Background: Medicinal plants are widely used in Nigeria because they are believed to be effective in the treatment of various medical conditions and are also easily accessable with minimal side effect. Aim: This study evaluates the prophylactic and therapeutic effects of different doses (200 mg/kg, 400 mg/kg and 800 mg/kg body weight) of Costus afer on lipid profile of 50 male albino rats. Methodology: The research study was divided into 2 phases with 25 rats used for each phases. The 25 rats used for each phase were randomly selected into 5 groups with each group containing 5 rats. The rats used for the prophylactic phase were induced with 800 mg/kg body weight paracetamol for liver toxicity after administration of the various concentrations of aqueous stem extract of C. afer for 28 days while those used during the therapeutic phase were administered with the various concentrations of aqueous stem extract of C. afer following confirmation of liver toxicity using 800 mg/kg body weight acetaminophen. The effect of the aqueous extract was assessed by measuring the serum concentration of total cholesterol, triglycerides and high density lipoprotein using Randox reagent, while low density lipoprotein was calculated from the other parameters. Atherogenic ratios were also computed. The result obtained from the experiment was subjected to statistical analysis using Graph pad prism version 5.3 and values were considered significant at p<0.05. Results: Total cholesterol, triglycerides and LDL levels were significantly (p<0.05) reduced and HDL significantly increased in the treatment groups (prophylactic and therapeutic phases) compared to the positive control. When both phases were compared, total cholesterol and triglycerides showed significant (p<0.05) difference in concentration in groups fed with 400 mg/kg, 200 mgkg while LDL-C showed significant (p<0.05) variation between the two phases only at 400 mg/kg body weight. The extracts were also found to significantly (p<0.05) reduce the atherogenic status of the albino rats in both phases of treatment and between each treatment phase. Conclusion: Findings from this study suggest that Costus afer possesses the ability to regulate paracetamol induced dyslipidaemia and improve the anti-atherogenic status of treated albino rats.

Abstract Background During the last few decades, patients worldwide have been interested in using alternative medicine in treating diseases to avoid the increased side effects of chemical medications. Green coffee is unroasted coffee seeds that have higher amounts of chlorogenic acid compared to roasted coffee. Green coffee was successfully used to protect against obesity, Alzheimer disease, high blood pressure and bacterial infection. Methods This study aimed to investigate the probable protective activity of the green coffee methanolic extract, silymarin and their combination on CCl4-induced liver toxicity in male rats. Thirty Sprague – Dawley male albino rats were divided into 5 groups; control negative (G1) just got the vehicle (olive oil) and the other four groups received CCl4 dissolved in olive oil through an intraperitoneal injection and were divided into untreated control positive group (G2), the third group (G3) was treated with green coffee methanolic extract, the fourth group (G4) was treated with silymarin, and the fifth group (G5) was treated with a combination of green coffee methanolic extract and silymarin. Results In the positive control group treated with CCl4 (G2), the CCl4-induced toxicity increased lipid peroxidation, IL-6, kidney function parameters, liver function enzymes, total cholesterol, triglycerides and low-density lipoproteins, and decreased irisin, antioxidants, CYP450 and high-density lipoprotein levels. Hepatic tissues were also injured. However, treating the injured rats in G3, G4 and G5 significantly improved the altered parameters and hepatic tissues. Conclusions Green coffee methanolic extract, silymarin, and their combination succeeded in protecting the male rats against CCl4 hepatotoxicity due to their antioxidant activity. Effect of green coffee methanolic extract mixed with silymarin in G5 was more efficient than that of green coffee methanolic extract in G3 or silymarin in G4.

