AbstractIntroductionThe experiment evaluated the effects of intravenous administration of polymyxin B on experimental endotoxaemia in sheep.Material and MethodsTwenty clinically healthy fat-tailed sheep were randomly divided into: a group treated with 6,000 U/kg of polymyxin B, a group at 12,000 U/kg, and positive and negative controls. Endotoxaemia was induced by intravenous administration of lipopolysaccharide (LPS) fromE.coliserotype O55:B5 at 0.5 μg/kg. polymyxin was infused intravenously along with 2.5 L of isotonic intravenous fluids at 20 mL/kg/h. The positive control group received LPS and 2.5 L of isotonic fluids, the negatives receiving just 2.5 L of isotonic fluids. Clinical signs were evaluated before and at 1.5, 3, 4.5, 6, 24, and 48 h after LPS administration. Blood was also sampled at the denoted hours and serum haptoglobin, tumour necrosis factor-α (TNF-α), and plasma lactate concentrations were assayed.ResultsThe serum concentration of TNF-α in the positive control group increased significantly up to 48 h after LPS administration. The concentration of TNF-α was significantly different from those of the polymyxin B and positive control groups from 3 to 48 h; also, the concentrations of haptoglobin at different times in the polymyxin groups were lower than those of the positive control group and were significant at hours 3 to 48 (P < 0.05). Following the LPS administration, haptoglobin and TNF-α concentrations changed without significant difference between the two polymyxin B groups.ConclusionPolymyxin B (6,000 U/kg) restrained blood lactate concentrations. Furthermore, it significantly improved the clinical signs in endotoxaemic animals, including rectal temperature and heart and respiratory rates. Polymyxin B may be an antiendotoxic in fat-tailed sheep.

Objective: To analyze the effect of curcumin and vitamin E on kidney function and inflammatory response of Wistar strain rats that received cisplatin. Material & Methods: An experimental laboratory study with a post-test only control design, using male Wistar strain rats (Rattus norwegicus). Rats were randomized using the simple randomized sampling method. Samples were treated with cisplatin 5 mg/kg (positive control group), vitamin E 100 mg/kg, curcumin 100 mg/kg body, and a combination of both (treatment group), to evaluate its effect on and kidney function and inflammatory response as measured by tumor necrosis factor-α (TNF-α), blood urea nitrogen (BUN) and serum creatinine. Results: There were differences in TNF-α levels in the positive control group (cisplatin 5 mg/kg) against each treatment group (p<0.05). Further analysis showed that there was a significant difference between the treatment group that received vitamin E and curcumin from the treatment group that received a combination of both (P<0.05). In addition, there were differences in BUN and serum creatinine levels in the positive control group (cisplatin 5 mg/kg) against each treatment group (p<0.05). However, there was no significant difference in BUN levels in the treatment group that received vitamin E with the treatment group that received curcumin or a combination of both (p>0.05). No differences were found in serum creatinine levels between treatment groups receiving vitamin E, curcumin, or a combination of both. Conclusion: Vitamin E 100 mg/kg, curcumin 100 mg/kg, and the combination of both have a nephroprotector feature in Wistar rats exposed to cisplatin.

Fly is one of the vectors of foodborne pathogenic vectors and causes myiasis in humans and animals. To prevent the contamination of food and myiasis, various chemical products are commonly used. However, eco-friendly is now a trend all over the world so natural products are the alternative ways to reduce the chemical residue in the environment. The present study was screened the 12 natural products in 4 groups; the essential oil plants, the alkaloid plants, the cyanogenic plant, and the inorganic compounds which use a simple extraction on the fly repelling and larvicide. The fly repelling efficacy with 2 criteria consist of the time of the first swarm and percent repelling (PR) in the first 15 min. The larvicidal efficacy was determined by the mortality rate within 24 h. The Kaffir lime peels aqueous crude extract exhibited the best result on fly repelling from both criteria with no significant difference. The bamboo shoots aqueous crude extract gave the best result on larvicide with significantly different from other groups (P < 0.001) which was faster than the positive control group (Coumaphos & Propoxur). Moreover, the high mortality rate reached 73.33 ± 15.28% within 24 h, higher than the positive control group. The present study indicates the power of common natural products on fly repellent and larvicide that may apply for fly repellant to reduce the food contamination especially fresh materials and prevention or treatment of animal and human myiasis in rural areas

