Oral fluid is an alternative biological material that confirms correlations with blood parameters in various pathological conditions of the body. In order to find a non-invasive approach to stratification of patients with COVID-19 disease, molecular biomarkers of the oral fluid have been determined in patients with moderate coronavirus infection in comparison with clinically healthy individuals. It has been shown that proteomic, carbohydrate, macro- and microelement profiles of the oral fluid in coronavirus infection can be used for diagnostics. The features of protein metabolism were revealed: an increase in the content of total protein, urea; increased activity of enzymes aspartate aminotransferase, gamma glutamyl transpeptidase, creatine phosphokinase, alkaline phosphatase; changes in carbohydrate metabolism, which is expressed by an increase in glucose and lactate levels, an increase in lactate dehydrogenase activity, sodium, chloride, calcium, magnesium, iron content.

The aim of this study was to evaluate steroid receptors’ status of tumor tissue in different molecular biological types of endometrial cancer (EC), subdivided according to the current classification, and their colonization by lymphocytic and macrophage cells, taking into account body mass index of the patients. Materials and methods: Material from treatment-naive patients with EC (total n = 229) was included; the number of sick persons varied depending on the method used. The average age of patients was close to 60 years, and about 90% of them were postmenopausal. It was possible to divide the results of the work into two main subgroups: a) depending on the molecular biological type of the tumor (determined on the basis of genetic and immunohistochemical analysis), and b) depending on the value of the body mass index (BMI). The latter approach was used in patients with EC type demonstrating a defective mismatch repair of the incorrectly paired nucleotides (MMR-D) and with a type without characteristic molecular profile signs (WCMP), but was not applied (due to the smaller number of patients) in EC types with a POLE gene mutation or with expression of the oncoprotein p53. According to the data obtained, when comparing various types of EC, the lowest values of Allred ER and PR scores were revealed for POLE-mutant and p53 types, while the “triple-negative” variant of the tumor (ER-, PR-, HER2/neu-) was most common in POLE-mutant (45.5% of cases) and WCMP (19.4%) types of EC. The p53+ type of EC is characterized by inclination to the higher expression of the macrophage marker CD68 and lymphocytic Foxp3, as well as mRNA of PD-1 and SALL4. In addition to the said above, for WCMP type of EC is peculiar, on the contrary, a decrease in the expression of lymphocytic markers CD8 (protein) and PD-L1 (mRNA). When assessing the role of BMI, its value of >30.0 (characteristic for obesity) was combined with an inclination to the increase of HER-2/neu expression in the case of MMR-D EC type and to the decrease of HER-2 /neu, FOXp3 and ER expression in WCMP type. Conclusions: The accumulated information (mainly describing here hormonal sensitivity of the tumor tissue and its lymphocytic-macrophage infiltration) additionally confirms our earlier expressed opinion that the differences between women with EC are determined by both the affiliation of the neoplasm to one or another molecular biological type (subdivided according to the contemporary classification), as well as by body mass value and (very likely) the associated hormonal and metabolic attributes.

This study investigated the influence of exercise and fasting state on haematologic and biochemical parameters in juvenile green turtles (Chelonia mydas). Animals were divided into two groups; one group was fasted for 72 h and one group was fed 1 h prior to exercise. Exercise was induced by repeated righting reflexes and blood values were measured prior to and post-exercise. Prior to exercise, fasted animals showed significantly decreased levels of urea, pH, PVCO2 and HCO3- and significant increases in Cl- and PVO2, compared to fed animals and fasted animals had significantly poorer exercise performance. Following exercise both fasted and fed animals had significant increases in Na+, K+, Cl-, PVCO2, PVO2, urea and lactate and significant decreases in pH and HCO3-. The magnitude of increase in lactate levels was significantly less in fasted animals. Prior to exercise, a significant correlation was calculated in fasted animals between pH and HCO3-. Following exercise, significant correlations were calculated in fed animals between pH and HCO3-, PVCO2 and lactate, and between pH and HCO3- in fasted animals. These results show that analytical method, fasting state and the physiologic changes induced during the intense exercise can affect haematologic and biochemical analytes and these factors should be considered when interpreting results from health assessment of wild animals.

The development of non-cellularized injectable suspensions of viscous chitosan (CHI) solutions (1.7–3.3% (w/w)), filled with cellulose nanofibers (CNF) (0.02–0.6% (w/w)) of the type nanofibrillated cellulose, was proposed for viscosupplementation of the intervertebral disc nucleus pulposus tissue. The achievement of CNF/CHI formulations which can gel in situ at the disc injection site constitutes a minimally-invasive approach to restore damaged/degenerated discs. We studied physico-chemical aspects of the sol and gel states of the CNF/CHI formulations, including the rheological behavior in relation to injectability (sol state) and fiber mechanical reinforcement (gel state). CNF-CHI interactions could be evidenced by a double flow behavior due to the relaxation of the CHI polymer chains and those interacting with the CNFs. At high shear rates resembling the injection conditions with needles commonly used in surgical treatments, both the reference CHI viscous solutions and those filled with CNFs exhibited similar rheological behavior. The neutralization of the flowing and weakly acidic CNF/CHI suspensions yielded composite hydrogels in which the nanofibers reinforced the CHI matrix. We performed evaluations in relation to the biomedical application, such as the effect of the intradiscal injection of the CNF/CHI formulation in pig and rabbit spine models on disc biomechanics. We showed that the injectable formulations became hydrogels in situ after intradiscal gelation, due to CHI neutralization occurring in contact with the body fluids. No leakage of the injectate through the injection canal was observed and the gelled formulation restored the disc height and loss of mechanical properties, which is commonly related to disc degeneration.

