Background: Yellow wood (Coscinium fenestratum) is one of the typical forest plants of East Kalimantan. The hallmark of this plant is the roots, stems and the fruits pulp have a yellow color. There have been many studies on its use and potential in treating liver disease. However, there is limited research about the use of their fruits peel. Objectives: The purpose of this study was as an initial screening of the antioxidant and antibacterial activity from yellow wood (Coscinium fenestratum) fruits peel. Material and Methods: The yellow wood fruits peel was extracted using methanol solvent to obtain the MeOH extract of their fruits peel. The DPPH test was carried out to determined its antioxidant activity (25,50 and 100 ppm). The agar well difusion method was carried out to determined its antibacterial activity. Phytochemical tests are also carried out to determined the secondary metabolites of that fruits peel. Results: The results of the phytochemical test showed that the fruits peel contained alkaloids, flavonoids, carbohydrates, and tannins, but there were no terpenoids or steroids. The fruits peel has an antioxidant activity (58%) at 100 ppm concentration. While the antibacterial test showed strong inhibition at 100 ppm concentration on S. mutans, S. aureus, P. acne and E. coli bacteria. Conclusions: The fruits peel of the yellow wood has compounds that have strong antibacterial activity and medium antioxidant activity.

Essential oils (EOs) from Citrus are the main by-product of Citrus-processing industries. In addition to food/beverage and cosmetic applications, citrus EOs could also potentially be used as an alternative to antibiotics in food-producing animals. A commercial citrus EO—Brazilian Orange Terpenes (BOT)—was fractionated by vacuum fractional distillation to separate BOT into various fractions: F1, F2, F3, and F4. Next, the chemical composition and biological activities of BOT and its fractions were characterized. Results showed the three first fractions had a high relative amount of limonene (≥10.86), even higher than the whole BOT. Conversely, F4 presented a larger relative amount of BOT’s minor compounds (carvone, cis-carveol, trans-carveol, cis-p-Mentha-2,8-dien-1-ol, and trans-p-Mentha-2,8-dien-1-ol) and a very low relative amount of limonene (0.08–0.13). Antibacterial activity results showed F4 was the only fraction exhibiting this activity, which was selective and higher activity on a pathogenic bacterium (E. coli) than on a beneficial bacterium (Lactobacillus sp.). However, F4 activity was lower than BOT. Similarly, F4 displayed the highest antioxidant activity among fractions (equivalent to BOT). These results indicated that probably those minor compounds that detected in F4 would be more involved in conferring the biological activities for this fraction and consequently for the whole BOT, instead of the major compound, limonene, playing this role exclusively.

The Indonesian forest is one of the most species-rich ecosystems in the world. Within such forests are plant species with secondary metabolites that have novel molecular structure and diverse biological activity with excellent potential to be used medicinally in prevention and cure of various diseases afflicting humans. Plant materials often contain various forms of antioxidants. Phenolic compounds found in plants have many biological effects. Flavonoids and other phenolics play a protective role against metabolic damage caused by disease and environmental stressors. The communities of Kutai Kartanegara in East Kalimantan Indonesia are representative of many traditional peoples who have evolved ways of treating human ailment and disease by use of specific plants sourced from their forests. The purpose of the research described in this paper was to identify significant medicinal plant species used by the Kutai ethnic community and to prepare extracts from these plants, mainly from the leaves, and to evaluate the extracts for bioactivity; namely by general identification of secondary metabolites, and by estimation of their antioxidant activity, toxicity, and antibacterial activity. Samples of ten plant species, used medicinally by the Kutai community, were extracted using ethanol solvent. Assay of antioxidant activity was carried out by the spectrophotometric method using DPPH (1,1-diphenyl-2-picrylhydrazyl radical) as the control. The degree of toxicity of the extracts was determined by the BSLT (Brine Shrimp Lethality Test) while anti-bacterial activity was evaluated using an in vitro assay of growth inhibition of cultures of the bacterium Escherichia coli. The result showed that nine of the plant species had strong antioxidant activity (IC50); extracts of two of the species were very toxic, while one other was toxic; and at least eight of the species had extracts that exhibited anti-bacterial activity. The phytochemical compounds identified in several of the ten species included flavonoids, tannins, saponins, steroids, triterpenoids, and alkaloids.

