Parijoto (Medinilla speciosa) is an endemic plant in Indonesia that its fruit have a purplish red color, which indicated that parijoto fruits contain anthocyanins, pigments of the flavonoid class that responsible for the red, blue and purple coloration of many flowers and fruits. This study was aimed to determine the total yield of extract and anthocyanins content of methanol extract of parijoto (Medinilla speciosa) pericarp and whole fruit and the effect of parijoto whole fruit extract on serum malondialdehyde (MDA) level of the hyperlipidemic rats. Extraction of parijoto fruit and pericarp were carried out by the maceration method with methanol as a solvent. Determination of the total anthocyanin was done by pH difference method. MDA serum levels were done in a pre-test and post-test Randomized controlled group design was used in this study. Rats were divided into five groups: normal rats, hyperlipidemic rats, hyperlipidemic rats treated with methanol extract of parijoto (500 mg · kg ˗1, 1000 mg · kg ˗1, and 1500 mg · kg˗1 body weight). Blood was collected at hyperlipidemia state and after treatment of methanol extract of parijoto for determination of MDA level in blood serum. The results showed that the total yield of parijoto pericarp extract and the whole fruits were 11.46 g (3.18% ) and 9.02 g (2.88%) respectively. Anthocyanins content of parijoto pericarp and whole fruit extracts  were 208.75 mg · L-1 and 173.7 mg · L-1. Administration of methanol extract of whole fruit parijoto  in hyperlipidemic rats reduced serum MDA levels significantly (p < 0.01), from 14.88 nmol · mL-1 to 8.63 nmol · mL-1.

Macaranga beccariana Merr. is an endemic plant of Kalimantan which is no report yet about its biological activity and isolated compounds. Therefore, in order to know the potency of M. beccariana, determination of antioxidant and antimalarial activities along with isolation of bioactive compounds from this plant must be carried out. In this research, the leaves of M. beccariana was extracted using methanol to obtain methanol extract. Furthermore, the extract was separated and purified to obtain compound 1 and compound 2. Based on structure elucidation using spectrometer analysis including UV, 1H and 13C NMR, compounds 1 and 2 which belong to phenolics were identified as 4-hidroxybenzoic acid and 3,4-dihidroxybenzoic acid, respectively. Moreover, antioxidant activity using DPPH (2,2-diphenyl-1-picrylhydrazyl) and antimalarial activity towards Plasmodium falciparum strain 3D7 using Giemsa staining were performed. Rutine and chloroquine difosfat were used as positive controls for antioxidant and antimalarial, respectively. The results showed that the methanol extract of M. beccariana and its isolated compounds (1 and 2) are active for those activities. It can be concluded that the leaves of M. beccariana has good potency as antioxidant and antimalarial agents.

Telang flowers is one of the medicinal plants that can grow well in Indonesia, but have not been widely used. Previous research stated that methanol extract of telang flowers had secondary metabolite compounds such as tannin, saponin, flavonoid and alkaloid. Flavonoid are natural compounds that have the potential as antioxidants that can capture free radicals that play a role in the emergence of degenerative diseases throught the mechanisme of damage to the body’s immune system, lipid oxidation and protein.The purpose of this study was to determine whether telang flowers content was determined whether by the AlCl3 method and measured by using UV-Vis Spektrophotometry from the methanol extract of telang flowers.The results of this study indicate that the methanol extract of telang flowers contains flavonoid with red color. The level of flavonoids intended is 4.65%.

Human prothrombin activation is unique in that, in addition to the release of fragment 1.2 (FI.2) from the NH-terminus of prothrombin by factor Xa during the generation of thrombin, an additional 13 residue polypeptide, fragment 3 (F3), is autocatalytically removed from the amino-terminus of the thrombin A chain. We have developed a rapid radioimmunoassay for human F3 which incorporates short incubation times and the use of a preprecipitated second antibody; the assay can be performed in three hours. Specificity studies in buffer systems show prothrombin and prethrombin 1 cross-reacting at a level of 0.001; purified thrombin does not cross-react. In the presence of 5% BSA, prothrombin displays considerably less cross-reactivity. No immunoreactive material to F3 antibodies could be detected in 400 μL of plasma. Serum, obtained from whole blood clotting, contained measurable quantities of F3 (40-100 ng/mL). This amount in serum represents only 5-10% of the theoretical amount available should all of the fragment be hydrolytically cleaved during the conversion of prothrombin to thrombin. This assay procedure is currently being utilized to monitor the activation of purified human prothrombin in the absence and presence of selected plasma inhibitors. This study was based on determination of the antiulcer activity from methanol extract was prepared by using barks of pergularia extensa linn.. Priliminary investigations showed presence of saponins, terpenes, cardiac glycosides, alkaloids and sterols. Based on OECD-423 Guidelines, the pharmacology and acute oral toxicity studies were conducted by using methanolic extract. Ulcer development was prevented by Tannins because of their vasoconstriction effects and due to protein precipitation. Similarly, the Methanolic extract of Pergularia extensa Linn shows triterpenoids and saponins. The phytoconstituents are present in the extract and these could be possible agents which are involved in order to prevent gastric lesions induced by aspirin. When compared to ulcerative control groups, this Pergularia extensa Linn., shows a dose dependent curative ratio. The extracts exhibited an inhibition percentage of 27.18, 45.47 and 61.28 at doses of 100, 200 and 400mg/kg doses respectively.

