The buffalo milk industry has economic and social relevance in Italy, as linked to the manufacture of traditional dairy products. To provide an overview of the current status of buffaloes’ performances on a large scale, almost 1 million milk test-day records from 72,294 buffaloes were available to investigate milk yield, energy corrected milk, fat, protein, and lactose content, and somatic cell score (SCS). Phenotypic correlations between milk traits were calculated and analysis of variance was carried out through a mixed model approach including fixed effect of parity, stage of lactation, sampling time, month of calving, and all their interactions and random effects of buffalo, herd-test-date, and residual. Third-parity buffaloes were the most productive in terms of milk yield, while the lowest solid content was detected in sixth parity buffaloes. A considerable gap between primiparous and multiparous buffaloes was observed for milk yield, especially in early- and mid-lactation. Overall, SCS progressively increased with parity and showed a negative correlation with milk yield in both primiparous (−0.12) and multiparous (−0.14) buffaloes. Results suggested that, at the industrial level, milk of primiparous buffaloes may be preferred for transformation purposes, since it was characterized by greater solid content and lower SCS. Results of this study provide a picture of the Italian population of buffaloes under systematic performance records and might be beneficial to both dairy industry and breeding organizations.

Objective of this study was to analyze the effect of the dairy farm on milk traits of cows in Vojvodina. The research was carried out on small farms with 10 to 20 cows, medium farms with 20 to 50 cows, and large farms with over 50 dairy cows. The study included registered animals of Simmental (SM) and Holstein-Friesian breed (HF; including Red Holstein) in the first lactation for traits of milk yield and yield and content of milk fat. Total of 1323 first lactations were analyzed. The average milk yield (both breeds) in the first lactation of 305 days was 6295 kg of milk with 234,3 kg of milk fat and average milk fat content of 3,74%. Milk performance of cows varied significantly (CV=22,9% and SD=1447,8), as well as milk fat yield (CV=21,6% and SD=50,8). Large farms produced in average 6534 kg of milk, medium farms 6347kg and small farms 4717kg. Size of the farm exhibited significantly high effect on all observed traits, and the tendency was that farms with higher number of animals realize also higher average of production. Farm management and various breeding-zootechnical conditions present on farms had significant effect on milk performance of cows.

Water buffaloes (Bubalus bubalis) are very sensitive to environmental stimulus before and during milking, and this explains why disrupted milk ejections due to blood oxytocin level instability are frequent in this species. According to the literature, the manual stimulation (MS) of teats before milking promotes oxytocin release and allows milk ejection to start within 2–3 min. However, the pre-milking stimulation of teats is not always part of the milking routine in Italian buffalo farms; moreover, buffaloes with unstable milk let-down are sometimes treated with exogenous oxytocin (OX). Different types of pre-milking stimulation can impact differently on the mammary gland epithelium and structures and, therefore, on milk yield. In this study, we observed the changes in teat morphological traits before and after the application of three types of stimuli, i.e., no manual stimulation (NS), MS, and OX, in buffaloes reared in an Italian dairy farm. In particular, measurements were available for 23 and 21 buffaloes for front and rear teats, respectively. Subsequently, the effect of the pre-milking stimulation type was estimated on teat morphological characteristics and on milk traits recorded after the application of stimuli. The results showed that the teat canal length was shorter (P < 0.05) after stimulation in the case of MS and OX compared to NS. Cistern diameter was overall greater for MS and lower for OX. On the contrary, teat wall thickness was greater and lower for OX and MS, respectively. Milk yield and quality (fat, protein, and somatic cell score) were similar across the three types of pre-milking stimulation. In perspective, the impact of these types of pre-milking stimulation should be evaluated on a large scale, and the focus might be put on mammary gland epithelium integrity, mastitis incidence, and other udder health indicators in milk, e.g., electrical conductivity, differential somatic cell count, lactose content, and sodium and chloride concentration.

