BACKGROUND: Mesenchymal stem cells (MSCs) may serve as immunoregulators by producing various anti-inflammatory molecules. Under sufficient level of TNF-α, MSCs become activated and adopt immune-suppressive phenotype (MSCs type-2) by releasing various anti-inflammatory molecule including TGF-β and IL-10. However, the ability of MSC itself to produce IL-10 under TNF-α stimulation and the correlation of TGF-β production of MSCs to IL-10 level remains to be elucidated. AIM: In this study, MSCs were activated with various TNF-α doses to determine the increase of IL-10 and TGF-β level as well as its correlation. MATERIAL AND METHODS: This study used post-test only control group design, by using 3 study groups, consist of 1 control (C) and 2 treatments (T) (TNF-α = 5 and 10 ng/mL) with triplicate induced in MSC for 24 hours, then the levels of IL-10 and TGF-β were measured by using ELISA assay. RESULTS: The results of this study showed a significant increase of TGF-β and IL-10 levels (p < 0.05) at TNF-α 5 and 10 ng/mL dose of TNF-α. Moreover, there was a significant negative correlation between TGF-β and IL-10 level on 5 and 10 ng/mL dose TNF-α treatment. CONCLUSION: Based on our study, we conclude that the 5 ng/mL dose of TNF-α is a sufficient dose for MSCs to suppress the inflammatory milieu. The higher increase of TGF beta is due to the controlled inflammation by IL-10.

Cadmium is  a toxic heavy metal. The present of cadmium caused inflammation in liver. This study aims to know the anti inflammatory of Centella asiatica extract on rat induced by cadmium. This research was an experimental study using post test only control group design. Twenty four rats divided into six groups  with four replications, i.e group of healthy control (C1), negative control induced by CdSO4 with dosage 56 mg/kg for 14 days and treatment (C3-C6) with dosage of 100, 200 ,300 and 400 mg/kg of C. asiatica. The blood Cd, GST, GSH, TNF- α and COx2 were measured after a 21 days administration of C. asiatica. The data were analyzed by ANOVA test followed by Duncan test with a significance level of 5%. The result showed that administrating C.asiatica can neutralized cadmium, improve inflammation in liver.The conclusion of our research that C.asiatica extract can decrease Cd level, TNF-α and COx2 levels and increase GST and GSH level in rat induced by cadmium. A 200 mg/kg was the effective dosage to reduce Cd, TNF-α and COx2 levels and increase GST and GSH levels. Key words : anti inflammatory, Cd, Centella asiatica, GST, COX2

Abstract Objective This study was designed to assess the anti-inflammatory effect of Musa acuminata through the expression of tumor necrosis factor-α (TNF-α) and nuclear factor kappa β (NF-κB) after 3 days of application of Musa acuminata stem extract (MASE) gel on oral mucosal wound. Materials and Methods An experimental study with post-test only control group design was conducted. Twenty male Rattus norvegicus (Wistar) were injured on their left buccal mucosa and treated three times a day with MASE gel of varying concentrations: 0% (as control), MASE 25%, MASE 37.5%, and MASE 50%. On day 3, a biopsy was performed on each mucosal wound for later immunohistochemical analysis for the expressions of TNF-α and NF-κB. Results The highest expression of TNF-α was observed in the control group (13.20 ± 1.79), while the lowest was in the treatment group using 50% MASE (6.40 ± 1.14). Meanwhile the comparison between treatment groups did not highlight any significant difference (p > 0.05). The highest expression of NF-κB was observed in the control group (13.20 ± 1.30), whereas the lowest was in the treatment group using MASE 50% (6.40 ± 1.14). NF-κB was significantly lower in the treatment group using MASE 50% when compared with other treatment groups (p < 0.05). Conclusion Application of MASE on mucosal wound reduces the expression of TNF-α and NF-κB at all concentrations. The anti-inflammatory effect of MASE 50% was the strongest one.

