The aim of the article is to study the peculiarities of antimicrobial immunity of the oral cavity of the industrial region’s inhabitants in order to optimize special preventive programs of major dental diseases. The object and research methods. 178 patients was examined from 21 to 50 years old, who turned to the University Dental Center of Zaporizhzhia State Medical University. The study group consisted of 126 patients with generalized periodontitis of the initial (8), I (32), II (68) and III (18) degree of chronic course, complicated by harmful factors of manufacturing. The comparison group consisted of 32 patients with periodontitis (5), I (10), II (11) and III (6) degrees who did not work in adverse conditions. The control group consisted of 20 relatively healthy individuals aged from 19 to 25 years without signs of generalized periodontitis. To study the factors of local immunity of the oral cavity, oral fluid was collected from each examined person, which was obtained without stimulation, spitting into sterile tubes. Then the oral fluid was centrifuged for 15 minutes at 8,000 rpm. The supernatant part of the oral liquid was poured into plastic tubes and stored at 30°C. The quantitative determination of markers in the oral fluid was performed by standard enzyme immunoassay kits according to the instructions of the "Lactoferrin-strip" ("VectorBest") manufacturers. The result was expressed in mµg / ml. The content of cathelicidin LL-37 was determined by enzyme-linked immunosorbent assay using NycultBiotechhuman LL-37 ELISA (Netherlands) firm reagent kit-manufacturers. The result was expressed in µg / ml. Statistica 13.0 licensed number JPZ804I382130ARCN10-J was used to process the results. The results of the study and their discussion. The hygienic condition in the examined patients of the study group was assessed as unsatisfactory: from 2.24 ± 0.5 to 2.99 ± 0.47 points depending on the age and length of service at the enterprise, but the PI and SBI values were increased. It was found, that the lactoferrin level in the oral fluid was higher in the all patients with periodontitis, than the same index in comparison with healthy control group and consistently increased with increasing severity of the process. In the patients’ study group, working in hazardous conditions of production, the lactoferrin content has increased in the oral fluid relative to the level of the healthy patients was more pronounced in comparison with the patients, suffering of periodontal tissue disease, but do not work in harmful manufacturing conditions. Increased lactoferrin in the oral fluid in the steelmaking workers can be considered as a means of compensation, that provides protection of the oral mucosa from colonization of microorganisms. In the steel industry workers, a statistically significant cathelicidin content decrease in the oral fluid was observed compared to healthy control patients’ group. In the clinical group of the patients without adverse factors of production, the cathelicidin’s concentration in the oral fluid was also reduced. The increasing level of lactoferrin in the oral fluid in parallel with the cathelicidin’s decrease in saliva are markers of the inflammatory phase, as well as the destructive phase of connective tissue. Conclusions. Thus, our observations have established a direct relationship between the lactoferrin’s concentration in the oral fluid and the severity of periodontal tissue diseases and the feedback between the cathelicidin’s content in the oral liquid and the activity of the pathological process. The determination of lactoferrin and cathelicidin in the oral fluid of the patients with harmful manufacturing factors allow us to identify the signs of pathological process in the oral cavity.

Objective: This study aims to investigate the expression and clinical significance of Th17 cells and related factors in peripheral blood of patients with Autoimmune Hepatitis (AIH). Methods: A retrospective selection of 100 patients with AIH were included as a study group, and 100 healthy volunteers in the outpatient clinic were selected as the control group. The levels of IL- 17, IL-6, IL-21 and TNF-α in peripheral blood of all subjects were detected by enzyme-linked immunosorbent assay and the frequency of Th17 cells and Treg cells was detected by flow cytometry. Results: Results showed that the study group had higher levels of serum total bilirubin (TBil), alkaline phosphatase (ALP), γ -glutamyltranspeptidase (γ-GT), immunoglobulin G (IgG), immunoglobulin M (IgM), alanine aminotransferase (ALT) and aspartate aminotransferase (AST) than the control group, as well as higher levels of IL-17, IL-6, IL-21 and TNF-α in serum. The frequency of Th17 cells in peripheral blood was higher in the study group, while the frequency of Treg cells was lower. Also, serum IL-17, TNF-α levels and Th17 cells frequency were positively correlated with ALT and AST, whereas Treg cells frequency were negatively correlated with ALT and AST levels. Conclusion: Our finding demonstrates that Th17 cell frequency and their related factors IL-17 and TNF-α, are associated with liver damage, which might be used to monitor AIH disease severity.