Ukrainian population of red dairy cattle is genetically active, its gene pool is significantly improved in each of the next generations, mainly due to artificial selection. Currently breeding structure of Ukrainian Red Dairy breed has the prospect of future expansion due to creating Podolian zonal type. Features of its formation are to use as the original breed, which being improved, Red Polish cattle. This breed, as Red Steppe cattle, was listed as a breed to preserve the gene pool under pure breeding. However, gene pool preservation of these breeds is impossible through lack of sufficient number of purebred bulls or their semen at breeding enterprises in Ukraine. The aim of our research was to study genesis and development prospects of red dairy cattle in Ukraine. Materials and methods of research. The materials of the research were the data of identifying breed composition of cattle in Ukraine by January 1, 2015, State Breeding List for 2005, 2010 and 2015, the electronic information database of leading breeding farms for 2007 and 2011 and database of bulls admitted to using during 2005, 2010 and 2015 of Institute of Animal Breeding and Genetics named after M.V. Zubets of NAAS. Results. Currently the group of cows of red breeds is about 365 thousand head, including 97.9 thousand head concentrated in agricultural enterprises. The largest share of this group takes the Red Steppe cattle (84.5%), the smallest one –Ayrshire and Angler breeds (0.7-0.8%). In Ukraine, the total number of Ukrainian Red Dairy cattle is about 40 thousand head, including 21.4 thousand head of cattle concentrated in agricultural enterprises. The results of these annual reports have shown that the average milk yield of the group of cows of red breeds was 3328 kg of milk in 2001. Then milk yield increased with each subsequent studied period and reached the highest figure of 4681 kg in 2014. So, increasing milk yield of cows was by 1353 kg of milk or 29% during fourteen years (2001-2014). The number of breeding animals of Red Steppe, Red Polish, Ayrshire, and Ukrainian Red Dairy breeds significantly decreased in the controlled part of population during the past fourteen years. However, level of milk production and quality increased slightly, except for Red Polish breed. Thus, 4688 cows of Ukrainian Red Dairy breed had milk yield in 5981 kg and milk fat content in 3.88% according to the appraisal of 2014, whereas, in 2013, 4902 cows had milk yield in 5837 kg with milk fat content in 3.86%. Up to 90% of the realized genetic progress in large-scale breeding is provided by using proven bulls, resulting most current need is to restore the national system of selection and testing of sires. The results show that during the past ten years, the number of proven bulls, admitted to using within the red breeds decreased almost seven times with a simultaneous increase 2.2 times in breeding value. Among 410 proven bulls, admitted to using in 2005 (bulls which being tested by progeny), unreasonably disproportionate share was for Holstein sires – 245 (59.8%), whereas, the share of bulls of the red breeds – 13.7%. In 2010, 123 bulls (65.4%) were of Holstein breed and only 9 ones (4.8%) – of Ukrainian Red Dairy among 188 bulls, admitted to using. In 2014, a similar situation was observed; there were 93 bulls, admitted to using, including 48 ones (51.6%) of Holstein and only 3 ones (3.2%) of Ukrainian Red Dairy breed. Given the current state of livestock breeding base on all the red breeds (Red Steppe, Ukrainian Red Dairy, Angler, Red Polish) we developed possible options of parameters of large-scale breeding. During the planning period the number of the controlled cows of the red breeds (mainly Ukrainian Red Dairy cattle) should be significantly increased. Calculations show that in the first stage of the program available number of cows in breeding farms, even if 30% of cows allocated to mating with laid-off bulls, is less than a third of the minimum at pressure of selection among laid-off bulls 1: 4 by productivity of 50 daughters. At the end of the program it is planned to increase the pressure of selection among laid-off bulls to 1:5 under increase of the controlled livestock number to 43700 cows (nearly 4.4 times against currently available one) Conclusion. Our results showed that, the number of breeding farms, in which were bred red cattle, significantly declined and the livestock number also decreased whereas milk production increased during 2001-2015. The high level of genetic potential of Ukrainian Red Dairy cows indicates the presence of highest-yielding cows with yield more than 10 000 kg of milk. At the same time, such animals at breeding farms for breeding Red Polish, Steppe, Ayrshire and Angler cattle weren’t found. During the past ten years, the number of proven bulls of red breeds, admitted to using, decreased almost seven times with a simultaneous increase of breeding value in 2.2 times. Predominance of Holstein bulls remains indisputable on reliability of assessment of breeding value. It causes the biggest request and the widest offer on the market of semen from bulls of the breed. The actual destruction of domestic system of sires breeding caused significantly lower rate of repeatability of breeding values in bulls of the red breeds. This is a concern and threatens possible loss or significant restriction of the gene pool of red breeds.

