This study was conducted to evaluate the effects of dietary lysozyme (LZM) supplementation on the vaginal microbiota, as well as the relationship between vaginal microbiota and the fecal microbiota of rectum and the reproductive performance of the sow. A total of 60 Yorkshire × Landrace sows (3–6 of parity) were arranged from day 85 of gestation to the end of lactation in a completely randomized design with three treatments (control diet, control diet + lysozyme 150 mg/kg, control diet + lysozyme 300 mg/kg). The results showed that sows fed with lysozyme increased serum interleukin-10 (IL-10, p < 0.05) on day 7 of lactation. The vaginal microbiota varied at different taxonomic levels with LZM supplementation by 16S rRNA gene sequencing. The most representative changes included a decrease in Tenericutes, Streptococcus, Bacillus and increase in Bacteroidetes, Actinobacteria, Enterococcus, and Lactobacillus (p < 0.05). There were 777 OTUs existing in both, vaginal and fecal microbiota. The addition of LZM also decreased the abundance of Tenericutes (p < 0.05) in the vagina and feces. The changes in the microbiota were correlated in some cases positively with the performance of the sow, for example, Bacillus in feces was positively correlated with the neonatal weight (p < 0.05). These results indicate that the addition of lysozyme to the diet of sow during perinatal period promote the change of vaginal bacterial community after farrowing. The variations in vaginal microbiota are also associated with the changes in the fecal microbiology of the rectum and the reproductive performance of the sow. Therefore, it is concluded that dietary supplementation with lysozyme in sows in late gestation stage until early lactation, is beneficial to establish vaginal microbiota that seems to promote maternal health and reproductive performance.

Abstract Most sows are fed a constant energy and amino acid supply throughout gestation, in line with the recommendations of most authorities and swine genetic companies. These recommendations for sow feeding have seen little change in decades, despite the many ways that sows have changed dramatically in reproductive performance. Beginning in about the year 2000, sow litter size has steadily increased as a result of genetic selection. With this increase in litter number has been a steady decline in birth weight, and the resulting negative effects of lower birthweight on subsequent piglet performance. Many experiments using so-called ‘bump’ feeding, or increased energy intake in late gestation, have been conducted in attempts to arrest this decline in birthweight and piglet performance. Generally, these experiments have shown little to no improvement in birthweight and often have negative effects on sow feed intake during gestation. These experiments have ignored the fact that the energy:amino acid ratios (lysine, threonine, isoleucine, tryptophan) in late gestation are different than during early and mid-gestation. In recent research in Australia we hypothesised that rapidly increasing essential amino acid levels in late gestation would increase birth weight and potentially improve subsequent reproductive performance. Three hundred and thirty-four multiparous PIC sows (average parity 3.6, average LW 261 kg) were housed in a dynamic gestation pen after mating and randomly assigned to one of two diet regimes. Two 13.5 MJ/kg DE gestation diets were formulated and created by blending in an ESF. The Control diet contained 0.48 g SID lysine per MJ DE and SID threonine, methionine+ cysteine, isoleucine and tryptophan at 68%, 65%, 58% and18% of SID lysine and offered at 2.2kg/day from d 28 to d 110. Sow were then moved to the farrowing house and placed on a lactation diet at 3.5kg/d. The Treatment diet contained 0.55 g SID lysine/MJ DE and SID threonine, methionine+cysteine, isoleucine and tryptophan at 78%, 65%, 60% and 20% of SID lysine and offered at 2.1kg/d from d 28 to d 85 and then increased to 2.4 kg/d to d 110 d. Increasing essential amino acid levels in late gestation increased gestational weight gain (5.6 kg, P=0.004), increased total litter birth weight (1.25 kg, P=0.003), and increased the birthweight of liveborn pigs from 1.286 to 1.329 kg, (P=0.04). There was no significant effect on the total number born or born alive. Piglet performance is not available because this commercial farm practices cross-fostering. Effects of continuation of this feeding regime in the same sows during subsequent parities is currently being evaluated.

AbstractThis study describes the associations between fecal microbiota and vaccine response variability in pigs, using 98 piglets vaccinated against the influenza A virus at 28 days of age (D28) with a booster at D49. Immune response to the vaccine is measured at D49, D56, D63, and D146 by serum levels of IAV-specific IgG and assays of hemagglutination inhibition (HAI). Analysis of the pre-vaccination microbiota characterized by 16S rRNA gene sequencing of fecal DNA reveals a higher vaccine response in piglets with a richer microbiota, and shows that 23 operational taxonomic units (OTUs) are differentially abundant between high and low IAV-specific IgG producers at D63. A stronger immune response is linked with OTUs assigned to the genus Prevotella and family Muribaculaceae, and a weaker response is linked with OTUs assigned to the genera Helicobacter and Escherichia-Shigella. A set of 81 OTUs accurately predicts IAV-specific IgG and HAI titer levels at all time points, highlighting early and late associations between pre-vaccination fecal microbiota composition and immune response to the vaccine.

ABSTRACTOne of the main challenges in understanding the composition of fecal microbiota is that it can consist of microbial mixtures originating from different gastrointestinal (GI) segments. Here, we addressed this challenge for broiler chicken feces using a direct 16S rRNA gene-sequencing approach combined with multivariate statistical analyses. Broiler feces were chosen because of easy sampling and the importance for pathogen transmission to the human food chain. Feces were sampled daily for 16 days from chickens with and without a feed structure-induced stimulation of the gastric barrier function. Overall, we found four dominant microbial phylogroups in the feces. Two of the phylogroups were related to clostridia, one to lactobacilli, and one toEscherichia/Shigella. The relative composition of these phylogroups showed apparent stochastic temporal fluctuations in feces. Analyses of dissected chickens at the end of the experiment, however, showed that the two clostridial phylogroups were correlated to the microbiota in the cecum/colon and the small intestine, while the upper gut (crop and gizzard) microbiota was correlated to the lactobacillus phylogroup. In addition, chickens with a stimulated gizzard also showed less of the proximate GI dominating bacterial group in the feces, supporting the importance of the gastric barrier function. In conclusion, our results suggest that GI origin is a main determinant for the chicken fecal microbiota composition. This knowledge will be important for future understanding of factors affecting shedding of both harmful and beneficial gastrointestinal bacteria through feces.

