concentration, mass motility and individual motility during the experimental period, testosterone and glutathione level, accompanied with a significant decrease in dead and abnormal sperm percentages and Malondialdehyde level compared with hydrogen peroxide. In addition to the improvement in the histological picture of the semineferous tubules, mitosis germ cells were observed through their arrangement in circular tubules. It was concluded from this study that Vitamin C and Sodium selenite reverse the adverse effects produced by hydrogen peroxide on certain physiological and reproductive aspects in adult male chickens.

The aim of this research was the visual characterization and investigating the effects of Alternaria spp. contaminated wheat grains in the starter stage of broilers nutrition on productive parameters and oxidative stress. The research was divided into two phases. Bunches of wheat in post-harvest period of year 2020 was collected from a various locality in Serbia and Albania. In the first phase, collected samples were visual characterized by Alternaria spp. presence by color measurement methods. Gained results are conferred in the range of the color properties of grain color properties of Alternaria toxins. Wheat grain samples were significantly different (p < 0.05) in terms of all measured color parameters (L*, a*, b*). Classification of field fungi in analyzed wheat grain samples showed that the significant field fungi were Rhizopus spp., followed by Alternaria spp., and Fusarium spp. In the second phase, biological tests with chickens were carried out during the broiler chickens’ dietary starter period in the first 14th days of age. At the beginning of the experiment, a total of 180-day-old Ross 308 strain broilers were equally distributed into three dietary treatments, with four replicates each. Dietary treatments in the experiments were as follows: basal diet without visual contamination of Alternaria spp. with 25% wheat (A1), a basal diet with visual contamination of Alternaria spp. with 25% wheat from Serbia (A2), basal diet with visual contamination of Alternaria spp. with 25% wheat from Albania (A3). The trial with chickens lasted for 14 days. After the first experimental week, wheat infected with Alternaria spp. in treatment A2 and A3 expressed adverse effects. The highest body weight of chickens of 140.40 g was recorded in broilers on control treatment A1 with statistically significant differences (p < 0.05) compared to treatments A2 (137.32 g) and A3 (135.35 g). At the end of the second week of test period, a statistically significant (p < 0.05) difference in body weight of broiler chickens could be noticed. The highest body weight of 352.68 g was recorded in control treatment A1, with statistically significant differences compared to other Alternaria spp. treatments. The lowest body weight of chickens was recorded in treatment A3 (335.93 g). Results of feed consumption and feed conversion ratio showed some numerical differences between treatments but without any statistically significant differences (p > 0.05). Alternaria spp. contaminated diet increased glutathione (GSH), glutathione reductase (GR), alanine aminotransferase (ALT), and aspartate aminotransferase (AST) and decreased peroxidase (POD) and superoxide dismutase (SOD) serum levels, respectively. Built on the achieved results, it can be concluded that the wheat contaminated with Alternaria spp. in broilers nutrition negatively affected growth, decreased oxidative protection and interrupted chicken welfare in the first period of life.

Abstract This study aimed to evaluate the effect of zinc supplementation in various diets on the performance and health status of IPB-D2 chickens. Subsequently, about 180 one-week-old IPB-D2 chickens were used in the study and were fed the treatment diets for 12 weeks. Also, this study employed a completely randomised factorial design with six treatments and three replications of ten birds each. These treatment diets included A1, which contained nutrients based on SNI (2013) without Zn supplementation, A2 which is a combination of A1 and 60 ppm of Zinc, B1 which contained nutrients 5% higher than SNI without Zn supplementation, B2 which is a combination of B1 and 60 ppm Zinc, C1 which contains nutrients 10% higher than SNI without Zn supplementation, and lastly, C2 which is a combination of C1 and 60 ppm of Zinc. Furthermore, the data were analysed using ANOVA, and the parameters observed were the body weight, body weight gain, feed consumption, feed conversion ratio, blood profile and the villi surface area of the jejunum. Consequently, the results showed that C2 significantly (P<0.05) increased the body weight and body weight gain when fed to the finisher and grower chickens, respectively. More so, when the finisher chickens were fed the 10% SNI diet, there was an increased body weight gain and also a significantly (P<0.05) decreased feed conversion ratio. Furthermore, the addition of 60 ppm zinc increased haemoglobin significantly (P<0.05), whereas C2 decreased leukocytes significantly (P<0.05) in chicken blood. However, the treatments had no effect on feed consumption in the villi surface area of the jejunum. Therefore, the study concluded that supplementing the IPB-D2 chickens with 60 ppm zinc in a 10% SNI diet resulted in the best performance and health status.