National data shows the prevalence of coronary heart disease is 3,6%. The prevalence of dyslipidemia is around 35,9%. Dyslipidemia has a causal relationship with cardiovascular disease. Based on previous research, 70% ethanolic extract of Ridge Gourd has been shown to have antihyperlipidemic activity. This study aimed to study the activity of 70% ethanolic extract fraction of Ridge Gourd in decreasing the levels of total cholesterol, low-density lipoprotein (LDL-C), and triglycerides in hyperlipidemic hamsters. A total of 32 hamster were divided into 8 groups. Negative control, normal control, positive control (using atorvastatin dose 1,24 mg/kg body weight), positive control (fenofibrate dose 12,35 mg/kg body weight), ethyl acetate fraction, n-hexane fraction, water fraction (fraction doses 36,75 mg/kg body weight), and ethanol extract group (extract dose of 240 mg/kg body weight). The results showed that fraction group (n-hexane, ethyl acetate, water) and ethanol extract group experienced a decrease in total cholesterol, LDL-C, and triglycerides levels. Statistical analysis with one-way ANOVA showed an effect on the treatment given (p = 0,005). Tukey's test showed that ethyl acetate fraction group had an effect comparable to positive control with a decrease in total cholesterol,  LDL-C, and triglycerides level by 56,03%, 52,14%, and 59,51% consecutively. Ethyl acetate fraction (doses 36,75 mg/kg body weight) was the most effective fraction in reducing total cholesterol, LDL-C, and triglycerides.

Aim: This study was targeted at valuing a claim by traditional herbal practitioners that the leaf of Eriosema psoraleoides possesses a hypoglycemic and hypolipidemic property by assessing the effect of aqueous leaf extract of Eriosema psoraleoides on antihyperglycemic and hypolipidemic potentials in alloxan-induced diabetic rats. Methodology: Thirty male albino rats weighing 180-220 g were divided into 5 experimental groups of six rats each; control, diabetic untreated, diabetic treated with glibenclamide and diabetic treated with Eriosema psoraleoides. Diabetes was induced by 130 mg/kg body weight (b.wt) of alloxan monohydrates. The control and diabetic groups received normal saline while the diabetic treated groups were administered with 0.3 mg/kg body weight glibenclamide, and 200 mg/kg, 400 mg/kg body weight of aqueous leaf extract of Eriosema psoraleoides respectively. The experiment period was 7 days, with the determination of their glucose level and body weight every two days.  At the end of the experimental period, the animal’s blood samples were taken from the animals for the determination of total cholesterol (TC), triglycerides (TG), high-density lipoprotein (HDL) and low-density lipoprotein (LDL). Statistical comparisons were performed by one-way analysis of variance with repeated measures and one-way analysis of variance followed by Duncan’s multiple range tests. Results: The results of the study show a significant reduction in blood glucose. The result also showed that in diabetic rats, body weight was decreased but the application of the extract increased (P<0.05, n=6). Similarly, the result showed a significant decrease in total cholesterol, TG and LDL level of the diabetic group when compared with the control, glibenclamide and extract treated diabetic groups (with the highest performance at 400 mg/kg). Also, Eriosema psoraleoides aqueous leaf extract treated diabetic rat’s shows a significant increase in HDL levels compared to the diabetic control. Conclusion: The study indicates that Eriosema psoraleoides possess hypolipidemic and antihyperglycemic potentials.

Abstract Objectives Protocatechuic acid (PCA) possesses numerous pharmacological activities, including antioxidative and anti-inflammatory activities. This study seeks to investigate its underlying mechanism of action in the liver and brain toxicity induced by CCl4 in male albino rats. Methods Rats were given PCA at 10 and 20 mg/kg daily and orally as a pretreatment for seven days. A single injection of CCl4 was given 2 h later to induce brain and liver toxicity. Results CCl4 moderately elevated the activities of alanine aminotransferase (ALT), aspartate aminotransferase (AST), and alkaline phosphatase (ALP). PCA lowered AST level significantly when compared to control. Total protein and albumin levels presented insignificant changes (p>0.05) in all groups while lipid profile showed increased total cholesterol level and reduced high-density lipoprotein (HDL) by CCl4. PCA (10 mg/kg) significantly reduced the cholesterol level while the 20 mg/kg dose moderately prevented HDL reduction. There was an increased MDA production with a corresponding low GSH level in the group treated with CCl4. Activities of superoxide dismutase, catalase, and glutathione-S-transferase in both organs also declined. PCA, especially at 10 mg/kg attenuated lipid peroxidation by increasing GSH level in the organs. Biochemical assays revealed the improvement of antioxidant enzyme activities by PCA in these organs. Furthermore, PCA lowered the level of proinflammatory cytokine COX 2 in the brain and liver while NF-kB expression was inhibited in the brain. Histopathology reports validated the effects of PCA. Conclusions PCA exhibited protection against toxicity in these tissues through antioxidant and anti-inflammatory activities and the potential mechanism might be through modulation of the NF-κB/COX-2 pathway.