The article presents the results of the influence of coniferous-energy additives on the morphological and biochemical blood parameters of cows of black-motley breed in different physiological periods. It was established that the inclusion of 17 g / kg of dry matter in the diets of cows feeding cows favorably contributed to the course of metabolic processes in the body.

Heavy metals as pollutants in the working and living environment are a serious health and environmental problem because they are toxic, non-biodegradable, accumulate in living systems and have a long half-life in soil. Sources of lead contamination are combustion products in the chemical industry and metallurgy, industrial waste water, landfills, traffic etc. Lead enters into the body via the food chain and drinking water. In the body lead is deposited in the liver, kidneys, brain and mineral tissues. Excretion of lead causes damage to the epithelial cells of certain organs. High level exposure to cadmium is usually the result of environmental pollution by human activities. Exposure to cadmium can lead to acute and chronic tissue damage of various organs, including liver and kidneys in humans and in animals. In this paper we analyzed the effects of lead and cadmium exposure, in working and living environment, on the model system of experimental animals, particularly the activity of certain liver enzymes, acid and alkaline DNase, and standard biochemical blood parameters. The study showed that lead and cadmium significantly affect the protein content, red blood cells, hemoglobin and hematocrit, and the activity of liver enzymes. This harmful effect of this toxic metal can be reduced by the supplements.

There were studied 92 somatically healthy persons of young age (18-25 years old), with generalized periodontitis (GP) of initial-I degree of development, among them: 30 patients with chronic generalized periodontitis (CGP), who were included into group І; and 32 patients with exacerbation of the chronic generalized periodontitis (ECGP) – into group II; and 30 healthy patients. The carbohydrate metabolism indexes were studied, namely: the content of glucose, pyruvate (pyruvic acid) and lactate (lactic acid) and lactate dehydrogenase activity (LDG) in the oral liquid.We have determined that in the presence of CGP of the initial-I degree of development, and especially in its exacerbation, there is a significant increase of indicators of carbohydrate metabolism. In young patients with CGP indicators of glucose, pyruvate, lactate and LDG activity in the oral liquid increased by 2.0, 1.34, 1.58 and 1.37 (p<0.001, p<0.01) times respectively, and in case of ECGP they grew even more: by 2.71, 1.98, 1.76 and 2.07 (p<0.001) times, respectively.Among all the indicators, that characterize the carbohydrate metabolism, in addition to the level of pyruvate, in case of different course of GP a significant difference was revealed: in patients with ECGP compared with the data in CGP, glucose and lactate levels and LDG activity in the oral liquid were significantly higher - at 35.75% (p<0.05), 34.29% (p<0.01) and 36.59% (p<0.001) respectively.Detected violations of carbohydrate metabolism indeces in the oral liquid indicate the involvement of these processes in the pathogenesis of GP and the necessity of their correction.

The purpose of the study was to analyze modern literature on the problems of dysbiosis in patients with COVID-19, to study the main mechanisms of systemic interaction between the intestine and lungs, as well as changes in the microbiota that occur under the influence of coronavirus infection. Materials and methods. A comprehensive selection of research methods was used for the work: systematization of the material, the method of generalization, methods of analysis and synthesis. Scientific works in the field of microbiology, epidemiology and infectious diseases were studied. Literature data for the last 2 years (2019-2021) were considered. The results of bacteriological studies from patients with COVID infection were described. The data obtained were processed using information-analytical and statistical-analytical methods. Results and discussion. As a result of this work, a complex of connections between intestine and lungs, which is called the "intestinal-lung axis", was considered. It is known that the interaction between these two biotopes occurs with the participation of microflora and its metabolites. Dysfunction of the intestinal barrier is accompanied by bacterial translocation. Bacteria from the intestinal lumen enter the liver through the portal vein system. The lymphatic pathway of bacterial translocation from the intestine to the lungs is also possible, which causes multiple organ failure syndrome in coronavirus infection. The COVID-19 virus is able to reduce the number of ACE2 receptors in the gastrointestinal tract, which leads to an imbalance in the intestines. At the same time, the infection process in the lungs promotes the growth of bacteria of the Enterobacteriacae family in the intestine, which also leads to dysbiotic disorders. The use of probiotics is an effective tool in the complex treatment of this infection, which facilitates the general condition of patients. In the course of treatment, it is important not only to eliminate the virus from the body, but also to restore normal intestinal microbiota after an infection. Conclusion. Thus, the use of probiotic drugs for the treatment of patients with coronavirus infection can significantly reduce the risk of developing dysbiosis and improve the condition of patients. A perspective direction is the development of new treatment regimens for dysbiotic conditions using probiotics, eubiotics, synbiotics and postbiotics to prevent the development of severe complications in COVID infection

