Infectious bovine keratoconjunctivitis (IBK) is a bacterial eye disease affecting cattle worldwide. The main etiological role in IBK belongs to Moraxella spp. (including in associations with Staphylococcus spp., Streptococcus spp., Pseudomonas aeruginosa, E. coli, Proteus). Persistence factors and antimicrobial resistance of these bacteria, reduce the effectiveness of traditional treatment measures for IBK. New strategies for combating bacterial infections set up prospects for the use of bacteriophages. The paper presents data substantiating the relevance of the development of a therapeutic and prophylactic method of phage processing and an agent based on lytically properties of bacteriophages to combat infectious keratoconjunctivitis in cattle.

The study was conducted to find out the prevalence of sub-clinical mastitis (SCM) and antibiogram of the causative bacteria in dairy cows at the Sylhet govt. Dairy Farm (SGDF) and Local Farms of Sylhet (LFS) during the period of July2014 to June2015. These farms were selected to assess the predominant types of bacteria involved in causing sub-clinical mastitis and to know the in vitro antibiotic sensitivity spectrum of these bacterial isolates against the commonly used antibiotics and to study the economic effects due to SCM. Use of California Mastitis Test (CMT) for the detection of sub-clinical mastitis showed 42out of 100 samples were test positive, among which +(Trace) 22.0%, ++(Distinct) 12.0%, +++(Strong) 3.0%, ±(Doubtful) 5.0%.  Bacteriological examination of milk samples of 100 milch cows (400 quarters) revealed that 42 cows (42.0%) had suffering from SCM with different bacterial infection with 95% confidence limit was 32.1574-51.8426. Among 42 positive samples31 (31.0%) cows had mono-bacterial infection and 11 (11.0%) cows had mixed bacterial infections. Statistical analysis of the result of single and mixed bacterial infections in the milk of apparently healthy milch cows revealed that the single infection was significantly (P < 0.05) higher than mixed bacterial infection. Of the 31 mono-bacterial isolates, of which 23 (23.0%) isolates were Staphylococci, 3 (3.0%) isolates were Escherichia coli, 5 (5.0%) isolated Streptococcus spp. The 11 cows had mixed infection, of which 5 had Staphylococcus spp. + Streptococcus spp., 3 had E. coli + Staphylococcus spp., and 3 had Streptococcus spp. + E. coli. Of 42 positive cases of SCM Staphylococcus spp. isolated from 23 samples. Among these 23 positive samples 13 obtained from SGDF (37.14%) and 10 obtained from LFS (15.38%). Escherichia coli isolated from 3 samples. Among these 3 positive samples 3 obtained from LFS (4.62%). Streptococcus spp. isolated from 5 samples. Among these 5 positive samples 1 obtained from SGDF (2.86%) and 4 obtained from LFS (6.15%). Ceftriaxone, ciprofloxacin and gentamicin were the best drug for treating sub-clinical mastitis.

Accumulation of opportunistic pathogenic bacteria in the aquatic environment may lead to the changes in the structure of the surface cover microbiocenosis as well as the fish intestinal tract. In its turn, it causes the growth of pathological processes in the fish organism, reduces the barrier functions of tissues and mucus and, as a consequence, may induce the spread of bacterial infections. Thus, assessing the state of fish organism, it is necessary to take into account the analysis of the epizootic situation in water (i.e. presence of infectious and invasive diseases of fish, death of fish) as well as condition of the skin, gills and gastrointestinal tract. The results of bacteriological studies of washes from the skin, gills and intestines showed that in the conditions of farming among rainbow trout of different age groups, E. coli, low fermented E. coli, Enterobacter spp., Klebsiella pneumoniae, Staphylococcus spp., Flavibacterium spp., Enterococcus spp. and Citrobacter spp. For the purposes of determining the sensitivity of isolated cultures to antibacterial drugs, it was found that Enterobacteriaceae are sensitive to enrofloxacin, cotrimoxazole, doxacillin, chloramphenicol, gentamicin, ceftriaxone, cefoperazone, ceftazidime and cefpiromycin and resistant to oxytetracycline, amoxicillin and ampicillin sulbactam. E. coli also demonstrated resistance to amoxicillin and cefpirome and sensitivity to enrofloxacin, oxytetracycline, cotrimoxazole, doxacillin, chloramphenicol, ampicillin sulbactam, gentamicin, ceftriaxone, cefoperazone, ceftazidime. Klebsiella pneumoniae was resistant to enrofloxacin, amoxicillin and ampicillin sulbactam, while Flavobacteriaceae were resistant to amoxicillin and ampicillin sulbactam and sensitive to all other antibacterial drugs. Through the direct detection of bacterial volatile organic compounds (VOCs), via surface enhanced Raman spectroscopy (SERS), we report here a reconfigurable assay for the identification and monitoring of bacteria. We demonstrate differentiation between highly clinically relevant organisms: <i>Escherichia coli</i>, <i>Enterobacter cloacae</i>, and <i>Serratia marcescens</i>. This is the first differentiation of bacteria via SERS of bacterial VOC signatures. The assay also detected as few as 10 CFU/ml of <i>E. coli</i> in under 12 hrs, and detected <i>E. coli</i> from whole human blood and human urine in 16 hrs at clinically relevant concentrations of 10<sup>3</sup> CFU/ml and 10<sup>4</sup> CFU/ml, respectively. In addition, the recent emergence of portable Raman spectrometers uniquely allows SERS to bring VOC detection to point-of-care settings for diagnosing bacterial infections.

