Effect of prolactin inhibition on thermoregulation, water and food intakes in heat-stressed fat-tailed male lambs

AbstractTwo groups each of six lambs of the fat-tailed Naeimi breed, well adapted to a semi-arid climate, were housed under semi-controlled temperature (43·2 to 43·8°C) with low relative humidity (<0·8). After a 1-week adaptation period, the first group (control) was injected subcutaneously with 1 ml vehicle solution of 40% ethanol in saline, twice daily at 08.00 and 17.00 hfor the 11-day experimental period. In the other group, each lamb was injected daily for 6 days with 0·18 mg/kg per day of the prolactin inhibitor, 2 bromo-a-ergocryptine (CB154) dissolved in the vehicle solution. Treatment during the next 5 days was as for the control animals. Rectal temperature (RT), respiratory rate (RR), water and food intakes were recorded before injections. Daily average data were used in the statistical analysis, except those of the 1st day of bromocryptine injection. Lambs of the control group were able to maintain their RT (39·5 (s.e. 0·04) °C) during heat exposure, but those treated with the bromocryptine could not (40·3 (s.e. 0·07) °C). However, the latter group continued to respire more rapidly (P < 0·01) after cessation of the CB154 treatment, reducing their body temperature slightly (39·9 (s.e. 0·06) °C). This shows that, following prolactin suppression, heat-stressed lambs were unable to regulate their body temperature despite their RR. Water intake did not change during bromocryptine treatment, but the ratio of water to dry-matter intake did increase due to lower food consumption. These results suggest that prolactin might have an important role in thermoregulation of the fat-tailed male sheep during elevated ambient temperature.

The experiment aimed to evaluate the feeding behavior of Santa Inês lambs fed diets containing different levels of detoxified castor meal in the concentrate. Twenty-four sheep with an average body weight of 18.5± 2.26 kg, at four months of age, were distributed in a randomized complete design with four treatment and six replications. Treatments consisted of four levels of castor meal (0, 33, 67, and 100%) replacing soybean meal. Animals underwent a 15-day adaptation period and an experimental period of 84 days. The diet was composed of 60% sugarcane silage and 40% concentrate, on a dry matter basis. Animal behavior (idle, rumination, and feeding activities) were observed visually for two 24-h periods with 5-min intervals, and recorded. The number of rumination chews and the time taken to ruminate each cud per day were counted using a digital stopwatch. Dry matter (DM) intake was not affected by inclusion of castor meal in the concentrate, averaging 884.02 g day?1. The feeding activity was not affected by addition of castor meal. Rumination time increased linearly, whereas the idle and rumination times decreased linearly with addition of castor meal. Feeding time, expressed in min per kg of DM and NDF, was not influenced by the inclusion of the meal in the diet, averaging 373.3 and 880.0 min, respectively. The number of chews and the time taken to ruminate each cud were not changed by inclusion of castor meal in the diet. Feed efficiency, expressed in grams DM and NDF per hour, was not influenced by castor meal inclusion in the diet. Rumination efficiency expressed in g DM h?1 decreased, but was not affected when expressed in g NDF h?1. Total replacement of soybean meal by castor meal in the concentrate does not affect the feeding time or feed efficiency of feedlot sheep, but leads to a small reduction of their rumination efficiency. Thus, we recommend the inclusion of castor meal in sheep diets at the levels tested in this study.

In 2010 and 2011 two experiments on gimmers of Šumava mountain sheep were performed. The first experiment was carried out on 12 animals (experimental period was 11 months); control group was fed with 3 mg I*kg−1 in dry matter per day and experimental group was fed with 5 mg I*kg−1 in dry matter per day. The second experiment lasted 10 months and it was carried out on 12 animals. Feed ration for control group contained 10 mg I*kg DM−1. Feed ration for experimental group contained 10 mg I*kg DM−1 too; moreover it included rapeseed meal and 1 g of sodium nitrate. The aim of the first experiment was focused on impact of high iodine intake on structure of thyroid gland. The aim of the second experiment was to discover effect of strumigens during high iodine intake on structure of thyroid gland. The animals were slaughtered after the experiment and there was executed the dissection of thyroid gland. The samples of thyroid gland were processed during classic paraffin method and dyed with haematoxylin and eosin. For finding of histometrical parameters was used program Leica IM 500 Version 4.0. The length, the width and the area of follicles were measured. The follicles were divided into three groups after this procedure (by the length); in each group were measured 20 thyreocytes. In group with intake 5 mg I*kg DM−1 (experiment from year 2010): higher weight of thyroid (p < 0.01), significant higher (p < 0.05) proportional representation of large and medium follicles, significantly (p < 0.05) higher average size of follicles, demonstrably lower (p < 0.05) height of epithelium were found. There were not found any differences in monitored parameters between the groups from experiment which was carried out during the year 2011. After comparing results from the both experiments significantly higher percentage representation of large follicles a significantly lower representation of small follicles, distinctly higher average size and higher height of epithelium in all size categories, in groups with iodine intake 10 mg I*kg DM−1was found (from second experiment – year 2011).

