Genetic diversity of nuts traits and fatty acids of pecan (Carya illinoinensis) germplasm resources

Abstract Pecan (Carya illinoinensis) is the most economically valuable nut tree growing in many countries of the world. 10 nut quantitative traits and 15 fatty acid components of 112 pecan accessions were determined to analyze the morphometric and fatty acids genetic diversity in this study. The measured nuts traits of single nut mass, nut transverse, longitudinal and lateral diameter, nut aspect ratio, single nuts kernel mass, kernel yield and shell thinness were found highly variable. 15 fatty acids were detected among 36 tested fat acids in the nut kernel of pecan, and 14 fatty acids were found high variation except for the C12:0. Plenty of these traits are significant economic importance and could be used as breeding targets to improve the pecan variety. The positive correlations were observed between each pair of single nut mass, nut transverse diameter, nut longitudinal diameter and nut lateral diameter. Single nuts kernel mass is significantly positively correlated with single nut mass, nut transverse diameter, nut longitudinal diameter and nut lateral diameter. The 2D PCA plot successfully grouped the samples according to their phenotypic resemblance and morphological characteristics. 112 accessions were grouped into 4 and 3 major clusters according to the nut quantitative traits and fatty acids components and contents, respectively. Based on these results, we suggest that multidisciplinary research team should be set for genetic breeding of pecan to promote the conservation of local genetic diversity and improve the nuts production and commercialization in China.

Macadamia is a recently domesticated Australian native nut crop, and a large proportion of its wild germplasm is unexploited. Aiming to explore the existing diversity, 247 wild accessions from four species and inter-specific hybrids were phenotyped. A wide range of variation was found in growth and nut traits. Broad-sense heritability of traits were moderate (0.43–0.64), which suggested that both genetic and environmental factors are equally important for the variability of the traits. Correlations among the growth traits were significantly positive (0.49–0.76). There were significant positive correlations among the nut traits except for kernel recovery. The association between kernel recovery and shell thickness was highly significant and negative. Principal component analysis of the traits separated representative species groups. Accessions from Macadamia integrifolia Maiden and Betche, M. tetraphylla L.A.S. Johnson, and admixtures were clustered into one group and those of M. ternifolia F. Muell were separated into another group. In both M. integrifolia and M. tetraphylla groups, variation within site was greater than across sites, which suggested that the conservation strategies should concentrate on increased sampling within sites to capture wide genetic diversity. This study provides a background on the utilisation of wild germplasm as a genetic resource to be used in breeding programs and the direction for gene pool conservation.

Abstract Background The Capparaceae family is commonly recognized as a caper, while Cleomaceae represents one of small flowering family within the order Brassicales. Earlier, Cleomaceae was included in the family Capparaceae; then, it was moved to a distinct family after DNA evidence. Variation in habits and a bewildering array of floral and fruit forms contributed to making Capparaceae a “trash-basket” family in which many unrelated plants were placed. Indeed, family Capparaceae and Cleomaceae are in clear need of more detailed systematic revision. Results Here, in the present study, the morphological characteristics and the ecological distribution as well as the genetic diversity analysis among the twelve species of both Capparaceae and Cleomaceae have been determined. The genetic analysis has been checked using 15 ISSR, 30 SRAP, and 18 ISTR to assess the systematic knots between the two families. In order to detect the molecular phylogeny, a comparative analysis of the three markers was performed based on the exposure of discriminating capacity, efficiency, and phylogenetic heatmap. Our results indicated that there is a morphological and ecological variation between the two families. Moreover, the molecular analysis confirmed that ISTR followed by SRAP markers has superior discriminating capacity for describing the genetic diversity and is able to simultaneously distinguish many polymorphic markers per reaction. Indeed, both the PCA and HCA data have drawn a successful annotation relationship in Capparaceae and Cleome species to evaluate whether the specific group sort individual or overlap groups. Conclusion The outcomes of the morphological and ecological characterization along with the genetic diversity indicated an insight solution thorny interspecies in Cleome and Gynandropsis genera as a distinct family (Cleomaceae) and the other genera (Capparis, Cadaba, Boscia, and Maerua) as Capparaceae. Finally, we recommended further studies to elucidate the systematic position of Dipterygium glaucum.

