Space maintainers have presented an increased interest due to their chemical composition which influences the electrochemical and electrolytic processes of the oral cavity, leading to important biological activity. The present study was purported to evaluate the biological in vitro activity of three types of space maintainers (S1, S2, and S3, differing from each other in terms of metal composition) used in pediatric dentistry, in terms of their antimicrobial effect and biosecurity profile using two types of keratinocytes (PGK: primary gingival keratinocytes, and HaCaT: human immortalized keratinocytes) by assessing the morphology, viability, cytotoxicity, and gene expression of the cells. Statistical differences were calculated by the one-way ANOVA test, followed by Tukey’s post-test. Antimicrobial screening highlighted a dilution-dependent influence that, in the case of all strains tested, did not show inhibition or stimulation of bacterial growth. The in vitro evaluations revealed that the test samples did not induce important cytotoxic potential on both keratinocyte cell lines (HaCaT and PGK), with the cells manifesting no morphological alteration, a good viability rate (above 90%: PGK–S1, * p < 0.05), and a low cytotoxic activity (less than 11%: PGK, S1 *** p < 0.001 and S3 * p < 0.05; HaCaT, S1 ** p < 0.01). The data obtained in this study highlight the fact that the samples analyzed are biocompatible and do not develop the growth of the studied bacteria or encode the gene expression of primary and immortalized keratinocytes.

Sodium hypochlorite (NaOCl), an effective endodontic irrigant against Enterococcus faecalis (EF), is harmful to periapical tissues. Natural pineapple-orange eco-enzymes (M-EE) and papaya eco-enzyme (P-EE) could be potential alternatives. This study aimed to assess the antimicrobial efficacy of M-EE and P-EE at different concentrations and fermentation periods against EF, compared to 2.5% NaOCl. Fermented M-EE and P-EE (3 and 6 months) at various concentrations were mixed with EF in a 96-well plate incubated for 24 h anaerobically. Minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of M-EE and P-EE were determined via EF growth observation. EF inhibition was quantitatively measured and compared between different irrigants using the one-way analysis of variance (ANOVA), and different fermentation periods using the independent-samples T-test. M-EE and P-EE showed MIC at 50% and MBC at 100% concentrations. There was no significant difference in antimicrobial effect when comparing M-EE and P-EE at 50% and 100% to 2.5% NaOCl. P-EE at 6 months fermentation exhibited higher EF inhibition compared to 3 months at concentrations of 25% (p = 0.017) and 0.78% (p = 0.009). The antimicrobial properties of M-EE and P-EE, at both 100% and 50% concentrations, are comparable to 2.5% NaOCl. They could therefore be potential alternative endodontic irrigants, but further studies are required.

Abstrak: Minyak cengkeh berguna sebagai antibakteri alami. Minyak esensial dari cengkeh mempunyai fungsi anestetik dan antimikrobial. Zat yang terkandung dalam cengkeh yang bernama eugenol dapat membunuh bakteri termasuk bakteri yang resisten terhadap antibiotika, salah satunya adalah bakteri Streptococcus mutans. Bakteri ini merupakan mikroorganisme penyebab utama terjadinya karies. Tujuan dari penelitian ini yaitu untuk mengetahui efek antibakteri ekstrak cengkeh terhadap bakteri Streptococcus mutans secara in vitro. Penelitian ini merupakan penelitian eksperimental menggunakan post test only control group design. Penelitian ini menggunakan bahan coba ekstrak cengkeh dengan konsentrasi 40%, 60%, dan 80%, Ciprofloxacin, aquades dengan pengulangan sebanyak lima kali. Data dikumpulkan dan dianalisis dengan one-way ANOVA dan post-hoc uji LSD ( = 0,05). Berdasarkan hasil uji statistik penelitian uji efek antibakteri ekstrak cengkeh terhadap bakteri streptococcus mutans secara in vitro, dapat disimpulkaan bahwa ekstrak cengkeh memiliki efek antibakteri dalam menghambat pertumbuhan bakteri Streptococcus mutans secara in vitro. Hasil uji lanjut post-hoc uji LSD menunjukan daya hambat ekstrak cengkeh 40%, 60%, 80%, lebih kecil (p<0,05) dalam menghambat Streptococcus mutans secara in vitro dibandingkan Ciprofloxacin. Kata Kunci: Ekstrak cengkeh, Streptococcus mutans.   Abstract: Clove oil is useful as a natural antibacterial agent, essential oil of clove has anesthetic and antimicrobial effect. Substances contained in clove called eugenol can kill bacteria including antibiotic resistant bacteria, one of which is the bacteria Streptococcus mutans. This bacteria is a major cause for caries. The purpose of this study was to mengetahui clove extrack antibacterial effects againts Streptococcus mutans bacteria in vitro. This study is an experimental study using a post test only control group design. This research try using clove extract with a concentration of 40%, 60%, and 80%, Ciprofloxacin, aquades repetition five times. Data collected and analyzed by one-way ANOVA and post-hoc LSD test (α = 0.05). Based on the results of the statistical test to test the effects of anti-bacterial research clove extracts against Streptococcus mutans bacteria in vitro, can disimpulkaan that clove extracts have antibacterial effects in inhibiting the growth of Streptococcus mutans bacteria in vitro.further test result post-hoc LSD test shoved its inhibitory clove extract 40%, 60%, 80% smaller (p<0,05)in hibiting Streptococcus mutans in vitro compared Ciprofloxacin. Background: Currently, there is a trend towards using natural and ethnopharmacological species with therapeutic potential. This investigation evaluated the antifungal activity of two species in the Ecuadorian Andes, which are used in treating dermatomycosis: Ambrosia arborescens Mill. (Marco) and Aristeguietia glutinosa Lam. (Matico). Methods: We worked with seven concentrations (100 to 700ppm) of Ambrosia arborescens Mill. extract and ten concentrations (0.5 to 5%) of essential oil (EO) of Aristeguietia glutinosa Lam. on Trichophyton mentagrophytes ATCC 9533, Trichophyton rubrum ATCC 28188, Microsporum canis ATCC 36299 and Candida albicans ATCC 10231. The methodology used was a modified version of the Kirby-Bauer method, using diffusion in agar wells. Results: The Tukey test, after the one-way Anova, determined effective concentrations of EO: 5% for Trichophyton mentagrophytes, 4.5% for Trichophyton rubrum, 5% for Microsporum canis and 2% for Candida albicans. In the extracts, the concentration of 700ppm was used for Trichophyton mentagrophytes, Trichophyton rubrum, and 600ppm for Microsporum canis and Candida albicans. Conclusions: The evaluation of the antifungal activity of the Ambrosia arborescens extract showed inhibition in the studied dermatophytes in each one of the planted concentrations (100 to 700ppm). The evaluation of the antifungal activity of Aristeguietia glutinosa EO showed inhibition in the studied dermatophytes in each of the planted concentrations (0.5 to 5%).