Hematopoietic stem cells (HSCs) cultured outside the body are the fundamental component of a wide range of cellular and gene therapies. Recent efforts have achieved more than 200-fold expansion of functional HSCs, but their molecular characterization has not been possible due to the substantial majority of cells being non-HSCs and single cell-initiated cultures displaying substantial clone-to-clone variability. Using the Fgd5 reporter mouse in combination with the EPCR surface marker, we report exclusive identification of HSCs from non-HSCs in expansion cultures. Linking single clone functional transplantation data with single clone gene expression profiling, we show that the molecular profile of expanded HSCs is similar to actively cycling fetal liver HSCs and shares a gene expression signature with functional HSCs from all sources, including Prdm16, Fstl1 and Palld. This new tool can now be applied to a wide-range of functional screening and molecular experiments previously not possible due to limited HSC numbers.

Multiple micronutrient deficiency (simultaneous insufficient supply of the organism with several vitamins at once (vitamin D, B group), as well as calcium, magnesium, zinc and iodine, and not an isolated deficiency of any one micronutrient is characteristic of a significant part of the Russian population throughout the year, regardless of places of residence. The growing need of the organism at different periods of life (puberty, pregnancy, breastfeeding, aging), various diseases, drug therapy, the period of convalescence, stress, the use of various diets, enrichment of the diet with polyunsaturated fatty acids and dietary fiber, religious observance, adverse environmental factors further increase the risk of multiple micronutrient deficiencies. Micronutrients (vitamins and essential minerals) are closely interconnected into complex metabolic networks, which provide maintaining homeostasis and health. Deficiency of each of the essential micronutrients is a risk factor for the development of a particular disease, and simultaneous multiple micronutrient deficiency creates a “causality network” of not just one, but several diseases at once. Conversely, an adequate or optimal sufficiency of the body with all micronutrients, respectively, creates a “network of conditions ensuring the prevention of the disease”, due to the full implementation of all processes dependent on them in the body. This means that replenishing inadequate food intake of scarce micronutrients is an important way to maintain health and longevity. Given the variety of vitamin-mineral supplements, it is important to choose the right supplement. The maximum effectiveness to maintain the body’s vitamin and mineral status at the optimum level will be ensured by the intake of not individual micronutrients, but vitamin and mineral supplements containing a complete set of vitamins in doses close to the physiological needs of the body and basic minerals (calcium, magnesium, zinc and iodine), the deficit of which is characteristic for the Russian population.

Ricinus communis L. of Euphorbiaceae family is a widespread plant in tropical regions. It is used in traditional medicines as an anti- fertility agent in India and different parts of the world. The ether soluble portion of the methanol extract of R. communis var minor possesses anti-implantation, anti - conceptive and estrogenic activity in rats and mice when administered subcutaneously.The study was conducted on 10 local breed male rabbits, 1-2 years old, of 1-2 kg body weight. The animals were divided into two groups, control non – treated group and treated group in which animals were treated with single daily dose of 50 mg /kg b. wt. P.O. of decorticated and defatted castor seeds (DDCS) for 14 days. 28th day post treatment, animals were anesthetized by diethyl ether, sacrificed, abdominal cavity was open. The sexual organ (testes, epididymis, prostate and seminal vesical) weighed. In addition to take a biopsy from each one for histopathological changes. The study also included clinical and hematological parameters, in addition to sperm counts and the changes in sperm morphology.Body weight, body temperature increased significantly in treated males. While in non- treated group there were no significant changes. Respiratory rates and heart rate were none significantly changed in treated and non- treated males.Bleeding time none significantly increased in treated males, but increased significantly in none treated males. Clotting times decreased none significantly in treated and non- treated males. The blood parameters including, total erythrocytes count, hemoglobin concentration, PCV% , MCV, MCH, MCHC, total leucocyte and differential leucocyte counts were either increased or decreased none significantly in both groups. The results revealed that the effects of exposure to extract of ricin for 14 days on reproductive efficiency of rabbits, exhibited Significant decrease in weights of testes, epididymis, tails, heads of epididymis, seminal vesicles and prostate in treated males in comparison with those of non- treated males. While the body of epididymis did not show a significant changes.Significant decrease in live sperm numbers, number of sperms in epididymal head, in addition to deformities in high numbers of sperm, including enlarged or small sperms. breaks head, and its detachment, presence of two heads in one sperm, bifurcation of tail and its breaking, sperm coiling in samples from treated males in comparison with those from non-treated males.Histological changes were hyperplasia of lining epithelial cells and vacuolar degenerative changes, loss of spermatogenesis, and spermatocytes necrosis in those from treated males.