The free-radical-scavenging property, antibacterial activity and brine shrimp toxicity of petroleum ether (PE), dichloromethane (DCM) and methanol (MeOH) extracts of Centaurea persica, a Turkish medicinal plant, were assessed using the 2,2-diphenyl-1-picryl-hydrazyl (DPPH) assay, the resazurin microtiter plate based assay, and the brine shrimp lethality assay, respectively. Additionally, the disc diffusion assay was also used to assess antibacterial activity. Only the MeOH extract of C. persica exhibited a significant free-radical-scavenging property in the DPPH assay, with an RC50 value of 0.025 mg/mL. However, in the TLC-based qualitative assay, petroleum ether and DCM extracts showed an extremely low level of free-radical-scavenging property. Among the solid-phase extraction fractions of the MeOH extract, the fractions eluted with 60% and 80% MeOH in water exhibited the highest level of free-radicalscavenging activity (RC50 = 0.010 and 0.015 mg/mL, respectively). While DCM extract showed reasonable antibacterial activity against five out of the nine test strains both in the disc diffusion assay and in the resazurin assay, the MeOH extract was highly active against both Escherichia coli and ampicillin-resistant E. coli strains. Among the solid-phase extraction fractions of the MeOH extract, fractions eluted with 80% MeOH in water and 100% MeOH displayed significant antibacterial potencies against both E. coli species. None of the extracts showed any significant toxicity towards brine shrimps (LD50 = >1.00 mg/mL).

Peach is a medicinal plant which has many traditional applications uses against various diseases. In this study we have evaluated differences in tannins and flavonoids in the composition of flowers and peach leaves and their antioxidant properties. Antibacterial activity of the peach flower and leaf extract was investigated using Mycobacterium tuberculosis and E. coli by the disk diffusion method. Total fractions of flavonoids and tannins were obtained using ethanol and aqueous extraction, respectively. The antioxidant activity was evaluated using the adrenaline autooxidation test. The results have shown that the peach flower extract contains many flavonoids, tannins that probably account for better antimicrobial effects as compared with the peach leaf extract. This shows perspectives for the use of peach flowers for the treatment of many diseases, especially for tuberculosis, and other diseases associated with overproduction of free radicals.

Background. Artemisia negrei L. (A. negrei) is a medicinal and aromatic plant belonging to the family Asteraceae that is more widespread in the folded Middle Atlas Mountains, Morocco. Materials and Methods. This study was run to investigate the phytochemical composition and antioxidant, antibacterial, and antifungal activities of Artemisia negrei L. essential oil. This oil was extracted from the fresh plant material by using the Clevenger apparatus. The phytochemical composition was characterized by GC-MS. The antioxidant activity was evaluated using different methods including DPPH, β-carotene bleaching, and total antioxidant capacity. The antibacterial activity was tested vs. multidrug-resistant bacteria including both Gram-negative and Gram-positive using inhibition zones in agar media and minimum inhibitory concentration (MIC) bioassays. The antifungal activity was conducted on Candida albicans, Aspergillus niger, Aspergillus flavus, and Fusarium oxysporum using a solid medium assay. Results. The chromatographic characterization of essential oils of A. negrei revealed the presence of 34 compounds constituting 99.91% of the total essential oil. The latter was found to have promising antioxidant activity by all bioassays used such as DPPH, β-carotene bleaching, and total antioxidant capacity. The results obtained showed that our plant oils had potent antibacterial activity towards Gram-negative (E. coli 57, E. coli 97, K. pneumonia, and P. aeruginosa) and Gram-positive (S. aureus), so that the maximum inhibition zones and MIC values were around 18–37 mm and 3.25 to 12.5 mg/mL, respectively. The oil also showed antifungal activity towards Candida albicans, Fusarium oxysporum, and Aspergillus Niger except for flavus species. Conclusion. The findings obtained in the work showed that A. negrei can serve as a valuable source of natural compounds that can be used as a new weapon to fight radical damage and resistant microbes.

