AbstractDairy farming in tropical climates is challenging as heat stress can impair reproduction in cows. Previous studies have demonstrated the beneficial effects of beta-carotene supplementation on bovine reproductive performance. This study was performed in Thailand, where the temperature-humidity index (THI) during the experimental periods was measured to range from 78.4 to 86.1. Lactating Holstein cows classified as repeat breeders (previous artificial insemination [AI] failures) were randomly assigned into two treatments, control treatment (T1; received placebo, n = 200) and test treatment (T2; received 400 mg/h/day of beta-carotene, n = 200). All cows were subjected to a protocol for synchronization of ovulation and timed artificial insemination (TAI). The day of the 1st ovulation synchronized protocol was defined as day 0, and the total experimental period was 160 days. Daily placebo or beta-carotene supplements were given orally on day 0 and each subsequent day of the experiment. Diagnosis of pregnancy was performed using ultrasound on day 30 after insemination. Non-pregnant cows were subjected to further ovulation synchronizations (maximum of four) and TAI over a period of 160 days. Milk samples were collected every ten days throughout the experiment. The samples were analyzed for beta-carotene concentration, superoxide dismutase (SOD) and glutathione peroxidase (GPx) activities. The pregnancies per AI of the cows in T2 were significantly greater than that of T1 from the 2nd to 4th TAI. During the entire experimental period, the pregnancies in T2 were significantly greater than that of T1. Cox's proportional hazards regression model data indicated a 44% greater probability of pregnancy for cows receiving beta-carotene. The concentrations of milk beta-carotene in T2 were significantly greater than T1 from the 2nd to 4th TAI. Significantly greater SOD and GPx activities were observed in T2 than T1, suggesting a reduction of oxidative stress in cows treated with beta-carotene. Dietary supplementation with beta-carotene thus improves the reproductive performance of repeat breeders exposed to heat stress, possibly by reducing oxidative stress.

Pulmonary arterial hypertension of newborns (PAHN) constitutes a critical condition involving both severe cardiac remodeling and right ventricle dysfunction. One main cause of this condition is perinatal hypoxia and oxidative stress. Thus, it is a public health concern for populations living above 2500 m and in cases of intrauterine chronic hypoxia in lowlands. Still, pulmonary and cardiac impairments in PAHN lack effective treatments. Previously we have shown the beneficial effects of neonatal melatonin treatment on pulmonary circulation. However, the cardiac effects of this treatment are unknown. In this study, we assessed whether melatonin improves cardiac function and modulates right ventricle (RV) oxidative stress. Ten lambs were gestated, born, and raised at 3600 m. Lambs were divided in two groups. One received daily vehicle as control, and another received daily melatonin (1 mg·kg−1·d−1) for 21 days. Daily cardiovascular measurements were recorded and, at 29 days old, cardiac tissue was collected. Melatonin decreased pulmonary arterial pressure at the end of the experimental period. In addition, melatonin enhanced manganese superoxide dismutase and catalase (CAT) expression, while increasing CAT activity in RV. This was associated with a decrease in superoxide anion generation at the mitochondria and NADPH oxidases in RV. Finally, these effects were associated with a marked decrease of oxidative stress markers in RV. These findings support the cardioprotective effects of an oral administration of melatonin in newborns that suffer from developmental chronic hypoxia.

Chronic vasopressin secretion induced by recurrent mild heat stress exposure is significantly enhanced by limited rehydration with a fructose-containing beverage both in rodents and in humans. Moreover, this effect has been associated with upregulation of the polyol–fructokinase pathway and increased renal oxidative stress. Previously, we have shown that pharmacological inhibition of both V1a and V2 vasopressin receptors with conivaptan improved such renal alterations. The aim of this study was to evaluate the independent contributions of V1a and V2 receptors to the renal damage caused by mild heat stress and limited rehydration with a fructose-containing beverage. Osmotic minipumps were used to deliver either relcovaptan (0.64 mg/day) or tolvaptan (0.25 mg/day) in male Wistar rats for two weeks. Corresponding dilution vehicles were used as controls. To induce dehydration, rats were exposed to mild heat stress (37 °C for 1 h, Monday to Friday). All groups received a 10% fructose solution as a rehydration fluid for 2 h after mild heat stress. For the remainder of the day and on weekends, rats received tap water. The independent blockade of either the V1a or the V2 receptor prevented renal damage, reduced oxidative stress, and decreased plasma cortisol and systemic inflammation. However, the beneficial effects were regulated by different mechanisms. Tolvaptan inhibited polyol–fructokinase pathway overactivation, while relcovaptan prevented upregulation of the renin–angiotensin system and SGK1 expression. These data suggest that both V1a and V2 receptors participate in renal damage caused by heat stress-induced dehydration when fructose-containing beverages are used as rehydration fluids.