IntroductionOxidative stress has been documented in chronic schizophrenia and in the first episode of psychosis, but there are very little data on oxidative stress prior to the disease onset.ObjectiveThis work aimed to compare serum levels of superoxide dismutase (SOD) and glutathione peroxidase (GPx) in young individuals at ultra-high risk (UHR) of developing psychosis with a comparison healthy control group (HC).MethodsThirteen UHR subjects and 29 age- and sex-matched healthy controls (HC) were enrolled in this study. Clinical assessment included the Comprehensive Assessment of At-Risk Mental States (CAARMS), the Semi-Structured Clinical Interview for DSM-IV Axis-I (SCID-I) or the Kiddie-SADS-Present and Lifetime Version (K-SADS-PL), and the Global Assessment of Functioning (GAF) scale. Activities of SOD and GPx were measured in serum by the spectrophotometric method using enzyme-linked immunosorbent assay kits.ResultsAfter adjusting for age and years of education, there was a significant lower activity of SOD and lower GPX activity in the UHR group compared to the healthy control group (rate ratio [RR]=0.330, 95% CI 0.187; 0.584, p<0.001 and RR=0.509, 95% CI 0.323; 0.803, p=0.004, respectively). There were also positive correlations between GAF functioning scores and GPx and SOD activities.ConclusionOur results suggest that oxidative imbalances could be present prior to the onset of full-blown psychosis, including in at-risk stages. Future studies should replicate and expand these results.

Introduction. The most popular type of dentures is permanent orthopedic structures. However, their use is a serious intervention that changes biological balance in the oral cavity through galvanic, reflex and toxic allergic effects. Because of this, galvanic currents appear in the oral cavity, which have an adverse effect on the oral mucosa and the physicochemical parameters of the oral fluid. As a result of increasing bioelectric potentials in the oral cavity, oral microbiocenosis disorders may occur, the periodontium pathology develops or worsens and an unpleasant odor arises, which in aggregate can cause a social maladjustment of the patient. It is known that the main intraoral factor in the development of halitosis is the unsatisfactory hygienic condition of the oral cavity. Non-removable dentures made from non-noble dental alloys can also significantly affect the development of intraoral halitosis. Objective. To study potentiometric parameters in patients with intraoral halitosis who have fixed orthopedic constructions made of various materials by various technologies. Methods and Materials. 158 people aged 35-55 years with fixed orthopedic constructions in the oral cavity in the amount of 5-8 units with a service life of 2-3 years were examined. Four groups were formed: with stamped-brazed dentures, with cast-denture prostheses from dental alloys, with cast-in-place titanium structures and a control group without disrupting the integrity of the dentition. All patients underwent a sanation of the oral cavity, professional hygiene and anti-inflammatory treatment. In addition, hygienic training of patients with quality control of personal hygiene was conducted. In the course of the study, 35 patients from the first and second groups after the complex treatment were replaced by replacing the existing dentures with orthopedic structures made of titanium. The effectiveness of treatment was assessed by the dynamics of indices of hygiene, PMA, organoleptic assessment of halitosis and potentiometric parameters. Results. As a result of the study, it was found that patients with dentures, manufactured with the help of the stamped-brazed technology, were characterized by high indices of hygiene (1.29 ± 0.18 points) and PMA (66.79 ± 2.48%). The I degree of halitosis was observed in 27 patients (41.5%) of this group, II degree was observed in 35 people (53.8%) and III degree in 3 people (4.6%). In the second group, the hygiene index was 0.97 ± 0.12, the PMA index was 65.57 ± 2.18%. In 33 patients (67.4%) I degree of halitosis was detected, in 16 patients (32.6%) – II degree. In the third group, the hygiene index was 0.84 ± 0.12, the PMA index was 56.23 ± 1.22. Unpleasant odor in the majority of patients in this group (40 people or 90.9%) was determined when approaching a distance of 10 cm, which corresponded to I degree of halitosis. Only 4 patients (9.1%) had halitosis of II degree. In the control group, the KPI index was 12.13 ± 0.24, the level of hygiene was on average 0.81 ± 0.12 points and 55.56 ± 1.13%, respectively. There were no organoleptic signs of halitosis in this group.