Postprandial hyperglycemia is a predisposing factor for vascular dysfunction and organ damage.α-glucosidase is a hydrolytic enzyme that increases the glucose absorption rate and subsequently elevates blood glucose levels. Garlic (Allium sativumL.) is a rich source of several phytonutrients, including thiosulfinate (THIO). The aim of this study was to evaluate the ability of THIO, a potent inhibitor of intestinalα-glucosidase, to reduce postprandial blood glucose. Male albino rats were randomly assigned to five different groups (n=10/group). Group 1 served as the control group. Groups 2–5 were injected intraperitoneally with a single dose of streptozotocin (STZ) to induce diabetes. Group 2 comprised untreated diabetic rats. Groups 3 and 4 contained diabetic rats that were given THIO orally (20 mg/kg body weight/day and 40 mg/kg body weight/day, resp.). Group 5 was the positive control having diabetic rats treated orally with acarbose (10 mg/kg body weight/day; positive control). Diabetic rats treated with THIO displayed a significant blood glucose reduction (p<0.001and < 0.01 by analysis of variance, resp.) and a significant elevation in insulin compared with that of untreated rats. THIO is an effective noncompetitive intestinalα-glucosidase inhibitor that promotes hypoglycemic action (p<0.001) in STZ-injected rats. THIO is a promising agent for the management of postprandial hyperglycemia.

Objective: The objective of this study was to determine if there were any harmful effects of monosodium glutamate (MSG) on the liver of Wistar albino rats chronically at three different doses, namely, low, mid, and high doses equivalent to human consumption doses in developing countries. Methods: The Wistar albino rats (n=24) were divided into four groups, namely control, Low dose MSG (180 mg/kg), Mid dose MSG (360 mg/kg), and High dose MSG (720 mg/kg). At the end of the experimental period (120 days), animal blood was collected retro-orbitally to analyze the liver enzymes such as aspartate transaminase (AST), alanine transaminase (ALT), alkaline phosphatase (ALP), Total protein, Albumin, and Total Bilirubin in blood serum. Lipid profiles, namely, Triglycerides, low-density lipoprotein (LDL), high-density lipoprotein (HDL), and Total cholesterol were subjected to analysis using blood serum. Results: Significant increase (p<0.05) in AST, ALT, ALP, and total bilirubin in serum of MSG induced low, mid, and high dose groups when compared to control group were recorded. There was a significant increase (p<0.05) in LDL, decrease in HDL, increase in total cholesterol and triglycerides of MSG-induced animal groups. Conclusion: The effects of MSG on serum liver enzymes and lipid profiles in this present animal study were not severely alarming even though the dosage was chronic which opens further discussion on the controversies revolving around MSG.

