Balanophora elongata (Balanophoraceae) is a tropical parasitic flowering plant 9 cm in height. Four known phenyl acrylic acid derivatives, methyl caffeate (1), caffeic acid (2), 1,6-di-O-caffeoyl-β-D-glucopyranose (3), and coniferin (4), were isolated from this plant. Structural elucidation of the isolated compounds was determined by IR, LC-ESI-MS, 1D and 2D NMR.  Extracts and isolated compounds were tested toward some standard human pathogenic bacteria using the agar disk diffusion method. Their inhibition zones were compared to that of chloramphenicol as positive control. Compound 1 showed inhibition toward Streptococcus mutans, while compound 3 and 4 inhibited Staphylococcus aureus.

Abstract Background: Bacopa monnieri (Linn) Pennell (Scrophulariaceae) is widely distributed in tropical regions of Asia, and used in the treatment of cough or as an antiseptic. The traditional use of this plant suggests its possible antibacterial properties, but its efficacy has not been examined yet. Objective: Evaluate the antibacterial efficacy against pathogenic bacteria using the disk diffusion method. Materials and methods: Five different concentrations (500 μg, 1, 2, 5, 10, and 15 mg/mL) of crude leaf extracts of Bacopa monnieri (L.) Pennell were tested for antibacterial efficacy against seven Gram-positive and 11 Gramnegative bacteria. The sensitivity of plant fractions was tested using the disk diffusion method. Results: Maximum activity was revealed by ethyl acetate and methanol extracts, followed by aqueous, benzene, and petrol extracts. Phyto-chemical analysis of the plant leaf showed the presence of alkaloids, flavonoids, and saponins. Conclusion: This plant may be effective for treatment of different pathogenic diseases.

An antibiotic-resistant and multidrug-resistant (MDR) issue open the role of researchers to continue to search for natural potential as a source of new antimicrobials. One of the potential fungi isolates that can produce antimicrobial active compounds from Indonesian tropical peat soils is Penicillium sp. LBKURCC34. In this study, the production of antimicrobial compounds from local isolates was carried out by batch fermentation method in liquid media with the addition of biotic elicitors to increase the extraction activity and yield. This study aims to optimize the results based on the time the elicitor is added. Staphylococcus aureus was used as a biotic elicitor, which was added on days 2, 3, and 4 in the production of antibiotics by fermentation incubation of 6-14 days. The antibiotic production media was extracted with ethyl acetate and evaporated. The antimicrobial test was carried out by the disk diffusion method against pathogenic bacteria Escherichia coli, Staphylococcus aureus, Bacillus subtilis, and Staphylococcus epidermidis using three crude extract contents (19; 38; and 57 µg/disc). Amoxicillin® was used as a positive control (10 µL/disc). The results showed that the addition of S. aureus biotic elicitor extended the log phase growth of the fungus Penicillium sp. LBKURCC34. The optimum condition of production was obtained by adding initiator treatment on the 3rd day for 14 days incubation with the highest yield and could inhibit the growth of all pathogenic microbes.