The research was conducted to study the effect of heat stress on blood parameters in indigenous sheep. Nine sheep were divided into three groups which were almost similar in age, sex and weight. Three groups were divided as zero hour (T0), four hours (T4) and eight hours (T8) heat exposure to direct sunlight. During experimental period temperature–humidity index (THI) value was calculated as 27.09 which indicate T4 and T8 groups were subjected to heat stress condition for at least four hours and eight hours respectively every day. The amount of red blood cell (RBC), hemoglobin (Hb%) and packed cell volume (PCV%) were increased significantly (p<0.05) with the increase of heat stress but the amount of white blood cell (WBC) had no significant difference (P>0.05) among the treatment groups. The objective of the present research was to describe the physiological and production responses of lactating dairy cows during and after sudden exposure to temperate-climate heat-wave conditions, compared with cows in thermoneutral conditions. Twelve lactating multiparous Holstein–Friesian dairy cows were housed in controlled-climate chambers for 4 days. Six were exposed to a short-term temperature and humidity challenge (THc, diurnal temperature and humidity fluctuations inducing moderate heat stress; temperature humidity index 74–84) and six cows were exposed to thermoneutral conditions (THn, temperatur humidity index 55–61). Cows were also measured during a 7-day pre-experimental and 14-day post-experimental period. Physiological indicators of heat stress were measured, including rectal and vaginal temperature and respiration rate, which indicated that the THc in controlled-climate chambers induced moderate heat stress. The cows exposed to the 4-day THc reduced their milk yield by 53% and their dry-matter intake by 48%, compared with the cows in the THn treatment. Milk yield of THc cows returned to pre-experimental milk yield by Day 7 and dry-matter intake by Day 4 of the post-experimental period. The short-term heat challenge induced metabolic adaptations by mobilising adipose tissue, as indicated by increased non-esterified fatty acids, and amino acids from skeletal muscle, as indicated by increased urea nitrogen to compensate for reduced nutrient intake and increased energy expenditure. Endocrine responses included greater prolactin concentrations, which is associated with thermoregulation and water metabolism. The cows exposed to THc displayed production and physical responses that facilitated lower metabolic heat production and greater heat dissipation in an attempt to maintain homeostasis during the short-term heat exposure. These results indicated that the conditions imposed on the cows in the controlled-climate chambers were sufficient to induce heat-stress responses and adversely affected production in the lactating dairy cow, and the delay between the return to normal feed intake and milk yield following the heat challenge suggests a period of metabolic recovery was occurring.

Stress emanating from environment is a factor limiting livestock productivity in the Tropics because of elevated temperature year round; hence this study took a look at Minna climate data for evaluation of Temperature-Humidity- Index (THI) as a way of identifying climate source of stress on livestock production. Climate Normals for Minna between years 1961 and 2018 were obtained, and the data were analyzed using general formulae for calculating Temperature-Humidity-Index for livestock production. Relationships between production parameters on commercial farms and the THI data indicated that heat stress is a potential cause of oxidative stress in the area. The THI showed that the environmental conditions in the study area has potential for heat stress on animals, and that it can aggravate oxidative stress in livestock under production in the study area, hence there is need for further studies to identify the pathophysiological mechanisms of heat stress so as to develop mitigation strategies for improved animal performance and productivity. The study suggested that instead of the penchant for importing exotic breeds of livestock with the aim of upgrading the indigenous breeds, the way forward could be the utilization of genetic expression of heat and oxidative stress genes in animals as candidate markers for improvement of their productive potentials.

