Abstract BackgroundSeed deterioration during rice seed storage will lead to seed vigor loss, which adversely affects agricultural production, the long-term preservation of germplasm resources, and the conservation of species diversity. However, the mechanisms underlying seed vigor maintenance remain largely unknown. ResultsIn this study, 16 hybrid rice combinations were selected from four sterile lines and four restorer lines. Following artificial aging and natural aging treatments, the metabolite markers that could accurately reflect the aging degree of the hybrid rice seeds were identified based on the germination percentage and metabolomics analysis by gas chromatography-mass spectrometry. Significantly differences in the degree of seed deterioration were observed among the 16 hybrid rice combinations tested, with each restorer and sterile lines after storage having the different germination percentage. The hybrid rice combination with the storage-resistant restorer line Guanghui122 exhibited the highest germination percentage under both natural and artificial storage. A total of 89 metabolic peaks and 56 metabolites were identified, most of which were related to primary metabolism. Interestingly, the content of galactose, gluconic acid, fructose and glycerol in the seeds increased significantly during the aging process. Absolute quantification indicated that galactose and gluconic acid were very significantly negatively correlated with the germination percentage of the seeds under the different aging treatments. The galactose content was significantly positively correlated with gluconic acid content. Additionally, while the relative content of raffinose did not change much during storage, a significant positive correlation between raffinose and the germination rate of the artificially aged seeds before storage was detected.ConclusionBased on the metabolomics, metabolite markers which could accurately reflect the aging degree of hybrid rice seeds were identified. Galactose and gluconic acid were very significantly negatively correlated with the germination percentage of the seeds which suggested that these metabolites could constitute potential metabolic markers of seed aging. These findings are of great significance for the rapid and accurate evaluation of seed aging, the determination of seed quality, and the development of molecular breeding approaches for high-vigor rice seeds.

The Effectiveness of Liquid Smoke and Electrotherapy to Eliminate Infection of Xanthomonas oryzae pv. oryzae in Rice SeedBacterial leaf blight caused by Xanthomonas oryzae pv. oryzae is a seed-borne disease that can reduce the productivity of rice. Alternative treatments that have not been widely developed is application of liquid smoke and electrotherapy. The aim of this research was to study the effectiveness of liquid smoke and electrotherapy and their combinations to eliminate X. oryzae pv. oryzae without damaging seed quality. The study consisted of three experimental stages: (1) Liquid smoke treatment on the viability of the bacteria and rice seeds; (2) Electrotherapy treatment on the viability of bacteria and rice seeds; (3) Combination of liquid smoke and electrotherapy treatment in bacteria-infected rice seed. The results showed that liquid smoke concentration (5% for 30 minutes) was able to reduce 83.33% population of X. oryzae pv.oryzae on rice seeds with 94.33% seed vigor, 98.00% germination, and time required to achieve 50% of total seed emergence (T50) of 3.13 days. Electrotherapy treatment (400 mA for 20 minutes) was not only eliminated X. oryzae pv. oryzae on rice seeds up to 100% but also maintained seed vigor 81.67%, germination 89.33%, and T50 3.47 days. Electrotherapy treatment (400 mA for 20 minutes) followed by immersion of infected rice seeds in liquid smoke concentration (1% for 30 minutes) could reduce 94.59% bacteria population while maintaining 86.00% seed germination. Treatment of electrotherapy (400 mA for 20 minutes) followed by immersion seeds into liquid smoke concentration (5% for 30 minutes) could reduce bacterial population up to 100%, but caused phytotoxicity effect to the seedling.

This study aimed to evaluate the efficiency of integrated biological and chemical control of pathogens in rice seeds and their effects on seed quality. The experiment was conducted in a 2 × 5 factorial completely randomized design. Fungicide-treated (carboxin/thiram) and untreated rice seeds were inoculated with distilled water (control), Azospirillum brasilense, Bacillus subtilis, Pseudomonas fluorescens, or Trichoderma harzianum. Seed vigor and viability, shoot and root length, and seedling dry weight were determined. The blotter test was carried out to assess seed health. Fungicide treatment improved seed vigor and viability and reduced the incidence of fungi. Biological treatment did not enhance the physiological quality of seeds but was able to control fungi. A. brasilense, B. subtilis, P. fluorescens, and T. harzianum controlled Phoma sorghina; B. subtilis, P. fluorescens, and T. harzianum were effective against Aspergillus flavus; P. fluorescens and T. harzianum controlled Pyricularia oryzae; and T. harzianum was effective against Gerlachia oryzae.

