Abstract Biochar has been used to alleviate the contamination of antibiotic resistance genes (ARGs) in soil and to inhibit ARGs transfer from soil to plants. However, the effect of aged biochar on ARGs abundance in soil and ARGs enrichment in plants are scarcely investigated. In this study, a pot experiment was conducted to compare the effects of fresh and aged biochars on the accumulation of five typical ARGs including tetX, tetW, sul2, ermB and intI1 in a chicken manure-fertilized soil and in pakchoi (Brassica chinensis L.). Results showed that both biochars significantly decreased the abundance of tetW, sul2, and ermB and increased the abundance of tetX and intI1 in soil. However, the accumulation of all tested ARGs in pakchoi were significantly decreased by both biochars. At the lower addition rate (1%), the fresh biochar was superior to the aged biochar in decreasing the accumulation of some genes (tetW, tetX, and sul2) in pakchoi, whereas an opposite tendency was observed for other genes (ermB and intI1). As the addition rate increased to 2%, the difference between the two biochars diminished and a similar capacity of decreasing ARGs transfer was observed. The reduction in ARGs accumulation in pakchoi was highly related to the type of ARGs, the biochar addition level and the aging of biochar. Our results provide insights into the naturally aged biochar on the fate of ARGs in a soil-plant system.

Abstract. Seawater intrusion in island aquifers was considered analytically, specifically for annulus segment aquifers (ASAs), i.e., aquifers that (in plan) have the shape of an annulus segment. Based on the Ghijben–Herzberg and hillslope-storage Boussinesq equations, analytical solutions were derived for steady-state seawater intrusion in ASAs, with a focus on the freshwater–seawater interface and its corresponding watertable elevation. Predictions of the analytical solutions compared well with experimental data, and so they were employed to investigate the effects of aquifer geometry on seawater intrusion in island aquifers. Three different ASA geometries were compared: convergent (smaller side is facing the lagoon, larger side is the internal no-flow boundary and flow converges towards the lagoon), rectangular and divergent (smaller side is the internal no-flow boundary, larger side is facing the sea and flow diverges towards the sea). Depending on the aquifer geometry, seawater intrusion was found to vary greatly, such that the assumption of a rectangular aquifer to model an ASA can lead to poor estimates of seawater intrusion. Other factors being equal, compared with rectangular aquifers, seawater intrusion is more extensive, and watertable elevation is lower in divergent aquifers, with the opposite tendency in convergent aquifers. Sensitivity analysis further indicated that the effects of aquifer geometry on seawater intrusion and watertable elevation vary with aquifer width and distance from the circle center to the inner arc (the lagoon boundary for convergent aquifers or the internal no-flow boundary for divergent aquifers). A larger aquifer width and distance from the circle center to the inner arc weaken the effects of aquifer geometry, and hence differences in predictions for the three geometries become less pronounced.

In psychotherapy, ambivalence may be conceptualized as a conflict between two distinct motivations: one that is favorable to change (pro-change) and another that favors the maintenance of a problematic pattern (pro status quo). Previous studies identified two processes by which clients resolve this conflict: imposing the innovative part and silencing the problematic one (dominance), and establishing negotiations between the innovative and the pro status quo parts (negotiation). The present exploratory study examined ambivalence resolution in a sample of clients diagnosed with complicated grief. Results revealed that, in recovered cases, negotiation increases and dominance decreases from the beginning until the middle sessions of therapy and the opposite tendency is observed from the middle to the final sessions. Unchanged cases reveal an overall high proportion of dominance and an overall low proportion of negotiation. These results are partially divergent from those reported in previous studies with samples of clients diagnosed with major depression.

Granulosa cell (GC) proliferation provides essential conditions for ovulation in animals. A previous study showed that DENND1A plays a significant role in polycystic ovary syndrome. However, the modulation of DENND1A in GCs remains unclear. Our previous integrated analysis of miRNA–mRNA revealed that the 3'-untranslated region of DENND1A could be a target of chi-miR-324-3p. In this study, we used quantitative reverse transcription polymerase chain reaction (RT-qPCR) to investigate DENND1A expression in ovarian tissues of high- and low-yielding goats. Furthermore, dual-fluorescent reporter vector experiments, Cell Counting Kit-8 (CCK-8) assay, and RT-qPCR were used to elucidate the regulatory pathway of chi-miR-324-3p-DENND1A in GCs. The results revealed an opposite tendency between the expressions of chi-miR-324-3p and DENND1A in the ovaries of high- and low-yielding goats. The CCK-8 assay indicated that chi-miR-324-3p overexpression significantly suppressed GC proliferation, whereas chi-miR-324-3p inhibition promoted GC proliferation. In addition, the expressions of GC proliferation markers LHR, Cylin D2, and CDK4 showed the same tendency. The dual-fluorescent reporter assay revealed that chi-miR-324-3p directly targeted DENND1A, and the RT-qPCR results revealed that DENND1A expression was inhibited by chi-miR-324-3p. In summary, chi-miR-324-3p inhibited the proliferation of GCs by targeting DENND1A.

AbstractIn order to explore the physiological reaction to hyperosmotic environment, Solenaia oleivora were exposed to 2.23‰ salinity. In 48h, the hemolymph osmolality kept increasing, and the hemolymph protein concentration increased in the first 6h and then decreased significantly, while the free amino acid content increased in the first 24h and then kept stable (P < 0.05). The activity of Na+/K+-ATPase at 0h was significantly higher than other times in most organs except intestine, which was highest at 3h (P < 0.05). The ions concentration were also influenced. The concentration of Na+ rose in haemolymph, axe foot and intestine, but decreased in gill and hepatopancreas. In hemolymph, gill, hepatopancreases and adductor muscle, the K+ concentration was the highest at 0h, while in axe foot and intestine, it showed a positive tendency. The concentration of Cl- in haemolymph, adductor muscle, intestine and axe foot were positively correlated with treatment time, while hepatopancreas showed opposite tendency. High salinity stress caused a difference in the gill histological structure, the gill structure shrunk, the gill lamellas space and shrinking degree showed an enlarging trend with salinity treatment time.

