Abstract Isocitrate dehydrogenase 1/2 (IDH1/2) mutations are common in astrocytic glioma and are frequently coupled with TP53 and ATRX mutations. Collectively, these alterations cause genomic instability leading to high basal DNA double strand breaks (DSBs). Understanding how IDH/TP53/ATRX mutant cells process endogenous DSBs may help exploit inhibitors of DNA damage response (DDR) for the treatment of patients with IDH mutant gliomas. Through systematic effort to uncover the mechanisms involved in repair of endogenous DSBs in IDH1/2 mutant GBMs, we have discovered that high basal phosphorylated DNA-PK (p-DNA-PK) was characteristic of an IDH1/TP53/ATRX mutant GBM164 patient derived xenograft (PDX) but not in another IDH1 mutant GBM196 PDX. Immunofluorescence (IF) studies in patient specimen from which GBM164 was derived showed that p-DNA-PK co-localized with g-H2AX, 53BP1 or H4K20me2 (but not p-RPA) the known surrogates of DSBs. In contrast, p-DNA-PK was absent in the patient specimen from which GBM196 was derived, which otherwise had equally intense g-H2AX immunostaining colocalized with p-RPA. An independent IF study involving 11 IDH1 wild-type (WT) and 11 IDH1 mutant GBM patient samples, the p-DNA-PK was observed in 3 (27%) of 11 IDH1 mutant samples while IDH1 WT tumors were negative for p-DNA-PK. A telomere specific fluorescence in situ hybridization (Tel-FISH) confirmed elevated alternative lengthening of telomere (ALT) activity in GBM196 (but not in GBM164) indicative of HR proficiency. Consistently, HR related genes, including BRCA1 and MRE11A, were found upregulated in ALT-positive GBM196 as compared to those in GBM164. Interestingly, ALT+ GBM196 cells were highly vulnerable to inhibitors of ATM and ATR pathways. In conclusion, IDH1/TP53/ATRX mutant gliomas can be subdivided into HR-mediated ALT-positive group, which repairs the endogenous DSBs by HR (e.g. GBM196) and an ALT-negative/p-DNA-PK group, which repairs DSBs by c-NHEJ (e.g. GBM164) and this subdivision can be developed as a prescient biomarker of sensitivity to DDR inhibitors.

Since its independence, Uzbekistan has often portrayed terrorism as one of the gravest threats to its sovereignty, integrity, unity, and internal stability. Uzbekistan’s authoritarian regime has been perceived endorsing and executing a series of counter-terrorism policies to exorcise this hazardous threat that includes all possible tactics for eliminating terrorism from the respective region. Unlike the state’s proclamation about the increasing gravity of the terrorist threat, some international human right organizations and Central Asian experts depict an opposite picture of it. They are seen to be very critical of the state’s exaggerated version of the terrorist threat and question the state’s intention behind such projection. This paper makes a systematic effort to critically examine how Uzbekistan’s authoritarian leaders have constructed official discourse on terrorism, taking into consideration the social, political and economic context of the region. The paper also examines the authenticity of the state’s continuous projection of terrorism to be one of the gravest threats to the sovereignty and integrity of the region by incorporating and analyzing a detailed account of the terrorist acts that have taken place in Uzbekistan since 1991 to 2018. Finally, the paper also explains why the Uzbek authoritarian regime is keen to construct terrorism to be one of the most dangerous threats to the state. Keywords: terrorism, Uzbekistan, Counterterrorism Act, security, non-state actor

The values of the Systematic of Revelation as the basic -/pattern according to what the Hidayatullah - understands, believes, and practices in carrying out the process of social creation of a miniature Islamic Civilization are the topic of this study. The purpose of this research is to examine the process of externalizing Systematic Wahyu ideals like the Hidayatullah -' - in carrying out social construction activities in a tiny Islamic civilisation. This research looks at the reality of the Hidayatullah - congregation's consciousness, which reveals things about reality and the interpretation of the Systematic of Revelation as a -/archetype that is employed as a social construction reference. The process of externalizing the values of Systematic Revelation is a conscious, structured, and systematic effort by Hidayatullah actors to create humans as abdullah (servants of Allah) and caliph (representatives) of Allah SWT on earth, based on revelation's guidance, in order to build Islamic civilization. The Islamic boarding School Campus is employed as a model of Islamic civilisation, with all cadres, members of the congregation, and their students demonstrating faith in all parts of everyday life. Internalized Hidayatullah players objected to the values that are consistently presented and then generate cadres capable of providing enormous advantages to society. This corresponds to Berger's notion that human selfhood cannot possibly remain silent within itself, in a closed sphere, before moving outside to express itself in the world around it. Externalization is essentially what human selfhood is accomplishing.