Objectives: To evaluate the efficacy of double manual cleaning (DMC) with enzymatic followed by alkaline detergent for removing biofilm on hinged surgical instruments compared to automated cleaning by the washer-disinfector. Methods: Biofilm of Staphylococcus aureus (ATCC 25923) was formed in vitro on hemostatic forceps (Fig. 1). Biofilm-covered forceps were rinsed in distilled water and subjected to one of the following cleaning regimes (n = 5 forceps each): Group 1 forceps were soaked in sterile water for 5 minutes. Group 2-DMC forceps were soaked in enzymatic detergent, brushed 5 times on each face, rinsed with filtrated water (0.2 µm), soaked in alkaline detergent, brushed 5 times each face, rinsed with filtrated water (0.2 µm), and dried with sterile cloth. For group 3-DMC plus hinge inner brushing (n = 5), the forceps were soaked in detergents and brushed as in group 2, including hinge inner brushing (2-mm lumen brush) (Fig. 1). In group 4 (automated cleaning in a washer/disinfector), forceps were prewashed, washed once, washed again, rinsed, thermally rinsed, and dried. After the treatments, forceps were evaluated for microbial load (counting of colony-forming units), residual protein (BCA protein assay kit), and biofilm (scanning electron microscopy). Results: There was no statistically significant differences between the microbial load and protein level contaminating the forceps subjected to DMC (group 2) and the positive control group. The DMC with hinge inner brushing group (group 3) and the automated cleaning group (group 4) demonstrated a significantly reduced microbial load: reduction averages of 2.8 log 10 (P = .038) and 7.6 log10 (P ≤ .001), respectively. The protein level remaining on the forceps also significantly decreased: 2.563 μg (P = .016) and 1,453 μg (P = .001), respectively, compared to the positive control group. There was no statistically significant difference between DMC with hinge inner brushing and automated cleaning (groups 3 and 4) for all of the tests performed. None of the cleaning methods completely removed biofilm and/or soil from the forceps hinge internal region (Fig. 1). Conclusions: Automated cleaning had the best efficacy for removing biofilm. However, DMC with hinge inner brushing was an acceptable alternative cleaning method for sterilizing service units with only manual cleaning available, as is the case in most low- and middle-income countries. Neither automated nor any manual cleaning regimes were able to completely remove biofilm and soil from the forceps hinged area, and the amount of protein left after automated and DMC plus hinge brushing was higher than the recommended. Cleaning is the most important step for the reprocessing of reusable medical devices; thus, efforts must be undertaken to improve cleaning in different social and economic realities and scenarios.Funding: This study was supported by Coordenação de Aperfeiçoamento de Pessoal de Nível Superior – CAPES.Disclosures: None

Chitosan is one of the most abundant and dispersed polymers in nature. It has a rapid healing effect for tissues and has an effect on bone remodeling. The purpose of this study was to prove that giving chitosan influenced the decrease of osteoclast number and increasing number of osteoblasts in female mice of wistar strain of ovariectomy menopause model.This study used a posttest only control group design. The total sample of 30 rats divided into three groups was randomized, osteoporosis was made with the menopausal model which was performed ovariectomy in the positive control group and the treatment group then in the treatment group was given chitosan extract of single dose of 45 mg / day via Nasogastric Tube (NGT). K1 was a negative control group (without ovariectomy and without chitosan), K2 was a positive control group (with no chitosan ovariectomy), K3 was the treatment group (ovariectomy and chitosan 45 mg / day).The results showed significant differences in the number of osteoblasts between K1 and K3, K2 with K3, and no significant differences were found in the K1 group with K2. While the number of osteoclasts showed that there were significant differences in K1 group with K2 and no significant difference was found in K1 group with K3, K2 with K3.Conclusions in this study showed that 45 mg / day of chitosan doses could significantly increase osteoblast counts in menopause female white rats and decrease the number of osteoclasts performed by ovariectomy with chitosan administration.