<p><strong>Background:</strong> <em>Corticosteroid has a strong anti-inflammatory effect, unfortunately long-term usege of corticosteroid drugs can suppressed immune system. This condition makes Candida albicans as an opportunistic phatogen caused Oral Candidiasis. Macrophages have an important role in the first defense against infection. Methanol extract of A.ilicifolius leaf  has anti-fungal, anti-inflammatory compounds to fight the growth of Candida albicans. <strong>Purpose:</strong> To determine the effect of methanol extract of A.ilicifolius leaf theraphy to the amount of macrophages in the healing pricess of oral candidiasis models with immunosuppressed conditions. <strong>Method:</strong> </em><em>This study was true experimental with post test only control group design. Thirty-six strain wistar, 3 months old, and 250 grams of weight which were given dexamethason 0.5 mg and tetracycline 0.5mg for 7days, after Candida albican(ATCC10231) was induced for 2 weeks (3 times/week). Rats were divided into 6 groups: untreated(K-), nystatin(K+), A.ilicifolius 8%(P1), A.ilicifolius 12%(P2), A.ilicifolius 16%(P3), A.ilicifolius 20%(P4 ) all groups were treated for 2 weeks. After being treated, the rats tongue biopsied to examine the macrophages with Haematoxylin-eosin staining and observed with microscope (400x magnification). Data analyzed statistically by One-way ANOVA. <strong>Results:</strong> the amount of macrophages at K+(47.60±4.16), P4(40.20±3.56), P3(36.20±1.92), P2(17.00±2.45) were greater than K-(15.60±4.56), P1(14.60±2.70). there are no significant difference between group K- with P1 and P2, P3 with P4. There are significant difference between group K- with K+, K- with P3 and P4. K+ with P1, P2, P3 and P4. Group P1 with P3 and P4. Group P2 with P3 and P4. </em></p><strong><em>Conclusion:</em></strong><em> A.ilicifolius extract can increase the amount of macrophages in oral candidiasis immunosuppression models</em>

Penelitian ini bertujuan untuk mengetahui pengaruh pemberian genistein terhadap penurunan kadar TNF-α studi pada (Mus musculus) model endometriosis. Penelitian dibagi menjadi 8 kelompok, yaitu : kontrol negatif, kontrol positif dengan perlakuan pemberian genistein dosis 50 mg/hari, 100 mg/hari, 200 mg/hari, 300 mg/hari, 400 mg/hari, 500 mg/hari. Pengukuran kadar TNF-α menggunakan ELISA kit. Data hasil pengamatan dianalisis dengan uji Kruskal Wallis dan uji Perbandingan berganda |Ri-Rj| 5%.Menggunakan mencit model endometriosis.Penelitian ini merupakan penelitian eksperimental murni (true experimental) dengan menggunakan rancangan percobaan dengan metode randomized post-test control group design., dilakukan di Laboratorium Fisiologi Reproduksi Embriologi Fakultas Kedokteran Hewan Universitas Airlangga Surabaya dan Laboratorium Fisiologi Fakultas Kedokteran Universitas Brawijaya.  Ada pengaruh pemberian genistein terhadap kadar TNF-α dalam cairan peritoneal mencit model endometriosis (p<0,05). Genistein mampu menurunkan kadar TNF-α terendah pada dosis 100 mg/hari (148.25±7.89 pg/ml) bila dibandingkan dengan perlakuan dosis yang lain.

Background: Several traditional medicines have been developed among communities as part of the efforts to improve thrombocytopenia in patients with Dengue virus infection (DVI). Among those efforts is the administration of Monascus jmbA rice (MJR), which is rice fermented with Monascus purpureus. The administration of MJR is believed to increase the platelet count in patients with DVI. Aim: The objective of this study is to elucidate the mechanism of the effects of MJR on increased levels of platelets in DVI serotype 3 through changes in IL-3, IL-6, IL-11 and TNF-α. Methods: It was a true experimental laboratory study using the randomized post-test only control group design. The study compared between groups of wistar rats were being treated only with group IVD wistar rats who experienced treatment followed by administration of MJR as well as groups of wistar rats without any treatment as a control group. Results: The increase in platelet count in the group treated with DVI + MJR was higher than that treated with only DVI and the difference was a significant (P<0.05). Increased levels of TNF-α in the group treated with DVI + MJR were lower than that treated with only DVI and the difference was significant (P<0.05). The significant levels of the causal relationship of TNF-α with IL-6, TNF-α with IL-11 and IL-6 with platelets were 0.044 (P<0.05), 0.029 (P<0.05) and 0.041 (P<0.05), respectively. Conclusion: MJR is capable of increasing platelet count through the role of TNF-α and IL-6 in Wistar rats infected with DVI serotype 3.