<p>Reproductive performance is greatly affected by environmental factors such as temperature. Heat stress (HS) during pregnancy and lactation can influence not only foetal growth but also postnatal development of kits. The aim of this study was to test the effect of HS during gestation and lactation on postnatal growth till Spanish commercial liveweight. To investigate this, 32 primiparous non lactating rabbit does were exposed to 1 of 2 environmental treatments: high temperature (between 25 and 36°C, HS group; n=16) or thermoneutral conditions (between 14 and 20°C, TN group; n=16). Does were allowed to acclimate 30 d before the artificial insemination. At birth, kits were allocated into 4 groups: HS was only applied during gestation (G group; n=54); HS was applied during gestation and lactation period (GL group; n=85); HS was only applied during lactation period (L group; n=60); and TN was applied during gestation and lactation period (C group; n=77). All litters were kept under each experimental environment until weaning at day 30. Then, litters were moved to TN temperatures until slaughter at day 63. Compared with TN does, the HS does presented lower litter size (9.7 and 11.4; P&lt;0.05), litter weight (503.0 vs. 630.5 g; P&lt;0.05) and kit weight at birth (56.6 vs. 61.4 g; P&lt;0.05), as well as a higher stillborn rate (25.4 vs. 9.9%; P&lt;0.05). The kits from does subjected to HS during gestation (G group) had similar postnatal growth compared to offspring from does gestated in TN conditions (C group), whereas kits from does that experienced HS during gestation and lactation (GL group) and during their lactation (L group) presented decreased postnatal growth. Together, these results demonstrate that kits from does that underwent HS during gestation did not alter postnatal growth until Spanish commercial liveweight, whereas HS during lactation resulted in decreased postnatal growth.</p>

Paraquat (PQ) is a dopaminergic neurotoxin and a well-known pneumotoxicant that exerts its toxic effect via oxidative stress-mediated cellular injuries. This study investigated the protective effects of Tualang honey against PQ-induced toxicity in the midbrain and lungs of rats. The rats were orally treated with distilled water (2 mL/kg/day), Tualang honey (1.0 g/kg/day), or ubiquinol (0.2 g/kg/day) throughout the experimental period. Two weeks after the respective treatments, the rats were injected intraperitoneally with saline (1 mL/kg/week) or PQ (10 mg/kg/week) once per week for four consecutive weeks. After four weekly exposures to PQ, the glutathione peroxidase activity and the number of tyrosine-hydroxylase immunopositive neurons in the midbrain were significantly decreased in animals from group PQ (p<0.05). The lungs of animals from group PQ showed significantly decreased activity of superoxide dismutase and glutathione-S-transferase. Treatment with Tualang honey ameliorated the toxic effects observed in the midbrain and lungs. The beneficial effects of Tualang honey were comparable to those of ubiquinol, which was used as a positive control. These findings suggest that treatment with Tualang honey may protect against PQ-induced toxicity in the rat midbrain and lung.


Intermittent Lighting Improves the Efficiency of Artificial Insemination in Cage Housed Laying Hens Kavtarashvili A.Sh., Kolokolnikova T.N. Federal Scientific Center “All-Russian Research and Technological Poultry Institute” of Russian Academy of Sciences Omsk Agrarian Scientific Center Summary: The effects of different lighting regimes on the oviposition schedule, productive performance, and reproductive efficiency in cage housed laying hens of layer parental flock (Hisex White-R) were studied; the reasonable regime of artificial insemination (AI) under intermittent lighting is proposed. It was found that intermittent lighting regime 1L:4D:4L:1D:4L:10D compared to the constant lighting significantly alters oviposition schedule: under this regime 82.3% of all daily eggs were laid until 9 am (vs. 66.6% in control). This regime and AI at 10 am improved the productive and reproductive performance compared to control (constant lighting 16L:8D and AI at 12 am): mortality by 1.9%, egg production per initial hen by 3.8%, egg weight by 1.1%, percentage of eggs suitable for incubation by 1.9%, egg fertility by 0.9%, hatchability by 2.3%, hatch of chicks by 2.9%, feed conversion ratio (kg of feed per 10 eggs) by 5.3%, the expenses of electric energy for lighting (per 1000 eggs suitable for incubation) by 54.5%. DM otherwise diabetes is now a days an epidemic with the percentage of patient population rising to almost 10% of the world population. Out of all the DM complications, cataract leads the way contributing to disabilities to about 60% of diabetic population. But the pathogenesis of DM cataract is still a half-understood area of medicine there by posing a problem in the therapy. The data that we have till now gives us enough evidence to advocate the oxidative stress has a major role for the pathogenesis of DM complications like DMnephropathy, DMneuropathy, and cardiac hypertrophy, which suggests the oxidative stress is a central feature of diabetes. In the current research, the pharmacological evaluation of Fisetin for its DM based anti-cataract property was performed. This research concentrates to estimate the possible involvement of Nrf-2 / heme oxygenase (HO)-pathway in the observed therapeutic effect, if any. The data obtained in this study also indicate that the observed beneficial effects mainly due to activation of Nrf2/HO-1 pathway. These effects probably result in increased tissue anti-oxidant status as well as decreased free radical production, which ultimately responsible for the observed beneficial effects of Fisetin against hyperglycemia-induced cataract.
