Environmental conditions in Uruguay favour the development of gastrointestinal nematodes (GIN) throughout the year, with clinical or subclinical manifestations that cause significant economic losses. The aim of this study was to evaluate the effect of the GIN on fertility, fecundity, reproductive rate and lamb growth in Australian Merino sheep grazing on basalt fields. The study was carried out in a farm located in north of Uruguay. Sixty-eight Merino ewes facing a natural parasite challenge were divided into two groups. Control group was doused with active anthelmintic that have proven efficacy to minimize the effects of parasitic and the parasitized group that did not receive anthelmintic, except rescue dosages. Stool sample of each sheep was collected monthly for egg counts (EPG) of GIN using McMaster technique and nematode genders were identified from infective larvae obtained in cultures by Roberts O’ Sullivan technique. Artificial insemination and ewes were mated for new service, and pregnancy condition was diagnosed by ultrasound. Lambing control was made and lambs were weighed at birth and marking. The evolution of EPG values showed significant differences between groups at the end of gestation and lactation. Haemonchus spp. was the main gender of nematodes found, followed by Trichostrongylus spp. Fertility was 95 % versus 90 %; fecundity 87 % versus 63 % and reproductive rate 73 % versus 47 % for the control group and the parasitized one respectively. No significant differences were recorded in birth-weight. However market-weight and lamb daily gain were lower in the parasitized group. The effect of natural infection GIN was evident in lower reproductive performance of Australian Merino sheep.

Helicoverpa armigera is one of the most important agricultural pests because it has a high reproductive rate and resulted in economic losses. One of the H. armigera control techniques that is in accord with IPM principle is the utilizing of entomopathogenic fungus. The aim of this research was to test the pathogenicity of Lecanicillium (Lecanicillium sp. PTN01) against H. armigera. Pathogenicity test was conducted by conidia application of Lecanicillium sp. PTN01 on both the larvae and eggs at density of 105, 106, 107 conidia/ml. The control group was only treated with steril water. Polymerase chain reaction (PCR) was performed for molecular identification Lecanicillium sp. PTN01. The results showed that Lecanicillium sp. PTN01 able to inhibit egg hatching 13.75%, and cause mortality of first instar larval survival 98.75%. The result of virulence test to the highest larval mortality at 107 conidia/ml density was 41,25%, with the values of LT25, 50, 75 were 3.95, 7.12, 12.82 (days) and LC25, 50, 75 were 4.6 x 105, 1.7 x 106, 4.6 x 109 (conidia/ml). The DNA sequence analysis of ITS 1 and ITS 4 primers showed that Lecanicillium sp. PTN01 was similar to fungus species L. kalimantanense strain BTCC F23 with 94% homology.

The experiment aimed to examine the impacts of an increased growth rate of ewes between three and seven months of age on udder development using ultrasound and to establish whether ultrasonography could be used to identify ewe mammary structures that may be indirect indicators of singleton growth to weaning. Udder dimensions, depths of gland cistern (GC), parenchyma (PAR) and fat pad (FP) were measured in late pregnancy (P107), early lactation (L29), and at weaning (L100) in 59 single-bearing yearling ewes selected from two treatments. The ‘heavy’ group (n = 31) was preferentially fed prior to breeding achieving an average breeding live-weight of 47.9 ± 0.38 kg at seven months of age. The ‘control’ group (n = 28) had an average breeding live-weight of 44.9 ± 0.49 kg. Udder dimensions, GC, PAR and FP did not differ between treatments. Lamb growth to L100 was positively associated (p < 0.05) with PAR at P107 and GC at L29. There was no evidence of negative effects of the live-weight gain treatments on udder development of yearling ewes as measured by ultrasonography. The results suggest that this ultrasound method has the potential to identify pregnant yearling ewes which would wean heavier singletons.

Several cases of animal and human salmonellosis caused by the Salmonella serotype Typhimurium have been reported. In animals, subclinical infection favors pathogen dissemination through feces. In this context, the domestic pigeon (Columba livia) with an asymptomatic condition may play an important role in the transmission of salmonellosis, through the elimination of contaminated feces in commercial aviaries or in poultry feed facilities, causing economic losses to the poultry industry and presenting a risk to public health. This study aimed to evaluate the mortality, clinical signs and the presence of Salmonella Typhimurium in the feces and organs of chicks previously inoculated with bacteria isolated from a pigeon. One-day-old chicks were distributed in two experimental groups (G1 and G2) of 32 birds each, and a control group of six birds. Two inocula of 0.4 and 0.7 mL with 105 and 106 colony forming units were used in G1 and G2 birds, respectively. At 1, 4, 7 and 14 days post-inoculation (dpi) fecal samples were pooled from each cage and individual cloacal swabs were collected. At 14 dpi, all chicks were euthanized and samples were collected from the liver, spleen, lung, cecum and intestine for microbiological analysis. Mortality was only observed among G2 birds (6.25%). Most birds presented clinical signs of diarrhea at 4 dpi and no symptom as observed at 14 dpi. The results from cloacal swabs demonstrated bacterial elimination in 68.8% and 53.1% of G2 and G1 birds, respectively at 1 dpi. Additionally, fecal samples had elevated bacterial shedding in all four periods of observation , with a higher excretion at 4 dpi (62.5%) for both groups. Among G2 birds, 74.2% were positive for the pathogen in the intestine; G1 birds presented the lowest rate of lung infection (29%), and both groups had more than 50% positivity for liver and caeca. The results revealed that infected chicks with a Salmonella Typhimurium strains isolated from pigeons may host the pathogen in several organs, and simultaneously present diarrheic disorders with significant levels of bacterial excretion in feces.