Abstract Background and aims: Cardiovascular diseases are widespread and causes many deaths in the world. The concentration of acute phase protein: C-reactive protein (CRP) and fibrinogen will rise dramatically when inflammation happens, which that can be used as an early marker of cardiovascular disease risk. Tempeh gembus contains fiber, unsaturated fatty acids and isoflavones are believed to reduce the inflammatory reaction. The aim of the study was to determinate the effect of tempeh gembus variations to levels of hcCRP and levels of fibrinogen of Sprague Dawley rats with atherogenic diet. Material and methods: This study was quasi-experimental with posttest only randomized control group design using 35 Sprague Dawley mice. The rats were randomized into 5 groups: negative control group given the standard diet, the positive control group given standard diet and atherogenic diet, and three treatment groups were given the standard diet, atherogenic diet and variation of tempeh gembus (tempeh gembus, heated tempeh gembus and tempeh gembus with bromelain enzyme) for 28 days. Serum levels of hsCRP and fibrinogen examined using ELISA (Enzyme-linked Immunosorbent Assay). Results and conclusions: The administration of tempeh gembus with bromelain enzyme is the most effective treatment for hsCRP serum level indicated a significant difference (p=0.028) between the negative control group, positive control group and first group with the third group. Fibrinogen serum levels showed significant differences in all treatment groups (p =0.042), administration of tempeh gembus with bromelain enzyme is the most effective treatment is shown by a significant difference between the negative control group and the positive control group with third group. The administration of tempeh gembus with bromelain enzyme for 28 days can reduce the serum levels of hsCRP and fibrinogen on rats significantly.

The experiment was conducted to investigate the protective effect of Petroselinum crispum leave extracted against gentamicin-induced nephrotoxicity in male rabbits by studying the body weight, clinical signs, haematological and biochemical parameters, gross lesion and histopathological changes. Twenty four rabbits male were used and divided into 4 groups. Group 1: rabbits served as a negative control, received distilled water 1 ml(orally). Group 2: rabbits served as a positive control group, received gentamicin at a dose of 80 mg/kg/day intramuscular for 15 days. Group 3: rabbits received gentamicin at a dose of 80 mg/kg/day then after one hour treated with ethanolic extract of Petroselinum crispum at dose 125 mg/kg orally for 15 days. Group 4: rabbits received gentamicin at a dose of 80 mg/kg/day then after one hour treated with ethanolic extract of Petroselinum crispum at dose 250 mg/kg orally for 15 days.The results of the gentamicin treated group( positive control group) showed clinical signs such as loss of body weight, loss of appetite and rough hair with hematuria. The body weight a significantly declined (p≤ 0.05) compared other groups. There was a significant decrease (p≤ 0.05) in WBC count, lymphocyte, GSH, SOD, CAT, and GPX levels, while it recorded a significant increase (p≤0.05) in weights of the kidneys, neutrophils, creatinine, urea, and MDA. Histological studies showed several kidney pathological changes such as pale colour, enlargement in size and weight and easy from detaching as opposed to negative control group. On the other hand, the group treated with ethanolic extractof Petroselinum crispum at dose 125 mg/kg induced improved of parameters as recorded significant increased(P ≤ 0.05) in body weight, WBC count, lymphocyte, GSH, SOD, CAT, and GPX, while significant decreased (P ≤ 0.05) in weights of the kidneys, neutrophils, creatinine, urea, and MDA compared with the positive control group whereas rabbits treated with ethanolic extract of Petroselinum crispum at dose 250 mg/kg restored the parameters and histological changes of the kidney to near normal status compared with the negative control group. These results showed that the dose-detected Petroselinum crispum extract (250mg / kg) acts as potential curative effect against gentamicin-induced nephrotoxicity in male rabbits.