Penelitian ini untuk mempelajari pengaruh nikotin terhadap kadar MDA serum dan keberhasilan fertilisasi in vitro pada Rattus novergicus. Rancangan penelitian ini adalah eksperimental dengan post test only control group design. Subjek penelitian terdiri dari 4 kelompok (36 ekor tikus) yang dipilih secara acak dan homogen. Kelompok penelitian terdiri dari kelompok kontrol (K0), kelompok perlakuan yang diberikan injeksi nikotin subkutan selama 7 hari dengan dosis 70 mg/Kg BB (K1), 52,5 mg/kgBB (K2) dan 35 mg/kgBB (K3). Hasil yang diamati adalah adanya badan polar II/sigot 2 sel. Hasil uji one way Anova adalah terdapat perbedaan yang bermakna secara statistik (p<0,05), kemudian dilanjutkan dengan uji BNT. Hasil uji BNT kadar MDA serum adalah terdapat perbedaan yang bermakna (p<0,05) antara K0 dan K1, kelompok K0 dan K2, K1 dan K2, serta K1 dan K3. Angka keberhasilan fertilisasi tidak dapat dilakukan uji statistik karena seluruh data menunjukkan 100% berhasil. Dengan demikian tidak dapat dilakukan uji hubungan antara kadar MDA serum dengan angka keberhasilan fertilisasi. Kesimpulan penelitian ini adalah nikotin dapat menyebabkan peningkatan kadar MDA serum tapi belum dapat mempengaruhi fertilisasi sehingga sehingga tidak dapat menghubungkan antara kadar MDA serum dengan angka keberhasilan fertilisasi.

Pneumonia is a lung parenchymal infection caused by Pseudomonas aeruginosa.It is Gram negative bacteria that have developed antibiotic resistance. Actinomycetes are Gram-positive bacteria that produce secondary metabolites which have the ability as antimicrobial. Objectives: To identified the ability of Actinomycetes isolates to inhibit the growth of the bacterium Pseudomonas aeruginosa ATCC 27853. The samples in this experiment were from Kebun Raya Bogor that had been rejuvenated on Starch Casein Agar (SCA). Methods: Six dilution series 10-1; 10-2; 10-3; 10-4; 10-5; 10-6 Actinomycetes isolates were used to observe the inhibition zone of P.aeruginosa growth on Mueller Hinton Agar (MHA) media by diffusion method. Results: The effective incubation time occurred at 24 hours, and then it resulted in the average clear zone diameter of 14.70 mm, 10.57 mm, 8.53 mm, 8.47 mm, 6.97 mm, and 5.30 mm. The results of the One – Way Anova test with p-value = 0.000 (p < 0.005) showed some differences at each concentration to inhibit the growth of P.aeruginosa ATCC 27853 at 24 hours incubation period. Conclusion: The most effective concentration of Actinomycetes isolates that can potentially be antibacterial was the concentration of 10-1 with potential solid inhibitory power.Keywords: Actinomycetes, antibacterial, Pseudomonas aeruginosa

This research was conducted to determine the effectiveness test of papaya leaf extract with a concentration of 10%, 20%, 30%, 40%, 50%, 60%, 70%, 80% and 90% and 100% against staphylococcus epidermidis bacteria. Staphylococcus epidermidis is a gram-positive bacterium in the form of cocci and is harmful to humans. This research was conducted with an experimental method with a post-test only design and sampling with a purposive sampling method. The investigation of the effectiveness of papaya leaf extract against staphylococcus epidermidis was done by diffusion using paper discs, namely by calculating the diameter of the bacterial inhibition zone of paper discs that have been moistened with papaya leaf extract. The results of this study were analyzed out with the One-Way ANOVA test followed by the Post Hoc Test. The test showed that there were significant differences from each treatment given with a 95% confidence index. The bacteria treated with papaya leaf extract 100% against staphylococcus epidermidis had the highest average inhibitory zone power compared to other concentrations of 14.97 mm. The papaya leaf extract 100% has the best antibacterial effect compared to other concentrations because the best inhibitory zone power is found on those extract concentrations.