Amyloid plaques found in the brains of Alzheimer’s disease (AD) patients primarily consists of amyloid beta 1-42 (Ab42). Commercially, Ab42 is synthetized using peptide synthesizers. We describe a robust methodology for expression of recombinant human Ab(M1-42) in Rosetta(DE3)pLysS and BL21(DE3)pLysS competent E. coli with refined and rapid analytical purification techniques. The peptide is isolated and purified from the transformed cells using an optimized set-up for reverse-phase HPLC protocol, using commonly available C18 columns, yielding high amounts of peptide (~15-20 mg per 1 L culture) in a short time. The recombinant Ab(M1-42) forms characteristic aggregates similar to synthetic Ab42 aggregates as verified by western blots and atomic force microscopy to warrant future biological use. Our rapid, refined, and robust technique to purify human Ab(M1-42) can be used to synthesize chemical probes for several downstream in vitro and in vivo assays to facilitate AD research.

The main factor in the treatment of suppurative and inflammatory processes is the timely optimization of treatment measures taking into account the nature of the microflora and its susceptibility to antimicrobial drugs. The purpose of the study is to monitor the spectrum of microorganisms – pathogens of purulent-inflammatory processes of soft tissues in surgical patients; study of the sensitivity of isolated strains to antibiotics. The material was collected in accordance with aseptic rules. The identification of a pure culture of bacteria was carried out according to morphological, culture, biochemical properties, and the presence of virulence enzymes. Sensitivity of bacteria to antibiotics was determined by the standard disks method (by Kirby-Bauer’s). 255 patients with purulent-inflammatory processes of soft tissues were examined for the period from 2014 to 2017. 229 strains of isolated bacteria were included to Escherichia coli, Citrobacer freundii, Enterobacter cloacae, E.aerogenes, Proteus vulgaris, Staphylococcus aureus, S.epidermidis, Streptococcus pyogenes, S.viridians, S.agalactiae, Pseudomonas aeruginosa. The main cause of purulent-inflammatory processes of soft tissues is Staphylococci (67,2%). Compared to previous studies, the number of P.aeruginosa isolated cultures increased (7.9%). In monoculture and in association with other microorganisms, E. coli (9.6% of cases), E.cloacae et aerogenes (3.9% of cases), P.vulgaris (3.9% of cases), C.freundi (2.5% of cases), S.agalactiae, S.pyogenes, S.viridans (3.5%). The number of associated sows reaches 12%. Clinical strains of microorganisms remain most sensitive to fluoroquinolones, cephalosporins, and also retains high sensitivity to gentamicin, lincomycin, rifampicin, which is important for empirical antibiotic therapy. To increase the effectiveness of antibacterial therapy, strict adherence to the mode of appointment of antibiotics, justification of indications, a combination of antibiotics of different spectrum of action, mandatory correction after determining the sensitivity of the pathogen.

