In-vitro antimicrobial efficacy of certian herbal seeds essential oils against important poultry microbes

Two in-vitro experiments were conducted to evaluate the antibacterial, antifungal and antiviral properties of essential oil of herbal seeds. In-vitro antimicrobial properties of essential oils of phytobiotics was determined by disc diffusion method against Escherichia coli, Staphylococcus aureus, Pasteurella multocida and Salmonella typhi and two fungi namely Aspergillus fumigatus and Candida albicans. Thyme oil had statistically similar or significantly (P<0.05) higher inhibition zone against all the bacterial and fungal species compared to standard antibiotic, chloramphenicol or antifungal drug, nystatin. Fenugreek oil was not found to be active against microbes compared to control. Fennel and cumin oils had statistically similar or significantly (P<0.05) higher inhibition zone against all microbes except Staphylococcus aureus and Salmonella typhi compared to control. Antiviral activity of essential oils was determined by anti-NDV assay against New Castle disease virus (LaSota). The results indicated that essential oils of herbal seeds do not possess antiviral activity against NDV LaSota virus at 1mg/ml concentration.

  Objective: The inhibitory properties of successive extracts from Dioscorea bulbifera (Dioscoreaceae) tubers have been evaluated for the presence of phytochemical constituents and antimicrobial efficacy against multidrug-resistant (MDR) clinical isolates was evaluated.Methods: The tuber of D. bulbifera was oven dried and extracted successively with n-hexane, chloroform, methanol, ethanol, and water. The antimicrobial potential of successive extracts against MDR isolates was studied by agar well-diffusion method. Qualitative phytochemical analysis was performed.Results: Qualitative phytochemical analysis demonstrated the presence of steroids, flavonoids, cardiac glycosides, saponins, and reducing sugars in almost all the extracts tested. Anthraquinones, phlobatanins, and tannins were not reported in any extracts tested. The in vitro antimicrobial activity of various solvents and water extracts of D. bulbifera was further investigated against ten MDR bacteria and three fungi, respectively. Aqueous and chloroform extracts were found to be more potent being capable of exerting significant inhibitory activities against the majority of the isolates such as Escherichia coli, Acinetobacter sp., Salmonella paratyphi, Klebsiella pneumoniae, and Candida albicans. The highest inhibitory activity was observed for K. pneumoniae with wide inhibition zone diameters (17 ± 0.15 mm), followed by E. coli 1(13 ± 0.11) mm, and Acinetobacter sp. (11 ± 0.12).Conclusion: Based on the present study, the extracts of D. bulbifera tubers have shown excellent activity against MDR microbial cultures tested. Further study is recommended for clinical evaluation, of the efficacy of crude extract in herbal medicine that can serve as a base for the development of novel potent drugs and phytomedicines.

A study was carried out to screen for phytochemical constituents and assess the antioxidant and antimicrobial activities of Senna alata and Senna tora leaf extracts. The leaves were first dried at room temperature and 50°C in an oven prior to solvent extraction using ethanol and methanol. The in-vitro qualitative assays showed that both S. alata and S. tora leaf extracts contained bioactive and secondary metabolites components such as tannins, steroids, saponin, terpenoids, glycosides, flavonoids and phenols. The antioxidant activity and capacity test were carried out by conducting free radical of 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity and Ferric reduction antioxidant plasma (FRAP) assays. Both assays showed S. tora leaf extract has higher antioxidant capacity than S. alata leaf extract. The efficacy of these leaf extracts were tested against skin pathogens through agar well diffusion method. S. alata extract showed an inhibition zone (1.15 – 1.59 mm) against Pseudomonas aeruginosa while S. tora extracts exhibited a strong antimicrobial activity against S. epidermidis (inhibition zone of 12 – 16.94 mm) followed by P. aeruginosa (inhibition zone of 1 – 1.59 mm). Nonetheless, no inhibition zone was observed for S. aureus by both leaf extracts. The phytochemicals and antioxidant constituents as well as inhibitory potential on skin pathogens possessed by S. alata and S. tora leave highlighted their potential utilization in the development of natural drugs or cosmetics to treat skin related diseases or infections.