<p>Seed is a component of production that contributes to increasing the national rice production. The use of low-quality seeds compared with production costs, non-uniform plant growth, yield reduction, and yield quality. One effort to improve the quality of seed vigor is through invigoration treatment. Invigoration technique among other hydration, thermal treatment, and coating. The hydration method is a water absorption system in seeds. Uncontrolled absorption (hydro-priming), by soaking and drying the seeds, hardening, on-farm priming, soaking for a certain period. The controlled system (osmo-priming) by immersing in osmotic solution, osmo-hardening (integration of hardening and osmo-priming), matri-priming with moist solid media, humidifications (with high humidity), and hormonal priming (with hormones. The second method is by temperature treatment, including by cooling and heating. The third technique is the coating treatment, namely coating the seeds. Hydration system hydro-priming for 30 hours can break seed dormancy with normal germination capacity of 91.3-99.3% in Memberamo, Cipunagara, and Maros varieties. The hardening technique is effective in increasing the germination of hybrid rice seeds for 12 hours with 2 cycles and for 24 hours with one cycle. The osmo-priming technique of soaking seeds in a 10 ppm GA3 solution or a mixture of 10 ppm GA3 + 15 ppm kinetin was effective in increasing the vigor of hybrid rice seeds. The matri-priming technique with rubbing ash as a medium plus the addition of 50 µ m GA3 was effective in breaking dormancy of the Membramo, Cipunagara, and Maros varieties of rice. It is can be applied, especially by small farmers.</p><p>Keywords: Rice, seed, invigoration, quality</p><p> </p><p><strong>Abstrak</strong></p><p><strong>PENERAPAN TEKNIK INVIGORASI DALAM MENINGKATKAN VIGOR BENIH PADI </strong></p><p>Benih merupakan komponen budi daya yang berkontribusi dalam meningkatkan produksi padi nasional. Penggunaan benih bermutu rendah berpengaruh terhadap penambahan biaya produksi, pertumbuhan tanaman tidak seragam, dan penurunan hasil dan mutu hasil. Salah satu upaya untuk meningkatkan mutu benih dari aspek vigor yang rendah adalah melalui perlakuan invigorasi. Teknik invigorasi dibagi menjadi tiga metode, yaitu hidrasi, perlakuan suhu, dan pelapisan (coating). Metode hidrasi merupakan sistem penyerapan air yang dapat terjadi pada benih. Penyerapan tidak terkontrol (hydro-priming) yaitu dengan merendam dan mengeringkan benih, pengerasan (hardening), on-farm priming yaitu merendam dalam jangka waktu tertentu. Sedangkan terkontrol (osmo-priming) dengan merendam pada larutan osmotik, osmo-hardening (integrasi hardening dan osmo-priming), matripriming dengan media padat lembab, humidifications (dengan kelembaban tinggi), dan hormonal priming (dengan hormon). Metode kedua adalah dengan perlakuan suhu, diantaranya dengan pendinginan dan pemanasan. Teknik ketiga adalah dengan perlakuan coating yaitu melapisi benih. Sistem hidrasi hydropriming selama 30 jam dapat mematahkan dormansi benih dengan daya kecambah normal 91,3-99,3% pada varietas Memberamo, Cipunagara, dan Maros. Teknik hardening efektif meningkatkan daya berkecambah benih padi hibrida selama 12 jam dengan dua siklus dan selama 24 jam dengan satu siklus. Teknik osmo-priming perendaman benih dalam larutan GA3 10 ppm atau larutan campuran GA3 10 ppm + kinetin 15 ppm efektif meningkatkan vigor benih padi hibrida. Teknik matri-priming dengan abu gosok sebagai media plus penambahan GA3 50 µm efektif mematahkan dormansi padi varietas Membramo, Cipunagara, dan Maros. Teknik invigorasi tersebut dapat diterapkan, khususnya oleh petani dengan skala produksi kecil.</p><p>Kata kunci: Padi, benih, invigorasi, mutu</p>