Context Genetic selection based on genetic markers for economically important traits in Sfakia sheep. Aims The aim of the present study was to investigate variation in the ovine myostatin gene (MSTN) and calpain 3 gene (CAPN3), and their association with milk-production traits. Methods Records for milk yield, milk fat content, protein content, lactose content, and non-fat solid content, pH and somatic-cell score (log), were obtained from 376 Sfakia ewes. Polymerase chain reaction–single-strand conformational polymorphism (PCR–SSCP) analyses were used to detect variation in intron 1 of MSTN and exon 10 of CAPN3. General linear models were then used to test for associations between the variation in MSTN and CAPN3, and milk-production traits. Key results The SSCP banding patterns for MSTN showed four variants (A1, A2, A3 and A4), which contained nine nucleotide sequence differences. Four SSCP banding patterns (C1, C2, C3 and C4) were observed for CAPN3 and these contained eight nucleotide-sequence differences. The MSTN variation was associated (P < 0.05) with variation in milk yield and non-fat milk solid content. Variation in CAPN3 was associated with milk yield (P < 0.001), fat content (P < 0.05) and lactose content (P < 0.05). Association analyses between the presence/absence of MSTN and CAPN3 variants and milk-production traits showed that a variant of MSTN that had previously between associated with muscle hypertrophy was associated with decreased milk yield (P < 0.05) and a lower non-fat milk solid content (P < 0.01). A CAPN3 variant that had previously been associated with increased sheep-carcass loin lean-meat yield was associated with a decreased milk yield (P < 0.01) and a decreased milk fat content (P < 0.05). Conclusions Our results have provided an insight into the effects of variation in ovine MSTN and CAPN3 on milk-production traits in sheep. Implications To preserve the dual-purpose characteristics of Sfakia sheep, breeding goals should take into account the possible antagonism between meat and milk traits.

The aim of this study was to evaluate relationship between milk progesterone concentration (P4) and milk traits at the start of estrus time and 12h after start of the estrus in dairy cows. The 96 milk samples of 48 Lithuanian dairy cows without reproduction disorders and 90–100 days after calving were evaluated. Cows were classified into two groups based on milk yield per day: less than 30 kg (n=20) and e”30 kg (n=28). Data were categorized by milk fat and protein content at the start estrus and 12h after start of estrus to evaluate relationship between P4 and milk traits examined. P4 at estrus time in dairy cows was significantly positively correlated with milk yield (P less than 0.001), whereas it was negatively correlated with milk protein (P less than 0.05-P less than 0.01) and fat at 12h after start of estrus. Dairy cows with F/P from 1.0 to 1.5 had the lowest P4 in milk. Results of the pregnancy in dairy cows were related with lower P4 and milk yield level (P less than 0.001), higher milk fat (P less than 0.05) and milk protein content (P less than 0.001). These cows had 1.90 times lower prevalence of the signs of subclinical ketosis (P less than 0.05) at estrus time when compared with non-pregnant cows. As a result, it was clearly demonstrated that P4 in dairy cows can help to evaluate and improve the reproductive properties of cows.

