Ethanolic Artichoke Extract improves outcome in a rodent model of 5- Fluorouracil Induced Cardio toxicity

Background: During treatment, many drugs may become cause of cardiac system toxicity, cytotoxic drugs therapy cause cardiac toxicity, including 5- Fluorouracil (5- FU). It is regarded as antimetabolite which cause its toxic effects during S phase of the cell cycle and got its activation by conversion of thymidine phosphorylase into fluorodeoxyuridylate (5 fluoro 2'deoxyuridine 5'monophosphate, 5-FdUMP) which cause inhibition of thymidylate synthase, which ultimately cause prevention of synthesis of DNA. Aim: To focus on evaluation of ethanolic artichoke extract (Cynara scolymus L) with respect to its cardio protective properties against 5-fluorouracil (5-FU) induced cardio-toxicity in rabbits by estimation of Alanine aminotransferase, creatine kinase and aspartate aminotransferase enzymes in serum. Methods: 4 groups consisting of 8 rabbits each were made for collected 32 rabbits who were albinos. Group I: (negative control) administered dimethyl sulfoxide (DMSO) (2 ml/kg /day) orally on daily basis for duration of 10 days. Group II: (positive control) administered DMSO (2 ml/kg /day) daily via oral route for duration of 10 days and subsequently received dose of 5-FU (150 mg/kg) (single) by intraperitoneal injection, on day 8th day in connection with DMSO. Groups III: administered ethanolic artichoke extract (200 mg/kg/day) orally on daily basis for duration of 10 days. Groups IV: administered ethanolic artichoke extract (200 mg/kg/day) daily on oral basis for 10 days with subsequently single intraperitoneal dose of 5-FU (150 mg/kg) on day 8th day. Results: Before intoxication via 5-FU, treatment of ethanolic artichoke extract note worthily reduces the increase serum levels of AST,CK & ALT enzymes due to cardio toxicity induced via 5-FU- in case of rabbits . Conclusions: With respect to present scenario, extracts of ethanolic artichoke serve as powerful modulator in reducing or masking cardiac toxicity cause by induction of 5-FU in case of rabbits.   The niacin in energy drinks has metabolic product that cause oxidative stress and liver damage, while the liver damage can be prevented by hepatoprotective agents. Scoparone in Artemisia vulgaris L. can act as a hepatoprotector by its antioxidant effect. This study aimed to investigate the effectivity of Artemisia vulgaris L. extract as a hepatoprotector in wistar hepatocytes induced by niacin. This study used 25 male rats which were divided into 5 groups: normal, the negative control, the positive control, the treatment group I, and II. Treatment was conducted for 28 days. The samples were terminated and the hepatocyte were prepared for histological examination. Histological appearance was catagorized as mild, moderate, and severe damage with or without inflamatory cells activity. The data analysis by Kruskal Wallis showed significant difference (p<0,001). Further analysis by Mann Whitney revealed significantly difference (p<0,05) between normal group and all groups, negative control group and positive control group, and positive control group and treatment group I, but not significantly difference between negative control group and treatment group I, negative control group and treatment group II, positive control group and treatment group II, and between treatment groups. The study concluded that the effectivity of Artemisia vulgaris L. extract has not been proven as a hepatoprotector but further study is needed to draw a definite conclusion.   Keywords: energy drink, niacin, Artemisia vulgaris L., hepatoprotector