Efforts to improve the yield and quality of cultivated chickpea (Cicer arietinum L.) are constrained by a low level of intraspecific genetic diversity. Increased genetic diversity can be achieved via the hybridisation of the cultivated species with the unimproved 'wild' relatives from within the 43 species of the Cicer genus. To date, the 8 species sharing an annual growth habit and chromosome number with C. arietinum have been the primary focus of screening and introgression efforts. Screening of these species has uncovered morphological characteristics and resistance to a number of abiotic and biotic stresses that are of potential value to chickpea improvement programs. Detailed analysis of protein and DNA, karyotyping, and crossability studies have begun to elucidate the relationships between the annual Cicer species. In comparison, perennial species have received little attention due to difficulties in collection, propagation, and evaluation. This review discusses the progress towards an understanding of genetic relationships between the Cicer species, and the introgression of genes from the wild Cicer species into the cultivated species.

AbstractEvaluation of the genetic diversity present within species is essential for conservation, management and utilization of the genetic resources. The objective of this study was to evaluate genetic variability of 70 common bean genotypes for seed storage proteins, grain morphological characteristics and agronomic traits. Two methods of extracting soluble seed proteins in salt were used.Positive correlations were observed among both seed morphological characters and developmental characters while yield components showed negative correlations with each other. Factor analysis for agronomic and grain morphological traits resulted in three factors were named yield components, seed morphology and phenology, respectively. Most genotypes had lower or medium scores for yield components and phenology factors. Considerable diversity was observed for seed morphology factor among the common bean genotypes.Nei’s diversity coefficient (He= 0.4), effective number of alleles (Ae= 1.69) and number of polymorphic loci (N = 17) indicated larger variation in the extraction method of soluble proteins in low salt (0.2 M NaCl) than high salt (1 M NaCl) condition. Considering that the centers of diversity for common bean are different in seed size, the result of Gst statistics showed that bands with relative mobility of 30, 32, 38 and 40 differentiated two weight groups more than other bands. Furthermore, significant differences were observed between these bands for number of pods per plant and number of seeds per plant.

The carpel is the female reproductive structure composed of a set of stigma, style and ovary. Normally passionflower have three carpels, however, some cultivars have this structure in greater quantity. The objective of this work was to identify relationships between the number of carpels and the physical characteristics of BRS Gigante Amarelo fruits. The flowers with three and four carpels were marked, pollinated and the fruits harvested from these flowers analyzed for transverse diameter, longitudinal diameter, pericarp thickness, fruit mass, pulp mass, pulp ratio, number and weight of seeds. A completely randomized design with 10 replicates per treatment was used and each plot had the average of 3 fruits. The data were submitted to variance analysis and the means were compared by the Tukey test at 5% of probability. The fruits generated by flowers with 4 carpels were 18,30% heavier, their pulp were on average 20.75% heavier, presented transverse diameter 7,53% superior to that of fruits generated by flowers with 3 carpels. Similar increments were identified regarding the number of seeds and the weight of seeds per fruit that increased 18,85% and 52.42%, respectively. This result indicates that cv. BRS Yellow Giant with 4 carpels generates heavier fruits, pulp and seeds with a more spherical shape.

The influence of humans on biodiversity at the genetic, species, community, and ecosystem levels is enormous. This is even more impactful on plant populations, which, being immobile, are at the mercy of their immediate environment. Digitaria horizontalis is a very common grass species in Benin City. Its ruderal nature accounts mostly for its widespread. The current study investigated the existence of possible genetic diversity of the grass in built environment in Benin City. Plant samples were randomly collected from 21 built environment locations within 6 Local Government Council Areas that make up Benin City. Test plants were allowed to acclimatize in experimental bowls and then allowed to grow for 4 weeks after which morphological characteristics were determined. Genetic diversity was determined by Random Amplification of Polymorphic DNA. Significant differences in plant height (33.0 – 59.0 cm) occurred when plants were compared based on location of collection. Changes in patterns of the association with weeds within the vicinity of the test plant was observed. The most prevalent plant species associated with the test plant was Pennisetum purpureum with a 12.18% prevalence. RAPD analysis using primer OPA 04 revealed a locus (about 600bp) that was absent in one or more of the locations with minimum disturbance. This 600pb fragment was however observed in most samples from highly built up and trampled environments. Primer OPA 03 showed monomorphic bands in 2 test samples with a band size of 400bp. It is therefore reported that there is an existence of the genetic diversity in a landscape of grass species (Digitaria horizontalis) found in different built environment in Benin City, Edo State, Nigeria.

