The purpose of this study is to examine whether brand image, food quality, and service quality have a significance effect towards customer repurchase intention on KFC Kakialy in Ambon. The sample was selected using purposive sampling method with total sample amount 100 valid respondent who had bought twice in that outlet. The data were collected and processed with SmartPLS v.3.2.8 program. The validity test was assessed using AVE and loading factor and reliability test was assessed using cross-loading. The formative construct was assessed by using sign of weight and multicollinearity test. The results of this study indicate that brand image does not have a positive and significant effect on repurchase intention. Food quality and service quality have a positive and significant effect on repurchase intention. Penelitian ini bertujuan untuk menguji apakah citra merek, kualitas makanan, dan kualitas layanan pengaruh signifikan terhadap niat pembelian kembali KFC Cabang Kakialy di Ambon. Pemilihan sampel dilakukan dengan menggunakan metode purposive sampling dengan total sebanyak 100 responden yang valid dan pernah melakukan pembelian minimal dua kali di gerai tersebut. Data yang terkumpul kemudian diolah menggunakan program SmartPLS v.3.2.8. Pengujian validitas menggunakan uji AVE dan loading factor dan pengujian realibilitas menggunakan cross-loading. Pengujian indikator formatif menggunakan uji sign of weight dan uji multikolinearitas. Hasil penelitian ini menunjukkan bahwa citra merek tidak memiliki pengaruh yang positif dan signifikan terhadap niat pembelian kembali. Pada kualitas makanan dan kualitas layanan memiliki pengaruh yang positif dan signifikan terhadap niat pembelian kembali.

Although many advanced insulation materials have been recently developed, very few are eco-friendly and their production requires a substantial amount of energy and complex manufacturing processes. To address this issue, a bio-based thermal insulation material was developed using short- and long-grained puffed rice. A set of experiments was subsequently carried out to identify the best rice type and the optimal range for the most influential parameters (sample amount, temperature, and moisture level). Our findings revealed that short-grained rice exhibited greater puffing ability and was thus adopted in further material optimization experiments. These assessments indicated that the most optimal thermal conductivity of the insulation material and the highest puffing ratio was attained at 12–15% moisture, 260–270 °C temperature, and 15–18 g sample weight. The thermal properties, including thermal conductivity and fire reaction, and thermal performance of samples obtained using these parameters were similar to those of common insulation materials.

Magnetic immunoassays employing Frequency Mixing Magnetic Detection (FMMD) have recently become increasingly popular for quantitative detection of various analytes. Simultaneous analysis of a sample for two or more targets is desirable in order to reduce the sample amount, save consumables, and save time. We show that different types of magnetic beads can be distinguished according to their frequency mixing response to a two-frequency magnetic excitation at different static magnetic offset fields. We recorded the offset field dependent FMMD response of two different particle types at frequencies f1 + n⋅f2, n = 1, 2, 3, 4 with f1 = 30.8 kHz and f2 = 63 Hz. Their signals were clearly distinguishable by the locations of the extremes and zeros of their responses. Binary mixtures of the two particle types were prepared with different mixing ratios. The mixture samples were analyzed by determining the best linear combination of the two pure constituents that best resembled the measured signals of the mixtures. Using a quadratic programming algorithm, the mixing ratios could be determined with an accuracy of greater than 14%. If each particle type is functionalized with a different antibody, multiplex detection of two different analytes becomes feasible.

One of the most common methods for purifying RNA is using TRIzol reagent because of its simplicity and economic feasibility. However, the drawback of this method is frequently the low quality of extracted RNA due to contaminants from the residue of phenol and guanidinium thiocyanate from the reagents. This study aimed to evaluate the improvement in the quality and concentration of RNA after the optimisation treatment. One-month-old tomato (Solanum lycopersicum) stem was used in this research. TRIzol or acid guanidinium thiocyanate-phenol-chloroform-based method was given optimisation treatments of the initial sample amount, twice chloroform extraction, overnight precipitation at low temperature, and three times final washing with ethanol. The results showed no significant improvement (p > 0.05) in the purity ratio A260/A280. At the same time, there was a significant improvement (p < 0.05) in RNA yield and purity ratio A260/A230. The quality of RNA was verified using agarose-formaldehyde electrophoresis gel. Eight of nine samples (89%) from the optimised group had better RNA integrity characterised by sharp bands for 28S and 18S rRNA. Furthermore, a representative sample from the optimised group was successfully synthesised into complementary DNA by reverse transcriptase-polymerase chain reaction (RT-PCR) with primers of the ubiquitin (UBI3) gene. To sum up, optimised TRIzol-based protocol provides meaningful insight to produce RNA with better quality and suitability for downstream applications.