BACKGROUND: Mastitis is an inflammation of the breast tissue, usually caused by bacteria. Mastitis stimulates pro-inflammatory cytokines. The cytokine interleukin-1β (IL-1β) is a crucial mediator of the inflammatory response. This cytokine has adverse effects of hosting immunity that mediates resistance to pathogens and also exacerbates damage during chronic disease and acute tissue injury. Scaevola taccada (Gaertn.) Roxb. has been used as an ethnomedicine for healing sores in several provinces in Indonesia. AIM: This study aimed to assess the profile of pro-inflammatory cytokine IL-1β through the treatment effect of leaf extracts of S. taccada (Gaertn.) Roxb. as adjuvant for healing mastitis. METHODS: This study was a true control group experiment using the pre-test-post-test control design aimed to measure the effect of hydroalcoholic compounds in leaf extracts of S. taccada on the systemic pro-inflammatory activity of interleukin-1β (IL-1β). The treated animals were 18 mice of Sprague Dawley strain induced by Staphylococcus aureus. These treated mice were divided into three groups in which each group consisted of six mice. The mice in the Group I (negative control) were given 1 ml aquabides/250 g body weight, those in the Group II (positive control) were delivered with 9.6 mg/ml amoxicillin/250 g body weight, and those in the Group III (experimental) were given 9.6 mg amoxicillin/250 g body weight + 400 mg/ml leaf extracts of S. taccada/g body weight for 5 days, respectively. Pathological examinations were carried out from the inflamed tissues to prove the healing process of the treated mice. IL-1Β levels were determined using the enzyme-linked immunosorbent assay method. Data were analyzed using ANOVA and post hoc tests. RESULTS: There were statistically significant differences of IL-1β levels after the administration of leaf extracts of S. taccada among all the treated mice groups at p < 0.05. The Group III had the lowest IL-1β level with the mean value ± 1.45 pg/ml compared to the IL-1β level in the Group II (positive control) with the mean value ± 3.82 pg/ml and the IL-1β level in the Group I (negative control) with the with mean value ± 5.22 pg/ml. The pathological analysis of breast tissues of the treated mice proved that leaf extracts of S. taccada (Gaertn Roxb.) could reduce damaged tissues, cellular infiltration, and subcutaneous edema induced by this pathogenic microorganism. CONCLUSION: Leaf extracts of S. taccada had a significant function as adjuvant for healing mastitis by reducing pro-inflammatory cytokine IL-1β.

The root of pasak bumi (Eurycoma longifolia, Jack) is one of plant from Indonesia known as aphrodisiac. The study about its standardized extract as aphrodisiac was show increasing libido, testosterone level, FSH and LH on male rats. This study was conducted to evaluate testosterone level and histopathological changes of the testes of male rats of standardized extract of pasak bumi root. There are 50 old male rats which ages 3-4 months old divided to five groups. Group I as negative control was administered aquadestilata. Group II as positive control was administered testosterone (Andriol®). Group III, IV and V ware given standardized extract of pasak bumi root at the dose 50, 100 and 200 mg/kg body weight Respectively. The extract was given orally twice a day for six days and forty nine days and then the testes was taken out on 7th and 50th day. The histopathological of the testes was evaluated using quantitative parameter by the presence of testes weight, number and diameter of Leydig cells. Testosterone level was taken on 7th and 50th day used ELISA method. The results were analyzed using parametric test, one way ANOVA (P<0,05), post hoc Dunnet (2-sided) (P < 0,05). The result of this study shows that the standardized extract of pasak bumi root was not effect on histopathological changes of the testes but it could increased the level of testosterone m at the dose 100 and 200 mg/kg body weight.  Key word : Eurycoma longifolia, Jack, aphrodisiac, histopathological testes testosteron

Hepatotoxic chemicals damage liver cells primarily by producing reactive oxygen species. The decoction of the leaves ofTamarindus indicaL. is used for liver disorders. In this work we evaluated the hepatoprotective activity of a tablet formulation of this plant. Thirty-five Sprague Dawley rats were randomly divided into five groups (n=7). First group (I) is control group, fed with standard diet. Groups II to V (hepatotoxic groups) were subjected to a subcutaneous injection of CCl4(0.5 mL/kg). Group II was negative control, fed with standard diet; group III was subjected to administration of Silymarin 150 mg/kg and groups IV and V were treated with tablets in dose of 100 mg/kg and 200 mg/kg, respectively. Lipid peroxidation and the activity of superoxide dismutase, catalase, and reduced glutathione were evaluated. Serum levels of alanine aminotransferase, aspartate aminotransferase, gamma-glutamine transferase, alkaline phosphatase, and a lipid profile were evaluated too. The tablets inhibit lipid peroxidation. The redox balance (SOD-CAT-GSH) remains normal in the experimental groups treated with tablets. The liver function using dose of 200 mg/kg of tablets was better than the other experimental groups. These results justify, scientifically, the ethnobotanical use of the leaves ofTamarindus indicaL.