The purpose of the research was to study the effectiveness of the use of rations for freshly calved cows during the increasing the milk yield period with the use of new mineral granulated complexes produced by Uralchem LLC. The results of research on the replacement of the mineral part of the vitamin and mineral premix in the rations of lactating cows with the mineral granular complex developed according to two recipes have been presented in the article. Research has shown that cows of the 1st and 2nd experimental groups during the experiment period gave more milk than their herdmates from the control group by 70,8 (P ≥ 0,95) and 106,2 kg (P ≥ 0,95). Due to the higher milk yield of animals that consumed the granulated complex, the yield of milk fat was greater than that of the control analogues by 3,21 (P ≥ 0,95) and 5,17 kg (P ≥ 0,95), protein by 2,70 and 4,30 kg (P ≥ 0,95). The milk yield of cows of the 1st and 2nd experimental groups for 305 days of lactation was higher than in the control group, respectively, by 205,3 (P ≥ 0,95) and 296,8 kg (P ≥ 0,95). In the milk of cows that consumed the granulated complex, there was a tendency for a higher content of dry matter (0,09 and 0,16 %), lactose (0,01 and 0,06 %), nonfat milk solids (0,09 and 0,07 %), ash (0,05 and 0,05 %) in comparison with analogues from the control group. A similar trend has been established between groups of cows in terms of milk density, acidity and rennet coagulability. The revenue from the sale of milk produced from cows of the 1st and 2nd experimental groups was more than in the control group for the experimental period (120 days) by 2074,6 and 3344,0 rubles and for the 305 days of lactation by 7055,4 and 9950,6 rubles.

120 selected layers (Bovans Goldline hybrid) aged 19 weeks with an average weight of 1 735 g were divided into two balanced groups: control group (Group C) and experimental (Group E). Layers were reared in three-tier cages with automatic watering, manual feeding, and at controlled light and temperature regimens. One cage accommodated two layers, the floor surface area was 0.1125 m<sup>2</sup> per layer. The experiment started after a 20-day adaptation period with layers aged 22 weeks (Week 22) and ended when layers were 68 weeks old (Week 68). In a period of initiation (i.e. Weeks 19–38), layers were fed the complete feeding mixture N1. Then they received the feeding mixture N2 until the end of the experiment. Feeding mixtures in both groups (C and E) had the same composition; the only difference between mixtures was that the feeding mixture in the experimental group was enriched with 1% clinoptilolite (i.e. the commercially available additive ZeoFeed). Layers received feeding mixtures and drinking water <I>ad libitum.</I> In the course of the experimental period, control layers laid 16 289 eggs while experimental layers laid 16 474 eggs. It follows from the results that the laying intensity in experimental layers was 1.7% higher as compared to control layers, i.e. the number of laid eggs in experimental layers increased by 5.6 eggs per layer. The mean weight of all laid eggs was 66.3 ± 6.25 g in the control group and 65.6 ± 5.44 g in the experimental group (<I>P</I> ≤ 0.01). Such performance was achieved at the consumption of feeding mixture being 141.7 g per laid egg in the control group and 137.6 g per laid egg in the experimental group. The consumption of feeding mixture in the experimental group was 4.1 g lower than that in the control group. The mean values of parameters monitored in blood plasma such as uric acid, cholesterol, glucose, lactose, calcium, phosphorus, ALP, and LDH in both groups of layers ranged within reference intervals, with no significant differences being detected between both groups.However, statistically significant differences between both groups were found in total plasma protein (<I>P</I> ≤ 0.01), triacylglycerol levels (<I>P</I> ≤ 0.05), and magnesium (<I>P</I> ≤ 0.01), which were elevated in the control group, and in AST (<I>P</I> ≤ 0.05) whose level in the control group was significantly lower than that in the experimental group. The results of haematological tests performed with layers’ blood revealed statistically significant changes in parameters such as the erythrocyte count (<I>P</I> ≤ 0.01), haemoglobin level (<I>P</I> ≤ 0.01), and MCHC (<I>P</I> ≤ 0.05), which were elevated in the experimental group, and in the leukocyte count (<I>P</I> ≤ 0.05), which was lower in the experimental group, as compared with the control. However, the values found varied within physiological ranges.