Latar Belakang. Indonesia adalah negara dengan populasi terbanyak di Asia Tenggara. Dalam mengantisipasi masalah laju pertumbuhan penduduk, Pemerintah mempersiapkan program Keluarga Berencana dan salah satu sasarannya adalah remaja.Tujuan. Penelitian ini bertujuan untuk mengetahui hubungan antara kebutuhan pelayanan keluarga berencana pada remaja belum kawin dengan niat menggunakan kontrasepsi di masa dewasaMetode. Penelitian menggunakan kuantitatif analitik dengan pendekatan cross sectional. Kuesioner yang digunakan Remaja Pria (RP) dan Wanita Usia Subur (WUS). Populasi dalam penelitian ini adalah seluruh remaja belum kawin di Indonesia dengan sampel berjumlah 18.464 responden.Hasil. 82,6% remaja yang berniat menggunakan kontrasepsi di masa dewasa. Hasil uji chi-square terdapat hubungan antara kebutuhan pelayanan KB pada remaja belum kawin dengan niat menggunakan kontrasepsi di masa dewasa. Hasil analisis multivariat, ada hubungan antara kebutuhan pelayanan KB pada remaja belum kawin dengan niat menggunakan kontrasepsi di masa dewasa setelah di kontrol variabel perancu.Kesimpulan. Remaja pria belum kawin yang tidak butuh pelayanan KB, terpapar media massa, memiliki pengetahuan <4 alat/cara KB lebih tidak berniat menggunakan kontrasepsi di masa dewasa. ABSTRACTBackground. Indonesia is the country with the most population in Southeast Asia. In anticipating a Population Growth Rate problem, the government is preparing a family planning program with youth as one of the targets. Objective. This study aimed to determine the relationship between the need for family planning services in unmarried adolescents and the intention to use contraception in adulthood.Method. The study is quantitative analytic with a cross-sectional approach. The questionnaire used by Adolescent Men (RP) and Women of Fertile Age (WUS). The population in this study were all unmarried adolescents in Indonesia, with sample amount 18,464 respondents. Results. 82.6% of adolescents intend to use contraception in the future. Chi-square test results show a relationship between the need for family planning services for unmarried adolescents with the intention to use contraception in adulthood. The result of the multivariate analysis shows a relationship between the need for family planning services for unmarried adolescents with the intention to use contraception in adulthood after being controlled for confounding variables.Conclusion: Unmarried male adolescents who do not need family planning services, are exposed to mass media, have knowledge of <4 methods/methods of contraception and do not intend to use contraception in adulthood.

ABSTRACT The purpose of research is for examine in empirical influence comprehension, trust, and transparency on zakat organization to society interest pay zakat to BAZNAS LUWU area. Subject to use in the research is independent subject (comprehension, trust, and transparency on zakat organization) but dependent subject is (interest). Research do this on Luwu Area with take population muzakki from Badan Amil Zakat Nasional (Baznas). Research kind to use this quantitative descriptive approach with survey method. Population in research is Muzakki it has name on registered BAZNAS Luwu Area. Interpretation technic sample use Accidental samping method is so easy sample to access. Amount determining sample use Slovin formulation. Sample amount to success process data has 68 people. Analysis data tecnic for use in research is Multiple Regresion linear with SPSS Version 23. Result from hypotheses test on research in a partial indication value t count for subject comprehension as big as -0,397. And for trust as 3,394 and for transparency on zakat organization as 4.114. so, result from t count show it in a partial subject comprehension nothinginfluenceto society interest pay zakat to Baznas Luwu Area but subject trust and transparency on zakat organization has influence to society interest pay zakat to Baznass Luwu Area. ABSTRAK Penelitian ini bertujuan untuk menguji secara empiris pengaruh pemahaman, trust, dan transparansi lembaga zakat terhadap minat masyarakat membayar zakat pada BAZNAS Kabupaten Luwu. Variabel yang digunakan dalam penelitian ini, yaitu variabel independen (pemahaman, trust, dan transparansi lembaga zakat) sedangkan variabel dependen (minat). Penelitian ini dilakukan di Kabupaten Luwu dengan mengambil populasi muzakki dari Badan Amil Zakat Nasional (BAZNAS). Jenis penelitian yang digunakan adalah pendekatan kuantitatif deskriptif dengan menggunakan metode survey. Populasi dalam penelitian ini adalah muzakki yang namanya terdaftar di BAZNAS Kabupaten Luwu. Teknik pengambilan sampel menggunakan metode Accidental Sampling yaitu sampel yang paling mudah untuk diakses. Penentuan jumlah sampel menggunakan rumus Slovin. Jumlah sampel yang datanya berhasil diolah yaitu sebanyak 68 orang. Teknik analisis data yang digunakan dalam penelitian ini adalah regresi linear berganda dengan menggunakan program SPSS Versi 23. Hasil dari pengujian hipotesis dalam penelitian ini secara parsial ditunjukkan nilai t hitung untuk variabel pemahaman sebesar -0,397, untuk variabel trust sebesar 3,394 dan variabel transparansi lembaga zakat sebesar 4,114. Jadi, hasil dari uji t membuktikan bahwa secara parsial variabel pemahaman tidak berpengaruh terhadap minat masyarakat membayar zakat pada BAZNAS Kabupaten Luwu. Sedangkan variabel trust dan transparansi lembaga zakat berpengaruh terhadap minat masyarakat membayar zakat pada BAZNAS Kabupaten Luwu. <b><i>Background:</i></b> Although serum carbohydrate antigen 19-9 (CA19-9) is widely used as a useful biomarker of pancreatic cancer for monitoring the response to therapy, it is not recommended for screening of early pancreatic cancer because of its limited sensitivity for small tumors. Thus, it is critical to discover novel serum biomarkers to complement CA19-9 in order to improve sensitivity. Although methylated runt-related transcription factor 3 (<i>RUNX3</i>) is a biomarker of pancreatic cancer, its detection by conventional bisulfite-based methylation assays from a small serum sample amount is very difficult. Therefore, we developed a new methylation assay, the combined restriction digital PCR (CORD) assay, that enables counting of even one copy of a methylated gene in a small DNA sample amount without DNA bisulfite treatment. <b><i>Objectives:</i></b> We evaluated the sensitivity and specificity of serum DNA testing of methylated <i>RUNX3</i> by the CORD assay in combination with and without CA19-9 for the detection of pancreatic cancer in 55 patients with pancreatic cancer, 12 patients with benign pancreatic disease, and 80 healthy individuals. <b><i>Results:</i></b> The CORD assay of methylated <i>RUNX3</i> had a sensitivity of 50.9% (28/55) and specificity of 93.5% (86/92). Combination of the CORD assay of methylated <i>RUNX3</i> and CA19-9 resulted in a sensitivity of 85.5% (47/55) and specificity of 93.5% (86/92) for all stages of pancreatic cancer and a sensitivity of 77.8% (7/9) for stage I pancreatic cancer. <b><i>Conclusions:</i></b> ombination of the CORD assay and CA19-9 may provide an alternative screening strategy for detecting early-stage pancreatic cancer.