Abstract Introduction The aim of obturation in the root canal is to completely seal the canal space to eliminate all the portals of entry and exit between root canal and periodontal space. Various techniques have been developed to achieve a hermetic seal. Materials and Methods As many as 150 extracted human maxillary central incisors were taken for the study. Biomechanical preparation was done up to F5 protaper file. According to different obturation techniques, samples were divided into six groups, keeping 30 samples in experimental and 15 samples in control groups. Group I–Lateral Condensation, Group II–Thermafil, Group III–Beefill, Group IV–GuttaFlow, Group V–Positive Control group, Group VI–Negative Control group. After obturation, the samples were immersed in 2% Rhodamine-B dye for 24 hours. Each sample was longitudinally sectioned to examine under confocal laser scanning microscope. Statistical Analysis The results were evaluated with ANOVA and posthoc Tukey honest significant difference (HSD) comparison test. Results The mean values of dye penetration of different groups were Group I (Lateral Condensation) 1.51 ± 0.451, Group II (Thermafil) 0.918 ± 0.399, Group III (Beefill) 1.30 ± 0.559. Group IV (GuttaFlow) 0.655 ± 0.396, Group V (Positive Control group) 1.96 ±0.046, Group VI (Negative Control group) 0 ± 0. The lowest mean value of apical microleakage was found in GuttaFlow amongst all experimental groups. Conclusion It can be concluded that the GuttaFlow obturating material exhibited better apical sealing ability with canal walls.

This article presents the results of a randomized clinical trial, designed to compare the efficacy and therapeutic profiles of YpozaneTM (osaterone acetate—OA) or SuprelorinTM (deslorelin acetate—DA) in male dogs with clinical signs of benign prostate hyperplasia (BPH). Forty-five intact male dogs were used in the study. The Group I (negative control) included 10 healthy dogs, the Group II (positive control) included 10 dogs with confirmed BPH and no treatment, whereas Group III and IV consisted of dogs with BPH and treated either with DA (15 dogs) or OA (10 dogs). The clinical response, testosterone and estradiol levels, hematology, biochemistry, and adverse effects incidence were evaluated. Both OA and DA proved to be effective for BPH treatment in dogs, as they allowed for the clinical remission in all treated dogs. The complete alleviation of BPH symptoms was noticed sooner with the use of OA (in 80% of dogs from day 7) compared to DA (in 40% of dogs within the first 21 days). The recurrence of clinical signs related to BPH was observed from week 24 in dogs treated with OA, whereas no relapse was noticed in dogs treated with DA at the end of the 36 weeks of the observation period. In 5 dogs (33%) treated with DA, a flare-up effect (increase in the clinical signs associated with BPH) was noticed on day 7. Despite individual differences in the clinical action, both medications were effective and safe options for the treatment of symptoms related to BPH in dogs.

Abstract Background and aims: Cardiovascular diseases are widespread and causes many deaths in the world. The concentration of acute phase protein: C-reactive protein (CRP) and fibrinogen will rise dramatically when inflammation happens, which that can be used as an early marker of cardiovascular disease risk. Tempeh gembus contains fiber, unsaturated fatty acids and isoflavones are believed to reduce the inflammatory reaction. The aim of the study was to determinate the effect of tempeh gembus variations to levels of hcCRP and levels of fibrinogen of Sprague Dawley rats with atherogenic diet. Material and methods: This study was quasi-experimental with posttest only randomized control group design using 35 Sprague Dawley mice. The rats were randomized into 5 groups: negative control group given the standard diet, the positive control group given standard diet and atherogenic diet, and three treatment groups were given the standard diet, atherogenic diet and variation of tempeh gembus (tempeh gembus, heated tempeh gembus and tempeh gembus with bromelain enzyme) for 28 days. Serum levels of hsCRP and fibrinogen examined using ELISA (Enzyme-linked Immunosorbent Assay). Results and conclusions: The administration of tempeh gembus with bromelain enzyme is the most effective treatment for hsCRP serum level indicated a significant difference (p=0.028) between the negative control group, positive control group and first group with the third group. Fibrinogen serum levels showed significant differences in all treatment groups (p =0.042), administration of tempeh gembus with bromelain enzyme is the most effective treatment is shown by a significant difference between the negative control group and the positive control group with third group. The administration of tempeh gembus with bromelain enzyme for 28 days can reduce the serum levels of hsCRP and fibrinogen on rats significantly.