Voluntary intake and dry matter digestibility of a mixture of 80 per cent spear grass hay and 20 per cent Townsville lucerne hay by sheep, in 'metabolism cages, supplemented with sodium sulphate, gluten, or sodium sulphate and gluten, were measured during a six-week experimental period. Apparent retentions of nitrogen, sodium, potassium, calcium, and phosphorus were also determined. All sheep receiving supplements gained weight whereas sheep on the basal ration lost weight. After four weeks supplementation, sheep receiving gluten alone or gluten + sodium sulphate consumed 64 per cent more feed than the control sheep, whose intake gradually declined over the experimental period. Sheep fed sodium sulphate alone consumed 48 per cent more feed than the control group. Dry matter digestibility of feed was increased by 5 to 7 percentage units when sheep were supplemented with either sodium sulphate or gluten.

ABSTRACT: In the present study it was evaluated the effect of the addition of linseed oil, inulin and horse chestnut added to a high fat (HF) diet on the content of fatty acids (FAs) in musculuss longissimus dorsi (MLD) of pigs. A 5d with adaptation period was followed by a 70 d experimental period, during which the pigs were fed with a HF basal diet. The HF basal diet which served as a control (group CG) was supplemented either with linseed oil (group LG) or with inulin and horse chestnut (group IG). All of the pigs were slaughtered at the end of the experiment and samples of MLD were taken for FA analysis. The concentration of α-linolenic acid in MLD of the LG group was 58 % and 61 % higher (P˂0.05) compared to CG and IG groups, respectively. The content of eicosapentaenoic acid (EPA) was 0.03 and docosahexaenoic acid (DHA) 0.07 in LG treatment. These FAs were not detected in CG and IG. The ratio of MUFA and PUFA n-6/n-3 in the MLD was the lowest (P˂0.05) in the LG (8.84) compared to CG (14.07) or IG (14.74) groups, representing a difference of 31.2%. The n-3/saturated FA ratio was highest (P˂0.05) in LG group (0.04) when compared to CG and IG groups (0.02). The supplementation of linseed oil, into the HF diet resulted in a higher concentration of α-linolenic acid, EPA, DHA and lower ratio of n-6/n-3 FA in MLD. Inulin and horse chestnut had no effect on FA profile of MLD.

AbstractThe aim of this experiment was to quantify the relationship between autumn grass supply and concentrate supplementation level on grass intake and animal performance. One hundred and ten continental steers (567 kg) were assigned to 10 treatments. The experimental design was a three grass allowances (6, 12 and 18 kg dry matter (DM) per head daily) by three concentrate levels: (0, 2·5 and 5 kg per head daily) factorial with a positive control group offered concentrates ad libitum and no grass. Grass allowance was offered daily and concentrates were given individually. The experiment began on 22 August and all animals were slaughtered after a mean experimental period of 95 days. Grass intake was calculated using the n-alkane technique and diet digestibility using ytterbium acetate as an indigestible marker. There was an interaction (P < 0·05) between grass allowance and concentrate level for grass intake. At the low grass allowance there was no effect of offering animals supplementary concentrates on grass intake, at the medium and high grass allowances, supplementary concentrates reduced grass intake by 0·43 and 0·81 kg DM respectively per kg DM concentrate offered. Increasing grass allowance increased (P < 0·001) complete diet organic matter (OM) digestibility at all concentrate levels and supplementary concentrates increased (P < 0·001) complete diet OM digestibility only at the low grass allowance. Both offering animals supplementary concentrates (P < 0·001) and increasing daily grass allowance (P < 0·001) increased their carcass growth rate. Relative to the animals offered the low grass allowance and no concentrate, supplementing with concentrate increased carcass growth by 116 g/kg concentrate DM eaten whereas increasing the grass allowance, increased carcass growth by 38 g/kg DM grass eaten. As a strategy for increasing the performance of cattle grazing autumn grass, offering supplementary concentrates offers more scope than altering grass allowance.