Prevention by utilizing medicinal plants can be used as an alternative treatment. This study aims to identify active antimicrobial substances and test the antimicrobial potential of traditional Balinese spices, namely Basa Selem, Basa Gede, Basa Wangen, Basa Bawang Jahe, and Basa Rajang against Salmonella sp bacteria in vitro. The True-experimental method with Posttest only-control design was used in this study, by intervening in the treatment group as well as the presence of positive and negative controls. The maceration method with 96% EtOH solvent was used to extract active substances and identify the levels of antimicrobial active substances. The TLC Spectrophotodensitometer instrument and the diffusion method (discs) were used to test antimicrobial potential. Data analysis was performed using the one-way ANOVA test. All five samples showed flavonoids, alkaloids, tannins, and phenols in qualitative tests. Based on quantitative test results of five samples, the highest compound content obtained in Basa Wangen (6.66 mg/ml of tannins), Basa Gede  (3.74 mg/ml of flavonoids), Basa Bawang Jahe (2.49 mg/ml of tannins), Basa Selem (2.87 mg/ml of tannin), and Basa Rajang (6.96 mg/ml of flavonoids). There are differences in the antimicrobial potential of various types of traditional Balinese spices against the growth of Salmonella sp in vitro (sig = 0.037). The traditional Balinese spices have the antimicrobial potential of the intermediate category with a range of inhibition (16-20 mm) based on the NCCLS standard.

<p>Excessive usage of synthetic pediculicides, such as permethrin, lindane, and malathion, can induce resistance, environmental pollution, health problems, and even death. Efforts to prevent the side-effects of synthetic pediculicides are needed, one of which is by shifting to powerful yet safe natural pediculicides, such as Citrus limon (lemon) and Allium sativum (garlic). This study aimed to observe the effective concentration of Citrus limon and Allium sativum in vitro on the mortality of Pediculus humanus capitis (P. h. capitis). This research was an experimental laboratory study conducted in July to September 2019 in the Laboratory of Parasitology at Faculty of Medicine, University of Riau, while the making of Allium sativum extract was done at the Faculty of Mathematics and Science, University of Riau. A total of 288 samples of adult P. h. capitis was taken from children in two orphanages in Pekanbaru. Citrus limon juice and Allium sativum extract of 25%, 50%, 75%, and 100% concentrations, permethrin 1% as the positive control, and distilled water as the negative control were used in this study, and repetition was done three times. P. h. capitis mortality was observed every 10 - 120 minutes, characterized by the absence of movement of the legs and antenna. Data analysis was performed after 120 minutes with the one-way ANOVA test, LSD (=0.05), and Lethal Concentration 50 test (LC50). The one-way ANOVA test results of this study showed that statistically Citrus limon juice and Allium sativum extracts showed differences in each treatment formulation of juice on P. h. capitis mortality (Sig 0.017&lt;0.05 and Sig 0.000&lt;0.05) and LC50 juice of Citrus limon and Allium sativum extract by 51.999% and 72.426%. In this study, Citrus limon juice and Allium sativum extract had effect as pediculicides.</p>

Escherichia coli (E. coli) dan Shigella dysenteriae (S. dysenteriae) merupakan bakteri penyebab diare. Salah satu tumbuhan yang dapat digunakan sebagai pengobatan untuk diare adalah daun pepaya. Daun papaya memiliki kandungan senyawa aktif seperti papain, tanin, alkaloid, flavonoid, fenol, saponin, dan steroid yang diduga memiliki efek antimikroba. Penelitian ini bertujuan untuk menganalisis efektivitas pemberian ekstrak daun pepaya (Carica papaya L.) dengan berbagai konsentrasi terhadap pertumbuhan bakteri  E. coli dan S. dysenteriae. Penelitian ini menggunakan rancangan post-test only with control group dengan metode difusi kertas cakram (Kirby-Bauer). Zona hambat yang terbentuk diukur menggunakan jangka sorong dan dianalisis menggunakan uji One Way ANOVA. Hasil penelitian menunjukkan bahwa ekstrak daun papaya dapat menghambat pertumbuhan bakteri E. coli dan S. dysenteriae secara in-vitro. Analisis uji statistika didapatkan (p<0,05) yang artinya terdapat perbedaan nyata antar kelompok perlakuan setelah pemberian ekstrak daun pepaya terhadap pertumbuhan bakteri E. coli dan S. dysenteriae.