AbstractDue to increased anthelmintic resistance, complementary methods to drugs are necessary to control gastrointestinal nematodes (GIN). Vaccines are an environmentally-friendly and promising option. In a previous study, a Teladorsagia circumcincta recombinant sub-unit vaccine was administered to two sheep breeds with different levels of resistance against GIN. In the susceptible Canaria Sheep (CS) breed, vaccinates harboured smaller worms with fewer eggs in utero than the control group. Here, we extend this work, by investigating the cellular and humoral immune responses of these two sheep breeds following vaccination and experimental infection with T. circumcincta. In the vaccinated CS group, negative associations between antigen-specific IgA, IgG2 and Globule Leukocytes (GLs) with several parasitological parameters were established as well as a higher CD4+/CD8+ ratio than in control CS animals, suggesting a key role in the protection induced by the vaccine. In the more resistant Canaria Hair Breed (CHB) sheep the vaccine did not significantly impact on the parasitological parameters studied and none of these humoral associations were observed in vaccinated CHB lambs, although CHB had higher proportions of CD4+ and CD8+ T cells within the abomasal lymph nodes, suggesting higher mucosal T cell activation. Each of the component proteins in the vaccine induced an increase in immunoglobulin levels in vaccinated groups of each breed. However, levels of immunoglobulins to only three of the antigens (Tci-MEP-1, Tci-SAA-1, Tci-ASP-1) were negatively correlated with parasitological parameters in the CS breed and they may be, at least partially, responsible for the protective effect of the vaccine in this breed. These data could be useful for improving the current vaccine prototype.

Abstract Bovine mastitis is one of the major diseases which directly affects the milk production performance and it causes huge economic losses in the dairy industry. Bacterial infection is the main risk factor of bovine mastitis and the antibiotic therapy is the primary choice to control the disease. However, persistence use of antibiotic increases the incidence of bacterial resistance and traces of antibiotic residues in animal products. Lactobacillus casei Zhang is one of the probiotics with multiple biological functions, which has certain bacteriostatic effect on pathogenic microorganism. The purpose of this study was to explore the effect of Lactobacillus casei Zhang (L. casei Zhang) on the prevention of E. coli-induced milk-blood barrier damage. Bovine mammary epithelial cells (BMECs) were used to establish a milk-blood model and Control group (PBS), E. coli group, and L. casei Zhang pretreatment plus E. coli group were set up respectively. The results showed that: L. casei Zhang could significantly reduce the increase of LDH release caused by E. coli treatment (P&lt; 0.05). And it can also significantly reduce the decrease of transmembrane resistance of monolayer cells caused by E. coli treatment (P&lt; 0.05). In addition, L. casei Zhang could significantly reduce the expression of tight junction proteins ZO-1, Claudin-1, Claudin-4 and Occludin (P &lt; 0.05). In conclusion, L. casei Zhang could effectively improve the damage of the blood-milk barrier caused by E. coli and could protect BMECs during bacterial infection.

Gastrointestinal nematode infections were evaluated in sheep raised in Botucatu, state of São Paulo, Brazil between April 2008 and March 2011. Every month, two tracer lambs grazing with a flock of sheep were exposed to natural infection with gastrointestinal nematodes for 28 consecutive days. At the end of this period, the lambs were sacrificed for worm counts. Haemonchus contortus presented 100% of prevalence. The seasons exerted no significant influence on the mean intensity of H. contortus, which ranged from 315 worms in November 2010 to 2,5205 worms in January 2011. The prevalence of Trichostrongylus colubriformis was also 100%, with the lowest mean intensity (15 worms) recorded in February 2011 and the highest (9,760 worms) in October 2009. In the case of T. colubriformis, a significant correlation coefficient was found between worm counts vs. rainfall (r = −0.32; P <0.05). Three other nematodes species were found in tracer lambs, albeit in small numbers. Their prevalence and mean intensity (in parenthesis) were as follows: Oesophagostomum columbianum 28% (25.2), Cooperia curticei 7% (4.5) and Trichuris spp. 2% (1). In conclusion, the environmental conditions of the area proved to be highly favorable for the year-round transmission of H. contortus and T. colubriformis.