The present study was aimed at a comparative study of the phytoconstituents, antioxidant and antimicrobial properties of four medicinal plants, Ficus capensis, Aristolochia ringens, Albizia zygia and Lannea welwitschii. The leaf of F. capensis and stem bark of A. ringens, A. zygia and L. welwitschii were extracted using methanol. Phytochemical analysis was done spectrophotometrically. Three in vitro antioxidant tests-hydrogen peroxide (H2O2), nitric oxide (NO) and 2, 2-diphenyl-1-picrylhydroxyl (DPPH) scavenging models were employed. Antimicrobial test was done by agar diffusion method against E. coli, Staphylococcus aureus, Klebsiela pneumonia, Bacillus subtilis, Pseudomonas aeruginosa, Salmonella typhi, Candida albicans and Aspergittus spp. Phytochemical analysis revealed that the most abundant phytoconstituents were flavonoids (F. capensis and A. zygia), reducing sugar, terpenoids, alkaloids and tannins. In the antioxidant models, A. zygia was found to produce the least IC50 in two of the models (NO and DPPH). Results of the antimicrobial tests showed that A. zygia showed a broader spectrum of activity than other plants. This study has shown that these plants possess antioxidant and antimicrobial activities which further justify their usage in traditional medicines. A. zygia featured prominently in these activities. Actinomycetes are found in soils with loose, humus, dry characteristics and around plant roots. Actinomycetes produce secondary metabolite compounds as antibacterial. Escherichia coli, Pseudomonas aeruginosa and Salmonella typhi are Gram negative bacteria that can cause infection in humans. This study aims to determine the ability of the Actinomycetes metabolite filtrate from Bogor Botanical Gardens as an antibacterial agent against the growth of E. coli, P.aeruginosa and S. typhi in vitro. This study used an experimental design with samples of Actinomycetes isolats originating from the Bogor Botanical Gardens soil using the disc diffusion method on Muller Hinton Agar (MHA) media by looking at the clear zone of bacterial growth around the disc paper. Of the three concentration groups, namely 50%, 60%, and 70%, the largest average inhibition zone is found at a concentration of 70% with the average for the three test bacteria E. coli, P.aeruginosa and S. typhi respectively: 4.23 mm; 3.0 mm and 8.43 mm. The results of the Kruskal - Wallis test with p value = 0.01 showed that there was an effect of the Actinomycetes metabolite filtrate on the growth of the three tested bacteria as antibacterials.