The presented research is focused on an investigation of the effect of the addition of polyvinyl alcohol (PVA) to a gelatin-based hydrogel on the functional properties of the resulting material. The main purpose was to experimentally determine and compare the properties of hydrogels differing from the content of PVA in the blend. Subsequently, the utility of these matrices for the production of an immobilized invertase preparation with improved operational stability was examined. We also propose a useful computational tool to predict the properties of the final material depending on the proportions of both components in order to design the feature range of the hydrogel blend desired for a strictly specified immobilization system (of enzyme/carrier type). Based on experimental research, it was found that an increase in the PVA content in gelatin hydrogels contributes to obtaining materials with a visibly higher packaging density, degree of swelling, and water absorption capacity. In the case of hydrolytic degradation and compressive strength, the opposite tendency was observed. The functionality studies of gelatin and gelatin/PVA hydrogels for enzyme immobilization indicate the very promising potential of invertase entrapped in a gelatin/PVA hydrogel matrix as a stable biocatalyst for industrial use. The molecular modeling analysis performed in this work provides qualitative information about the tendencies of the macroscopic parameters observed with the increase in the PVA and insight into the chemical nature of these dependencies.

To identify the microwave drying characteristics of corn, microwave drying tests were conducted on corn. By taking the moisture content, drying rate, and drying temperature as indices, this research revealed the effects of different microwave powers and loads on the microwave drying characteristics of corn. Moreover, energy consumption and quality of dried corn were analysed under different drying conditions. The results demonstrate that microwave drying has significant energy-saving effects. The energy consumption by microwave drying is less than 0.3 times that used by electrothermal drying under the same load. Both microwave power and load exert significant influences on drying characteristics. Higher microwave power results in a greater average drying rate, wherein shorter periods of time are required to reach the maximum drying rate and higher temperatures of the corn. However, the load shows the opposite tendency. The smaller the load, the higher the temperature of the corn in the early stage of drying. However, as drying continues, the temperature curve changes significantly, and the temperature rises with the increase in load in the later stage of drying. In consideration of energy consumption and dried quality, the load of corn should be increased as appropriate, and the microwave intensity should be limited to no higher than 0.7 W/g in the experiment.

Abstract Practitioners usually include change of direction (COD) and linear speed measurements in the testing batteries of soccer players; however, despite being a commonly occurring action, curve sprint (CS) ability is rarely assessed in soccer. The aims of this study were to analyze the association between linear sprint, CS, and COD speed performances, and compare the association and direction of asymmetries between these skills. Thirty-three male soccer players performed linear sprint (17 m), CS (17 m), and COD-90⍛ speed tests (COD [8.5 + 8.5 m]). Our main findings were (a) a large relationship between linear and multidirectional tasks (COD-90⍛ and CS tests) (r = from 0.6 to 0.64, p < 0.05), (b) a moderate relationship between CS and COD-90⍛ tests (r = from 0.33 to 0.41, p < 0.05), with a certain opposite tendency (higher relationships between opposing directions [CurveLEFT - CODRIGHT; r = 0.41] than between equal directions [CurveLEFT – CODLEFT; r = 0.33]), and (c) no relationship (p > 0.05) between COD and CS asymmetries, with opposing directional dominance in ~70% of players (e.g., curve left and COD right dominance). These results indicate that performance in linear sprints is strongly related to performance in multidirectional trajectories, whereas CS and COD-90⍛ seem to be more independent actions. Additionally, the direction of asymmetry or dominance is generally opposite between the non-linear tasks measured.

The antioxidant activity of beers comes mainly from phenolic compounds and melanoidins. The aim of this research was to evaluate the effect of technological operations, especially the ethanol fermentation process using top fermentation brewer’s yeast Saccharomyces cerevisiae, on the antioxidant activity of dark dry hopped beers with a high xanthohumol content. Four beers were produced using different varieties of hops. The polyphenol content during beer processing increased at the stage of hopping and fermentation, while it decreased during aging. The ability to reduce iron ions increased for all beers compared to hopped wort. The opposite tendency was noted for the antioxidant capacity expressed as the ability to reduce the radical cation ABTS•+ generated from 2,2′-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid). Fermentation and aging caused a decrease in beer color intensity. The content of 5-hydroxymethylfurfural (5-HMF) increased with the color intensity of wort, therefore in beers no presence of 5-HMF was observed. The beers were characterized by a distinctly high content of xanthohumol in the range of 1.77–3.83 mg/L and 0.85–1.19 mg/L of isoxanthohumol. The content of prenylflavonoids and bitterness of beer depended on the variety of hops used.

The collision effects on the low-frequency ion-acoustic Trivelpiece–Gould wave are investigated in weakly and completely ionized plasma waveguides by using the normal mode analysis. In weakly ionized plasma waveguides, it is found that the dependence of the harmonic mode on the absolute value of the scaled damping rate shows the opposite tendency for large and small radii of the cylindrical waveguide. It is also is found that the scaled damping rates for both weakly and completely ionized plasma waveguides decrease with an increase of the electron temperature. It is interesting to note that the scaled damping rate for weakly ionized plasma waveguides shows anti-symmetric behavior when the Trivelpiece–Gould wave propagates in the negative-z direction. However, it is found that the scaled damping rate for completely ionized plasma waveguides shows the symmetric behavior when the Trivelpiece–Gould wave propagates in the negative-z direction.