To investigate the antihyperglycemic and antihyperlipidemic effect of Momordica charantia (Karala), the aqueous extract of the Karala fruit was tested on streptozotocin (STZ)-induced diabetic rats. Thirty six albino rats were used in the experiment, 30 diabetic and the remaining six as negative control (T1). Diabetes was induced by administering (injecting) STZ at dose of 55mg/kg body weight. Thirty diabetic animals were randomly divided into five groups such as diabetic control group (T2) without any application of treatment, and groups T3,T4,T5 and T6 were treated with aqueous extract of Karala fruits daily at the doses of 250,    500 and 750mg/kg and glibenclamide (at a dose of 5mg/kg body weight) respectively. The body weight was taken and blood samples were collected from individual animal to determine glucose levels at 15 day interval up to 90 days. In addition, Asparate  Transaminenase(AST), Alanine Transaminenase(ALT), Alkaline Phosphatase(ALP), Total cholesterol (TCh) and Triglyceride (TGA) were determined at day 15 and at the end of the experiment. All three doses of Karala extracts reduced diabetic induced blood sugar and the reduction is comparable with standard glibenclamide (GLM) dose particularly with higher doses Karala extracts (500 and 750mg). Karala also prevented body weight loss due to induced diabetes as did by GLM treatment.. The treatment also resulted in a significant reduction of Asparate Transaminenase(AST), Alanine Transaminenase(ALT), Alkaline Phosphatase(ALP), Total cholesterol (TCh) and Triglyceride (TGA) activities of treated rats when compared to the STZ induced  diabetic rats. Higher doses of Karala (500 and 750mg/kg) are as effective as standard GLM dose on measured variables. This study demonstrated that Karala has hyperglycemia and antihyperlipidemic effect against STZ induced diabetic rats. These findings open the possibility of using Karala extract to treat diabetic animal and human patients although further research is warranted. DOI: http://dx.doi.org/10.3329/jesnr.v5i1.11550 J. Methamphetamine intoxication can cause acute hepatic failure. Chlorogenic and caftaric acids are the major dietary polyphenols present in various foods. The aim of this study was to evaluate the protective role of chlorogenic and caftaric acids in liver toxicity and oxidative stress induced by methamphetamine in rats. Thirty-two male albino rats were divided into 4 equal groups. Group 1, which was control group, was injected (i.p) with saline (1 mL/kg) twice a day over seven-day period. Groups 2, 3, and 4 were injected (i.p) with methamphetamine (10 mg/kg) twice a day over seven-day period, where groups 3 and 4 were injected (i.p) with 60 mg/kg chlorogenic acid and 40 mg/kg caftaric acid, respectively, one day before methamphetamine injections. Methamphetamine increased serum aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, bilirubin, cholesterol, low-density lipoprotein, and triglycerides. Also, malondialdehyde in serum, liver, and brain and plasma and liver nitric oxide levels were increased while methamphetamine induced a significant decrease in serum total protein, albumin, globulin, albumin/globulin ratio, brain serotonin, norepinephrine and dopamine, blood and liver superoxide dismutase, and glutathione peroxidase levels. Chlorogenic and caftaric acids prior to methamphetamine injections restored all the above parameters to normal values. In conclusion, chlorogenic and caftaric acids before methamphetamine injections prevented liver toxicity and oxidative stress where chlorogenic acid was more effective.

This study investigated the hypoglycaemic and hypolipidaemic effects of Treculia africana plant used in Nigeria as medicinal plant. Diabetes mellitus was induced by a single dose intraperitoneal injection of alloxan 150 mg/kg body weight. Twenty five (25) male albino rats were divided into five groups, five (5) rats per group; normal control, diabetic control and diabetic groups treated with aqueous leaves extract of 200,400 and 800 mg/Kg body weight respectively for 21 days orally. The effects of the extract on some biochemical parameters were evaluated; fasting blood glucose level was assayed using glucose oxidase method, total cholesterol and HDL –cholesterol were assayed using enzymatic method while LDL- cholesterol was determined by Friedewald equation. The results showed that, extract significantly (p<0.05) decrease the elevated fasting blood glucose levels, total cholesterol, triglyceride and LDL- cholesterol when compared with the diabetic control rats. The extract also caused significant (p<0.05) increased in HDL –cholesterol and body weight when compared with diabetic control rats. Aqueous leave extract of Treculia africana possess hypoglycemic effect and the most effective dose was 800 mg/Kg body weight in amelioration of hyperglycaemia and most all toxicity effects of alloxan on lipid profile.