Aim: This study was conducted to develop liquid enhancer containing KCl, TU, GA, and SA for germination of drought-stressed Oryza sativa subsp. indica cv. MR284 seed. Study Design: All experiments were conducted in a completely randomized design. Two steps were involved in the development process which are to select an ideal concentration for each KCl, TU, GA, and SA, and to find an ideal combination of chemicals from the selection of ideal concentrations acquired in step 1 to form liquid enhancer. There were 20 treatments for step 1 and 9 treatments for step 2. All of these treatments with 6 replicates. Place and Duration of Study: Department of Biology, Faculty of Science, University Putra Malaysia, between June 2018 and December 2018. Methodology: The sterilized rice seed cv. MR284 was stressed in the -1.2 Mpa PEG 6000 solution for three days and germinated in the KCl, TU, GA, and SA solution in a series of concentration for 10 days, in a controlled room. Seed germination was observed daily. Results: In the first step, drought-stressed rice seed showed the best germination performance in the 30 mM of KCl, 2.0 mM of TU, 0.24 mM GA, and 0.5 mM SA. Meanwhile, in the second step, the drought-stressed rice seed showed the best germination performance in the combination of 30 mM KCl + 2.0 mM TU + 0.24 mM GA + 0.5 mM SA. The best germination performance was evaluated by the highest germination percentage (%), germination index, seed vigor, leaf length, root length and biomass. Conclusion: Therefore, the combination treatments of 30 mM KCl + 2.0 mM TU + 0.5 mM SA was found to be the most effective and simplest liquid enhancer formula that has an ability to enhance seed germination of drought-stressed rice cv. MR284 seed.

In this study, a comparative analysis of seed quality indicators of 1196 hybrid maize seed samples from the main maize-producing areas in China from 2013 to 2018 was carried out. The results showed that the maize seed quality in China had changed obviously in the past six years, and was mainly as follows: The percentage of samples with coated seed in 2015–2018 was higher than 62.8% in 2013 and all exceeded 97%; the sample rate of packaging according to seed number was from 24.5% in 2013 to 58.6% in 2018, and the percentage of samples which met the prescribed quality standards was from 89.2% in 2013 to 98.4% in 2018. Principal component analysis indicated that standard germination energy (SGE), standard germination percentage (SGP), cold test germination percentage (CTGP), accelerated aging test germination percentage (AATGP), and mean field seedling emergence (FSE) were the primary predictors of seed germination and seedling emergence. Meanwhile, combining other statistical methods, regression models of SGE, SGP, CTGP, and AATGP were established to predict the field seedling emergence. Furthermore, seed bulk density and total starch content were correlated with seed vigor, which needs to be further studied. This study offered a theoretical basis and data support to better understand the changes of maize quality in China over the past six years, and provided an important reference to further improve the maize seed quality in the future.

Seed treatment with growth regulators, especially salicylic acid, is a promising alternative to the seed industry because it is an important inducer of resistance to diseases and pests, as well as acting significantly on quality and seed yield. The objective of this study was to evaluate the performance of rice seed treated with different concentrations of salicylic acid, as well as assess the crop yield and seed quality. The treatments consisted of increasing levels of 0, 50, 100, 150 and 200 mg.L-1 salicylic acid. To this was prepared a stock solution of salicylic acid and the highest concentration by successive dilution in distilled water, the other concentrations were obtained. The physiological quality of seeds produced was treated and evaluated by tests of vigor and germination, and after harvest were evaluated seed yield. It follows that treatment of rice seeds with salicylic acid concentrations up to 130 mg.L-1 at a dose of 2 mL.kg-1 seed does not affect the germination and affects the strength, however provides substantial increases in the yield of seeds. The seed treatment with salicylic acid has no influence on seed quality produced.

