This study investigated the immunomodulatory effect of Salvia plebeia R. aqueous extract (FIE-SP, SPW) in forced swimming exercise-induced mice and the immunostimulatory effects on Raw264.7 cells. Mice were randomly assigned to four groups: the control group (CON), the forced swimming test group (FST), and two FIE-SP groups (low and high dose of FIE-SP). Compared with the control group, the FIE-SP groups showed significantly increased ratios of T lymphocyte surface markers CD4+/CD8+ and major histocompatibility complex (MHC)I/MHCII, as well as increased concentrations of immunoglobulin (Ig)A and IgG. FIE-SP groups significantly increased Th1 cytokines and decreased Th2 cytokines compared with negative control exercise-induced mice. Conversely, the immunostimulatory effects of FIE-SP significantly increased phagocytic activities, nitric oxide (NO) production, and pro-inflammatory cytokines tumor necrosis factor alpha (TNF-α) and interleukin (IL)-1β in Raw264.7 cells. Furthermore, FIE-SP increased natural killer (NK) cell activities and cytokines (IL-12) in splenocytes compared with the CON group. These results indicated that FIE-SP supplementation could prevent imbalanced immune states and produce immunostimulatory effects to support innate immunity.

The objective of this study was to investigate the protective role of polysaccharide fromGynostemma pentaphyllumMakino (PGP) supplementation against exhaustive swimming exercise-induced oxidative stress. A total of 48 mice were randomly divided into four groups: control, low-dose, medium-dose, and high-dose PGP supplementation groups. The control group received distilled water and the supplementation groups received different doses of PGP (50, 100 and 200 mg/kg body weight) by gavage once a day for 28 consecutive days. After 28 days, the mice performed an exhaustive swimming exercise, and some biochemical parameters related to oxidative stress, including superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase (CAT) and malondialdehyde (MDA), were measured. The results showed that PGP supplementation could increase SOD, GPx and CAT contents, as well as decrease MDA contents in the liver and skeletal muscle of mice, which suggests that PGP supplementation has a protective role against exhaustive swimming exercise-induced oxidative stress.

The purpose of the present study was to investigate the effects of curcumin on physical fatigue and oxidative damage induced by forced swimming exercise in mice. The mice were divided into quiet group and exercise group, and then the above two groups were divided into control group and medication group. The medication group received curcumin (100 mg/kg), while the control group received physiological saline for 28 days. The exercise group performed swimming exercise for 4 weeks and then performed exhaustive exercise. Exhaustive swimming time and superoxide dismutase (SOD) and malondialdehyde (MDA) contents in liver, as well as serum alanine aminotransferase (ALT) contents were measured. The data showed that curcumin could extend exhaustive swimming times of the mice, as well as increase the SOD contents in liver, while decreasing MDA contents in liver and ALT contents in serum. This study indicated that curcumin has anti-fatigue and protective effects against exercise induced oxidative damage.

Polysaccharides, the mainly bioactive ingredient ofRadix Astragali, were evaluated for its effects on the oxidative stress induced by exhaustive swimming exercise of mice. A total of 48 mice were randomly divided into four groups: control group, low-dose polysaccharide fromAstragali radix(RAP) treated group, medium-dose RAP treated group, and high-dose RAP treated group. The control group received only distilled water ig, and the RAP treated groups received different doses of RAP (50, 100, and 200 mg/kg, ig) for 28 days. After the final treatment with RAP, the mice were subjected to swimming to exhaustion and the superoxide dismutase (SOD), glutathione peroxidase (GPx) and catalase (CAT) were measured. The data showed that RAP promote increases in the activities of SOD, GPX and CAT in liver and muscle of mice, and the high-dose RAP (200 mg/kg) presented the best effect. These results indicated that RAP possessed protective effects against exercise-induced oxidative stress.

Irisin is a myokine primarily secreted by skeletal muscles and is known as an exercise-induced hormone. The purpose of this study was to determine whether the PGC-1α -FNDC5 /Irisin-UCP1 expression which is an irisin-related signaling pathway, is activated by an acute swimming exercise. Fourteen to sixteen weeks old male C57BL/6J mice (n = 20) were divided into control (CON, n = 10) and swimming exercise groups (SEG, n = 10). The SEG mice performed 90 min of acute swimming exercise, while control (non-exercised) mice were exposed to shallow water (2 cm of depth) for 90 min. The mRNA and protein expression of PGC-1α, FNDC5 and browning markers including UCP1 were evaluated by quantitative real-time PCR and western blotting. Serum irisin concentration was measured by enzyme-linked immunosorbent assay. An acute swimming exercise did not lead to alterations in the mRNA and protein expression of PGC-1α in both soleus and gastrocnemius muscles, the mRNA and protein expression of UCP1 in brown adipose tissue, mRNA browning markers in visceral adipose tissue and circulating irisin when compared with the control group. On the other hand, an acute swimming exercise led to increases in the mRNA and protein expressions of FNDC5 in the soleus muscle, the protein expression of FNDC5 in the gastrocnemius muscles and the protein expression of UCP1 in subcutaneous adipose tissue.