The purpose of this study was to evaluate the effects of dietary supplementation of Quillaja saponin (QS) on growth performance, nutrient digestibility, fecal microbiota, and fecal gas emission in growing pigs. A total of 50 crossbred growing pigs [(Yorkshire × Landrace) × Duroc] with an initial body weight of 23.83 ± 1.95 kg were randomly assigned to 1 of 2 treatments for a 56-day trial with 5 replicate pens per treatment and 5 pigs (2 barrows and 3 gilts) per pen. Dietary treatments including control diet and control diet supplemented with 200 mg/kg QS. The average daily gain was significantly increased during days 0-56, while the fecal ammonia emission on day 56 and fecal coliform bacteria counts on day 28 were significantly decreased in pigs fed with QS containing diet. However, dietary supplementation of QS had no significant effects on apparent total tract digestibility. In conclusion, dietary supplementation of 200 mg/kg QS had beneficial effects on growth performance, fecal microbiota, and fecal gas emission in growing pigs. Considering the carry-over effects, the adaption period should be at least 28 days when supplementing 200 mg/kg QS to the diet of growing pigs for improving the growth performance.

Abstract Background The causes of gestational diabetes mellitus (GDM) are still unclear. Recent studies have found that the imbalance of the gut microbiome could lead to disorders of human metabolism and immune system, resulting in GDM. This study aims to reveal the different gut compositions between GDM and normoglycemic pregnant women and find the relationship between gut microbiota and GDM. Methods Fecal microbiota profiles from women with GDM (n = 21) and normoglycemic women (n = 32) were assessed by 16S rRNA gene sequencing. Fasting metabolic hormone concentrations were measured using multiplex ELISA. Results Metabolic hormone levels, microbiome profiles, and inferred functional characteristics differed between women with GDM and healthy women. Additionally, four phyla and seven genera levels have different correlations with plasma glucose and insulin levels. Corynebacteriales (order), Nocardiaceae (family), Desulfovibrionaceae (family), Rhodococcus (genus), and Bacteroidetes (phylum) may be the taxonomic biomarkers of GDM. Microbial gene functions related to amino sugar and nucleotide sugar metabolism were found to be enriched in patients with GDM. Conclusion Our study indicated that dysbiosis of the gut microbiome exists in patients with GDM in the second trimester of pregnancy, and gut microbiota might be a potential diagnostic biomarker for the diagnosis, prevention, and treatment of GDM.

AbstractGut microbiota dysbiosis has a significant role in the pathogenesis of metabolic diseases, including obesity. Nuciferine (NUC) is a main bioactive component in the lotus leaf that has been used as food in China since ancient times. Here, we examined whether the anti-obesity effects of NUC are related to modulations in the gut microbiota. Using an obese rat model fed a HFD for 8 weeks, we show that NUC supplementation of HFD rats prevents weight gain, reduces fat accumulation, and ameliorates lipid metabolic disorders. Furthermore, 16S rRNA gene sequencing of the fecal microbiota suggested that NUC changed the diversity and composition of the gut microbiota in HFD-fed rats. In particular, NUC decreased the ratio of the phyla Firmicutes/Bacteroidetes, the relative abundance of the LPS-producing genus Desulfovibrio and bacteria involved in lipid metabolism, whereas it increased the relative abundance of SCFA-producing bacteria in HFD-fed rats. Predicted functional analysis of microbial communities showed that NUC modified genes involved in LPS biosynthesis and lipid metabolism. In addition, serum metabolomics analysis revealed that NUC effectively improved HFD-induced disorders of endogenous metabolism, especially lipid metabolism. Notably, NUC promoted SCFA production and enhanced intestinal integrity, leading to lower blood endotoxemia to reduce inflammation in HFD-fed rats. Together, the anti-obesity effects of NUC may be related to modulations in the composition and potential function of gut microbiota, improvement in intestinal barrier integrity and prevention of chronic low-grade inflammation. This research may provide support for the application of NUC in the prevention and treatment of obesity.

Dietary phenolics or polyphenols are mostly metabolized by the human gut microbiota. These metabolites appear to confer the beneficial health effects attributed to phenolics. Microbial composition affects the type of metabolites produced. Reciprocally, phenolics modulate microbial composition. Understanding this relationship could be used to positively impact health by phenolic supplementation and thus create favorable colonic conditions. This study explored the effect of six stilbenoids (batatasin III, oxyresveratrol, piceatannol, pinostilbene, resveratrol, thunalbene) on the gut microbiota composition. Stilbenoids were anaerobically fermented with fecal bacteria from four donors, samples were collected at 0 and 24 h, and effects on the microbiota were assessed by 16S rRNA gene sequencing. Statistical tests identified affected microbes at three taxonomic levels. Observed microbial composition modulation by stilbenoids included a decrease in the Firmicutes to Bacteroidetes ratio, a decrease in the relative abundance of strains from the genus Clostridium, and effects on the family Lachnospiraceae. A frequently observed effect was a further decrease of the relative abundance when compared to the control. An opposite effect to the control was observed for Faecalibacterium prausnitzii, whose relative abundance increased. Observed effects were more frequently attributed to resveratrol and piceatannol, followed by thunalbene and batatasin III.