Анализ использования генотипирования по полиморфным системам групп крови и белкам молока в племенном и промышленном скотоводстве позволил выявить уменьшение общего количества аллелей полиморфных локусов групп крови крупного рогатого скота. Наибольшей частотой встречаемости, как и в более ранних исследованиях, отличался аллель G2Y2E'1Q', уровень которого у коров-первотелок исследуемой популяции составил 22,3%. Полученные результаты в свою очередь свидетельствуют о снижении генетического разнообразия в результате крупномасштабной селекции в племенных хозяйствах Московской и Рязанской областей. Так, снижение генетической изменчивости повлекло за собой увеличение фенотипической однородности стада по основным хозяйственно-полезным признакам. Результаты исследований помогли выявить тенденцию к положительной корреляции между уровнем гомозиготности и величиной удоя за 305 дней первой лактации при начальном уровне гомозиготности в группе более 10%. Средняя гомозиготность по локусу каппа-казеина (CSN3) у исследуемых групп животных составила примерно 63%, по локусу бета-лактоглобулина (LGB) – 43%. Также была отмечена тенденция увеличения частоты встречаемости гетерозиготных генотипов на 6,8% в группе помесных животных. Было выявлено, что гетерозиготные животные характеризовались стабильностью показателей массовой доли жира в течение трех лактаций от 3,68 до 3,70%, а с увеличением удоя наблюдалось и незначительное повышение жирномолочности. По итогам третьей лактации у коров-гомозигот массовая доля жира в молоке понизилась на 0,01-0,32% соответственно. Динамика выхода молочного жира у подопытных животных была аналогична изменению удоев. Analysis of the use of genotyping for polymorphic systems of blood groups and milk proteins in breeding and industrial cattle breeding revealed a decrease in the total number of alleles of polymorphic loci of bovine blood groups. The highest frequency of occurrence, as in earlier studies, differed allele G2Y2 '1Q', the level of which in the first-calf cows of the study population was 22,3 %. The results obtained, in turn, indicate a decrease in genetic diversity as a result of large-scale selection in breeding farms in the Moscow and Ryazan regions. Thus, the decrease in genetic variability resulted in an increase in the phenotypic homogeneity of the herd on the main economic and useful features. The results of the research helped to reveal a positive correlation between the level of homozygosity and the value of milk yield for 305 days of the first lactation with an initial level of homozygosity in the group of more than 10 %. The average homozygosity for the Kappa-casein locus (CSN3) in the studied groups of animals was approximately 63 %, for the beta-lactoglobulin locus (LGB) – 43 %. There was also a tendency to increase the frequency of heterozygous genotypes by 6,8 % in the group of crossbred animals. It was found that heterozygous animals were characterized by a stable mass fraction of fat during three lactation from 3,68 to 3,70 %, and with an increase in milk yield, there was also a slight increase in fat content. According to the results of the third lactation in homozygous cows, the mass fraction of fat in milk decreased by 0,01-0,32 %, respectively. The dynamics of milk fat yield in experimental animals was similar to the change in milk yields

Accurate inference of gene genealogies from genetic data has the potential to facilitate a wide range of analyses. We introduce a method for accurately inferring biobank-scale genome-wide genealogies from sequencing or genotyping array data, as well as strategies to utilize genealogies within linear mixed models to perform association and other complex trait analyses. We use these new methods to build genome-wide genealogies using genotyping data for 337,464 UK Biobank individuals and to detect associations in 7 complex traits. Genealogy-based association detects more rare and ultra-rare signals (N = 133, frequency range 0.0004% - 0.1%) than genotype imputation from ~65,000 sequenced haplotypes (N = 65). In a subset of 138,039 exome sequencing samples, these associations strongly tag (average r = 0.72) underlying sequencing variants, which are enriched for missense (2.3×) and loss-of-function (4.5×) variation. Inferred genealogies also capture additional association signals in higher frequency variants. These results demonstrate that large-scale inference of gene genealogies may be leveraged in the analysis of complex traits, complementing approaches that require the availability of large, population-specific sequencing panels.

The objective of this research was to examine heritabilities and genetic, phenotypic and permanent environmental relationships between milk dry matter (DM) and milk traits such as milk, fat, protein and lactose yields, milk urea nitrogen (MUN) and somatic cell score (SCS) in extended (to 395 days) lactations of Holstein cows from a big farm in Poland. The data set consisted of 78 059 test day records from the first, second and third lactations of 3 792 cows, daughters of 210 sires and 1 677 dams. Single- or two-trait random regression models were used with fixed effects of calving year, calving month, dry period and calving interval and random additive genetic and permanent environmental effects. The last two fixed effects were not included in the analysis of first lactation data. The highest values of heritabilities for all traits, except DM, were observed in the second lactation. First lactation heritabilities for all traits – except milk yield and SCS – were smaller than those in the third lactation. Lactose yield was highly heritable, with average h<SUP>2</SUP> equal to 0.25, 0.29 and 0.28 in lactations 1, 2 and 3, respectively. Heritability for DM was slightly lower than that for lactose (0.22, 0.26 and 0.28 for lactations 1, 2 and 3, respectively). In all lactations heritabilities for SCS were below 0.1. Genetic correlations between DM and milk yield (0.64–0.74) were lower than those between MUN and milk yield (0.67–0.79) as well as between lactose and milk yield (0.72–0.82). In general, DM was much more closely correlated with fat or protein yield (0.55–0.79) than with MUN or lactose (0.38–0.76). Only in the third lactation the correlation between DM and protein (0.72) was lower than between lactose and protein (0.76). For all lactations there were very high genetic correlations between DM and lactose (0.96–0.98) and high correlations between DM and MUN (0.63–0.83) and between lactose and MUN (0.70–0.85). The results suggest that further research is needed, focused on DM and its relationship with other traits in larger populations. &nbsp;

