Seaweeds are an important ingredient of functional foods recommended for daily food, due to their unique compositions and nutritional value. Padina tetrastromatica is a brown edible seaweed that is commonly found along the coastal regions of Peninsular Malaysia and consumed as food by some coastal communities. This study investigates the nutritional and antihyperglycaemic potential of P. tetrastromatica extracts, which is generally accepted as an important functional food. In our methodology, we induced diabetes intraperitoneally in experimental animals with a dose of 65 mg kg−1 body weight of streptozotocin. Oral treatment with 200 and 400 mg kg−1 of P. tetrastromatica ethanolic and ethyl acetate extracts were initiated, respectively, to experimental rats once daily for 18 days. Metformin was used as the positive control. Biochemical estimations and histopathological analysis were included in this study. Treatment with P. tetrastromatica extracts significantly lowered the plasma glucose levels in Streptozotocin-induced diabetic rats. In addition, P. tetrastromatica extract treatment also showed a significant reduction in serum alanine transaminase levels. However, no significant changes were observed in serum aspartate transaminase levels. The ethyl acetate extract of P. tetrastromatica at 400 mg kg−1 dose shows some nephroprotective effect, which is observed from the significant increase in the plasma albumin levels. Histopathological evaluation revealed no marked morphological changes in tissues of the isolated organs of the ethyl acetate extract-treated group, revealing the safe nature of P. tetrastromatica.

Background: The only remedy for up surging problem of antibiotic resistance is the discovery of antibacterial agents of natural origin. Objective: The present study was aimed at finding antibacterial potential of crude and solvent extracts of mature leaves of Plumeria pudica. Methods: Antibacterial activity of three different solvent extracts were evaluated in four human and four fish pathogenic bacteria by measuring the zone of inhibition and determining Minimum Inhibitory Concentration and Minimum Bactericidal Concentration values. Standard antibiotics were used as positive control. Preliminary phytochemical screening of most effective extract i.e., ethyl acetate extract, Fourier Transform Infra Red analysis and GC-MS analysis of the Thin Layer Chromatographic (TLC) fraction of ethyl acetate extract were done meticulously. All experiments were done thrice and analyzed statistically. Results: Crude leaf extracts and solvent extracts caused good inhibition of bacterial growth in all selected bacteria. Ethyl acetate extract showed highest inhibition zones in all tested strains with maximum inhibition (19.50±0.29 mm) in Escherichia coli (MTCC 739). MBC/MIC of the extracts indicated that all three solvent extracts were bactericidal. Preliminary phytochemical tests revealed the presence of tannins, steroids and alkaloids and FT-IR analysis revealed presence of many functional groups namely alcoholic, amide, amine salt and aldehyde groups. From the GC-MS analysis of TLC fraction of ethyl acetate extract five different bioactive compounds e.g., 2,4-ditert –butylphenyl 5-hydroxypentanoate, Oxalic acid; allyl nonyl ester, 7,9-Ditert-butyl-1-oxaspiro(4,5)deca-6,9-diene-2,8-dione, Dibutyl phthalate and 2,3,5,8-tetramethyl-decane were identified. Conclusion: Leaf extracts of P. pudica contain bioactive compounds that can be used as broad spectrum bactericidal agent.

Background: In recent years, the study of the structure and biological activity of medicinal plants has a particularly important to search for diabetes medicine. Ruellia tuberosa is used to treat various diseases such as diabetes by inhibiting the activity of α-glucosidase. Objective: In this study, experiment was designed to isolated isolate and identified identify α-glucosidase inhibitory extracts and compounds from Ruellia tuberosa L. through bio-assay guided isolation. Method: Dry powder of Ruellia tuberosa L. was extracted with 70% ethanol, then liquid-liquid partition with n-hexane, ethyl acetate and butanol, respectively. The extracts were evaluated with α-glucosidase inhibition. The potential extracts were isolated and identified compounds. The effects of these compounds on the α-glucosidase inhibitory were evaluated. Results: The a-glucosidase inhibitory activities showed that the n-hexane, ethyl acetate and the butanol extract had the α-glucosidase inhibition with an IC50 of 46.2 0.9, 6.6 2.9 and 8.9  μg/mL, respectively. From the n-hexane and ethyl acetate extracts, the structures of four compounds were elucidated by NMR spectroscopic method, including lupeol (1), syringaresinol (2), apigenin (3), verbascoside (4). The a-glucosidase inhibitory activities showed that all isolated compounds were more active than the positive control - acarbose with an IC50 of 37.5  0.4; 19.5  0.2; 20.1  0.3; 69.3  0.2 µg/mL, respectively. Conclusion: The ethyl acetate extract showed strong activity about 19 times more than positive control - acarbose. The compound syringaresinol (2) was the most powerful α-glucosidase inhibitor. Therefore, Ruellia tuberosa L. contains many compounds that can inhibit α-glucosidase activity.

