The aim of the present study was to compare the effects of unsaturated and saturated oils on the performance, hormonal levels and hsp gene expression in broiler chickens exposed to heat stress. 300 one-day male broiler chicks were assigned to 4 treatments (Diets containing palm, corn, linseed or olive oils) with 5 replicates. At day 28 of age, 2 chickens were removed from each replicate, then blood samples and liver tissue samples were collected for analyses. Feeding linseed and olive oil reduced feed conversion ratio compared to corn and palm oils. The lowest level of insulin was for chickens fed linseed oil and corn oil. The highest level of corticosterone was found in chickens fed palm oil and the lowest level was for those received linseed oil. Chickens received linseed and corn oils had the highest levels of T3 and T4 and those fed palm and olive oils had the lowest levels. The highest HSP 70 gene expression was for chickens fed diet containing olive and linseed oils and the lowest one was for those fed corn and palm oils. It was concluded that olive oil and linseed oil could improve performance and heat tolerance of chickens under heat stress.

A total of 300 day old broiler chicks (Hubbard) were assigned to 30 floor pens (10 birds per pen) under cyclic heat stress. Three diets including a control, as well as two levels of grape seed powder (GSP) and zinc (OZ) at the rates of 2.5 g/kg GSP + 50 mg/kg OZ and 5 g/kg GSP + 50 mg/kg OZ, were supplied to the broilers for 35 days. According to the results, broiler feed intake improved (p < 0.05) in GSP + OZ groups from 3–5 weeks and on an overall basis compared to the control diet. Body weight increased (p < 0.05) in GSP-5 + OZ-50 during weeks 2–5 and on an overall basis. The findings indicated that feed conversion ratio (FCR) decreased (p < 0.05) during week 5 in broilers supplemented with GSP-5 + OZ-50. The antibody titer (HI) against Newcastle disease (ND) was higher (p < 0.05) in GSP + OZ groups compared to control treatment. The value of malondialdehyde (MDA) decreased (p < 0.05) under GSP + OZ diets compared to control. Moreover, paraoxonase (PON1) was higher (p < 0.05) in GSP + OZ groups compared to untreated broilers. In conclusion, GSP + OZ positively supported growth traits, reduced MDA, and augmented PON1 and HI titer against ND in broilers exposed to heat stress.

Abstract This study was conducted to explore the effect of the zinc oxide nanoparticles (ZnONPs) supplement on the regulatory appetite and heat stress (HS) genes in broiler chickens raised under high or normal ambient temperatures. In this study, 240 one-day-old male broiler chicks (Cobb 500) were randomly assigned to 48 battery cages. From day 1, these 48 cages were randomly subjected to four different treatment strategies: Control (wherein, their basal diet included 60 mg/kg of ZnO), ZNONPs 40 (wherein basal diet included 40 mg/kg of ZnONPs), ZnONPs 60 (basal diet included 60 mg/kg of ZnONPs), and ZnONPs 100 (basal diet included 100 mg/kg of ZnONPs). Thereafter, from day 22 to 42, the chickens from each dietary treatment group were subjected to different temperature stresses either normal (23 ± 1 °C constant) or HS (34 ± 1 °C for 6 h/d), which divided them into eight different treatment groups. Our findings revealed that dietary ZnONPs altered the gene expression of cholecystokinin (ileum), heat stress proteins (HSP) 70 (jejunum and ileum), and HSP 90 (duodenum, jejunum, and ileum). The gene expression of ghrelin was affected by the interaction between the ZnONPs concentration and temperature in the duodenum and stomach. More studies are required to elucidate its complex physiological and biochemical functions of the regulation of gene expression within the intestine in heat-stressed broiler chickens.

Objective: This study was conducted to investigate the effects of normal and heat stress environments on growth performance and, selected physiological and immunological parameters, caecal microflora and meat quality in Cobb 500 and Ross 308 broilers.Methods: One-hundred-and-twenty male broiler chicks from each strain (one-day-old) were randomly assigned in groups of 10 to 24 battery cages. Ambient temperature on day (d) 1 was set at 32°C and gradually reduced to 23°C on d 21. From d 22 to 35, equal numbers of birds from each strain were exposed to a temperature of either 23°C throughout (normal) or 34°C for 6 h (heat stress).Results: From d 1 to 21, strain had no effect (p>0.05) on feed intake (FI), body weight gain (BWG), or the feed conversion ratio (FCR). Except for creatine kinase, no strain×temperature interactions were observed for all the parameters measured. Regardless of strain, heat exposure significantly (p<0.05) reduced FI and BWG (d 22 to 35 and 1 to 35), immunoglobulin Y (IgY) and IgM, while increased FCR (d 22 to 35 and 1 to 35) and serum levels of glucose and acute phase proteins (APPs). Regardless of temperature, the Ross 308 birds had significantly (p<0.05) lower IgA and higher finisher and overall BWG compared to Cobb 500.Conclusion: The present study suggests that the detrimental effects of heat stress are consistent across commercial broiler strains because there were no significant strain×temperature interactions for growth performance, serum APPs and immunoglobulin responses, meat quality, and ceacal microflora population.

