AbstractChaperonins are homo- or hetero-oligomeric complexes that use ATP binding and hydrolysis to facilitate protein folding. ATP hydrolysis exhibits both positive and negative cooperativity. The mechanism by which chaperonins coordinate ATP utilization in their multiple subunits remains unclear. Here we use cryoEM to study ATP binding in the homo-oligomeric archaeal chaperonin from Methanococcus maripaludis (MmCpn), consisting of two stacked rings composed of eight identical subunits each. Using a series of image classification steps, we obtained different structural snapshots of individual chaperonins undergoing the nucleotide binding process. We identified nucleotide-bound and free states of individual subunits in each chaperonin, allowing us to determine the ATP occupancy state of each MmCpn particle. We observe distinctive tertiary and quaternary structures reflecting variations in nucleotide occupancy and subunit conformations in each chaperonin complex. Detailed analysis of the nucleotide distribution in each MmCpn complex indicates that individual ATP binding events occur in a statistically random manner for MmCpn, both within and across the rings. Our findings illustrate the power of cryoEM to characterize a biochemical property of multi-subunit ligand binding cooperativity at the individual particle level.

Abstract Purpose A real-time automatic cataract-grading algorithm based on cataract video is proposed. Materials and methods In this retrospective study, we set the video of the eye lens section as the research target. A method is proposed to use YOLOv3 to assist in positioning, to automatically identify the position of the lens and classify the cataract after color space conversion. The data set is a cataract video file of 38 people's 76 eyes collected by a slit lamp. Data were collected using five random manner, the method aims to reduce the influence on the collection algorithm accuracy. The video length is within 10 s, and the classified picture data are extracted from the video file. A total of 1520 images are extracted from the image data set, and the data set is divided into training set, validation set and test set according to the ratio of 7:2:1. Results We verified it on the 76-segment clinical data test set and achieved the accuracy of 0.9400, with the AUC of 0.9880, and the F1 of 0.9388. In addition, because of the color space recognition method, the detection per frame can be completed within 29 microseconds and thus the detection efficiency has been improved significantly. Conclusion With the efficiency and effectiveness of this algorithm, the lens scan video is used as the research object, which improves the accuracy of the screening. It is closer to the actual cataract diagnosis and treatment process, and can effectively improve the cataract inspection ability of non-ophthalmologists. For cataract screening in poor areas, the accessibility of ophthalmology medical care is also increased.

Abstract This paper focuses on identifying the impact of technical and financial obstacles to the applications and expansion of the smart branches of banks in Jordan. This, however, depends on the Automation of Robotic Processes to identify the reality of applying smart branches in the world, including Jordan. The study employed a sample of 100 employees selected in an equal random manner from 25 Jordanian banks. With its spreading strands, multiple linear regression was used to test the hypotheses of the study. The study concluded that the expansion of banks of smart branches (robots) contributes to reducing operational cost and increasing the bank's profits by reducing the cost of banking service, enhancing its quality, increasing the market share of banks, and reducing the technical and financial obstacles that have a significant impact on the expansion of the smart branches of banks. The study recommended the importance of the development of smart branches of banks in Jordan due to its benefits in reducing the costs of the provided banking service. This contributes to increasing its profits. Also, banks should hold specialized training courses for bank workers on how to use the new technology. Banks should also allocate funds needed to create the essential infrastructure for the smart branch Applications of Artificial Intelligence, as this has an impact in achieving competitive advantage.

An annotated photoplethysmogram (PPG) is required when evaluating PPG algorithms that have been developed to detect the onset and systolic peaks of PPG waveforms. However, few publicly accessible PPG datasets exist in which the onset and systolic peaks of the waveforms are annotated. Therefore, this study developed a MATLAB toolbox that stitches predetermined annotated PPGs in a random manner to generate a long, annotated PPG signal. With this toolbox, any combination of four annotated PPG templates that represent regular, irregular, fast rhythm, and noisy PPG waveforms can be stitched together to generate a long, annotated PPG. Furthermore, this toolbox can simulate real-life PPG signals by introducing different noise levels and PPG waveforms. The toolbox can implement two stitching methods: one based on the systolic peak and the other on the onset. Additionally, cubic spline interpolation is used to smooth the waveform around the stitching point, and a skewness index is used as a signal quality index to select the final signal output based on the stitching method used. The developed toolbox is free and open-source software, and a graphical user interface is provided. The method of synthesizing by stitching introduced in this paper is a data augmentation strategy that can help researchers significantly increase the size and diversity of annotated PPG signals available for training and testing different feature extraction algorithms.