Kefir is one of milk products, produced by the addition of bacteria and yeast-containing kefir starter. This study aimed to evaluate the microbial characteristic, antibacterial activity, chemical and physical characteristics, antioxidant activity, and organoleptic characteristic of goat milk kefir which treated with angkak (red yeast rice) extract supplementation during storage. The study was initiated with milling to produce angkak flour, which then was diluted on sterilized distilled water on 1:2 ratio. Kefir were obtained by fermenting goat milk using 3% kefir grain for 18 hours at room temperature. Different treatments on the study were angkak extract supplementations on level of 0; 2; 4; and 6% for three different storage periods, i.e., 0, 7, and 14 days. The parameter of microbial characteristics observed on this study were total of lactic acid bacteria, TPC, total yeast, and antibacterial activity. Chemical tests performed on this study included the value of acidity, pH value, alcohol content, and water content. Meanwhile, the physical evaluations included viscosity, color test (brightness value, red value, and yellow value), and followed with antioxidant activity test on DPPH method and organoleptic test. Data from chemical, microbial, and physical evaluation were analyzed on one way ANOVA for these following parameters: total count of lactic acid bacteria, TPC, antibacterial activity, value of acidity, pH value, alcohol content, water content, viscosity, brightness value, red value, yellow value, antioxidant activity, and sensory values (alcoholic taste and acceptability). The analysis were further continued on two way ANOVA for total count of lactic acid bacteria, TPC, antibacterial activity on E. coli and S. aureus, alcohol content, viscosity, brightness value, red value, yellow value, and antioxidant activity. Meanwhile, the organoleptic characteristics were analyzed on non-parametric Kruskal Wallis, followed by Duncan’s multiple range test (DMRT).  Data were analyzed on computer program of Software Statistical Product and Service Solution (SPSS) version 18 for windows. The study revealed that angkak extract supplementation on goat milk kefir during 0, 7, and 14 days of storage had significant effects (P<0.05) on total count of lactic acid bacteria, TPC count, antibacterial activity, value of acidity, pH value, alcohol content, and sensory qualities (alcoholic taste and acceptability). There was an interaction between angkak extract supplementation and storage period on total count of lactic acid bacteria, TPC, antibacterial activity on E. coli and S. aureus, alcohol content, viscosity, brightness value, red value, yellow value, and antioxidant activity. As summary, supplementation of angkak extract at 6% level and 14 days storage periods increase the microbiological quality, viscosity, alcohol content, antioxidant activity, as well as its alcoholic taste-associated acceptability, and inhibit the growth of pathogens (E. coli and S. aureus).

Biodiversitas 21: 1751-1758.  Plants produce a plethora of natural compounds of medicinal value. Although most plant-derived drugs have been identified in angiosperms, some species of mosses from the genera Leucobryum and Campylopus also exhibit antioxidant and antibacterial activity. The research aimed to evaluate the antioxidant and antibacterial activities of two species of these genera, Leucobryum aduncum and Campylopus schmidii, that are common in Taman Wisata Alam Gunung Tangkuban Perahu, West Java. Plants were collected, dried, and extracted in n-hexane or methanol. Antioxidant activity was evaluated by determining the concentration of extract that resulted in 50% reduction of 1,1-diphenyl-2-picrylhydrazine (DPPH) in the DPPH assay. These concentrations were >2000 mg/L for L. aduncum extracts and 1329.02 ± 7.8 mg/L for C. schmidii extracts; these large values are consistent with little antioxidant activity. Antibacterial activity was quantified as the ability of the extracts to inhibit the growth of the Gram-negative and Gram-positive bacteria Escherichia coli and Staphylococcus aureus, respectively. Nonpolar extracts of L. aduncum inhibited the growth of E. coli >50% at a concentration of 32 mg extract/L. Based on combined gas chromatography-mass spectrometry, the antibacterial activity in the non-polar extracts of L. aduncum was attributed to the lipophilic compounds 1-nonadecene, 5-eicosene, and cyclotetracosane. Thus, L. aduncum is a promising candidate as a source of new antibiotics.