The effect of dietary 18:2n-6/18:3n-3 ratio (by the replacement of corn oil with linseed oil) on n-3 polyunsaturated fatty acids (PUFA) enrichment in breast muscle of broiler chickens and the expression of lipogenic genes were investigated. Broiler chickens were fed ad libitum with diets containing 5% corn oil (CO), 3.75% corn oil + 1.25% linseed oil (CL1), 2.5% corn oil + 2.5% linseed oil (CL2), and 5% linseed oil (LO) based on the basic diets, respectively. Dietary 18:2n-6/18:3n-3 ratio did not affect 42-day body weight and 0&ndash;42-day feed conversion efficiency (feed/gain, P &gt; 0.05) of broiler chickens, however, 5% linseed oil significantly increased 0&ndash;21-day feed conversion efficiency (feed/gain, P &lt; 0.05) and decreased breast muscle weight (by 16%, P &lt; 0.05) of broiler chickens. With the decrease of dietary 18:2n-6/18:3n-3 ratio, the enrichment of total n-3 PUFA, 18:3n-3, 20:5n-3, and 22:5n-3 increased linearly (P &lt; 0.01), while the enrichment of total n-6 PUFA and 18:2n-6 decreased linearly (P &lt; 0.01) in breast muscle of broiler chickens. Dietary corn oil increased the enrichment proportion of 20:4n-6 in a dosage-independent manner. Replacing 1.5% corn oil with linseed oil increased the enrichment proportion of 22:6n-3 (P &lt; 0.05), but continuing to increase dietary linseed oil could not further elevate its deposition. Real-time quantitative RT-PCR was used to determine the expression of the mRNA levels of related genes. Dietary PUFA had insignificant effect on the expressions of LPIN2, WD and tetratricopeptide repeats 1 (WDTC1) and △-6 fatty acid desaturase (FADS2) in both breast muscle and abdominal fat. The effect of dietary PUFA on the expression of LPIN1 gene showed clear tissue dependence. Equivalent adding of corn oil and linseed oil could up-regulate the mRNA level of LPIN1 in abdominal fat (P &lt; 0.01). This study demonstrated that decreasing dietary 18:2n-6/18:3n-3 ratio promoted the deposition of desirable n-3 long chain PUFA in the edible tissue and influenced the expression of LPIN1 in a tissue-dependent manner. &nbsp;

Some vegetable oils show beneficial effects in modulating neurodegeneration; in this work, we evaluated the therapeutic potential of corn and olive oils against neurodegenerative processes using the acute parkinsonism murine model induced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) in C57BL6 mice. The effects of corn and olive oils were quantified by the performance of mice in the open field and rotarod, and grasp strength tests and neuronal survival in the substantia nigra and striatum were determined by immunohistochemistry. Extra-virgin olive oil decreased the toxicity induced by MPTP administration judged by the performance in the behavioral motor tests and the number of total neurons in the analyzed brain regions. In contrast, corn oil only produced discrete changes in the behavioral and histological evaluations. Despite the numerous benefits of olive oil, its active substances that confer desirable effects and their mechanism of action remain unclear. Our observations can help to understand the ameliorative effects of some natural oils on neurodegeneration induced by some toxins, particularly the attenuation of neural damage related to toxin-induced parkinsonism or other pathologies that comprise neuronal death and motor disruption.