The aim of the present study is to evaluate the effect of aqueous extract of Thonningia sanguinea on blood electrolytes of broiler chickens. To do this, two hundred (200) 15-day-old Hubbard broiler chickens were divided into 2 batches of 100 chickens each. The chickens in the control group (Batch C) received unsupplemented water while those in the test group (Batch E) received water supplemented with 10 g / L of aqueous extract of T. sanguinea for one week. Then, the zootechnical parameters such as weight growth, weight gain, consumption index and the level of serum electrolytes were determined. The results of the study showed that the aqueous extract of Thonningia sanguinea improved weight growth, weight gain and consumption index. At the same time, the results also showed that T. sanguinea did not significantly (p <0.05) affect the level of blood electrolytes of broilers chickens. From the results obtained in the present study, the aqueous extract of Thonningia sanguinea could be used to improve poultry productivity without damage on blood electrolytes. Background: There are few biomarkers that can be easily accessed in clinical settings and may reflect refractory Th2-eosinophlic inflammation and remodeling of the asthmatic airways. Serum periostin may be one such biomarker to aid our understanding of the patho-bio-physiology of asthma and exercise induced asthma. The aim of the study is to explore the relationship between serum periostin level and exercise induced bronchoconstriction in asthmatic children. Materials and Methods: This cross-sectional study was carried out on (90) children both sexes aged from 6 to 15 years including, (60) children with bronchial asthma and (30) children were enrolled as control group in the period from January 2018 to January 2019. Patients were randomly classified into two groups: I) Patient group: divided into 2 groups according to standardized treadmill exercise challenge test: Group A: (30) asthmatic children with positive test. Group B: (30) asthmatic children with negative test. II-Control group: (30) children apparently healthy with no personal or family history of asthma. All children were subjected to the following Investigations: Chest x-ray, pulmonary functions tests (FEV1 & PEFR) except controls, Laboratory investigations as CBC and Serum periostin level. Results: The mean values of both the percentage of PEFR and FEV1 after exercise in group A were significantly lower than those in group B and the percentage of PEFR and FEV1 after exercise in each group were significantly lower than the percentage before exercise in the same group. The mean value of eosinophilic count in group A was significantly higher than (group B and control group) and the mean value of eosinophilic count in group B was significantly higher than control group. The mean value of serum level of periostin in group A was significantly higher than (group B and control group), however, there was no significant difference between group B and control group as regard to serum level of periostin. Chest tightness, cough and wheezes after exercise and eosinophilic count in patients with high serum periostin level were significantly higher than patients with low serum periostin level, and both PEFR and FEV1 after exercise in patients with high serum periostin level were significantly lower than patients with low serum periostin level. Also the normal serum periostin levels vary among different age groups. Conclusion: Serum periostin level can be considered as a useful biomarker for diagnosis of Exercise induced bronchospasm (EIB) in asthmatic children especially when lung function test cannot be done However, cautious is required in evaluating serum periostin levels in children because it varies with age.

Aim: The aim of this study was to evaluate the supportive effect of human chorionic gonadotropin (hCG) on the quality of spermatogenesis, including count, motility, morphology, viability and apoptosis of sperm following forced swimming exercise. Materials and methods: Twenty-four adult male Sprague-Dawley rats were used in this study. All rats were divided into four groups: control group; swimming exercise group (S); hCG administration group and swimming (SG) with hCG administration group (G). The experimental group was trained to force swimming stress for 10 min for 6 days. Then the sperm quality parameters were measured after dissection and epididymis removal. Spermatogenesis and germ cell apoptosis were evaluated by using Miller &amp; Johnsen&rsquo;s score and TUNEL staining respectively. Results: Results showed the count (control: 113&plusmn;3.1, S: 74&plusmn;1.9, G: 111&plusmn;3, and SG: 103&plusmn;2.4), motility (control:&nbsp; 93&plusmn;2, S: 67&plusmn;2.8,G: 90&plusmn;2.7, and SG: 78&plusmn;1), morphology (control: 89&plusmn;3%, S: 47&plusmn;2.4%, G: 90&plusmn;3.1%, and SG: 67&plusmn;1.1%), and viability of sperm (control: 91&plusmn;2.9, S: 50&plusmn;2, G: 91&plusmn;1.9, and SG: 70&plusmn;1.3) in swimming exercised-hCG administered group, significantly enhanced by hCG treatment compared to the swimming exercise group (p&le;0.01). Also the number of apoptotic germ cells significantly decreased in swimming exercised-hCG administered group compared to the swimming exercised group. Conclusions: These results suggest that administration of hCG can protect the testes against the detrimental effect of forced swimming exercise in adult male rats.