The teacher-centered mindset and teaching methods are still teacher-centered, closing the space for students to develop their creative thinking abilities. This study aims to determine the thinking skills of students in physical education learning using discovery learning models. This study used a quasi-experiment method and a pretest and posttest control group design. The research sample was obtained by means of cluster random sampling with a sample amount of 70 people. The instrument that used to measure students' creative thinking skills used the Torrance Test of Creative Thinking (TTCT) which before be tested, it have conducted the validity and reability. The t test was used as the analysis of this study. Discovery learning model is proven to be able to develop students' creative thinking skills when learning physical education. This is proven by the sig value. 0,000. So the discovery learning model in physical education learning will be able to develop creative thinking on students. This learning model must be applied in various fields of education. Research on the same topic should be carried out in the future, with the variability of different educational levels of subjects or educational institutional settings

SAMPLE INJECTION METHODS IN ELECTROPHORETIC MICROSYSTEMS. Electrophoresis is by far the most popular separation method implemented in microscale, most probably due to its instrumental simplicity, low cost and portability. Due to the increasing use of miniaturized electrophoretic systems, the study of fundamental aspects can improve the development of methodologies for several applications. One of the major challenges related to electrophoresis chips refers to the sample injection mode, in this way, this study presents a review on sample injection methods for microchip electrophoresis covering electrokinetic and hydrodynamic approaches, describing theoretic and instrumental aspects. Since the sample volume affects the analytical performance, the precise and reproducible control of the sample amount to be introduced into the separation channel is highly desirable to ensure reliable chemical analysis. Electrokinetic modes based on floating, pinched and gated protocols are presented and discussed providing an overview about the electrokinetic control of the sample through three methodologies. In the same way, hydrodynamic techniques including the use of microfabricated valves and pumps, syringe pumps, electronic micropipettes, rubber suction bulb and acupuncture needle are approached, thus expanding the view of hydrodynamic injectors based on classical and alternative methods.

The purpose of this research is to determine the effect of queue, service quality on customer satisfaction of Biznet Home in Kelapa Gading Branch partially and simultaneously. The population in this research is the customers of Biznet Home in the Jakarta Utara region. Total sample amount of this research is 32 respondents, which for the sampling here by using the Purposive Sampling technique. Data collected by using survey method with questionnaire instrument. For the data analysis method used by the researcher, is double regression analysis, t test (partial) and F test (simultaneous). From the result of this research shows that: (1) The effect of Queue positively affects the customer satisfaction, with tcount 3.912 > from ttable or 3.912>2.045 then H1 is accepted. (2) The Service Quality significantly affects the customer satisfaction, tcount 6.653 > from the ttable or 6.653 > 2.045 then H2 is accepted. From the result of those hypothesis test then the decision taken is simultaneously all of the independent variable has positive and significant effects on customer satisfaction.