Malacca is one of traditional medicine that possesses a potent antimicrobial activity. This study aims to determine the inhibitory activity of Malacca leaf extract on the growth of Salmonella sp in vitro. The bacteria was obtained from Microbiology Laboratory of the Faculty of Veterinary Medicine, Universitas Syiah Kuala. The study was conducted using n-hexane extract, ethyl acetate extract and ethanol of malacca leaves with dilution concentrations of 5%, 25%, and 50%.The inhibitory property of malacca leaf was tested using Kirby-Bauer method. Data were analyzed descriptively. The results of this study indicate that n-hexane extract, ethyl acetate extract and ethanol extract of malacca leaves can inhibit the growth of Salmonella sp. The n-hexane extract of malacca leaves showed a greater inhibition than the ethyl acetate and ethanol extract of malacca leaves. n-hexane extract with a concentration of 5%, 25%, and 50% showed average diameter inhibition of 1.35 mm (weak), 4.97 mm (moderate), and 12.87 mm (strong), respectively ethyl acetate extract with a concentration of 5%, 25%, and 50% showed average diameter inhibition of 2.00 mm (weak), 5.72 mm (moderate), and 7.58 mm (moderate), whereas in ethanol extract were 0.47 mm (weak), 2.58 mm (weak), and 4.35 mm (weak), repectively. The clear zone areas in negative and positive control were 0.00 mm 20.00 mm, respectively. Malacca leaf extract possess inhibitory property against the growth of the Salmonella sp.

Abstract Investigations were carried out to determine the antiulcerogenicity of the flavonoid fraction (ethyl acetate extract) of Erica andevalensis Cabezudo-Rivera on gastric ulceration induced by different experimental models. Oral treatment with the ethyl acetate extract and the major flavonoid (myricetin 3-O-ᴅ-galactoside) were found to be effective to prevent gastric ulceration induced by cold-restraint stress in rats . Statistically significant ulcer index values with respect to the control group were observed. Mucus content was not increased although it was accompanied by an increase in proteins and hexosamines. In pyloric-ligated animals flavonoids showed a significant reduction in the number and severity of the ulcers. Under the same conditions acidity did not decrease with the flavonic extract and myricetin 3-O-ᴅ-galactoside significantly as compared to control. Gastric ulcers induced by oral administration of absolute ethanol were reduced by pretreatment with the flavonoid extract of doses from 125 to 250 mg/kg and the isolated flavonoid of 25 mg/kg p.o. However neither the flavonic extract nor the isolated flavonoid induced changes in the amount and glycoprotein content of gastric mucus.

Objective: The aim of the present study was to investigate the effect of antihyperlipidemic activity of ethyl acetate extract of Commiphora mukul (EECM) gum resin and compares its efficacy with that of atorvastatin against atherogenic diet-induced hyperlipidemia model using rat as an experimental animal.Methods: Wistar albino rats were divided into five groups. Control animals received normal diet, hyperlipidemic control; rats treated with EECM 100 and 200 mg/kg and positive control, receiving standard drug atorvastatin 2.70 mg/kg.Results: Increase the level of serum cholesterol, phospholipids, high-density lipoproteins, low-density lipoproteins, and triglycerides causing hyperlipidemia further leading to the development of atherosclerosis. On the other hand, oral administration of ethyl acetate extract of C. mukul at dose of 200 mg/kg for 15 days resulted in the prevention of above abnormalities.Conclusion: The results suggest that EECM could be beneficial in the treatment of atherosclerosis, characterized by atherogenic lipoprotein profile and abnormalities in lipid metabolism.

Caulerpa racemosa (Forsskal) J. Agardh is a green seaweed used as food and folk medicine since ancient times in the Indo-Pacific region, particularly in southeast Asia. In this study, the proximate nutrient composition, phytochemical, anti-oxidant and anti-diabetic properties of sea grape C. racemosa collected from culture fishponds in Johor, Malaysia were analysed. The contents (dry weight basis) of carbohydrate, crude protein, crude lipids, ash and caloric value obtained were 33.42 ± 1.34%, 20.27 ± 0.14%, 4.20 ± 0.32%, 28.25 ± 0.27% and 2544.67 ± 7.04 cal g−1, respectively. The amino acid score (AAs) and biological protein value (213.43 mg g−1) indicated that C. racemosa presented a better protein quality. The most abundant fatty acids were C16:0 (palmitic acid: 63.27%), followed by C18:1 (oleic acid: 5.80%), and C18:2 ῳ6 (linoleic acid: 5.33%). The analysis of the ash content indicated that essential minerals and trace elements, such as Ca, Fe, and Mn, were present in the seaweed. The total phenolic content (TPC) and total flavonoid content (TFC) observed in the ethyl acetate extract were 17.88 ± 0.78 mg GAE g−1 and 59.43 ± 2.45 mg QE g−1, respectively. The ethyl acetate extract of C. racemosa demonstrated notable anti-diabetic activity in diabetic induced rats. The low (100 mg kg−1) and high (200 mg kg−1) doses of cultivated C. racemosa extract exhibited a significant decrease (p < 0.05) in blood glucose levels while preventing weight loss, reducing plasma AST, ALT levels as a sign of hepatoprotective effect and recording albumin levels similar to positive control in diabetic induced rats. The results support the usefulness of cultivated C. racemosa as a potential functional food.