ABSTRACT Aim The present study was conducted to evaluate the efficacy of commercially available herbal toothpastes against the different periodontopathogens. Materials and methods Six herbal toothpastes that were commonly commercially available were included in the study. Colgate herbal, Babool, Meswak, Neem active, Dabur red toothpastes were tested for the study whereas sterile normal saline was used as control. Antimicrobial efficacies of dentifrices were evaluated against Streptococcus mutans and Actinobacillus actinomycetemcomitans. The antimicrobial properties of dentifrices were tested by measuring the maximum zone of inhibition at 24 hours on the Mueller Hinton Agar media inoculated with microbial strain using disk diffusion method. Each dentifrice was tested at 100% concentration (full strength). Results The study showed that all dentifrices selected for the study were effective against the entire test organism but to varying degree. Neem active tooth paste gave a reading of 25.4 mm as the zone of inhibition which was highest amongst all of the test dentifrices. Colgate Herbal and Meswak dentifrices recorded a larger maximum zone of inhibition, measuring 23 and 22.6 mm respectively, compared to other toothpastes. All other dentifrices showed the zone of inhibition to be between 17 and 19 mm respectively. Conclusion The antibacterial properties of six dentifrices were studied in vitro and concluded that almost all of the dentifrices available commercially had antibacterial properties to some extent to benefit dental health or antiplaque action. The use of earphones has increased in recent times throughout the world especially among the different level of students such as school, college or university who have a higher tendency of sharing these among them. Unlike airline headsets, headphones and stethoscope ear-pieces, ear phones are often shared by multiple users and can be a potential medium for transmission of pathogens, which can give rise to various ear related infections. The objective of this study was to detect the pathogenic bacteria from the ear-phones used by the students of Stamford University Bangladesh. A total of 16 ear-phone swabs were collected by sterile cotton swabs. The swabs were inoculated onto blood agar and incubated aerobically overnight at 37oC. Microscopic observation and standard biochemical tests were performed to confirm the identification of all the bacterial isolates. Six presumptively identified Staphylococcus spp. (38%) were tested against six different types of antibiotics following Kirby-Bauer disk diffusion method. Isolates were found to be 84% resistant against Cotrimoxazole and demonstrated 100% sensitivity to Vancomycin and Ciprorofloxacin. The findings of this study suggest the users to disinfect their respective ear phones and not to exchange them as they may act as a potential source to transfer pathogenic and antibiotic resistant bacteria among the ear phone users. During a screening for antifungal secondary metabolites, six new mono-/bis-alkenoic acid derivatives (2–7) and one known alkenoic acid derivative (1) were isolated from an endophytic fungi Scopulariopsis candelabrum. Their chemical structures were identified by 1H-NMR, 13C-NMR, 2D NMR, and high-resolution mass spectrometry, as well as comparisons with previously reported literatures. Among them, fusariumesters C‒F (2–5) are bis-alkenoic acid derivatives dimerized by an ester bond, while acetylfusaridioic acid A (6) and fusaridioic acid D (7) are alkenoic acid monomers. All the isolates were submitted to an antifungal assay against Candida albicans and the corn pathogen Exserohilum turcicum using the filter paper agar diffusion method. As a result, only compound 1 decorating with β-lactone ring turned out to be active against these two tested fungi. The broth microdilution assay against Candida albicans showed the minimum inhibitory concentration (MIC) value of 1 to be 20 μg/ml, while the minimum inhibitory concentration value of the positive control (naystatin) was 10 μg/ml. And the half maximal inhibitory concentration (IC50) value (21.23 μg/ml) of 1 against Exserohilum turcicum was determined by analyzing its inhibition effect on the mycelial growth, using cycloheximide (IC50 = 46.70 μg/ml) as the positive control.

Streptococcus pyogenes is one of the pathogenic bacteria that causes pharyngitis. Natural treatment to overcome these problems is to use cardamom seeds. The purpose of this study was to test the antibacterial activity, determine the concentration of inhibitory zone and phytochemical compounds from  ethanol 96% extract of Java cardamom seeds (Amomum compactum Soland. Ex Maton) against Streptococcus pyogenes. The method is used   solid dilution and paper disc diffusion method. The solid dilution method is used for the Minimum Inhibitory Concentration (MIC) test with a concentration of 1.25%, 2.5%, 5% and 7.5% while the paper diffusion method is used for the Inhibition zone Diameter (IZD)  using five treatments namely three concentrations of ethanol 96% extract of Java cardamom seeds (7.5%, 10% and 12%), one positive control of amoxicillin 0.01 mg/mL and one negative control of sterile distilled water. IZD data were  analyzed using ANOVA with a confidence level of 95% and α = 0.05 and Duncan's further tests to determine differences between treatments. The results showed that the MIC  was at a concentration of 7.5% while for the IZD test which had the highest activity there was a concentration of 12% with an average inhibition diameter of 12.03 ± 0.14 mm. In addition, ethanol 96% extract of Java cardamom seeds contain alkaloids, flavonoids, terpenoids and tannins which function in antibacterial activity.