Background: Areca nut (Areca catechu Linn.) is the seed of the fruit of the oriental palm that is commonly used among Southeast Asian communities. Chrysanthemum (Dendrathema grandiflora) is a flowering plant originating from East Asia and dominantly grows in China. Both of these plants have strong antioxidant activities. To investigate the mechanism of their wound healing activities, we prepared areca nut and chrysanthemum polyethylene oral gel and performed several in vivo assays using Sprague–Dawley rats. Methods: Sprague–Dawley rats were divided into five groups: Negative control group (rats with base gel treatment), positive control group (rats treated with triamcinolone acetonide), F1 (treatment with 20% areca nut:80% chrysanthemum), F2 (treatment with 50% areca nut:50% chrysanthemum), and F3 (treatment with 80% areca nut:20% chrysanthemum). Traumatic ulcers were performed on the buccal mucosa of all experimental animals that received topical oral gel and triamcinolone acetonide twice a day for seven days. The clinical and histological characteristics were analyzed and scored. Results: During the six days, the ulcerated area receded linearly over time and was completely cicatrized in F2 and positive control group (Dependent t-test, p<0.05). There were significant increases in body weight in F2 and positive control groups. There were no significant differences between groups in histology examination (Kruskal Wallis test, p<0.05). The moderate score of TNF-α levels was seen in F2 and positive control groups (ANOVA/Tukey test). Similar results were seen in the collagenases assay. Conclusions: A balanced combination of areca nut and chrysanthemum extract in the oral gel can optimize the healing of traumatic oral ulcers in rats through the increase of TNF-α and collagen deposition.

Background: Ivory palm water (Cocos capitata) has a high potassium content. Potassium ion can depolarized excited dental nerve on dentin hypersensitivity. This study was to determine the effect of water gel ivory palm against dentine hypersensitivity by the anxiety level of mice using RGS (Rat Grimace Scale).Method: This research has been through ethical clearance which researchmethod was quasy experimental in vivo. The sample was 16 Wistar rats (Rattus novergicus) male 4-5 months with 4 mice in each group. Group I as a positive control. Group II as the ivory palm water gel 10% treatment group. Group III as the ivory palm water gel 70% treatment group. Group IV as a negative control. Research data analysis used Kruskal Wallis test with p <0.05 then continued by Mann Whitney test.Result: Kruskal Wallis test obtained significant difference between the four study groups (p <0.05). Mann Whitney test results in comparison either between positive control group and ivory palm water gel 10% and 70%, and comparison group of ivory palm water gel 10% and 70% weren’t significantly different.Conclusion: This study conclusion there was no significant difference between ivory palm water gel 10 % and 70 % in reducing anxiety level in male Wistar rats with dentinal hypersensitivity

Wound is considered as a serious health problem, because it can interfere with quality of life due to pain, weakness, and decreased mobility and quality of life. This study aims to determine the effect ofj jathropha multifida, linn to the number of PMN and selt mast in the process of wound healing in rats with acute injury models. This study is an experimental research laboratory using the design of post test control group design Wistar rats were used as research subjects. Based on Tukey HSD test results, the average number of PMN leukocytes third day the negative control group had no significant difference in the positive control group (0.037) and treatment group (0004). On the seventh day the number of PMN leukocytes negative control group had no significant difference in the positive control group (0.069) and had a significant difference to the treatment group (0.00). Meanwhile, on the fourteenth day the number of PMN leukocytes negative control group had no significant difference in the positive control group (0.000) and had a significant difference to the treatment group (0.00). Based on the results of data analysis Annova one direction on the third day obtained a significant level of 0:50 means there is no significant difference in the number of mast cells in each group on the seventh day. While on day three significant values ​​of 0:08 was also no significant difference, as well as on the fourteenth day found no significant difference between the group with a value of 0.68. Conclusions there was a decrease in the number of Leukocyte PMN in rat wounds given a 10% stem extract ointment, and there was a decrease in the number of mast cells in the wounds of rodents given a 10% stem extract ointment. Suggestions need to be investigated regarding the use of distance stem extract in various concentrations to obtain effective results for wound healing and the need to examine other factors related to wound healing.