Background : Pain and Postoperative Nausea and Vomiting (PONV) are surgical side effects. Pain management can use opioids and non-opioids. Opioids have side effects such as nausea, vomiting, pruritus, somnolence, and respiratory depression, so other alternatives are needed to reduce pain. Paracetamol is an analgesic while dexamethasone is antiinflammatory and anti-emetic. This study was to determine whether the combination of paracetamol and dexamethasone was more effective in dealing with pain and PONV after cesarean section than paracetamol.Materials and Methods : This was an experimental study employing post-test only control group design, using two study groups. Group PD (combined paracetamol and dexamethasone) was treated using dexamethasone 8 mg and paracetamol 1 gram intravenously, while the group P (paracetamol) was treated using paracetamol 1 gram intravenously. Data of Pain and PONV were collected using the Wong Baker Faces Scale, Numeric Rating Scale, and a frequency table for PONV at the 4th, 6th, 12th, and 24th hours of postoperative administration. The data were analyzed using Mann Whitney test.Results : The results of this study found that the multimodal administration of combined paracetamol and dexamethasone was better in reducing the pain compared to administration of a paracetamol at 4th, 6th, 12th, and 24thhours of post cesarean section(p <0.05). Multimodal administration of combined paracetamol and dexamethasone was better reduced the incidence of PONV compared tothat of paracetamol at the 4thhour of post cesarean section(p <0.05). The multimodal use of combined paracetamol and dexamethasone had resulted fewer use of opioid and anti-emetic frequencies compared to that of paracetamol.Conclusion : The combined paracetamol and dexamethasone was more effective to reduce pain and PONV in samples with post cesarean section.International Journal of Human and Health Sciences Vol. 04 No. 03 July’20 Page : 184-188

Background. Andrographolide as an anti-inflammatory inhibit activation of NF-κβ, the production of TNF-α, IL-12, pressing the release inducibele nitic oxide synthase (iNOS), inhibiting the release of COX-2 in human fibroblast cells and also prevents the production of oxygen radicals. TNF- α stimulates the migration of immune cells to get to the site of infection, contribute to the formation of granulomas, and can control the disease progresificity. Methods. The study aims to analyze the effect of andrographolide on the expression of TNF-α and tuberculosis granuloma in mice infected with CFA. Laboratory tests in the laboratory of histology and pathology anatomy medical faculty of UNS. Samples are 30 individuals’ mice, consist of 10 rats as control, 10 rats injected with CFA, and 10 rats injected with CFA+ andrographolide. Histopathology and immunohistochemistry of lung tissue granuloma examinated in the laboratory of pathology anatomy medical faculty of UNS. Results. The study design was purely experimental or (true experiment, with randomized post test only control group design). Andrographolide lowering the average expression of TNF-α compared to the CFA group, based on the description of the variable expression of TNF-α. The results of the analysis of three different variations or the average using Kruskal Wallis test showed that there are differences in the average number 3 granuloma with P=0.003. It means different average number of granulomas in the control group, CFA and CFA+ Andrographolide completely different convincingly. Compared with the average number of granulomas in the control group, the group CFA has a tendency average number of granuloma higher (increased), then the average number of granuloma in group CFA + Andrographolide has an average lower than the group CFA or mean average granuloma amount can be reduced by giving Andrographolide paniculata extract. Conclusion. Andrographolide 14.8% paniculata extract shown to decrease the expression of TNF-α induction of pulmonary tuberculosis in mice the CFA. Andrographolide 14.8% paniculata extract shown to reduce the number of lung granuloma in mice induced tuberculosis CFA. There is a positive correlation between the expression of strong TNF-α by the number of granulomas in the lungs in mice induced tuberculosis CFA. (J Respir Indo 2018; 38(2): 75-82)

Preeklampsia dan eklampsia adalah penyakit pada kehamilan yang menjadi salah satu penyumbang angka kematian Ibu khususnya di Indonesia. Tahapan pertama dari preeklampsia ini adalah terjadi abnormalitas dalam remodelling vaskular trofoblastik mengakibatkan disfungsi endotel, tahap kedua pelepasan faktor plasenta kedalam sirkulasi darah seperti soluble fms-like tyrosine kinase (sFlt-1), TNF-α, IL-6, IL-8, angiotensin II tipe 1 reseptor antibodies (AT1-AA), dan thromboxan (TX) menyebabkan respon inflamasi dan peningkatan aktivasi sel endotel. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh pemberian ekstrak kakao terhadap kadal IL-8 pada model HUVECs Preeklampsia. Penelitian true experimental dengan pendekatan yang digunakan  post test only control group design yang dikerjakan dilaboratorium dengan invitro. Kultur HUVECs dibagi menjadi 5 kelompok masing-masing dilakukan replikasi 5x. Pengukuran kadar IL-8 menggunakan ELISA (Enzyme-Linked Immunosorbent Assay). Analisis data yang digunakan pada penelitian ini adalah dengan  uji Anova one way pada kadar IL-8, selanjutnya dilakukan uji perbandingan berganda (Multiple Comparisons) dengan uji Beda Nyata Terkecil/BNT (Least Significant Difference/LSD) Hasil penelitian pemberian ekstrak kakao pada kelompok kontrol positif dan 3 dosis kelompok perlakuan ternyata dapat menurunkan secara bermakna (p-value 0.000< α 0.05, Anova One way) kadar IL-8.