Development of a vaccine against congenital cytomegalovirus (CMV) infection is a public health priority, but CMVs encode immune evasion genes that complicate live virus vaccine design. To resolve this problem, this study employed guanosyl phosphoribosyl transferase (gpt) mutagenesis to generate a recombinant guinea pig CMV (GPCMV) with a knockout of a viral chemokine gene, GPCMV MIP (gp1). MIP deletion virus replicated with wild-type kinetics in cell culture but was attenuated in nonpregnant guinea pigs, demonstrating reduced viremia and reduced inflammation and histopathology (compared to a control virus with an intact GPCMV MIP gene) following footpad inoculation. In spite of attenuation, the vaccine was immunogenic, eliciting antibody responses comparable to those observed in natural infection. To assess its protective potential as a vaccine, either recombinant virus or placebo was used to immunize seronegative female guinea pigs. Dams were challenged in the early 3rd trimester with salivary gland-adapted GPCMV. Immunization protected against DNAemia (1/15 in vaccine group versus 12/13 in the control group,P<0.01). Mean birth weights were significantly higher in pups born to vaccinated dams compared to controls (98.7 g versus 71.2 g,P<0.01). Vaccination reduced pup mortality, from 35/50 (70%) in controls to 8/52 (15%) in the immunization group. Congenital GPCMV infection was also reduced, from 35/50 (70%) in controls to 9/52 (17%) in the vaccine group (P<0.0001). We conclude that deletion of an immune modulation gene can attenuate the pathogenicity of GPCMV while resulting in a viral vaccine that retains immunogenicity and demonstrates efficacy against congenital infection and disease.

The effect of different nematophagous fungi [Duddingtonia flagrans (AC001 and CG722) and Monacrosporium thaumasium (NF34)] with regard to controlling infective larvae (L3) of nematodes after gastrointestinal transit in female cattle (3/4 Holstein × Zebu) was evaluated. A total of 24 pubescent female cattle were used, weighing approximately 320 kg each one. There were three treatment groups, each contained six animals that received 150 g of pellets (0.2 g of mycelium), orally in a single dose, in a sodium alginate matrix containing mycelial mass of the fungus D. flagrans (AC001 or CG722) or M. thaumasium (NF34); and one control group (without fungi). Fecal samples were collected from the animals at intervals of 12, 15, 18, 21, 24, 48, and 72 hours. At the end of 17 days, the L3 not subjected to predation were recovered by means of the Baermann method. The fungal isolates tested were capable of destroying the L3 after gastrointestinal transit. It was observed that within 72 hours, the isolates AC001, CG722, and NF34 showed a higher predatory activity (81.2%, 97.3%, and 98.3%, respectively). The results justify the need for studies in the field, and over longer intervals, in order to observe the efficiency of the fungus D. flagrans, or even M. thaumasium, for environmental control over nematodes in naturally infected cattle.

This study aimed to evaluate the efficacy of ivermectin, levamisole HCl and albendazole against gastrointestinal nematodes in naturally infected goats of government goat development farm, Sylhet, Bangladesh. The study included 52 black Bengal breed of which 30 were naturally infected and randomly selected 20 on the basis of their weight and egg count. Twenty black Bengal goats of 12-13 month old irrespective of sex infested with gastrointestinal nematodes were selected for this experiment and randomly divided into four equal groups (group A, B, C and D) where each group consisted of 5 goats and goats of group D were kept as control group. One injectable ivermectin (200?gkg-1 body weight, S/C) preparations (Techno Drugs Ltd. Bangladesh) and two solid levamisole HCl, albendazole (7.5 mgkg-1 body weight, orally) preparations (The ACME Laboratories Ltd. and Square Pharmaceuticals Ltd. Bangladesh) were used for positive control of gastrointestinal nematodes as group A, B and C. Goats of group D was kept as control without giving any treatment. Before trials (day 0), total egg count, blood samples and initial body weight were recorded. During the study period the fecal and blood samples were collected directly from rectum and examined on 7th, 14th, 21st and 28th day using McMaster fecal egg counting method. Body weight was recorded on day 28 following the treatments. The results of the comparative efficacies of different anthelmintic of ivermectin was 86.75%, followed by levamisole HCl 85.07% and albendazole 92.81%. McMaster fecal egg counting method discloses the percentage of Haemonchus spp. (15.38%), with Trichostrongylus spp, Strongyloides spp., and Cooperia spp. also present. After treatment with ivermectin, levamisole HCl and albendazole, Total Erythrocyte Count (TEC), Hemoglobin (Hb) content and Packed Cell Volume (PCV) were increased significantly (p<0.01 and p>0.05) in goats but Erythrocyte Sedimentation Rate (ESR) and Total Leukocyte Count (TLC) were decreased significantly (p<0.05 and p>0.01) in all treated goat and body weight was increased significantly (p<0.01) on day 28.International Journal of Natural Sciences (2015), 5(1) 26-34
