Abstract The aim of this study was to investigate the effect of omega-3 supplementation on some haematological parameters like red blood cells (RBC), haemoglobin (Hgb), haematocrit (Hct), white blood cells (WBC), neutrophils (Neu) and lymphocytes (Lym) that may have a direct effect on the performance of athletic horses. Ten regularly trained Standardbred horses (6 geldings and 4 females, 4-5 years old, mean body weight 500 ± 25 kg) were used for the study. They were randomly divided into two groups. The experimental group received an omega-3 dietary supplementation every day for 30 days. The control group received no supplementation. Every 10 days, horses took part in a 1660 metre harness race and blood samples were collected from each horse: one in the morning before race (pre) and one after race (post). The application of one-way analysis of variance for repeated mesures (ANOVA) showed a statistically significant difference due to the race in the two different groups. The results obtained in the present study show a discordant effect of supplementation with omega-3 on RBC, HCT and Hgb, while omega-3 supplementation has been shown to have a better effect on WBC, Neu and Lym, but further studies should be performed to better evaluate the benefits of these on the performance of the athletic horse.

Benzo[a]pyrene (B[a]P) is a major polycyclic aromatic hydrocarbon (PAH), it can bind the DNA to produce DNA-adducts, which has major carcinogenic potential. Enzyme-linked immunosorbet assay (ELISA) is the method used to detect these DNA adducts of B[a] P diolepoxide (BPDE) within the living cells.The aim of this study is to evaluate exposure to bitumen fumes, and to B[a]P in asphalt plant workers by measuring the BPDE-DNA adducts in their peripheral white blood cells (WBC), which are considered biological markers for exposure risk assessment.In this study, Hemostatic blood (CBC, AST) were measured, and the levels of BPDE-DNA adducts were measured in DNA samples of WBC obtained from asphalt plant workers in Syria and compared to those measured from a control group. The measurement was performed using BPDE-DNA Adducts ELISA.kit. The sample size was determined to be 50 with 25 asphalt plant workers and 25 healthy volunteers with no occupational exposure to PAHs.The results showed some diseases associated with exposure to asphalt fumes among the workers in the study group and a statistically significant difference in the values of (CBC; WBC, leukocytes, HCT, MCHC and AST) between the study group and the control group. BPDE-DNA adducts were detected in WBC of 11 asphalt plant workers with concentrations ranging between 0 and 2.75 ng/ml and only one individual in the control group with concentration of 0.75 ng/ml. These results indicate significant positive relationship between exposure to the bitumen fumes and formation of BPDE-DNA adducts.BPDE-DNA adducts is potential biomarker for PAHs exposure and likely helpful indicator of PAH-induced DNA damage and possibly carcinogenesis.

The effect of scent leaf (Ocimum gratissimum) as feed additive in rabbit diet on growth and hematological parameters of rabbits was investigated using a number of 24 non-descript rabbits in an eight (8) weeks feeding trial. Four experimental diets were formulated to contain 0.0, 2.0, 4.0 and 6.0g scent leaf (SL) per kg of feed. The diets were randomly assigned to four treatment groups of 6 rabbits per treatment in a completely randomized design (CRD) experiment. Feed and water were offered ad libitum. Feed intake and weight gain was taken and used to calculate other growth performance parameters while blood sample (2 ml) was collected aseptically between 6:30 and 7:30 am from three rabbits per treatment and transferred into heparinised tubes which was used to determine the haematological parameters (PCV, Hb, red blood cells and white blood cells). The results showed that there was significant (P<0.05) differences in body weight gain and feed conversion ratio (FCR) across the four (4) treatment groups with treatment group fed scent leaf at 4.0g/kg diet having the highest weight gain and the best FCR. There was also significant difference (P<0.05) in albumin and cholesterol levels. Haematological parameters showed significant difference (P<0.05) across the treatment groups with treatment group fed 2.0g scent leaf having the lowest white blood cell (8.00 x 1012 /L). In conclusion treatment group fed 4.0g scent leaf gave the best result in terms of weight gain and FCR.