&nbsp;A 2 &times; 3 factorial design experiment was conducted to evaluate the effect of adding lycopene <br />(0 and 75 mg/kg) and vitamin E (0.50 and 100 mg/kg) to the diet of chickens. Moreover, the study investigated growth traits, oxidative stability and chemical composition of leg meat and the vitamin content of meat and liver. The study was conducted using five hundred and forty Ross 308 male broilers that were assigned to one of the six dietary treatments. Significant interactions between lycopene and vitamin E additions affected the body weight of 21-days-old chickens (P = 0.005), the malondialdehyde content in fresh leg meat (P &lt; 0.001) and leg meat stored for 3 days at temperatures of 2.5 to 4&deg;C (P = 0.032), the cholesterol content in leg meat (P &lt; 0.001) and the lycopene content in liver (P = 0.006). The chickens with the highest body weight were fed 75 mg/kg of lycopene and 50 mg/kg of vitamin E. The vitamin E supplement increased the oxidative stability of fresh and stored leg muscle (P &lt; 0.001). The lowest mean cholesterol value (3.49 g/kg of dry matter) was found out in the meat from broilers that were fed 75 mg/kg of lycopene in contrast to broilers fed the control treatment without lycopene (3.93 g/kg of dry matter). Dietary vitamin E significantly reduced the fat content (P = 0.033) and increased the ash content of&nbsp;leg meat. The highest lycopene concentration in liver (2.82 mg/kg of dry matter) was in chickens that were fed the highest levels of vitamin E and lycopene in contrast with the control group (0.28 mg/kg of dry matter). &nbsp;

The fungicide carbendazim (CBZ) and insecticide chlorpyrifos (CPF) are currently applied together by farmers for the control of pests. Here, we investigated the impacts of 7 days oral co-exposure to 10 mg/kg body weight of CPF and 50 mg/kg body weight of CBZ on selected oxidative stress and antioxidant biomarkers in the liver, kidney, and spleen of female rats. The results showed that while the body weight gain and relative organ weights were not significantly affected after separate exposure to CPF and CBZ, there was a significant decrease in the body weight gain with concomitant increases in the relative kidney and spleen weights of rats treated with the mixture. Also, CPF and CBZ co-exposure significantly increased the levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT), urea, and creatinine ( p < 0.05) when compared with the groups treated with CBZ or CPF alone and the control. The significant decreases in both antioxidant enzymes activities and nonenzymatic antioxidant level following individual administration of CPF and CBZ to rats were intensified in the co-exposure group ( p < 0.05). Additionally, the marked increases in the levels of oxidative stress indices in liver, kidney, and spleen of rats treated with CPF or CBZ alone were intensified in the co-exposure group ( p < 0.05). Histopathologically, co-exposure to CPF and CBZ exacerbates their individual effects on the liver, kidney, and spleen. These findings showed that co-exposure to CPF and CBZ in rats elicited more severe oxidative damage on the liver, kidney, and spleen of the rats, indicative of an additive effect compared to CPF or CBZ alone and as such, may pose a greater environmental risk to humans.

The clinical use of doxorubicin as a potent chemotherapeutic agent is limited due to its dose-dependent cardiotoxicity. Oxidative stress and inflammatory pathways have a pivotal role in doxorubicin-induced cardiotoxicity. Sumatriptan, a 5-hydroxytryptamine (5-HT)1B/1D agonist that is mainly used to relieve migraine pain, has suggested exerting protective effects in numerous pathological conditions through antiinflammatory properties. The aim of the present study was to investigate the effects of sumatriptan on doxorubicin-induced cardiotoxicity and the contribution of anti-inflammation and antioxidative responses. Cardiotoxicity was induced by the administration of doxorubicin three times a week (2.5 mg/kg i.p) for two consecutive weeks on male rats. The animals were divided into four groups, including Control, Sumatriptan (0.1 mg/kg) received group, doxorubicin received group, and Doxorubicin+Sumatriptan (0.1 mg/kg) received group. Sumatriptan was administered 30 min before every injection of doxorubicin. On the last day of the second week, the body weight, mortality rate, electrocardiogram (ECG) and histopathological changes, cardiac inotropic study, and biochemical factors were evaluated. The loss of body weight, mortality rate, ECG parameters, reduction of papillary muscle contractility force as well as histopathological scores following administration of doxorubicin indicated severe cardiac damage. However, treatment with sumatriptan inhibited the functional and structural impairment induced by doxorubicin. In addition, sumatriptan could significantly reduce cardiac tissue levels of malondialdehyde (MDA) and tumor necrosis factor-alpha (TNF-α), which were increased in the doxorubicin-treated rats. This study illustrated the protective effects of sumatriptan on decreasing doxorubicin-induced cardiac toxicity and mortality rate in part through inhibition of inflammatory and oxidative stress pathways.