Penelitian ini bertujuan untuk mengetahui kemampuan menulis surat dinas siswa kelas VIII di SMP Negeri Se-Kecamatan Panyingkiran Kabupaten Majalengka, mengetahui kemampuan menulis surat dinas dengan menggunakan model pembelajaran STAD (Student Teams-Achievment Divisions) siswa kelas VIII di SMP Negeri Se-Kecamatan Panyingkiran Kabupaten Majalengka, serta menjelaskan efektifitas model pembelajaran STAD (Student Teams-Achievment Divisions) untuk meningkatkan  pengajaran menulis surat dinas siswa Kelas VIII di SMP Negeri Se-Kecamatan Panyingkiran Kabupaten Majalengka.Penelitian ini menggunakan pendekatan kuantitatif dan kualitatif dengan metode eksperimen Pre test-Post test Control Group Design, populasinya yaitu SMP Negeri Se-Kecamatan Panyingkiran Kabupaten Majalengka tahun ajaran 2013/2014, sedangkan  sampelnya ditentukan melalui teknik simple random sampling yaitu kelas VIII G SMP Negeri 2 Panyingkiran sebagai kelas eksperimen dan kelas VIII B SMP Negeri 1 Panyingkiran sebagai kelas kontrol. Instrumen pengumpulan data yang digunakan tes tertulis tes awal dan tes akhir, kuesioner, lembar observasi guru dan siswa, setelah data diperoleh maka dilakukan analisis data. Data kuesioner dan observasi dianalisis dalam bentuk uraian sedangkan data tes dianalisis dengan statistik dan di deskripsikan.Berdasarkan perhitungan statistik program SPSS 16.0 diperoleh hasil uji t pretes pada kedua kelas tersebut memiliki nilai t=5.429 derajat kebebasan (df)=n-1=38 nilai probability (sig 2-tailed) sebesar 0,000.  Hal ini berarti t hitung <0,005. Artinya 0,000<0,05, pada uji t ini bahwasanya pretes pada kedua kelas terdapat perbedaan yang signifikan antara kelas eksperimen dengan kelas kontrol.  Apabila ditinjau dari segi rata-rata pretes eksperimen lebih efektif daripada kelas kontrol. Hal ini terbukti dari rata-ratanya pretes kelas eksperimen 64,8 kelas kontrol 62,3 selisih rata-rata 2,5. Sedangkan rata-rata postes kelas eksperimen 81,2 dan kelas kontrol 69 terjadi selisih postes 12,2.

Tujuan dari penelitian ini adalah menganalisis pengaruh model pembelajaran kooperatif tipe Number Head Together (NHT) terhadap keterampilan problem posing matematis siswa. Penelitian ini dilakukan di SD Al- Zahra Indonesia, untuk tahun akademik 2014/2015. Metode yang digunakan dalam penelitian ini adalah metode kuasi eksperimen dengan Post-test Only Control Group Design, melibatkan 60 siswa sebagai sampel. Untuk menentukan sampel digunakan teknik cluster random sampling. Pengambilan data menggunakan instrumen seperti tes esai tertulis. Keterampilan problem posing matematis siswa yang mengajar dengan model pembelajaran kooperatif tipe Number Head Together (NHT) lebih tinggi daripada keterampilan problem posing matematis siswa yang diajarkan dengan model pembelajaran klasik (t hitung = 2,01 table t tabel = 2,00 ). Persentase jawaban siswa dari problem posing berdasarkan gambar, tabel dan diagram yang diajarkan dengan model pembelajaran kooperatif tipe Number Head Together (NHT) lebih tinggi daripada siswa yang mengajar dengan model pembelajaran klasik. Kesimpulan hasil penelitian ini bahwa pembelajaran matematika Pecahan dengan model pembelajaran kooperatif tipe Number Head Together (NHT) memiliki pengaruh yang signifikan terhadap keterampilan problem posing matematika siswa.