Background: During pregnancy the women undergo several structural and physiological changes almost in all body systems including the hematological profile. The main reported hematological change is the plasma expansion and haemodilution which affect the red blood cells(RBCs) and white blood cells (WBCs) count in addition to other hematological indices including packed cell volume (PCV). The objective of this study was to assess the effect of pregnancy on packed cell volume and white blood cells among Sudanese pregnant women and derive a formula for the normal values in pregnant women in relation to the packed cell volume.Methods: An analytical case control study was conducted in Khartoum state, Sudan 2017. The study included20 Sudanese pregnant women in the second and third trimesters attending the antenatal care at The National Ribat university hospital and 20 Sudanese non-pregnant women in the same age group as control. A questionnaire was used to collect socio-demographic and clinical data.5ml of venous blood was collected in a container containing EDTA. Complete blood count (CBC) was measured by using automated cell counter Sysmex.Results: There was statistically significant decrease in PCV value during 2nd and 3rd trimesters of pregnancy compared to the non-pregnant control group with p values (0.00 and 0.04 respectively), and statistically significant increase in WBCs count during 2nd and 3rd trimesters of pregnancy compared to the non-pregnant control group with p values (0.04 and 0.00 respectively). Also, a progressive increase in WBCs count with gestational age was demonstrated (second trimester 6.65×103/uL, third trimester 7.43×103/uL). The results showed mean WBCs of 7.79×103/uL, when compared to the estimated values the results showed no significant difference with the mean WBCs in 2ndand 3rdtrimesters (p value 0.06 and 0.39 respectively).Conclusions: During pregnancy there was statistically significant decrease in PCV values, statistically significant increase in WBCs count and by considering the haemodilution factor there was a higher increase in WBCs count.

Objective: To assess miR-155’s effect on aplastic anemia (AA) rats. Methods: In the present study, the healthy rats (control group) and AA rats including AA rats with miR-155 overexpression (experimental group I) and those with miR-155 deficiency (experimental group II), were selected. The levels of miR-155, STAT3 (a key gene in STAT3 signaling pathway) and phosphorylated STAT3 (p-STAT3) in control group and experimental group were detected via qPCR and Western blotting. Moreover, the number of white blood cells (WBCs), red blood cells (RBCs), platelets (PLTs) and hemoglobin (HGB) level were also measured. Results: The level of miR-155 in AA rats was significantly declined compared with that in healthy rats (p < 0.05). STAT3 mRNA level was significantly declined in AA rats with miR-155 overexpression compared with that in AA rats with miR-155 deficiency (p < 0.05). STAT3 and p-STAT3 protein expression in AA rats with miR-155 overexpression were significantly lower than those in AA rats with miR-155 deficiency (p < 0.05). Besides, it was found that the number of WBC, RBC, PLT and HGB level were significantly elevated in AA rats with miR-155 overexpression compared to those in AA rats with miR-155 deficiency (p < 0.05). Conclusion: miR-155 can improve the AA symptoms of rats through inhibiting the STAT3 signaling pathway.

Purpose:To investigate the acute changes in leukocyte number and cortisol after a single bout of soccer training.Methods:Ten elite female national-team soccer players and 8 nonathletes participated in the study. The duration of the exercise was 2 h, and it was performed at an intensity of 75% of maximal heart rate (HRmax). Blood samples were taken before, immediately after, and 4 h after a soccer training session to determine total white blood cells; the subsets of neutrophils, lymphocytes, monocytes, eosinophils, and basophils; and cortisol. At the same time, blood samples were obtained from nonathletes who refrained from exercise.Results:Data analysis indicated a significant increase in total white blood cells in the athletes postexercise (P < .001). The leukocytosis was still evident after 4 h of recovery (78% higher than the preexercise values), and there was a significant difference between athletes and nonathletes (P < .001). This leukocytosis was primarily caused by neutrophilia—there were no significant differences in lymphocytes after the end of exercise or between the 2 groups (P > 0.05). In addition, there was a statistically significant difference in cortisol concentration between athletes and nonathletes after the exercise (P < .001).Conclusion:These findings revealed that the single bout of soccer training at an intensity of 75% of HRmax induced leukocytosis without affecting the lymphocyte count in elite female athletes and probably the effectiveness of cellular components of adaptive immunity. Coaches should provide adequate time (>4 h) until the next exercise session.

Tartrazine is a synthetic organic azo dye widely used in food and pharmaceutical products. The current study aimed to evaluate the possible adverse effect of this coloring food additive on renal and hepatic structures and functions. Also, the genotoxic potential of tartrazine on white blood cells was investigated using comet assay. Twenty adult male Wistar rats were grouped into two groups of 10 each, control- and tartrazine-treated groups. The control group was administered orally with water alone. The experimental group was administered orally with tartrazine (7.5 mg/kg, b.wt.). Our results showed a marked increase in the levels of ALT, AST, ALP, urea, uric acid, creatinine, MDA and NO, and a decreased level of total antioxidants in the serum of rats dosed with tartrazine compared to controls. On the other hand, administration of tartrazine was associated with severe histopathological and cellular alterations of rat liver and kidney tissues and induced DNA damage in leucocytes as detected by comet assay. Taken together, the results showed that tartrazine intake may lead to adverse health effects.