Abstract:Kasturi as a typical plant in South Kalimantan is one fruit that has many benefits. The barks of kasturi has proved to have benefits, especially to inhibit the activity of Staphylococcus aureus. Staphylococcus aureus is a gram positive bacteria that cause pneumonia, mastitis, and urinary tract infections. The bark of kasturi can be used in extract form. The objective of this research is to know the difference between the preparations of inhibition kasturi’s bark extract and ampicillin 30μg in inhibiting the growth of Staphylococcus aureus in vitro. This study used an experimental method consisting of 9 treatments with 3 repetitions. Treatment test in the methanol extract of the bark of kasturi 25%, 37.5%, 50%, 62.5%, 75%, 87.5% and 100%. The control group used ampicillin and 70% methanol. Bacterial test was done by using a diffusion method. The parameter measured was the amount of inhibition zone (mm) which grown on media MH. Analysis of study data used One way Annova test and Post Hoc LSD test at α=0,05. The results showed that there were significant differences between the treatment kasturi’s bark extract 25%, 50%, 62.5%, 75%, 87.5% and 100% concentration different compared to ampicillin. Meanwhile, at the concentration of 37.5% (p <0.05) was not significant. The antibacterial effectiveness was obtained from the concentration of 100%. Key words: the bark of kasturi, methanol extract, Staphylococcus aureus, inhibition zone. Abstrak: Kasturi sebagai salah satu tanaman khas yang ada di Kalimantan Selatan merupakan salah satu buah yang memiliki banyak khasiat. Kulit batang kasturi terbukti memiliki manfaat terutama dapat menghambat aktivitas Staphylococcus aureus. Staphylococcus aureus merupakan bakteri gram positif yang menyebabkan penyakit pneumonia, mastitis, dan infeksi saluran kemih. Kulit batang kasturi dapat digunakan dalam bentuk ekstrak. Tujuan penelitian ini mengetahui perbedaan daya hambat antara sediaan ekstrak kulit batang kasturi dengan ampisilin 30µg dalam menghambat pertumbuhan bakteri Staphylococcus aureus secara in vitro. Penelitian ini menggunakan metode eksperimental yang terdiri dari 9 perlakuan dengan 3 kali pengulangan. Perlakuan yang di uji adalah ekstrak metanol kulit batang kasturi 25%, 37,5%, 50%, 62,5%, 75%, 87,5% dan 100%. Dan kontrol perlakuan dengan ampisilin dan metanol 70%. Uji bakteri dilakukan menggunakan metode difusi. Parameter yang diukur adalah besaran zona hambat (mm) yang tumbuh pada media MH. Analisis data penelitian mengunakan uji One way ANNOVA dan uji Post Hoc LSD pada α=0,05. Hasil penelitian menunjukan bahwa terdapat perbedaan bermakna diantara perlakuan ekstrak kulit batang kasturi 25%, 50%, 62,5%, 75%, 87,5% dan 100% konsentrasi yang beda di bandingkan dengan ampisilin. Sedangkan pada konsentrasi 37,5% (p<0,05) tidak bermakna. Efektivitas antibakteri yang terbesar di peroleh dari konsentrasi 100%.Kata-kata kunci: kulit batang kasturi, ekstrak metanol, Staphylococcus aureus, zona hambat.

Introduction: Staphylococcus aureus infection can cause pyoderma. These days there is a shift in Staphylococcus aureus sensitivity to antibiotics that is marked by the existence of Methicillin-Resistant Staphyloccus aureus, so alternative therapy that is herbal medicine might be needed. There are a lot of medicinal plants in Indonesia, but until today the usage is still minimum. One of the plant genus that is already being used is allium that had been known to have antimicrobial activity. The genus allium that is consumed a lot by Indonesians especially people of North Sumatra is bawang batak (Allium chinense G.Don.). This study is conducted to assess the antimicrobial activity of bawang batak bulbs in several concentrations to MRSA. Methods: This research is an experimental research with agar diffusion method. The extracts were divided into six groups: extracts in concentration level of 6.25%, 12.5%, 25%, 50%, 100% and a negative control (DMSO). The sample size is four per group with total sample of 24. Results: The results showed that the inhibition of bacterial growth occured at the concentration levels of 50% and 100% with inhibition zone diameters of 8.695 mm and 10.545 mm respectively. Extracts with concentration levels of 25%, 12.5% and 6.25% did not show any inhibition. Conclusion: Bawang batak bulbs extract can be used to inhibit the growth of MRSA