Belimbing wuluh (Averrhoa bilimbi Linn.) is a tropical plant that has antibacterial properties. The purpose of this study was to test the antibacterial activity of bark Belimbing wuluh against Klebsiella pneumoniae and Staphylococcus epidermidis and their bioautography. Extraction methods used to research is method maceration with a solvent ethanol 96 %. Fractinations done by method partition liquid-liquid with a separating funnel. Test performed in this research covering identi� cation bacteria, the sensitivity bacteria, antibacterial activity, thin layer chromatography, bioautography. The result of antibacterial activity ethanol extract of disk diffusion method with concentrations 400 μg/disk, 800 μg/disk, 1600 μg/disk is 8±0,5; 10,34±0,58; 12,17±0,76 on Klebsiella pneumoniae, 10,17±0,29; 11±0; 11.5±0 on Staphylococcus epidermidis, n-hexane fraction with concentration 400 μg/disk, 800 μg/disk, 1600 μg/disk is 8,34±0,29; 9,34±0,29; 10,84±0,76 on Klebsialla pneumoniae, 8,5±0,5; 9,34±0,29; 10,67±0,29 on Staphylococcus epidermidis, ethyl acetate fraction with concentration 400 μg/disk, 800 μg/disk, 1600 μg/disk is 9,17±0,29; 10,34±0,29; 11,17±0,29 on Klebsiella pneumoniae and 9,5±0,5; 10,67±0,29; 12,67±1,26 on Staphylococcus epidermidis, ethanol-water fractions with concentration 400 μg/disk, 800 μg/ disk, 1600 μg/disk is 8,17±0,29; 9,17±0,29; 10±0 on Klebsiella pneumoniae, 9±0; 9,67±0,29; 10,34±0,29 on Staphylococcus epidermidis. The TLC show chemical compounds contained in the ethanol extract, n-heksan fraction, ethyl acetate fraction, and ethanol-water fraction is a compound of the saponins, alkaloids, � avonoids and phenolic. Antibacterial activity of different types of P. odorata leaf extracts was evaluated in combination with standard antibiotics. Persicaria. odorata leaves were extracted with n-hexane (n-hex), dichloromethane (DCM) and methanol (MeOH).  Each extract was applied on vancomycin (30µg), erythromycin (15µg) and gentamicin (10µg) discs, respectively. Disk diffusion method was used to evaluate the synergistic activity of each combination on Staphylococcus aureus, Staphylococcus epidermidis, Streptococcus pyogenes, Streptococcus pneumoniae, Pseudomonas aeruginosa, Salmonella typhi, and Escherichia coli. Minimum inhibitory concentration (MIC) and gas chromatography mass spectrometry (GCMS) analysis was performed on the active extract. Synergistic effects seen were mainly from the n-hex+antibiotics combinations, mainly on the Gram-positive bacteria (7 additive, 5 antagonistic), with MIC range from 50 µg/ml to 100 µg/ml, as well as Gram-negative bacteria (2 additive, 2 indifferent, 5 antagonistic). In particular, synergism showed by the combination of n-hex+van were all additive against the susceptible bacteria. DCM extract combination showed synergistic effects on three Gram-positive species (S. aureus, S. epidermidis, S. pyogenes). Meanwhile, MeOH+antibiotics combination showed significant additive synergistic effects (p<0.05) on S. aureus and S. epidermidis.  The major compounds of leaves extract were decanal and β-citral. n-Hex extract superiorly inhibited Gram-positive bacteria growth as compared to DCM and MeOH extracts. The additive synergistic property of the n-hex P. odorata extract could be further studied for possible use as an antibacterial agent.