Accumulating evidences have reinforced the use of medicinal plants in the treatment of various ailments as a result of negative side effects associated with conventional drugs. Plant components such as phenols and flavonoids with antioxidant potential have confirmed protective roles against oxidative stress-induced degenerative diseases like diabetes mellitus (DM). The current study was carried out to investigate the effect of seed pod ethanol extract from Copaifera salikounda (SPEECS) in alloxan-induced diabetic rats. SPEECS was obtained by maceration of seed pod powder in absolute ethanol for 72 h, filtered, concentrated and dried in-vacuo. Gas chromatography-mass spectrometry (GC–MS) technique was used to quantitatively elucidate the chemical constituents of SPEECS. Twenty-four male albino rats were randomly allocated into four groups (n=6): normal control, DM control, DM + 200 mg/kg SPEECS and DM + 400 mg/kg SPEECS groups. DM was induced in the Wistar albino rats through intraperitoneal injection of 200 mg/kg body weight of alloxan. After 14 days of treatment, the body weight changes and the fasting blood glucose level were determined in the different groups. Also, serum biochemical parameters such as alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), albumin (ALB), total protein (TP), malondialdehyde (MDA), superoxide dismutase (SOD) and catalase (CAT) were estimated. The GC-MS results confirm nine bioactive compounds with 9-octadecenoic acid (55.75%) being most abundant. SPEECS (200 and 400 mg/kg) administration significantly (P 0.05) caused gain in weight, decreased fasting blood glucose and reversed the elevated liver function enzymes (ALT, AST, ALP) while total TP and ALB were markedly elevated relative to DM control group. Furthermore, SPEECS attenuated the activities of SOD and CAT while the level of MDA was significantly (P 0.05) decreased in dose dependent manner in comparison to the DM control. This study indicated that SPEECS can alleviate hyperglyceamia of DM. Key words: Copaifera salikounda; oxidative stress; medicinal plants; diabetes mellitus; phytochemicals; orthodox ANTIDIABETIČNI UČINEK EKSTRAKTA ETANOLA Copaifera salikounda (HECKEL) NA SLADKORNO BOLEZEN, SPROŽENO Z ALLOXAN-om, PRI PODGANAHIzvleček: Obstaja vedno več dokazov, ki poudarjajo uporabnost zdravilnih rastlin pri zdravljenju različnih bolezni, tudi zaradi različnih negativnih stranskih učinkov, povezanih s konvencionalnimi zdravili. Rastlinske sestavine kot so fenoli in flavonoidi z antioksidativnim potencialom, imajo po nekaterih raziskavah zaščitno vlogo pred degenerativnimi boleznimi, ki jih povzroča oksidativni stres, kot je sladkorna bolezen diabetes mellitus (DM). Študija je bila izvedena z namenom raziskovanja učinka etanolnega semenskega ekstrakta iz rastline Copaifera salikounda (SPEECS) pri podganah s sladkorno boleznijo, ki jo je povzročil alloxan. SPEECS je bil pridobljen z maceracijo praška semen v prahu v absolutnem etanolu 72 ur ter nadaljnjo filtracijo, koncentracijo in sušenjem v vakuumu. Za kvantitativno ugotavljanje kemijskih sestavin SPEECS je bila uporabljena tehnika plinske kromatografije in masne spektrometrije (GC-MS). Štiriindvajset samcev podgan Wistar je bilo naključno razporejenih v štiri skupine (n=6): normalna kontrola, kontrola DM, DM + 200 mg/kg SPEECS in DM + 400 mg/kg SPEECS. DM je bil pri podganah sprožen z intraperitonealno injekcijo 200 mg/kg telesne mase alloxana. Po 14 dneh zdravljenja so bile pri različnih skupinah določene spremembe telesne teže in nivo glukoze v krvi (na tešče). Poleg tega so avtorji raziskave izmerili še nekatere serumske biokemične parametre kot so ravni alaninske aminotransferaze (ALT), aspartatne aminotransferaze (AST), alkalne fosfataze (ALP), albumina (ALB), skupnih proteinov (TP), malondialdehida (MDA), superoksiddismutaze (SOD) in katalaze (CAT). Rezultati GC-MS so v izvlečku SPEECS pokazali devet bioaktivnih spojin, v katerih je največ 9-oktadecenojske kisline (55,75%). SPEECS (200 in 400 mg/kg) je povzročil znatno (P 0,05) povečanje telesne mase, znižanje glukoze v krvi na tešče in znižal raven encimov pokazateljev jetrne funkcije (ALT, AST, ALP), medtem ko je bila raven TP in ALB pri podganah, ki so prejemale SPEECS izrazito povišana v primerjavi z DM kontrolno skupino. Zdravljenje s  SPEECS je tudi oslabilo aktivnosti SOD in CAT, medtem ko se je raven MDA znatno zmanjšala (P 0,05) v primerjavi s kontrolno skupino DM. This study was to study the effect of adding Lepidium sativum to the lamb diets of Awassi sheep. Nine male lambs (their age is 3 months old and their body weight is 19±0.5 Kg) were distributed randomly into 3 treatments (3 lambs per treatment). The coarse feed (hay) was provided freely to lambs, while the concentrated feed was provided based on 3% of the body weight. All treatments were fed on similar diet and differed only in the weight of seeds, where 7.5 g was added to the second treatment (T2) and 15 g of Garden Cress to the third treatment (T3) while the control treatment (T1) left without adding seeds. The experiment continued for 60 days. The results showed a significant increase in pH at 0 hour in treatments T2 and T3 with Garden Cress of 7.5 and 15 g respectively, but there was a significant decrease at 3 hours in treatment T3 and also at 6 hours in T2 and T3 as well. For total phenols there was a decrease at 6 hours in treatment T2.When adding Garden Cress to concentrated rations, it did not significantly affect N-NH3, but improved somewhat of rumen fermentation and microorganism’s numbers at 3 hours for T2 and T3 with Garden Cress of 7.5 and 15 g respectively. There was a significant increase in fatty acids at 6 hours in T2 and T3 while there was no significant effect at other times and treatments.