The aim of our study was to find out the effect of our proposed treatment and prevention complex, which included oral administration of «Kvertulin» complex drug, «Imupret» drops, «Pikovit» multivitamin drug and «Exodent» local irrigation of the oral cavity with a solution of tooth elixir that influence on the rate of salivation and viscosity of oral fluid in children with chronic catarrhal gingivitis and diabetes mellitus.The treatment and prophylactic measures had positive effect on the homeostasis of the oral cavity, which assisted to reduce the viscosity of the oral fluid, increase the rate of salivation, as evidenced by observation of patients for 6 months. The treatment and prevention complex are recommended to use 2 times a year, as the studied indicators deteriorate over time.

Abstract PURPOSECarbonic anhydrase-2 (CA-2) plays a role in mineralization and calcification in organism. Strong evidence suggests that CA-2 is associated with urolithiasis. However, the relationship between CA-2 and urinary stone remains unclear. The study aimed to assess the association of urine CA-2 (uCA-2) level and the potential risk of urinary stone.METHODSFrom March 2017 to November 2019, a prospective cohort study was conducted on patients with urinary stones and healthy subjects to determine the pretreatment uCA-2 level detection by Enzyme linked immunosorbent assay (ELISA). The difference of uCA-2 level between patients with urinary stones and healthy subjects was compared. Then comparison between stone patients with complications and without complications was carried out as well as correlation analysis to detect factors associated with biomarker expression.RESULTS118 patients with urinary stones were into urinary stones group and 42 healthy subjects were into healthy control group. The mean pretreatment uCA-2 level was significantly higher in patients with urinary stones group than healthy controls group (P=0.028). Furthermore, The uCA-2 level was positive correlation with urinary stones complications (R=0.379, P=0.000), especially pain complications (R=0.524, P=0.000) and hematuria complications (R=0.374, P=0.000). Receiver operating characteristic curve (ROC) analysis that a uCA-2 level threshold of 10.94 ng/mL had 83.67% sensitivity and 68.12% specificity for predicting urinary stones complications. CONCLUSIONSExcessive uCA-2 excretion is a major risk factor for urinary stone. Our findings suggested that uCA-2 may be used as an unappreciated biomarker for the diagnosis urinary stone in patients and to predict its complications.

Abstract Background: We investigated bilateral tear cytokine levels including interleukin (IL)-1β, IL-10, IL-17, tumor necrosis factor (TNF)-α and Matrix metalloproteinase-9 (MMP-9) in patients with fungal keratitis(FK). Meanwhile, we evaluated the relationship between the changes of tear cytokines with corneal perception and pain in infected eyes, and the relationship between tear cytokines and tear film function in contralateral uninfected eyes .Methods : A total of 60(20 FK, 20 contralateral, 20 healthy controls) tear samples were collected prospectively and analyzed by enzyme linked immunosorbent assay(ELISA). Approximately 50 to 60 ul of tear samples in each case were collected. Meanwhile ,we analyzed the changes of visual analogue scale(VAS), tear breakup time (TBUT), Schirmer I test (SIT) and corneal perception compared with healthy controls. Results :The concentrations of IL-1β, IL-10 and IL-17 increased in bilateral eyes compared with healthy controls(P<0.05). The tear concentrations of MMP-9 , TNF-α only significantly increased in affected eyes (P <0.05). Patients with FK showed significant reduction in corneal perception of infected eyes compared with controls(P<0.05). Corneal perception of the normal eyes in FK patients was slightly lower than that of control group, but there was not statistical difference (P>0.05).TBUT and SIT of contralateral uninfected eyes were significantly lower than that of control group(P<0.05), which were significantly correlated with levels of IL-1β, IL-17(P<0.05). SIT were also negatively correlated with MMP-9(P<0.05), while the levels of IL-1β, IL-10, IL-17, TNF-α and MMP-9 in the tears of the healthy control group had no significant correlation with TBUT and SIT indicators(P>0.05).The corneal perception and VAS score of the affected FK eyes showed correlation with IL-1β, IL-17 and TNF-α(P<0.05).In addition, concentration of IL-10 inversely was correlated with VAS (P<0.05). Conclusion: Proinflammatory tear cytokines are elevated in bilateral eyes with unilateral FK as associated with tear film function ,pain and corneal sensitivity.