AbstractThis work aimed at determining whether high temperature intermittent drying followed by tempering at ambient temperature could preserve the seed viability and vigor.JaponicaandIndicarice seeds with 21.2 % and 22.6 % wet moisture contents (w.b.) were dried at 50 ºC and 60ºC for either 5, 10, 15 or 20 minutes, followed by tempering at 25ºC for 45 minutes. Each drying cycle was repeated until the rice seeds were dried to 12.0 % (w.b.). The drying rate was improved and the total in-dryer time was reduced in the intermittent drying when compared to continuous drying, due to the tempering process. The seed vigor was significantly reduced by intermittent drying at 60 °C with all exposure times, although the seed still kept the ability to germinate for both rice cultivars. The intermittent drying at 50 ºC for 5 minutes per drying cycle preserved the seed vigor ofJaponicarice well, while inIndicarice, the intermittent drying at 50 ºC up to 20 minutes could still be able to maintain the seed vigor.

Thiamethoxam is a systemic insecticide that is transported within the plant through its cells and can activate various physiological reactions such as protein expression. These proteins interact with defense mechanisms against stress in adverse growing conditions. The objective of this study was to evaluate the effect of thiamethoxam in rice seeds and the potential benefits that it can provide. Two experiments were carried out and, in both, seeds were treated with commercial product containing 350 g of thiamethoxam active ingredient per liter of product, at doses 0, 100, 200, 300 and 400 mL.100 kg-1 of seeds: 1) it was conducted with three lots of IRGA BR 424 cultivar rice seeds, which were submitted to the following laboratory tests: germination, cold test, accelerated aging test, as well as field assessment: total seedling length, root system length, number of panicles and productivity; 2) four lots of IRGA BR 424 cultivar rice seeds, two high and two low-vigor, were subjected to the following tests: germination, cold test and greenhouse seedling emergence test. Thiamethoxam rice seed treatment positively favors the seed quality.

ABSTRACT To evaluate seed vigor, electrical conductivity and ethanol tests are fast and efficient methodologies. They have the potential to be used in several species, such as red rice. However, there are no protocols or information about their efficiency. Thus, the objective was to evaluate the efficiency, and define parameters of execution for electrical conductivity and ethanol tests, to evaluate the vigor of red rice seeds. The study was conducted using four lots of ‘BRS 901’ red rice, which was subjected to a germination test, as well as first count, accelerated aging, and field seedling emergence tests. The electrical conductivity test was performed with 25, 50, and 100 seeds soaked in 50 mL and 75 mL of water, at 25 °C and 30 °C, for 3, 6, 20, and 24 hours, respectively. The ethanol test was performed with 50 and 100 seeds soaked in a volume of water equivalent to 1.0, 1.5, 2.0, 2.5, and 3.0× the mass of the seed sample. To assess the vigor of red rice seeds, the electrical conductivity test is an efficient method when conducted with 50 seeds soaked in 50 mL of water at 25 °C for 20 hours. Meanwhile, the ethanol test is most effective when performed with 50 seeds, in a volume of water that is 2.5× the mass of the sample, at 40 °C for 24 hours.

Maize (Zea mays dis.L.) is one of the most important staple crops in the world. Sindhupalchok, Kavre and Nuwakot were the three districts selected for the maize CBSP groups under the respective Agriculture Development Offices identified by CDD for the present study. Altogether 2x6x3 samples for the study were collected. Seed damage as broken seeds, damages on embryo, seed quality assessments like seed moisture content, pure seed percentage, seed vigor measured by root and shoot length, seed germination, and embryo damages were carried in seed testing laboratory in Seed Science & Technology Division (SSTD) Khumaltar following the ISTA Rules, 2007. For the embryo damage test, tetrazolium salt test (TZ) was conducted following the procedures prescribed for maize seed in ISTA Rules, 2007. All the samples possessed of 99 % of pure seed. It ranged from 99.51 % in Nuwakot to 99.96 % in Kavre for samples using corn sheller. They were found within the standards for pure seed and were of acceptable range. The corn sheller was found as efficient as that of hand shelling. There was not found the effect on the quality of seed that they were within the standard values. The germination percentage decreased from initial 96 to 85 (Kavre); 94 to 87 (Sindhupalchwok) and 91 to 83 (Nuwakot). This study concludes that the corn sheller techniques offer Wooden corn sheller is a simple but traditional device made locally for shelling the maize kernels and distributed to Community based seed production (CBSP) farmers contribute to minimizing the impacts of hand sheller on agricultural systems.DOI: http://dx.doi.org/10.3126/ijasbt.v1i2.8198 Int J Appl Sci Biotechnol, Vol. 1(2): 68-71