Introduction: Bacterial meningitis is an acute central nervous infection with high mortality or permanent neurological sequelae if remained undiagnosed. However, traditional diagnostic methods for bacterial meningitis pose challenge in prompt and precise identification of causative agents. Aims: The present study will therefore aim to set up in-house PCR assays for diagnosis of six pathogens causing the disease including H. influenzae type b, S. pneumoniae, N. meningitidis, S. suis serotype 2, E. coli and S. aureus. Methods: inhouse PCR assays for detecting six above-mentioned bacteria were optimized after specific pairs of primers and probes collected from the reliable literature resources and then were performed for cerebrospinal fluid (CSF) samples from patients with suspected meningitis in Hue Hospitals. Results: The set of four PCR assays was developed including a multiplex real-time PCR for S. suis serotype 2, H. influenzae type b and N. meningitides; three monoplex real-time PCRs for E. coli, S. aureus and S. pneumoniae. Application of the in-house PCRs for 116 CSF samples, the results indicated that 48 (39.7%) cases were positive with S. suis serotype 2; one case was positive with H. influenzae type b; 4 cases were positive with E. coli; pneumococcal meningitis were 19 (16.4%) cases, meningitis with S. aureus and N. meningitidis were not observed in any CSF samples in this study. Conclusion: our in-house real-time PCR assays are rapid, sensitive and specific tools for routine diagnosis to detect six mentioned above meningitis etiological agents. Probiotics are live microorganisms, which when ingested in sufficient amounts, confer health benefits to the host by improving the gut microflora balance. The purpose of this research was to determine whether commercial probiotic products containing multitude of commensal bacteria would reduce the growth rate of pathogenic bacteria, specifically Escherichia coli and Salmonella typhimurium. Growth curves were established, and the growth rates were compared for samples of E. coli, S. typhimurium, Nature’s Bounty Controlled Delivery probiotic, Sundown Naturals Probiotic Balance probiotic, and cocultures of the pathogenic bacteria mixed with the probiotics. The findings of this research were that the commercial probiotics significantly reduced the growth rate of E. coli and S. typhimurium when combined in cocultures. Probiotics containing multiple strains may be taken prophylactically to reduce the risk of bacterial infections caused by E. coli and S. typhimurium. Probiotics could be used to reduce the high global morbidity and mortality rates of diarrheal disease.

In the present study, twenty atopic dogs were investigated for secondary skin infections using different diagnostic techniques. The majority of dogs were suffering from bacterial infections, especially Staphylococcal infection (90 %). Different commensals on skin like Staphylococcus spp., Aspergillus spp. (50 %), Malassezia spp. (45 %), Demodex spp. (25 %) etc. caused secondary or concurrent infections in many atopic dogs indicating the importance of atopy in recurrent or non-responding dermatitis with those commensal organisms.

This paper presents the implementation of an agent-based architecture suitable for the coordination of power electronic converters in stand-alone microgrids. To this end, a publish-subscribe agent architecture was utilized as a distributed microgrid control platform. Over a distributed hash table (DHT) searching overlay, the publish-subscribe architecture was identified based on a numerical analysis as a scalable agent-based technology for the distributed real-time coordination of power converters in microgrids. The developed framework was set up to deploy power-sharing distributed optimization algorithms while keeping a deterministic time period of a few tens of milliseconds for a system with tens of converters and when multiple events might happen concurrently. Several agents participate in supervisory control to regulate optimum power-sharing for the converters. To test the design, a notional shipboard system, including several converters, was used as a case study. Results of implementing the agent-based publish-subscribe control system using the Java Agent Development Framework (JADE) are presented.

AbstractThe broiler industry in the Middle East (ME) faces many challenges related to bacterial infections, including M. gallisepticum, M. synoviae, E. coli, and other gram-negative bacteria, exacerbated by various errors in the brooding process. Antibiotics use in the first three days of life, such as Linco-Spectin 100 SP, tilmicosin, enrofloxacin, tylosin, colistin, and doxycycline, is the trend in the market to control such challenges. This study aimed to evaluate the efficacy of the newly introduced aroA E. coli vaccine (Poulvac E. coli) and its ability to reduce over-reliance on the heavy use of antibiotics in the ME. The study was conducted on 160 broiler chicks, divided into eight even groups. Each group was treated differently in terms of antibiotic therapy and ages at the time of Poulvac E. coli administration and the challenge of virulent avian pathogenic E. coli (APEC), serotype O78. Spray application of Poulvac E. coli at seven days of age plus Linco-Spectin 100 SP during the first three days provided the best results for zero mortality after challenge with APEC, while Poulvac E. coli at seven days with enrofloxacin during the early three days resulted in 10% mortality. Poulvac E. coli hatchery vaccination protected birds against mortality but reduced body weight gain compared to the 7-day group vaccinated with Linco-Spectin 100 SP during the first three days. Poulvac E. coli given on day one or day seven did not affect the immune response to concurrent respiratory viral vaccines and, in some cases, improved response. This study shows that Poulvac E. coli at seven days of age, together with Linco-Spectin 100 during the first three days, has produced the best results in terms of protection and performance in the ME high presence of avian pathogenic E. coli field challenge.