Vibration disease is one of the leading in the structure of occupational diseases. A more detailed study of the pathogenesis, expanding the possibilities of diagnosis of vibration disease, is necessary. The aim of the study is to determine the diagnostic value of stabilometry, quantitative sensory testing and neuroenergocarting in vibration disease associated with the combined effects of local and general vibration. Materials and methods. Two groups of patients were examined: with the diagnosis of vibration disease associated with the combined effect of local and general vibration – 50 people (age 48.7 ± 3.1 years), not in contact with vibration – 30 people (age 49.1 ± 2.8 years). The study of the equilibrium function, the determination of the level of the constant potential of the brain by 12 standard leads and the determination of temperature and pain sensitivity. Statistical processing of the results was carried out using the software package “Statistica 6.0” (StatSoft Inc., USA). Differences were considered statistically significant at p < 0.05. Results. In vibration disease, the vertical stance of patients is more unstable than in patients with no contact with vibration. In patients with vibration disease, there is a decrease in the threshold of cold sensitivity and an increase in the threshold of thermal sensitivity and thermal pain compared to the control group. According to our data, the increased level of permanent potential in patients with vibration disease is observed in the central (22.7 (12.6; 30.7) mV) and right central (20.4 (11.5; 27.1) mV), also the average level of permanent capacity in this group is increased – 17.1 (8.4; 25.8) mV.Conclusion. The results obtained indicate the involvement of the neurosensory complex united by single thalamic and cortical centers in the pathological process and confirm the diagnostic value of the methods used.

Βackground: angiogenesis is seen during the multiple stages of carcinogenesis, aswell as during the process of surgical wound healing, a fact which has led tosubstantial debate over the last decades about the potential impact of surgery upon thefinal outcome of ceratin patients treated for breast cancer.Aim: the present research aims at investigating the potential effect of surgery on theprocess of angiogenesis, by studying a number of factors that are related to the latter,in patients suffering from breast cancer before and after the time of the procedure,whilst comparing these results with those of patients that were operated on their breastfor non-malignant disease.Material-Methods: blood from 10 female patients with breast adenocarcinoma(Study Group) was collected via venipuncture before surgery (labeled as PRO), aswell as on post-operative day 3 (labeled as D3) and day 7 (labeled as D7). Moreover,blood samples were also taken from 6 female patients with fibroadenoma (ControlGroup) before surgery (PRO) and on day 3 afetr surgery (D3). These samples weremeasured for detection of circulating levels of three established angiogenesisbiomarkers using ELISA (Enzyme-Linked ImmunoSorbent Assay): VascularEndothelial Growth Factor-A (VEFG-A), Interleukin-8 (IL-8) and basic FibroblastGrowth factor (bFGF or FGF-2). In addition, circulating transcripts of 84 agiogenesirelatedgenes were determined using RT-PCR (Real Time Polymerase ChainReaction). The two groups of patients were firstly compared to each other regardingtheir results. Also, patients belonging to the Study Group were analized at differenttime points regarding surgery. Finally, the results were investigated againstclinicopathological data and patient outcome.Results: using ELISA we were able to detect increased levels of circulating VEGF-Aand IL-8 in the Study Group patients compared to the Control Group patientspreoperatively (p=0,0381 and p=0,0218 respectively), while for bFGF there was nostatistically significant difference documented. Surgery resulted in a significantincrease in VEGF-A levels on D3 (p=0,0389) and D7 (p=0,0172) as compared toPRO levels. Perioperative kinetics of IL-8 showed a mild trend towards increase,which, however, was not statistically significant. Postoperative levels of bFGF wereslightly increased on D3, but on D7 they were even lower than preoperative values(p=0,0205). Using RT-PCR certain differences between the Study Group and theControl Group were recorded regarding the circulating transcripts of a great numberof angiogenesis-related genes preoperatively: upregulation of VEGF-C, EGF, IL-8,FGF-1, SPHK1, NRP1, LAMA5, COL4A3, TEK, EFNA3, EFNB2. AKT1, ITGB3,THBS1, CCL11, TIMP3 and downregulation of CXCL10. Moreover, mastectomyinduced an altered expression in several key-genes in breast cancer patients:upregulation of THBS1, COL4A3, BAI1, ITGB3 and downregulation of EREG,SERPIFN1, CXCL9, CXCL10, IL1B, CCL2, CXCL1, HIF1A, NOTCH4. Conclusions: patients suffering from breast cancer have a different angiogenic profilein comparison to patients with fibroadenoma, as documented through their differencesin circulating levels of angiogenic factors. These levels are greatly changed after thesurgical procedure. VEGF showed a transient increase, while bFGF initially increasedbut only to finally decrease to levels that were even lower than the preoperative ones.Moreover, mastectomy promoted a shift in the expression pattern of a broad panel ofangiogenesis-related gene transcripts.