Background Adverse health effects among agricultural workers due to chronic heat exposure have been characterized in the literature as not only due to high ambient temperatures but also due to intensive manual labor in hot and humid conditions. The aim of this study was to use biomonitoring equipment to examine the effectiveness of selected cooling devices at preventing agricultural workers from exceeding the core body temperature threshold of 38.0°C (Tc38) and attenuating heat-related illness symptoms. Methods A convenience sample of 84 agricultural workers in Florida was randomized to one of four groups: (a) no intervention, clothing as usual; (b) cooling bandana; (c) cooling vest; and (d) both the cooling bandana and cooling vest. Biomonitoring equipment worn by the participants included core body temperature monitor and an accelerometer to capture physical activity. Findings A total of 78 agricultural workers completed one intervention workday trial. Compared with the control group, the bandana group had lower odds of exceeding Tc38 (odds ratio [OR] = 0.7, 90% confidence interval [CI] = [0.2, 3.2]) and the vest group had higher odds of exceeding Tc38 (OR = 1.8, 90% CI = [0.4, 7.9]). The simultaneous use of cooling vest and bandana showed an effect little different from the control group (OR = 1.3, 90% CI = [0.3, 5.6]). Conclusion/Application to Practice This is the first field-based study to examine cooling intervention among agricultural workers in the United States using biomonitoring equipment. This study found that using a bandana while working in a hot agricultural environment has the potential to be protective against exceeding the recommended Tc38 threshold.

Industrialization of the Babassu coconut for edible oil production generates many co-products and has great potential to be used in ruminant nutrition. The aim of this study was to evaluate the effect of different inclusions of endocarp meal type II of Babassu coconut (BEMII) on intake and digestibility of sheep diets. Twenty non-castrated male sheep with an undefined breed were distributed in a completely randomized design with four treatments and five replications. The animals received an experimental diet with increasing inclusions of BEMII (0.0%, 7.5%, 15.0%, and 22.5%). Increasing additions of the coproduct negatively influenced dry matter intake (g/BW and g/kg0.75). Crude protein intake (in grams) per day decreased linearly with the inclusion of BEMII, presenting a reduction of 49.35 g/d. The inclusion of 7.5% showed the best response for energy balance among all inclusions and presented the closest nitrogen balance value when compared to the control group; therefore, this inclusion percentage is the best for endocarp meal utilization.

AbstractThe aim of this work was to study the effect of the contemporary administration, in separate troughs, of two diets differing in digestible energy (DE) and crude fibre (CF) content, on the performance of lactating rabbit does. At the first mating (130 days of age and 3·8 kg), 54 nulliparous rabbit does (New Zealand) were assigned to the control group offered a conventional lactation diet (diet C: 188 g crude protein (CP) per kg dry matter (DM) and 11·43 MJ DE per kg DM); whilst another 54 rabbit does were offered at the same time, in separate troughs, diet E (203 g CP per kg DM and 12·66 MJ DE per kg DM) and diet F (178 g CP per kg DM and 10·73 MJ DE per kg DM). The contemporary administration of two diets did not significantly affect food intake. However, the rabbit does in group F + E, due to the greater consumption of diet E (approx. 0·76 of the total), consumed more digestible protein (DP) both during the first (14·40 v. 13·44 g/day per kg M0·75 and 80·1 v. 73·5 g/day per kg M0·75, respectively for days 1 to 21 and days 22 to 35) and subsequent lactations (15·37 v. 14·35 g/day per kg M0·75 and 82·6 v. 76·5 g/day per kg M0·75 respectively for days 1 to 21 and days 22 to 35). Also the DE intake was higher for the rabbit does of group F + E during the 22- to 35-day period (6708 v. 6361 kJ/day and 6918 v. 6620 kJ/day respectively for primiparous and multiparous). The improved performance of rabbit does given two different diets does not justify the extra time involved in such a technique, as the intake of diet E was constant during the experimental period and was not influenced by the different energy requirements during lactation.