AbstractThe desire to extend the wisdom of traditional health systems has motivated the trade of many phytomedicine on a global scale for centuries, especially some dietary herbs, making a great overlap exits between western and eastern phytomedicine. Despite the communication since ancient times, a key disconnect still exists in the dialog among western and eastern herbal researchers. There is very little systematic effort to tap into the friction and fusion of eastern and western wisdom in utilizing phytomedicine. In this review, we analyzed the similarities and differences of three representative phytomedicine, namely Rhodiola, seabuckthorn, and fenugreek, aiming to open up new horizons in developing novel health products by integrating the wisdom of the east and the west.

The transformation from agriculture to industrial business has led to radical socio-economic shifts, while the related employment issues have taken place. Certain problems have been illustrated by a critical view of attain the education, employability, patterns and political outlook for people with disabilities. While paradigms in their employment facilities have changed dramatically, a number of shortages and obstacles make a more concentrated and systematic effort to achieve maximum benefits essential for them. Hence the government taking more steps for these people those who struggling for primary education and their employment opportunity. The educational policies is the supportive stone of their employment opportunity of differently abled people and this lack of education is the informal influence of them become a self-employer in the environment for their survival. This study indicates the educational policy of differently abled people and various ministries allocated for the educated and uneducated person with disability employment opportunities in India.

Kuwait, has participated in international TIMSS test for many years, however, there has been little systematic effort to compile and assess changes between genders. This study attempts to look at the results of Kuwaiti students on the TIMSS mathematics assessments in general and according to gender in particular and the views of supervisors on the relatively low performance of Kuwaiti students. The study employed a mixed method approach in which data analysis of test results, IEA-issued documents, and statistics and official reports were used. Two focus group interviews were conducted with a convenience sample of nine educational supervisors from mathematics and science. The results show that the performance of Kuwaiti students in both the fourth and eighth grades was extremely low on the TIMSS mathematics assessments in general and in content areas in particular, since the first participation in 1995, even though the performance of eighth graders showed a slight improvement in 2015 in all areas. The TIMSS results also show that the higher the level of thinking that was assessed, the lower the performance of Kuwaiti students was. The data indicate that throughout all years, Kuwaiti girls outperformed boys, considering the slight improvement especially in 2015, yet both performances lagged behind international norms. Focus group transcript analysis reveals that supervisors perceived that students’ low performance on the TIMSS assessment test is related to a number of reasons as lack of interest in TIMSS test, unfamiliarity with TIMSS questions, and students’ weakness in the Arabic language. The paper concludes the need to systematically evaluate the TIMSS results, and develop interventions and a competent national curriculum in Kuwait.

Hemoglobin disorders (thalassemia and sickle cell disease) are a group of hereditary anemias that today occur across the world. The recent population movement has led to a steady increase of carriers and patients in all countries of the European Union. Requiring complex monitoring and treatment and, as a consequence, well-organized and nationally coordinated, supported and funded services, these lifelong conditions are now visible to healthcare services in the EU. The purpose of this study is to provide an overview of the current situation pertaining to these disorders, as perceived by the patient/parent community that the Thalassemia International Federation (TIF) represents. The aim is to establish a comprehensive understanding of the situation and unmet needs faced by migrants with thalassemia. The implementation of activities by TIF in 2018-2020 to identify and address these challenges, paves the way to increased awareness, education and policy changes building on international expertise and knowledge that will enable the provision of state-of-art clinical management services thus guaranteeing an improved quality of life. A bird’s eye view of the prevalence of these disorders is presented contributing to the further understanding of challenges met by both patients and healthcare professionals in the receipt and provision of quality healthcare respectively.

Aberrant alternative splicing is a hallmark of cancer, yet the underlying regulatory programs that control this process remain largely unknown. Here, we report a systematic effort to decipher the RNA structural code that shapes pathological splicing during breast cancer metastasis. We discovered a previously unknown structural splicing enhancer that is enriched near cassette exons with increased inclusion in highly metastatic cells. We show that the spliceosomal protein small nuclear ribonucleoprotein polypeptide A′ (SNRPA1) interacts with these enhancers to promote cassette exon inclusion. This interaction enhances metastatic lung colonization and cancer cell invasion, in part through SNRPA1-mediated regulation of PLEC alternative splicing, which can be counteracted by splicing modulating morpholinos. Our findings establish a noncanonical regulatory role for SNRPA1 as a prometastatic splicing enhancer in breast cancer.