Introduction. Improper eating habits among children and adolescents often lead to overweight and obesity. Research indicates that these are risk factors for many diseases, including diseases in the oral cavity. Aim. To analyse the correlations of selected parameters of dental and periodontal health as well as oral hygiene status in overweight and obese children and adolescents versus control group. Material and methods. The study included 120 patients aged 11-18 years, who were classified into a study group with 60 overweight/obese individuals and a control group of 60 individuals with normal body weight based on BMI (Body Mass Index). Each patient underwent anthropometric measurements, such as height (cm), body weight (kg), which were used to calculate BMI (Body Mass Index). Additional measurements were taken of waist circumference (WC) and hip circumference (HC), to obtain the waist-hip ratio (WHR). Clinical examination of the oral cavity included an assessment of dentition (DMF), oral hygiene API (Approximal Plaque Index) and periodontal tissue (PD – Pocket Depth, CAL – Clinical Attachment Level, %BOP – % Bleeding On Probing) and CPITN (The Community Periodontal Index for Treatment Needs). Results. Statistically significant differences were found between patient groups with BMI ≥ 25 and BMI < 25 in the values of the following parameters: DMF p = 0.005, API p < 0.001, %BOP p < 0.001, PD p < 0.001, CPI p < 0.001. Conclusions. Overweight and obese children were found to have have worse parameters of dental and periodontal health as well as oral hygiene status compared to the group with normal weight. Our observations indicate that overweight and obesity may be potential risk factors for periodontal diseases in the study group.

AIM: This work investigated associations between tissue inhibitor metalloproteinase-1 and diabetic cardiovascular diseases in type 2 diabetic patients; also it investigated the role of osteopontin in the diagnosis of type 2 cardiovascular diabetes complications.SUBJECTS AND METHODS: These were examined on eighty subjects, divided into three groups as follows: twenty volunteer healthy control subjects, thirty type 2 diabetes mellitus (DM) patients, and thirty cardiovascular, diabetic patients. Full clinical measurements were carried out, and the expression level of tissue inhibitor metalloproteinase-1 in blood samples was analysed by real-time PCR, using gene-specific primer pairs. Also osteopontin concentrations had been measured by the enzyme-linked immunosorbent assay. Data were tested statistically by parametric tests.RESULTS: The concentrations of osteopontin and the expression levels of tissue inhibitor metalloproteinase-1 were significantly increased in diabetic and cardiovascular diabetic groups compared to control group also they were significantly increased in the cardiovascular diabetic group compared to the diabetic group.CONCLUSION: Tissue inhibitor metalloproteinase-1 and osteopontin concentrations were significantly increased in diabetic patients with cardiovascular complications than other groups.