Abstract This study investigated the short-term metabolic and endocrine responses to different levels of concentrate formulated to particularly provide one major macronutrient source. Based on parity number, lactational stage, milk yield and BW, 9 mid lactating cows (211 ± 19 d in milk) were grouped into 3 blocks of 3 animals each. Concentrate supplements (aminogenic: rich in crude protein and nitrogen sources; glucogenic: high content of carbohydrates and glucogenic precursors; lipogenic: high lipid content) were fed in a factorial arrangement at 3 increasing levels for 3 d each. Milk yield, BW and feed intake were recorded daily. Blood and milk were sampled every 3 d at the end of each concentrate level. Statistical analysis was carried out using a mixed model with concentrate supplement and level as fixed, and block and animal as random factors. Milk fat, protein, lactose and urea contents were determined. In blood, concentrations of various metabolites, endocrine factors and enzyme activities (e.g. glucose, non-esterified fatty acids (NEFA), β-hydroxybutyrate, urea, cholesterol, triglycerides, insulin, glucagon, ASAT, GGT, GLDH) were measured. Milk yield, milk composition and BW were not affected by diets and concentrate levels. Feed intake increased in cows receiving the aminogenic and lipogenic diet. Milk und plasma urea concentrations were clearly elevated in the aminogenic and to a lesser extent in the lipogenic treatment compared to the glycogenic diet (P &lt; 0.05). Glucose concentrations in plasma were not affected by treatments, whereas insulin and glucagon increased only in the aminogenic treatment with concomitantly reduced NEFA concentrations (P &lt; 0.05). Activities of ASAT, GGT and GLDH as well as the total antioxidant capacity were not affected by diets (P &gt;0.05). To conclude, immediate metabolic and endocrine responses occurred due to the short-term dietary changes. Particularly a surplus of nitrogen supply affected metabolic responses and stimulated insulin and glucagon secretion.

Background: Several single nucleotide polymorphisms had been detected in goats and for the researchers it is important to reveal candidate genes with substantial effects on the traits of economic importance. The aim of this study was to investigate the association between LPL, STAT5 and AGPAT6 gene polymorphisms and milk traits of goats. Methods: We investigated about 204 goats, four different breeds (Czech White Shorthaired, Saanen, Anglo Nubian, Alpine) and two crossbreeds (Saanen and Alpine, Saanen and Anglo Nubian). The milk samples were analysed using spectrophotometers LactoScope 550 and LactoScope FTIR (Delta Instruments, the Netherlands). The somatic cell count (SCC) in milk was determined by flow cytometry method using the Somascope CA-3A4 (Delta Instruments, the Netherlands). Variations of the goat AGPAT6, STAT5, LPL genes were detected by PCR-RFLP method. Result: Analysis of AGPAT6 gene revealed that goats with GC genotype had higher milk yield, fat content and lactose content; goats with CG genotype had higher protein content. STAT5 gene analysis showed that goats with CT genotype had higher milk yield; goats with CC genotype had higher fat content and lactose content; goats with TT genotype had higher protein and urea content and also SCC. According to LPL gene analysis, goats with CC genotype had higher milk yield while goats with GG genotype had higher fat and protein content. The highest milk yield was estimated in Saanen and Alpine crossbreed goats while the highest milk fat and protein content was estimated in Anglo Nubian goats.