The objective of the current study is represented by the determination of silica and a phytochemical screening of phenolic derivates of some Equisetum species. The antioxidant and antimicrobial activity for Equisetum pratense Ehrh.,, Equisetum sylvaticum L. and Equisetum telmateia Ehrh. (sin. Equisetum maximum Lam.) were also investigated. The concentration of silicon (Si) in plants was determined by the spectrophotometric method using previous treatment with NaOH 50% both for the stem and the nodal branches [1]. Results obtained varied from 95.12 to 162.10 SiO2 mg/g dry plant which represents 4.44% to 7.58% Si/100g dry sample. Two types of total extracts were obtained using different solvents and were subjected to qualitative and quantitative chemical analysis considering total phenolic content [2]. The highest concentration of investigated compounds was found in the methanolic extract, E. sylvaticum, 196.5mg/g dry sample. Antioxidant activity was monitored spectrophotometrically and expressed in terms of IC50 (�g/mL) [3]. Values gathered ranged from 261.7 to 429.5 �g/mL. The highest capacity to neutralized DPPH radicals was found in E. sylvaticum. In vitro antimicrobial activity was determined using difusimetric method [4]. Testing was performed on four microorganisms: three strains of bacteria and one species of fungi. Different effects were noticed against the bacteria, furthermore the methanol extract appeared to be most efficient. All extracts showed significand antimicrobial activity against Staphylococcus aureus (ATCC 25923) and Candida albicans (ATCC 90028) and weak to no activity against Pseudomonas aeruginosa (ATCC 27853) and Escherichia coli (ATCC 25922).

Abstract A procedure for the enzymatic determination of lecithin and sphingomyelin in aqueous solution is described. The phospholipids are first dissolved in chloroform:methanol (2:1 by vol), the solvent is evaporated, and the residue is redissolved in an aqueous zwitterionic detergent solution. The enzymatic reaction sequences of both assays involve hydrolysis of the phospholipids to produce choline, which is then oxidized to betaine, thus generating hydrogen peroxide. The hydrogen peroxide is subsequently utilized in the enzymatic coupling of 4-aminoantipyrine and sodium 2-hydroxy-3,5-dichlorobenzenesulfonate, an intensely red color being formed. The presence of a non-reacting phospholipid enhances the hydrolysis of the reacting phospholipid. Thus we added lecithin to the sphingomyelin standards and sphingomyelin to the lecithin standards. This precise procedure may be applicable to determination of lecithin and sphingomyelin in amniotic fluid.

Summary Background Circulating serum sclerostin levels are supposed to give a good estimation of the levels of this negative regulator of bone mass within bone. Most studies evaluating total serum sclerostin found different levels in males compared to females and in older compared to younger subjects. Besides an ELISA detecting total sclerostin an ELISA determining bioactive sclerostin has been developed. The aim of this study was to investigate serum levels of bioactive sclerostin in an Austrian population-based cohort. Methods We conducted a cross-sectional observational study in 235 healthy subjects. Using the bioactive ELISA assay (Biomedica) bioactive sclerostin levels were evaluated. Results Serum levels of bioactive sclerostin were higher in men than in women (24%). The levels correlated positively with age (r = 0.47). A positive correlation could also be detected with body mass index and bone mineral density. Conclusion Using the ELISA detecting bioactive sclerostin our results are consistent with data in the literature obtained by different sclerostin assays. The determination of sclerostin concentrations in peripheral blood thus appears to be a robust parameter of bone metabolism.