Background: This study evaluated the effect of Beta vulgaris (beetroot) smoothie on some biochemical parameters on dimethyl 2,2-dichlorovinyl phosphate (DDVP, known as dichlorvos)-exposed albino Wistar rats. Methods: A total of 30 rats of both sexes were grouped into five groups of six animals each. Group I served as the negative control and were not exposed to dichlorvos. Group II served as the positive control and were exposed to dichlorvos but received no smoothie. Group III received 500 mg/kg body weight beetroot smoothie and was not exposed to dichlorvos. Groups IV and V were exposed to dichlorvos but received beetroot before and after exposure, respectively. At the end of the 6-week experiment, the animals were euthanized, the blood samples collected for some biochemical assays while the organs (kidney and liver) were harvested and subjected to histopathological examination. Results: From the biochemical assay, it was observed that the beetroot smoothies regulated and significantly reduced the elevated levels of AST, ALT, urea and creatinine observed in the animals that were exposed to dichlorvos. Additionally, the beetroot was able to regenerate the liver and kidney organs that were damaged on exposure to dichlorvos. Conclusion: This study concluded that beetroot smoothie possesses hepato-protective, hepato-curative as well as nephro-curative properties.

Acute hepatitis is an acute disease caused by viruses, alcohol and drugs, which is characterized by an increase in SGOT SGPT serum. The purpose of this study was to determine the effectiveness of boiled comfrey leaves (Symphytum Officinale L) levels of SGOT and SGPT serum in male wistar strain rats with acute hepatitis models. The objects in this study were 30 male Wistar strain rats aged 2-3 months with a weight of 180-200 grams. The objects were randomly divided into 3 groups: the treatment group, positive control and negative control. The treatment group and positive control group were induced paracetamol 120 mg / day orally for 7 days. Boiled water of 2.8 grams of comfrey (symphytum officinale L) leaves was given as much as 2.7 cc orally for 7 days to the treatment group. Data were analyzed with SPSS version 24, one way ANOVA test was performed to compare serum levels of SGOT & SGPT. The results showed there were significant differences in serum levels of SGOT between the treatment group, positive control and negative control (p <0.05) and there were significant differences in SGPT results between the treatment group and positive control group (p <0.05) but there were no significant differences between the SGPT levels of the treatment group and the negative control group (p = 0.173). As Conclusion, boiled Comfrey leaf has an effect in decreasing serum SGOT & SGPT in male wistar strain rats with acute hepatitis model.

Trigona honey (TH) is well known for its therapeutic characteristics. To date, the study of Trigona honey as a prophylactic or immune booster prior to the bacterial infection of the invivo model is not well covered. This study aims to investigate anti-inflammatory and immune activities in Wistar rats infected with respiratory infection following with Trigona honey. 25 Wistar rats were assigned to possitive groups, negative control group, positive control group was fed TH (5 g / kg body weight) orally, the untreated group was infected with Staphylococcus aureus to induce respiratory infection, the treated group has been infected with S. aureus followed by treatment with TH at a dose of 1.5 ×108 CFU / mL and the preventive group ingested TH one week before S. aureus infection. Blood was obtained for biochemical analysis. Lung tissues have been collected for molecular examination. The results showed a significant decrease in serum levels of ALT, AST, urea and creatinine in the preventive and treated groups, serum IgG increased significantly (P<0.05) in the preventive and treated groups, IFN-y increased in the preventive group while decreased in the treated group, and IL-8 increased in the treated group while decreased in the preventive group. The mRNA expression of AGP is up-regulated in the positive control, preventive and treated groups. The α2-MG, TNF-α , and mRNA expressions showed lower regulation after administration of TH in preventive and treated groups. The results show the ability of TH to counteract immune and inflammatory changes in serum levels and gene expressions.