Variation in cultivars can influence plant biological activities. This study aimed to identify superior cultivars while determining the variability in the phytochemical content, antioxidant, alpha-glucosidase inhibitory and antibacterial activities of cladode extracts from selected spineless Burbank cactus pear (Opuntia ficus-indica and Opuntia robusta) cultivars. Total phenolic and flavonoid contents were determined using the Folin-Ciocalteu and aluminum chloride spectrophotometric methods, respectively. Antioxidant activity was investigated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging and β-carotene linoleic acid assays. Alpha-glucosidase inhibition was determined using a spectrophotometric method and antibacterial activity using a non-polar (petroleum ether) and polar (50% methanol) extracts against two Gram-positive and two Gram-negative bacteria. Significant variation in phytochemical content, antioxidant, antidiabetic and antibacterial activities was observed amongst the cultivars. Alpha-glucosidase inhibitory activity varied widely with IC50 values ranging from 0.06 to 1.85 mg/mL. Radical scavenging activity of Polypoly cultivar was about seven fold higher than that recorded in other cultivars with low activity. Turpin and Berg x Mexican cultivars had the highest total phenolic and flavonoid contents, whilst the non-polar extract of Turpin also exhibited higher antibacterial activity against Bacillus subtilis and Escherichia coli. Sicilian Indian Fig was amongst the cultivars with a higher antioxidant activity, whilst also showing a strong inhibition against B. subtilis and E. coli. Polypoly cultivar demonstrated strong antioxidant and antidiabetic activities while its polar extract showed the highest total antibacterial activity against B. subtilis. The cultivar Malta was superior in terms of its antibacterial potency and efficacy against B. subtilis, Staphylococcus aureus and E. coli. The potential of using spineless cactus pear cladodes as a functional food with antioxidant, antidiabetic and antibacterial properties against pathogenic food spoilage bacteria in place of synthetic compounds was established. The significance of cultivar selection to increase this potential was highlighted.

Endophytic fungi are reported to be potential as antibacterial, antifungal, antioxidant, anticancer, and antivirus. One of the source of endophytic fungus is ulin (Eusideroxylon zwageri Teijsm & Binn.) which produced the same antimicrobial compound with ulin wood itself. The purpose of this research was to isolate endophytic caps from ulin wood branch, and tested the antimicrobial and antioxidant activity. The isolates were fermented on PDB media (Potato Dextrose Broth), and then tested the antimicrobial activity on Eschericiacoli, Pseudomonas aeruginosa, and Enterococcus aerogenes). In this research, there were two endofit isolates (white/PT and greenish black/HT). HT endophytic isolate exhibited strong antibacterial activity against the two pathogens (P. aeruginosa and E. coli) shown in 12 mm inhibition zone, while PT endophytic isolate exhibited the E. coli in 11.5 mm inhibition zone. Rosmarinus officinalis L. is known for its antioxidant and antibacterial properties against a variety of microorganisms. In this work, research has been carried out on pharmacy rosemary. Its antioxidant and antimicrobial properties have been determined. This study showed that the ethanol extract of R. officinalis L. has a pronounced antimicrobial activity against strains of both gram-positive and gram-negative bacteria. The extract also showed good antioxidant activity. Rosemary showed significantly more potent antibacterial activity against E. coli. The diameter of the zone of oppression was 17 mm. Thus, Rosemary Pharmaceutical has demonstrated potential as a dietary supplement due to its stronger antibacterial activity.