This study aimed to investigate phytochemical screening and antioxidant activity of n–hexane, ethyl acetate and ethanol extract from lakoocha leaves. The powdered simplicia was macerated with n–hexane, ethyl acetate and ethanol 96% successively, filtered, then concentrated using rotary evaporator to obtain n–hexane extract, ethyl acetate extract and ethanol extract. Phytochemical screening and antioxidant activity was performed against these extracts. Antioxidant activity was determined by DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging method using ultraviolet-visible spectrophotometer at wavelength of 516 nm after incubated for 60 minutes in dark place. Quercetin was used as positive control. The result of phytochemical screening showed n-hexane extract contains steroid, ethyl acetate extract contain steroid, tannin, glycoside, flavonoid and saponin, whereas ethanol extract contain tannin, glycoside, flavonoid and saponin. The IC50 value of n–hexane, ethyl acetate and ethanol extract was 1062.03±1.42 ppm, 323.18±0.02 ppm and 99.23±0.07 ppm respectively, whereas for quercetin was 2.32±0.01 ppm. This study showed that ethanol extract had antioxidant activity with strong category whereas n-hexane extract and ethyl acetate extract had inactive antioxidant activity with very weak categories.       Keyword: Antioxidant Activity, DPPH, Lakoocha leaf

Fifty 1-2-month-old Guinea pigs were divided into 5 equal groups, 10 each. Control (Gp1) did receive neither viable bacteria nor treatment. Each animal from the other groups (Gp2–5) was challenged with (1-2 × 108) viable E. coli in 200 μL normal saline (0.9%) through IP route. GP2 infected group was treated with 200 μL saline IP and kept as positive control group. Gp3-4 are infected and treated with Withania somnifera (ethanol root extract) with doses 50 and 100 mg/kg. BW, respectively. Gp5 infected treated group was treated with cefoperazone antibiotic at dose 35 mg/Kg BW. The treatment by drug or the extracted medicinal plant was started 72 h after infection for 7 successive days. Serum and whole blood sample were collected from all groups 14 days after treatment to evaluate some hematological and biochemical changes as well as immunomodulatory cytokine tumor necrosis factor-alpha (TNF-α). Oral treatment of the plant extract caused significant benefit results in infected Guinea pig appeared in the correction of some hematological and biochemical parameters also try to suppressed inflammatory cytokine response represent in TNF-α. It could be concluded that W. somnifera extract has potent antibacterial activity, and this appears in the correction with hematological, biochemical, and immunological results.

Abstract Rheumatoid arthritis (RA) is a multifactorial disease which is complicated by apoptosis resistance. Autophagy is one of the key mechanisms which are involved in the development of resistance to apoptosis as well as to the standard therapies against RA. Aberration in autophagy and apoptosis homeostasis results in the development of oxidative stress thus complicates the pathogenesis of RA. In the given study, tomorou, an indigenous herb of Hunza-Nagar Valley, has been evaluated for its pro-apoptotic, anti-inflammatory, and anti-rheumatic activity. Several major classes of bioactive phytochemicals including steroids, terpenoids, phenols, flavonoids, and essential oils have been detected in the aqueous and ethyl acetate extracts of tomorou through phytochemical analysis. Plant extracts depicted enhanced free radical scavenging activity through di-phenyl-2-picryl hydrazyl hydrate (DPPH) assay and ameliorated the symptoms of arthritis in collagen induced arthritic (CIA) mice model. Moreover, the 6 week extract treatment resulted in the reduction of IL-6 serum levels thus making it an effective anti-inflammatory agent. Upregulation of microtubule-associated proteins light chain 3b (LC3b) and downregulation of UNC51-like kinase 1 (ULK-1) in arthritic mice proposed a ULK-1 independent non-canonical autophagy pathway. Treatment with extracts upregulated the expression of caspase 3 which in turn inhibited the activity of LC3b thus altering the autophagy pathway. However, ULK-1 expression was restored to normal in aqueous extract treated group whereas it was upregulated in ethyl acetate extract treated group. On the other hand, a novel LC3b-independent autophagy pathway was observed in mice treated with ethyl acetate extract due to ULK-1 upregulation. Despite of significantly high IL-6 levels, the arthritic symptoms waned off which suggested the participation of IL-6 in LC3b-independent autophagy pathway in the extract prepared in ethyl acetate. Conclusively, the study established pro-apoptotic, antioxidant, anti-inflammatory and anti-rheumatic activity of tomorou and suggested an intricate autophagy pathway shift.

Apoptosis is now widely accepted as playing a role in tumorigenesis. An effective compound which can kill tumors via apoptonic pathway appears to be a relevant strategy to suppress various human tumors. The ethyl acetate extract from the stem bark of Cudrania tricupidata (EACT) showed dose- and time-dependent cytotoxic effects on human leukemia HL-60 cells. DNA fragmentation and morphological changes, accompanied by condensed and fragmented nuclei, were observed in the cells cultured for 6 hr with EACT. These results suggest that the cytotoxicity of the crude extract from Cudrania tricuspidata against HL-60 cells is due to apoptosis.