Background: Liver plays an important role in energy homeostasis. Intense liver diseases are accompanied with lower concentrations of n-3 and n-6 poly unsaturated fatty acids (PUFA). It has been found that n-3 PUFA play importantly protective roles in the liver. There was limited information about the effects of lipid sources on serum lipid components and liver sterol regulatory element binding-1 (SREBP-1) gene expression. Therefore, the aim of this study was to evaluate and compare the effects of fish oil, corn oil, olive oil and tallow, respectively, as dietary sources of n-3, n-6, n-9 and saturated fatty acid on serum lipid compound and liver SREBP-1 gene expression in broiler chicks.Materials, Methods & Results: In a completely randomized design, 240 one-day-old broiler chicks were allocated to five dietary treatments, four replicates and 12 chicks per each. Dietary treatments included of: control (diet without lipid supplementation) and diet supplemented with fish oil as a n-3 fatty acid source, corn oil as a n-6 fatty acid source, olive oil as a n-9 fatty acid source and tallow as a saturated fatty acid which were added to diets at 1.5, 3 and 4% in the starter, grower and finisher, respectively. At days 28 and 42 of age, liver tissue was dissected out and samples were placed in liquid nitrogen, also blood samples were collected. The SREBP-1 mRNA expression in liver tissue was quantitated using RT-PCR. Broilers fed diets containing fish oil, corn oil and olive oil as unsaturated fatty acid sources had lower concentrations of triacylglycerol than those fed other diets. Also, diets containing fish oil and tallow showed the lowest low density lipoprotein (LDL) levels at day 28 of age. Diets containing corn oil and olive oil showed higher levels of cholesterol and high density lipoprotein (HDL) than other diets but, these differences were not significant at days 28 and 42 of age  (P ≥ 0.05). In both sampling periods, the chickens fed diets containing tallow expressed higher (P < 0.05) SREBP-1 gene as compared to those fed other dietary lipids. Broilers fed diet containing corn oil had higher (P < 0.05) SREBP-1 gene expression than those fed diet containing fish oil and olive oil.Discussion: Serum lipids were affected by dietary fat source. Lower concentration of serum triacylglycerols was found in broilers fed fish oil and corn oil diets as dietary n-3 and n-6 fatty acid sources. It was shown that dietary PUFA, especially n-6 and n-3 fatty acids could reduce hepatic fatty acid and triacylglycerol synthesis. Broilers fed corn oil diet showed elevated LDL levels respect with those fed fish oil or tallow at day 28 of age. The results showed that dietary polyunsaturated fatty acids and saturated fatty acids substantially affected on SREBP-1 gene expression at days 28 and 42 of age. The birds fed fish oil as n-3 PUFA source expressed lower SREBP-1 gene compared with other groups at day 28 of age. Longer chain length, high number of double bonds, and the presence of the first double bond gives these fatty acids distinct and unique properties that separate them and their metabolic products from the more common n-6 and n-9 fatty acids. The finding of this study indicated lipogenic effects of tallow and then corn oil. The results confirmed that each of dietary lipid sources had different effect on serum lipid components. It was also found that SREBP-1 gene expression is age-dependent and it increased as age of broilers increased.

<p class="awabstrak2"><span lang="EN-US">Heat stress is an important issue in broiler chicken farms in tropical countries, such as Indonesia. Heat stress is very detrimental to broiler chickens because reducing production performance, health, and causing mortality. In the condition of heat stress, broilers synthesize Heat Shock Protein (HSP) quickly as the body's response to heat stress. HSP 70 is the most studied HSP group related to heat stress. The objective of this study was to review the nutritional approach that has been done to mitigate heat stress in broiler chickens with the HSP70 gene expression as its indicator. Based on some studies, nutritional approaches that can be taken are through the management of feed availability, supplementation of vitamin C, vitamin E, plant bioactives, amino acids (taurine and glutamine), probiotics, prebiotics, synbiotics, manan oligo saccharide (MOS) and minerals (selenium, zinc, manganese, chromium). By these approaches, HSP70 gene expression decreased indicating that the heat stress level of broiler chicken also reduced. It can be concluded that the nutritional approach can be used as a method for heat stress mitigation in broilers with the HSP70 gene expression indicator. </span></p>