Ayurveda is the science of life. The main aim of Ayurveda is to maintain the health of a healthy person and to cure the diseased person. For the maintenance of the health of a healthy person, rasayan chikitsa is advised in Ayurve- da. Rasayan chikitsa is the separate branch of Ayurveda, which deals with various aspects of preventive health care. In today’s modern era, the incidence of lifestyle disorders like hypertension, obesity, diabetes mellitus and cardiovascular diseases increased, because of altered dietary and living patterns. Oxidative stress may be respon- sible for most of the diseases. Lack of dietary antioxidants supplements and generation of free radicals induces disease associated with oxidative stress. Oxidative stress is essentially an imbalance between the production of free radicals and the ability of the body to counteract or detoxify their harmful effects through neutralization by antioxidants. Free radicals contain unpaired electrons, and they are unstable and highly reactive. They donate or accept an electron from other molecules, therefore behaving as oxidants. An antioxidant is a stable molecule, enough to donate an electron to rampaging free radical and neutralize it, thus reducing its capacity to damage. Ac-cording to Ayurveda, Amalaki is the best rasayan, tridoshhar. As per modern science, it contains the largest amount of vitamin C (6oomg), and vitamin C has the best antioxidant property. Amalaki rasayan plays an im- portant role as an antioxidant and thus relives oxidative stress, thus helpful in the prevention of many diseases. The aim of the study was to determine the effect of vitamin C on indicators of gilts and sows reproductive value. The study was conducted using 64 gilts of the Pulawska breed. The experimental gilts received a supplement of vitamin C in an amount of 2.4 g daily throughout the whole experiment. Sixteen gilts from both control and experimental groups were mated in their third estrus. In order to determine the number of corpora lutea and the state of development of the reproductive organs, other gilts from the control and experimental groups were slaughtered after third estrus between the 5th and 10th day of the following estrous cycle. The results showed that gilts from the experimental group were characterized by higher potential fertility, as well as reared more piglets per litter. In the body weight and backfat thickness of sows during the first reproduction cycle in the control and experimental groups were not statistically significantly different. The piglets in the experimental group had higher body weight in the 1st and 21st day of rearing. Larger contents of protein, immunoglobulins, and vitamin C were recorded in the colostrum and milk from experimental sows.