The anti-fatigue effect was investigated of the probiotic supplement, OM-X®, on forced swimming capacity in mice. Mice were administered either vehicle (distilled water; DW) or OM-X® (85 mg/kg body weight) by gavage for 4 weeks. Forced swimming tests were conducted weekly using the Ishihara-modified Matsumoto swimming pool. The endurance swimming time of the final forced swimming exercise in mice fed with OM-X® group showed an approximately 2-fold increase compared with the vehicle control group. Biomedical parameters, including blood lactate, blood superoxide dismutase (SOD) activity, serum triacylglycerol (TG), hepatic total lipids (TL), TG and phospholipid (PL) were significantly lower in mice fed with OM-X® than those in the vehicle control group. Furthermore, the mRNA expression levels of carbamoyl phosphate synthetase 1 (Cps1) and arginase 1 (Arg1), in the urea cycle, were increased by OM-X® feeding. Thus, our findings suggest promotion of lipid metabolism and up-regulation of the urea cycle, at least in part, for the anti-fatigue effect mediated by OM-X®.

In the present study, 30 male rats (age 3 mos, Wt 128.6 ± 3.7 g) were randomly divided into Control group (CG), Experimental group (EG), and Supplemented group (SG), 10 per group. An exercise protocol (3 hrs swimming per day, 5 days a week for 4 weeks) was followed in EG and SG, with no exercise in CG. In SG, α-tocopherol succinate was injected subcutaneouly at a dose of 50 mg•kg−1 per body weight per day. After 4 weeks of exercise, significant diminutions (p <  0.05) were noted in somatic indices of testes and accessory sex organs; seminiferous tubuler diameter (STD); testicular Δ5, 3β-hydroxysteroid dehydrogenase (Δ5, 3β-HSD) 17 β-hydroxysteroid dehydrogenase (17 β-HSD) activities; plasma levels of testosterone (T), luteinizing hormone (LH); preleptotine spermatocytes (pLSc), mid-pachytene spermatocytes (mPSc), and Stage 7 spermatids (7 Sd); testicular α-tocopherol and glutathione (GSH) content; superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPX), and glutathione-s-transferase (GST) activities in EG when compared to CG. Moreover, a significant elevation (p <  0.05) in malondialdehyde (MDA) was found in testes of EG compared to CG. No significant alteration was noted in body weight among the groups. Co-administration of α-tocopherol succinate restored the above parameters. Intensive swimming exercise-induced oxidative stress causes dysfunction in the male reproductive system, which can be protected by α-tocopherol succinate. Background: Ayurvedic literature indicated use of Bryonia laciniosa (BL) for metabolic abnormalities. However only one study evaluated Bryonia laciniosa seed ethanolic extract in streptozotocin-induced diabetes in rats and was found to have anti-hyperglycemic and anti-hyperlipidemic effects. The authors looked at the renal function test in addition and found that it prevents deterioration of renal function in rats. Hence it was interesting to find if Bryonia laciniosa had any protective action in streptozotocin induced diabetic nephropathy in rats.Methods: Streptozotocin induced diabetic nephropathy model in wistar rats was standardised in phase 1. In phase 2 animals were divided into 4 groups and diabetic nephropathy was induce with STZ. Vehicle Control group animals received CMC 0.5%, Positive control group received enalapril 5 mg/kg, Test group 1 received ethanolic extract of Bryonia laciniosa (EEBL) 250 mg/kg and Test group 2 received (EEBL) 500 mg/kg from 3rd day onwards till 6 weeks. Renal function, morphology and histopthological changes were evaluated.Results: There was a significant reduction (p <0.001) in the serum creatinine and BUN in both low and high dose EEBL groups. High dose of EEBL lowered raised MDA values significantly (p<0.001) and raised the GSH values (p<0.001). On histopathology, the high dose EEBL reversed nephropathic changes which were equivalent to enalapril.Conclusions: Bryonia lacinosa in high doses 500 mg/kg has potential to reverse diabetic nephropathic changes induced in wistar rats.