Consumption of ochratoxin A (OTA) contaminated diets by broilers results in economic losses to the poultry industry. This experiment evaluated the effects of quantity and time of exposure to dietary OTA on performance, organ weights, serum biochemistry, and renal gene expression of chicks. Determination of genes expressed in response to OTA will allow for the identification of pathways that are influenced by OTA. 180-day old male broiler chicks were randomly assigned to a 3×3 factorial arrangement of treatments (3 levels of OTA; 0, 1 and 2 mg OTA/kg diet and 3 time periods; 7, 14 and 21 days) with 4 replicate pens of 5 birds each per treatment. For RNA-sequencing analysis (RNA-Seq), kidney samples were collected weekly from 3 controls and 3 chicks fed 1 mg OTA/kg. NextGENe software was used for read alignment and transcript quantification. Birds fed 2 mg OTA/kg diet had decreased feed intake and body weight gain, and increased serum uric acid on days 14 and 21. Compared to controls, birds fed 2 mg OTA/kg diet also had poorer feed conversion and increased kidney weights. On day 21, birds fed 1 mg OTA/kg diet had decreased albumin, and aspartate aminotransferase concentrations. Genes associated with carbohydrate and amino acid metabolism were downregulated, and genes associated with the immune system were upregulated at days 7 and 14. Genes associated with lipid metabolism and xenobiotic biodegradation were also downregulated on day 14. These changes disappeared on day 21 suggesting that the kidney and other related organs were repaired or the damage was contained. In conclusion, decreased performance and increased kidney weight and serum uric acid in birds fed 2 mg OTA/kg confirmed the effects of OTA. Supplementation of 1 mg OTA/kg diet caused time-dependent alterations in renal gene expression in chicks.

Abstract A total of 700 day old broiler chicks (Arbor Acres) were weighed and allocated to 28 floor pens (25 birds per pen). Five diets were prepared and allocated in a completely randomized design consisting of a control (basal diet) and four levels of dried Tamarind seeds pulp (DTSP) powder at the rate of 5 (DTSP-5), 7.5 (DTSP-7.5), 10 (DTSP-10) and 12.5 (DTSP-12.5) g/kg of feed for 35 days under heat stress condition. Body weight and feed conversion ratio (FCR) increased significantly (P<0.05) in the DTSP fed broilers compared to the control. Blood concentration of paraoxonase-1 (PON1) was significantly (P<0.05) higher in the DTSP-10 and DTSP-12.5 compared to the control. However, antibody titre against New Castle disease (ND) was significantly (P<0.05) higher in DTSP-7, DTSP-10 and DTSP-12.5 compared to the control. Blood cholesterol and triglyceride were significantly (P<0.05) lower in DTSP-7, DTSP-10 and DTSP4-12.5 compared to the control. However, blood HDL was significantly (P<005) higher in DTSP-10 and DTSP-12.5 compared to the control. Blood LDL was significantly (P<0.05) lower in DTSP-7, DTSP-10 and DTSP-12.5 compared to the control. Significantly (P<0.05) higher total leucocytes count (TLC) was found in DTSP-12.5, however, heterophils were significantly (P<0.05) lower in the same group. In contrast, lymphocytes increased significantly (P<0.05) in DTSP-7, DTSP-10 and DTSP-12.5 while monocytes was significantly (P<0.05) decreased in the same group. In conclusion, DTSP-5 supplementation improved growth, however, redox balance, cholesterol profile and cellular and humoral immunity increased in DTSP-5, DTSP-7.5 and DTSP-12.5 in broilers exposed to heat stress.

Sulfur amino acids are typically the first-limiting amino acids (AA) used in protein metabolism in poultry. Therefore, we hypothesized that their utilization in the pre-hatch period would affect embryonic development, IGF-I and TLR4 gene expression, antioxidant status, serum biochemical profile, and jejunum histomorphometry of newly hatched Ross broiler chicks incubated under heat stress conditions. A total of 150 fertile broiler eggs were subjected to heat stress (39.6 °C for 6 h/d) from d10 until d18 and injected at d 17.5 of incubation with methionine and cysteine (Met-Cys) at a dose of 5.90 mg l-methionine plus 3.40 mg l-cysteine. The effects of Met-Cys administration were examined and compared with the control (Non-injected group) and 0.75% NaCl injected group. The results showed that no significant differences among all groups in serum protein profiles (total protein, albumin, globulin, and albumin/globulin ratio) and creatine kinase were observed. The level of heat shock protein-90 was decreased with Met-Cys In ovo injection. The In ovo injection of Met-Cys also improved the values of total antioxidants capacity and glutathione in examined tissues. At the same time, an increase in fold change mRNA abundance of IGF-I and TLR4 was observed after Met-Cys injection in tested tissues. Finally, an increase of 29% in villus area was found after Met-Cys injection compared to the control group. In conclusion, the In ovo injection of Met-Cys resulted in improved embryonic development, IGF-I and TLR4 gene expression, antioxidant status and jejunum histomorphometry of newly hatched broiler chicks exposed to heat stress during incubation.