AbstractBackground Staphylococcus aureus is one of causes infection and this bacteria have been resistence for many antibiotic. Bay leaf have antibacterials substance, which stew leaves can be treat infection caused Staphylococcus aureus. The purpose of this study was to determine differences in growth inhibition zone of Staphylococcus aureus at various concentrations of water stew of bay leaf.Method The method of this study is true experiment with posstest only control design, and used Kirby Bauer disk diffusion method with various concentrations of water stewed of bay leaf (20%, 40%, 60%, 80%,100%), positive control (chloramfenicol 30 μg) and negative control (sterile distilled water). Result The result showed that the average diameter of inhibition zone in concentration 20%, 40%, 60%, 80%, and 100% is 7 mm, 8,4 mm, 9,6 mm, 10,5 mm and 11,5 mm. Based on statistical analysis using oneway ANOVA available the value of p (0,000) α (0,05), so the inhibition zone is significant difference of growth inhibition zone of Staphylococcus aureus at various concentrations of stewed water bay leaf. Conclusion Water stew of bay leaf can inhibit the growth of Staphylococcus aureus, and there are differences in growth inhibition zone of Staphylococcus aureus at various concentrations of water stew of bay leaf. Keywords: stew of bay leaf; Staphylococcus aureus; inhibition zone

This study aimed to determine the inhibitory effect and the concentration of fingerroot extract to inhibit Staphylococcus aureus growth. The data was analyzed descriptively. Ampicillin was used as positive control, distilled water was used as negative control, and the treatments were given fingerroot extract with a concentration of 5%, 15%, 25%, 35% and 45%. This study was conducted with three replications. The parameter measured was the diameter of inhibition zone formed by diffusion method. The average diameter of inhibition zone of the fingerroot extract were 15% : 10.3 mm; 25% : 13,6mm; 35% : 18,7mm; 45% ± 21,1mm, and at a concentration of 5% the inhibition zone is not formed. The final conclusion is that the fingerroot extract can inhibit the growth of Staphylococcus aureus. The higher concentration of fingerroot extract, the more extensive inhibition zone formed.

ABSTRACTBackground Staphylococcus aureus is a pathogenic bacteria that can cause infection. Biduri leaf has antibacterial compound such as tannin, flavonoid, saponin, and polyphenol.Objective This study aimed to know the difference of growth inhibition zone of Staphylococcus aureus at the various concentration of ethanol extract of biduri leaf and to determine an effective concentration.Methods This study was the true experiment with posttest only control design, used Kirby-Bauer disk diffusion method with five concentrations (20%, 40%, 60%, 80%,100%), positive control (chloramphenicol 30 µg) and negative control (ethanol 96%).Result The result of this study showed the average of inhibition zone diameter of each concentration consecutively 26,2 mm, 28,3 mm, 29,7 mm, 31 mm and 31,5 mm. One Way Anova statistic analysis showed that the value of p is 0.000 so there is the difference of growth inhibition zone of Staphylococcus aureus at various concentrations of ethanol extract of a biduri leaf.Conclusion The conclusion of this study is there are differences in growth inhibition zone of Staphylococcus aureus at various concentrations of ethanol extract of biduri leaf and an effective concentration that can inhibit of Staphylococcus aureus is concentration 20% with mean inhibition zone is 26,2 mm. Keywords: ethanol extract of biduri leaf; Staphylococcus aureus; inhibition zone

Our research work was designed to investigate the curative and preventive effects of Carthamus oxycantha root extract against diarrhea and microorganisms. For the antibacterial experiment, the agar well diffusion method was used against standard bacteria Staphylococcus aureus, Escherichia coli, Pseudomonas aeroginosa, and Salmonella typhi, while for the assessment of antidiarrheal activity, castor oil and the magnesium sulfate-induced diarrhea method was used on albino, laboratory-bred (BALB/c) mice at a dose rate of 200 and 400 mg/kg (body weight, b.w) orally. The methanol extract of C. oxycantha significantly (p < 0.001) decreased the frequency of defecation, and wet stools in a dose depended on the manner of after receiving magnesium sulfate (2 g/kg (b.w)) and castor oil (1.0 mL/mice). Furthermore, the extract of C. oxycantha showed concentration-dependent antimicrobial properties against S. aureus followed by S. typhi, E. coli, and P. aeroginosa bacterial strains, with inhibitions ranging from 10.5–15 mm. These findings show significant results that C. oxycantha is effective as an antidiarrheal and antibacterial agent. However, further works are needed to establish its mode of action.