ABSTRACT Characterization of cocoa clones produced in the semiarid region is necessary to enlarge the database about these implanted clones and thus enhance the quality of their by-products. Therefore, this study aims to evaluate physical, chemical, and physicochemical characteristics of the CCN 51, CEPEC 2004, CEPEC 2005, and PS 1319 clones, produced in the region of Vale do Jaguaribe in the state of Ceará (Brazil), and to suggest food processes or products for them. The clones were evaluated according to their physical traits (total fruit mass, rind, pulp, seeds, and placenta and pulp with seeds), fruit transverse diameter (FTD), fruit longitudinal diameter (FLD), the ratio FTD/FLD; rind external thickness (ERT), rind internal thickness (IRT), the ratio ERT/IRT, number of seeds, seed thickness, seed transverse diameter (STD), seed longitudinal diameter (SLD), and the ratio STD/SLD, yield, pulp color, chemical traits (humidity, lipids, proteins, ashes, crude fiber, and carbohydrates), and physicochemical traits (titratable acidity, pH, soluble solids, and reducing sugars) were evaluated. The CCN 51 and CEPEC 2005 clones are the most suitable for the process of cocoa fermentation. For desserts, jams, pulp, and nibs for fat-restricted diets, the most suitable clones are CCN 51, CEPEC 2005, PS 1319 and CEPEC 2004, respectively.

Abstract Background: The agaves are plants of cultural importance that have been used by humans for about 10,000 years for about 40 specific uses, of which the most culturally and economically important are fermented (Pulque) and distilled beverages (Mescal). Pulque continues to be produced in nearly all of Mexico, and the agaves used for this propose have shown domestication syndrome and selected morphological characteristics, such as larger plant size (since size is positively related to sap production) and less dentition of the leaves.Methods: In this study, we carry out an ethnobotanical, morphological and genetic analysis of the traditional varieties of pulque agave used in the production of aguamiel (agave sap) and pulque in two localities of the state of Hidalgo. We did semi-structured interviews and tours with crop owners, tlachiqueros (sap harvesters), and people related to the production system. We analyzed the morphology of 111 plants, and we studied the diversity and genetic structure of 130 individuals using 16 nuclear microsatellites.Results: We found wild, tolerated, transplanted, and cultivated varieties of agave. Asexual reproduction was the main method of propagation. We recorded 19 traditional varieties of pulque agave, 12 of them in production during the study, which correspond to the species Agave americana, A. salmiana y A. mapisaga with five intraspecific entities. The varieties were grouped according to a management gradient; the wild varieties were the smallest, with more lateral teeth and a larger terminal spine. The cultivated varieties clearly exhibited domestication syndrome, with larger plants and smaller dentition. The expected heterozygosity (He) of the varieties ranged between 0.204 to 0.721. Bayesian clustering suggested the existence of three genetic groups (K = 3), both at the level of traditional varieties of pulque agaves and for management categories, a result that matches multivariate clustering.Conclusions: Pulque producers in the studied localities, maintain high agrobiodiversity. The cultivated varieties exhibit domestication syndrome, as has been reported for other species of the genus with the same selection purposes. Our results support the hypothesis of a decrease in genetic diversity in crops compared to wild agaves, which seems to be due to vegetative propagation, among other factors.

Body fatty acids are important in health and disease. Previously, we reported a positive association between % AA (20:4 n6) and relative amounts of EPA (20:5 n3) and of some other fatty acids. We now study positive and negative correlations in general between %AA and percentages of other fatty acids, as observed in chicken breast muscle. Two groups of fatty acids were identified: Group 1) with relative amounts correlating negatively with %AA, and Group 2) with relative amounts correlating positively with %AA. With the positive correlations, but not with the negative ones, we obtained similar scatterplots using true and random numbers. This apparent discrepancy is probably related to differences in skewness of the concentration distribution of some fatty acids. Most of Group 2 fatty acids are eicosanoid or docosanoid precursors. The overall correlation outcome may be largely explained by the particular concentration ranges of the fatty acids. We therefore suggest Distribution Dependent Correlations to be an evolutionary regulatory principle, possibly ensuring balance between various eicosanoids and docosanoids.