Abstract Zearalenone (ZEN) widely contaminates animal feed of plant origin. The recommended safe concentrations of ZEN in feeds for various animal species are set mainly based on the mycotoxin’s hormonal properties (NOEL). Our growing knowledge about biologically active concentrations of ZEN, molecular mechanisms and cells/tissues targeted by ZEN indicates that the harmful effects exerted by this mycotoxin on animals may be far greater than previously believed. This experiment was performed on pre-pubertal gilts divided into a control group (n=9) and an experimental group (ZEN, n=9). The control group received placebo, whereas the experimental group was administered ZEN at a dose of 0.1 mg/kg feed (equivalent to 5 μg/kg BW/day) for 42 days. On days 14, 28 and 42 blood samples were collected from the animals to determine the concentrations of selected zearalenols, serum biochemical and haematological parameters. Conjugated ZEN was found in the blood serum of the experimental gilts. Changes in the analysed biochemical parameters included a transient increase in albumin and cholesterol levels. A statistically significant increase in the concentrations of neutrophilic and acidophilic granulocytes was observed in the white blood cell system. The results indicate that long-term per os exposure of pre-pubertal gilts to low doses of ZEN (below NOEL) has a modulatory effect on liver function and white blood cells.

Abstract Adaptation mechanisms as response to water content, oxygen level and pollutants are very important and they can be interpreted by hematological analysis. The aim of this study was the analysis of hematological and immune adaptations of common carp (Cyprinus carpio Linnaeus, 1758) to thermal stress. All specimens were divided into a control and experimental group. The control group of fish was exposed to a constant water temperature of 10°C. We induced thermal stress in experimental fish by gradually heating water to 28°C, held for 30 minutes and then comparing the obtained results with the control fish. Short-term hyperthermia lead to an increase of the number of leukocytes, especially pseudoeosinophilic granulocytes and monocytes, while the number of neutrophils and lymphocytes was reduced. The analysis of the leukocyte number and differential blood count in the control group showed high individual variation of segmented granulocytes, monocytes and pseudoeosinophilic granulocytes. Statistically significant differences (p=0.00) were found for the white blood cells, nonsegmented neutrophils and pseudoeosinophils between the control and experimental group. The experimental group of males had an increased number of white blood cells, monocytes and pseudoeosinophils, where significant differences were found for nonsegmented and total neutrophils and also for pseudoeosinophils (p=0.00), lymphocytes (p=0.01) and monocytes (p=0.03). Females had an increased total number of white blood cells, lymphocytes, monocytes and pseudoeosinophils, while significant differences (p=0.00) were obtained in the number of white blood cells, nonsegmented and total neutrophils and pseudoeosinophils between the control and experimental group. Adaptation mechanisms in carp after water temperature heating are mostly reflected in the increase of pseudoeosinophils and the decrease of neutrophils.

The effect of exposure to inhaled lead acetate in guinea pigs was evaluated. The present study comprised of five groups of guinea pigs including control (C), sensitized to ovalbumin (OA; S) and three groups exposed to 0.1, 0.2, and 0.4 M inhaled lead (Pb; n = 6 for each group). Tracheal responsiveness to methacholine and OA, total and differential white blood cells (WBCs) count in lung lavage, serum levels of cytokines (interferon γ (IFN-γ) and interleukin 4 (IL-4)), histamines, and immunoglobulin E (IgE), and Pb concentration in lung were measured. Tracheal responsiveness to methacholine, OA, total and differential WBC types as well as IL-4, IFN-γ, histamine, and IgE were significantly increased but IFN-γ/IL-4 were significantly decreased in sensitized animals as well as those exposed to high Pb concentrations when compared with the control group (from p < 0.05 to p < 0.001). In addition, there was not a significant difference in most measured values between animals exposed to high Pb concentration and group S. The Pb concentration in lung tissues of animals exposed to all three Pb concentrations was significantly higher than that of group C ( p < 0.001 for all cases).These results showed that inhaled lead acetate exposure can induce lung inflammatory changes similar to sensitized animals. Therefore, exposure to environmental Pb pollution may cause asthma-like changes.

Hematological studies were conducted in calves in dynamics during the prophylaxis of hypo-elementosis in the Amur region. The object of the study was calves (breed-Holstein, age-up to two months). The selected animals were divided into two equivalent groups: control and experimental. In the control group, planned measures were taken to prevent hypo - elementosis; in the experimental group, complex injectable drugs were used: Se-containing and tissue. Laboratory tests of calves ‘ blood were performed at the beginning and end of the experiment with determining the number of red blood cells, white blood cells, hemoglobin level, calculating the color index, differential counting of white blood cells and calculating the color index. It was established that the quality of blood oxygenation in calves of the experimental group improved. This is confirmed by the regeration to the standard values of hemoglobin level and color index as a result of their growth by 28 and 17%, respectively. At the end of the experimental period, the percentage of conditionally healthy calves was 80 % in the experimental group and 40 % in the control group. Thus, it should be noted that the proposed scheme of complex application of Se-containing and tissue preparations contributed to the normalization of erythropoiesis processes and increased the resistance of calves to diseases by 40 %.