Essential oils (EOs) and honeybee products (e.g., honey and propolis) are natural mixtures of different volatile compounds that are frequently used in traditional medicine and for pathogen eradication. The aim of this study was to evaluate the antibacterial properties of tea tree (Melaleuca alternifolia) EO (TTEO), Rosmarinus officinalis EO (ROEO), manuka-based gel, and propolis against 23 strains of Staphylococcus pseudintermedius (SP) isolated from canine pyoderma. Antimicrobial resistance screening was assessed using a panel of nine antimicrobial agents coupled with a PCR approach. An aromatogram was done for both EOs, using the disk diffusion method. The minimum inhibitory concentration (MIC) was determined for all the compounds. Among the 23 SP strains, 14 (60.9%) were multidrug-resistant (MDR), 11 strains (47.8%) were methicillin-resistant (MRSP), and 9 (39.1%) were non-MDR. The mean diameter of the inhibition zone for Melaleuca and Rosmarinus were 24.5 ± 8.8 mm and 15.2 ± 8.9 mm, respectively, resulting as statistically different (p = 0.0006). MIC values of TTEO and ROEO were similar (7.6 ± 3.2% and 8.9 ± 2.1%, respectively) and no statistical significances were found. Honeybee products showed lower MIC compared to those of EOs, 0.22 ± 0.1% for Manuka and 0.8 ± 0.5% for propolis. These findings reveal a significant antibacterial effect for all the tested products.

Abstracts Background Resistance to antimicrobial agents of pathogenic bacteria has become a major problem in routine medical practices. Carbapenem resistance has long been increasing. The production of carbapenem- hydrolysing β-lactamases (carbapenamases), which include NDM, KPC, OXA-48, IMP-1 and VIM is the most common mechanism. Case presentation A 56 years old male presented with fever and mental changes with progressively decreasing sensorium for the last 3 days. He was admitted to Intensive care unit (ICU) with a diagnosis of meningoencephalitis. On day seven, he developed ventilator associated pneumonia due Klebsiella pnemoniae and Acinetobacter baumannii. He was on meropenem, but the isolates were susceptible to colistin, tigecyclin and amikacin solely. Hence, amikacin was started with addition of intravenous and nebulized colistin. Subsequently, vital signs improved with resolution of fever. However, on day 18, he developed fever once again with a drop in blood pressure. Inotropic support was maintained, and echinocandins and tigecycline were added to the regimen. Repeat blood and urine culture grew Providencia species, which were resistant to most of the drugs on phenotypic Kirby-Bauer disk diffusion method and are intrinsically resistant to colistin and tigecycline. Phenotypic detection of ESBL (combined disk method), MBL, KPCs, AmpC and co-producer were tested according to updated CLSI guideline and all were negative. But the Modified Hodges test was found to be positive. Consequenty, OXA-48 drug resistance pattern was brought into action by blank disc method according to A Tsakris et al., which revealed indentation of growth toward both EDTA and EDTA/PBA disk indicating production of OXA-48 carbapenamase. To confirm the resistance pattern we processed the isolated colonies for Xpert Carba-R (Cepheid) assay, which detected blaOXA-48 gene and confirmed the OXA-48 drug resistance pattern. Hence, the infecting organism was not susceptible to any of the antibiotics. The patient was kept under isolation and on 31th day of admission, he died of septic shock. Conclusions Carbapenamase production along with intrinsic colistin resistance in infecting bacterial pathogens can cause fatal outcomes in the resource limited countries like Nepal where new antibiotic combinations ceftazidime+ Avibactam, or aztreonam +avibactam are not available. Drug resistance patterns including OXA 48 producer should be characterized in all cases by standard phenotypic methods or by Xpert Carba-R assay and larger studies are required to know the exact burden of OXA 48 producer in Nepal.
